      • RT-LAMP를 이용한 콩황화일반모자이크바이러스의 진단

        Soybean yellow common mosaic virus (SYCMV) has been recently reported, it has been occurred a lot with Soybean mosaic virus (SMV) and Soybean yellow mottle mosaic virus (SYMMV) in soybean field. SYCMV belongs to genus of Sobemovirus and induced viral symptoms with yellowing, mottle and mosaic. A reverse transcription loop-mediated isothermal amplification (RT-LAMP) method allowed one-step detection of gene amplification by simple procedure and needed only a simple incubator for isothermal template. This RT-LAMP method allowed direct detection of RNA from virus-infected plants without thermal cycling and gel electrophoresis. In this study, we designed RT-LAMP primers named SYCML-F3/B3/FIP/BIP from coat protein gene sequence of SYCMV. After the reaction of RTLAMP, products were identified by electrophoresis and with the detective fluorescent dye, SYBR Green I. under daylight and UV light. Optimal reaction condition was at 63 for 60min and the primers of RTLAMP showed the specificity for only SYCMV tested in this study.

        조선업 도장작업자의 유사노출군과 노출변이; 크실렌 노출을 중심으로

        목적 : 조선업 도장 작업자의 크실렌 노출을 평가하기 위하여, 수행직무 및 밀폐특성에 따라 유사노출군으로 구분하여 노출 변이 및 동질성을 파악하고자 하였다. 방법 : 3회 이상 반복측정이 이루어진 근로자 중동일공정에서 직무상 변동이 없는 근로자 총 28명을 대상으로 하였다. 노출농도 값을 대수전환(lOgtransformation)하여 근로자간, 근로자내 노출 분산요소을 추정하기 위하여, nested randomeffects ANOVA model을 활용하였다. 선정된 유사노출군 내 구성원의 측정 평균이 동일하다고 할수 있는지를 검정하기 위하여 분산요인(9f)을 산출하였고, 쿨즈0.031일 경우 동일하다고 생각하였다.또한 노출 값을 대수전환하여 95 %의 근로자(b-R9s)가 2배수 범위 내에 측정값을 가질 경우 동일하다고 생각하였다. bwRSS의 값을 유사노출군내의 근로자들간(between worker)의 기하표준편차(bwsy)를 구한 후 exp(3.92 bwsy)로 계산하여 산출하였고, 그 값이 2이하이면 동일하다고 생각하였다. 결과 : 일반적으로 유사노출군은 노출농도가 비슷한 것이라는 생각과는 달리, 직무에 근거한 유사노출군은 동질성 검정상 이질적이었다. 분무도장과 붓도장 모두 bwRss은 2를 초과하였고, 룰 또한 0.031를 초과하여 변이 폭이 컸다. 그러나 밀폐특성에 따른 노출 분포의 변이성은 직무만으로 구분했을 때의 변이지표 보다 그 수치가 낮아 밀폐특성으로 직무의 변이성을 설명 할 수 있었다. 결론 : 직무에 근거한 유사노출군이 동질적이지알다는 결과는 유사노출군으로 구분된 근로자의 업무특성이 하나의 유사노출군으로 설정할 수 없다는 점을 시사한다. 따라서 향후 작업환경 측정 시 직무뿐만 아니라 특정 업무(specific task) 또는 세부작업현황까지 파악할 필요성을 제시하고 있다. Objectives : This study was conducted to assess the concentration of xylene exposure and exposure variability of a similar exposure group (SEG) in accordance with job title and confined space. Method : The study subjects included 28 workers working in the painting process in the ship-building industry. Measurement of subjects were repeated three times. Within and between-worker exposure valiance components were estimated from log-transformed exposure concentrations employing a one-way nested random effects analysis of variance model. A uniformly exposed group was defined as one where the between-worker variance(σB2) was less than or equal to 0.031 or bwR95 ≤2, where bwR95 =exp(3.92 bwSy) , representing the radio of the mean exposures of the 97.5th and 2.5th percentile groups of the workers sampled. Result : The results indicated that, contrary to popular belief, similar exposed groups based on job title were heterogeneous. The between-worker variance(σB2) is large(≥0.031) and the ratios of the 97.5th and 2.5th percentile workers of the log normally distributed exposures of each group of worriers was more than 2. However dividing job title into confined space (in-block, out-block, in/out-block) decreased the between-worker exposure variability. Conclusion : This study focused on the quantitative aspects of exposure in painters in the ship-building industry. We cannot accept the assumption that SEG are uniformly exposed, as a major component of variation in exposure relates to differences in tacks or other factors. Therefore, the observational schemes can be improved by investigating Job-specific task and practice.

