        Human kidney organoids model the tacrolimus nephrotoxicity and elucidate the role of autophagy

        Background/Aims: Tacrolimus has been used as an immunosuppressive agent in organ transplantation. Despite the therapeutic benefits, tacrolimus’s use is limited due to its nephrotoxicity. To reduce tacrolimus nephrotoxicity, effective humanized experimental models may be helpful. Here, we modeled tacrolimus nephrotoxicity using kidney organoids derived from human inducible pluripotent stem cells (iPSCs) in vitro. Methods: Kidney organoids were differentiated from the CMC11 iPSC cell line, re-seeded in 96-well plates, and treated with tacrolimus at doses of 0, 30, or 60 μM for 24 hours. This in vitro model was compared to a mouse model of tacrolimus nephrotoxicity and the associated mechanisms were investigated. Results: The size of the kidney organoids and cell viability decreased in dose-de-pendent manners after treatment with tacrolimus. The number of tubular cells decreased with a loss of polarity, similar to the effects seen in mouse tacrolimus nephrotoxicity. Ultrastructural analysis showed numerous vacuoles in the proximal tubular cells of the kidney organoids treated with tacrolimus. Tacrolimus treatment induced oxidative stress and mitochondrial dysfunction, and autophagic activity was enhanced in the kidney organoids. Rapamycin, an autophagy inducer, accelerated cell death in the kidney organoid model of tacrolimus nephrotoxicity, which was attenuated by treatment with 3-methyladenine, an autophagy inhibitor. These findings indicate that the augmentation of autophagy by rapamycin treatment accelerated tacrolimus nephrotoxicity. Conclusions: Our data suggest that human kidney organoids are an effective in vitro model of tacrolimus nephrotoxicity and that autophagy plays a critical role in tacrolimus nephrotoxicity.

        RNA-seq Gene Profiling Reveals Transcriptional Changes in the Late Phase during Compatible Interaction between a Korean Soybean Cultivar (Glycine max cv. Kwangan) and Pseudomonas syringae pv. syringae B728a

        Soybean (Glycine max (L) Merr.) provides plant-derived proteins, soy vegetable oils, and various beneficial metabolites to humans and livestock. The importance of soybean is highly underlined, especially when carbonnegative sustainable agriculture is noticeable. However, many diseases by pests and pathogens threaten sustainable soybean production. Therefore, understanding molecular interaction between diverse cultivated varieties and pathogens is essential to developing disease-resistant soybean plants. Here, we established a pathosystem of the Korean domestic cultivar Kwangan against Pseudomonas syringae pv. syringae B728a. This bacterial strain caused apparent disease symptoms and grew well in trifoliate leaves of soybean plants. To examine the disease susceptibility of the cultivar, we analyzed transcriptional changes in soybean leaves on day 5 after P. syringae pv. syringae B728a infection. About 8,900 and 7,780 differentially expressed genes (DEGs) were identified in this study, and significant proportions of DEGs were engaged in various primary and secondary metabolisms. On the other hand, soybean orthologs to well-known plant immune-related genes, especially in plant hormone signal transduction, mitogen-activated protein kinase signaling, and plant-pathogen interaction, were mainly reduced in transcript levels at 5 days post inoculation. These findings present the feature of the compatible interaction between cultivar Kwangan and P. syringae pv. syringae B728a, as a hemibiotroph, at the late infection phase. Collectively, we propose that P. syringae pv. syringae B728a successfully inhibits plant immune response in susceptible plants and deregulates host metabolic processes for their colonization and proliferation, whereas host plants employ diverse metabolites to protect themselves against infection with the hemibiotrophic pathogen at the late infection phase.