      • A new mid-late maturing rice variety「Subo」with a good grain quality and for direct seeding

        ‘Subo’ is a new japonica rice variety developed by a cross breeding between HR21124-B-59 and HR19567-B-70-3 having a good Phenotype with a multi-resistance and high yield elite line. The new variety developed for direct seeding by the rice breeding team in Rice Breeding and Cultivation Research Division, Department of Rice and Winter Cereal Crop, NICS, RDA in 2012. The heading date of this variety is August 13 and earlier than that of check variety, Nampyeongbyeo, by two days. ‘Subo’ has 72cm of culm length and 104 spikelets per panicle in direct seeding cultivation. This variety showed resistance to bacterial leaf blight and rice stripe virus and have germinating ability in sumerged soil conditions. The milled rice of this variety exhibits translucent and very clear non-glutinous endosperm. ‘Subo’ has much better palatability of cooked rice than that of Nampyeongbyeo. The whole grain rate of milled rice are 86.9% and milled rice recovery of are 74.6%. The yield of ‘Subo’ in direct seeding cultivation is 5.55MT/ha in milled rice. ‘Subo’ could be adaptable to the southern part plain area of Pyeongtaek and southwestern in Korea.

      • Development of salt-tolerant transgenic rice using OsCBF4 cDNA

        This study was conducted to isolate a salt tolerant gene and to develop salt tolerant rice for reclaimed-saline areas through genetic transformation. A rice c/DRE binding factor 4(OsCBF4) cDNA was isolated from rice using RT-PCR. The full-length cDNA of the CBF4 gene consists of 1,429 nucleotides and 274 amino acid residues. In order to develop salt tolerant rice, transgenic rice plants containing the OsCBF4 gene were obtained via Agrobacterium-mediated transformation. The stable incorporation of the OsCBF4 gene into rice genome was confirmed by PCR and Southern analysis. The stable expression of introduced gene was also validated by RT-PCR analysis in T2 plants. Biological assay of T3 progeny of the transgenic plants in Yoshida solution containing 120mM Nacl for 2weeks, confirmed that the OsCBF4 confers salt tolerance to transgenic rice plants. OsCBF4 transgene in the transgenic line CBF4-10 was markedly expressed up to over three-fold in the leaf by 120 mM NaCl treatment. Real-time PCR analysis revealed that the levels of the transgene expression were markedly increased under salt treatment. The transgenic line CBF4-10 which showed highest ability to recover from the saline stress could be used as a potential source for salt tolerance in rice breeding programs

      • A new mid-late maturing japonica rice variety Hyeonpum with a good grain quality and eating quality

        ‘Hyeonpum’ is a new japonica rice variety developed by a cross breeding between Iksan469 having a good canopy architecture and Sindongjin and Musashino 7 having a good eating-quality with a view to develop a new variety having high quality of grain and palatability by the rice breeding team of Rice Breeding and Cultivation Research Division, Department of Rice and Winter Cereal Crop, NICS, RDA in 2012. The heading date of this variety is August 18 and later than that of check variety, Nampyeongbyeo, by four days. ‘Hyeonpum’ has 74cm of culm length and 101 spikelets per panicle. This variety showed resistance to bacterial leaf blight and rice stripe virus, but susceptible to leaf blast and planthoppers. The milled rice of this variety exhibits translucent and very clear non-glutinous endosperm. ‘Hyeonpum’ has much better palatability of cooked rice than that of Nampyeongbyeo. The whole grain rate of milled rice and milled rice recovery of ‘Hyeonpum’ are slightly higher than those of Nampyeongbyeo as 91.8% and 75.2% respectively. The yield of ‘Hyeonpum’ is 5.6 MT/ha in milled rice. ‘Hyeonpum’ could be adaptable to the southern part plain area of Pyeongtaek and southwestern costal areas in Korea.