        이현화 〈 불가불가 〉의 기호학적 연구-"극중극"의 보조행위소 기능을 중심으로

        부조리극이란 20세기 후반 서양 연극에서 가장 영향력 있는 연극의 한 형태로, 전통적인 글쓰기와 세계 인식을 동시에 전복한다. 그리고 희극적인 구성에도 불구하고 인간의 본질과 실존의 문제를 치열하게 추적하며, 인과율을 따르는 논리적 플롯을 거부하고, 단편적인 일화들을 나열하는 방식을 취한다. 1960ㆍ70ㆍ80년대의 한국적 상황은 정치적으로는 유신체제로부터 군사 독재 체제의 연장인 제5공화국의 출범으로 문화ㆍ예술이 위축되는 상황하에서 연극쪽에서는 일그러진 정치ㆍ사회현실에 대한 객관적 통찰과 심미적 인식을 향한 연극적 효능이 주된 관심사로 부상하면서 일군의 작가들은 부조리극을 들고 나온다. 한국의 대표적인 부조리극 작가로 이현화를 꼽지 않을 수 없다. 주로 1970~80년대에 집중되어 있는 그의 작품은 초기에는 인간 실존의 문제를 천착하다가 〈 카덴자 〉(1978), 〈 불가불가 >(1982)를 발표하면서 역사인식에로의 전환을 한다. 부조리극 작가들이 즐겨 사용한 연극 기법 중의 하나가 ``극중극`` 양식인데 ``연극성``과 ``거울``로서의 분신이라는 ``구조적 이분``과 ``주제의 이분``에 따른 말하기의 독특한 방식으로 부조리한 현실을 표현해내는 데 적합하다는 이유일 것이다. 극중극은 포괄 구조(전체틀)와 내적 구조로 이루어져 있다. 이현화의 〈 불가불가 〉는 극중극의 형식으로 연극 연습의 과정이라는 포괄 구조와, 역사적 중요 사건들이 "어전회의"라는 내적 구조로, 포괄 구조와 내적 구조가 병렬ㆍ교차되는 구성방식을 취하고 있다. 극중극 즉 내적구조는 포괄구조의 배우①로 하여금 내적구조의 배우⑤를 살해하게끔 한다. 이처럼 내적 구조의 보조적 기능은 관객으로 하여금 연극과 현실의 모호한 경계 형성을 유발시켜 현실에서의 방관자적인 태도를 버리고 실천적 행위를 유도하는 것이다. 본고에서는 그 역학적 가능성을 극중극 기능의 하나인 보조행위소로서의 기능을 그레마스(Greimas)의 행위소 모델(models actantiels)을 분석틀로 삼아 분석해 보고자 한다. "Theatre of the absurde" is a type of the most influential plays in Western Play in the second half of the 20th century, and overthrow equally both the traditional writing and the cognition(understanding) about the world. And in spite of the comic plot, it is drastically in pursuit of the human essential and existential problems and it denies the logical plot under the law of causality, and it takes a way to arrange some fragmentary stories. In 1960s, 70s and 80s, the Korean culture and art had been atrophied because of Korean political situation which had been appeared the 5th republican government extended to the caudillismo (soldier dictatorship) from Yusin organization: The Revitalizing Reforms, and in the field of plays, some theatrical effect for the objective insight and the aesthetic cognition about the present had raised to the surface to the main concerns and a group of writers have appeared with some theatre of the absurd. I think Lee Hyun-hwa is one of the most representative writers in the theatre of absurd. His works that had been centered chiefly between the 1970s and 1980s had been early seeking for the problem of human existence and then publishing "Kadenza(1978)", and "Bulgabulga(1982)", he had started to make a change of the historical cognition One of the theatric techniques that the writers of ``theatre of absurd`` have enjoyed is a form of a play within a play, which is a unique form of speaking in ``the structural dichotomy`` and ``the thematic dichotomy`` which are the incarnations of the nature of theatre and mirror, and the reason for using the technique is proper to express the absurd present. A play within play consists of inclusive structure and internal structure. "Bulgabulga" written Lee Hyun-hwa takes the technique of a play within a play, which composes of the inclusive structure and internal structure, one is a course of exercise in play and the other is composed of the historical events in "the meeting in the presence of the King", which arrange in a row and crosse each other. A play within a play, that is, the internal structure is that the actor 1 of the inclusive structure is forced to kill the actor 1 of the internal structure. Like this, the accessorial function in the internal structure induce the audience to form the vague border between the theatre and the present and then eventually throw away the attitudes of an onlooker and cause the practical behavior. In this treatise, I would like to analyze the dynamic possibilities with the functions of the supplementary actantiels on the basis of Greima`s models actantiels.

        초등 교사가 인식하는 학교의 조직문화와 공동체성이 관계만족도에 미치는 영향

        The purpose of this study is to investigate the influence of organization culture and educational community on teachers perceived satisfaction with members like directors, young children, parents, and peer teachers. The subjects consisted of 148 elementary school teachers working in the P city and data were collected and analyzed by means of descriptive statistics, correlation, and multiple regression. As a result, teachers tend to perceive subcultures of school culture and educational culture as above average. And there were significant correlations between variables of organizational culture and relational satisfactions with members(r =.16~.88), between educational communality of the school and relational satisfactions with members(r =.58~.84). As a result of Regression analysis total explanation of the independent variables, excepting Rational culture on relational satisfactions with members was 92% and there were differences in the explanations in case of each members. Based on this, we discussed the leadership of the principal and the improvement of the sharing of core values between the organizational culture and the members of the school.

        현대 사회의 제조업 트렌드(trend)가 빠르게 변화함에 따라, 제품의 수명주기 감소 및 소비자들의 니즈(needs)가 다양화되면서 개인 맞춤형 생산이 요구되고, 경제 구조가 제조업에서 정보기술(IT)를 포함한 서비스업 중심으로 전향되면서, 전통 제조업은 큰 타격을 받아 제조업의 혁신이 요구되고 있으며 현재 진행형이다. 따라서, 본 연구에서는 스마트팩토리가 4차 산업의 핵심 개념으로 등장함과 동시에, 제조업에서 전 세계적으로 관심이 높아지고 있음에 착안하였으며, 스마트팩토리 설비를 위한 중요 기술의 하나인 ‘예지 보전’(PdM, Predictive Maintenance) 기술에 대하여 깊게 연구를 진행할 필요성이 있다고 판단하여, 스마트센서를 사용한 실험 데이터 취득 후 이를 분석한 결과에 대한 그래프 해석을 연구 방법으로 선정하여, 예지 보전시스템 구축 가능성에 대하여 연구를 진행하였다. As the manufacturing trend in modern society rapidly changes, personalized production is required as the life cycle of products and consumer needs are diversified, and the economic structure has shifted from manufacturing to service industries, including information technology, and manufacturing innovation is required. Therefore, this study focused on the growing interest in the manufacturing industry as well as the core concept of the 4th industry. It was judged that it was necessary to conduct a deep study on PdM (Preditive Maintenance) technology for smart factory facilities.

        In various multiphase system, development of recent technologies are often limited by interfacial breakup phenomena. Interfacial phenomena with dynamic conditions has investigated intensively by numerous researchers. However, previous researchs have typically been conducted on dynamic contact angle. In our previous research, it was investigated the interfacial break-up phenomena of a water-ethanol mixture slug flow in a hydrophobic minichannel. However, due to the limitation of the visualization techniques based on light ray, it was difficult to visualize clearly interfacial behavior and shapes. Therefore, in this study, we introduce the synchrotron X-ray imaging with high spatial and temporal resolutions. We confirm the previous flow patterns (steady, loss, separation) in previous research via synchrotron X-ray imaging. On the basis of visualization data, we observe the gas creates/destroys the liquid-gas interface continuously due to the speed differences with the high-speed flow conditions.