        Antibody response to pneumococcal vaccination in children with chronic or recurrent rhinosinusitis

        Purpose: Although chronic and recurrent rhinosinusitis is prevalent in children, little is known about its causes. Here, we investigated the humoral immunity in children with chronic or recurrent rhinosinusitis. Methods: We examined 16 children attending the outpatient clinic at the CHA Bundang Medical Center including 11 boys and 5 girls, aged 3-11 years (mean age, 5.6 years), who had rhinosinusitis for >3 months or >3 times per year. The complete blood count with differential and total serum concentrations of Immunoglobulin (Ig) E, IgA, IgD, IgM, IgG, and IgG subclasses ($IgG_1$, $IgG_2$, $IgG_3$, and $IgG_4$) of all children were measured. All subjects received 23-polysaccharide pneumococcal vaccination (PPV), and the levels of antibodies to 5 serologic types (4, 6B, 14, 18C, and 23F) of pneumococcal capsular polysaccharide antigens were measured before and after vaccination. Post-PPV antibody titers ${\geq}0.35{\mu}g/mL$ or with a ${\geq}4$-fold increase were considered as positive responses. Results: The titers of IgG, IgA, IgD, and IgM were within normal range in all 16 children, whereas the total IgE concentration was higher than normal in 2 children. $IgG_1$ deficiency was observed in 1 patient and $IgG_3$ deficiency in 3. After PPV, 1 patient failed to respond to all 5 serologic types, 2 failed to respond to 4 serologic types, and 2 failed to respond to 3 serologic types. Conclusion: Clinicians should consider the evaluation of humoral immune functions in children with chronic or recurrent rhinosinusitis who do not respond to prolonged antibiotic treatment.

      • A Study on localized corrosion susceptibility of the welded STS 304 and 316L in acidic solution

        In case of stainless steel, uniform corrosion is not of importance. The increased corrosion resistance, compared to low alloy steel, is explained by the chromium content which affects the formation of passivation. However the sensitization could be caused by lack of Cr on the welded area of stainless steel. In this study, therefore, the corrosion resistance characteristics of STS 304 and 316L were estimated to determine the extent of its use in acidic environment (pH 2). From the results, repassivation potentials(Ep) of STS 304 and 316L were below or above to the natural corrosion potential(Ecorr). Therefore, using STS 304 and 316L materials in acidic solution according to the circumstances should be carefully considered.

        Soybean mosaic virus (SMV) is a prevalent pathogen that causes significant yield reduction in soybean production worldwide. SMV belongs to potyvirus and causes typical symptoms such as mild mosaic, mosaic and lethal necrosis. SMV is seed-borne and also transmitted by aphid. Eleven SMV strains, G1 to G7, G5H, G6H, G7H, and G7a were reported in soybean varieties in Korea. A reverse transcription loop-mediated isothermal amplification (RT-LAMP) method allowed one-step detection of gene amplification by simple procedure and needed only a simple incubator for isothermal template. This RT-LAMP method allowed direct detection of RNA from virus-infected plants without thermal cycling and gel electrophoresis. In this study, we designed RT-LAMP primers named SML-F3/B3/FIP/BIP from coat protein gene sequence of SMV. After the reaction of RT-LAMP, products were identified by electrophoresis and with the detective fluorescent dye, SYBR Green I. under daylight and UV light. Opmtimal reaction condition was at 58℃ for 60min and the primers of RT-LAMP showed the specificity for nine SMV strains tested in this study.
