우울증 어머니에 의해 양육된 우울하면서 소극적인 아동을 위한 아동중심 놀이치료 사례 연구

본 연구는 어머니의 우울로 인하여 적절한 양육을 받지 못해서 우울하면서 소극적인 성향을 보이는 아동에게 아동중심 놀이치료를 실시한 놀이치료 사례이다. 아동에게는 총 45회의 놀이치료가 실시되었고, 어머니 교육 및 상담을 병행하였다. 놀이치료의 목표는 안정된 분위기를 제공하여 치료자와의 신뢰관계를 형성하고, 감정을 표현하고 부정적인 감정을 해소하며, 놀이상황에서 재양육경험을 통해 자율성을 회복 하고 자신감을 증진시키고자 하였다. 놀이치료를 통해 아동의 우울증상이 감소하고, 자아 존중감과 자신감이 증가하였으며, 가정 및 학교에서도 적응적인 모습을 보여 아동중심 놀이치료가 우울하고 소극적인 아동에게 효과적인 치료적 개입으로 밝혀졌다. The purpose of this study was to investigate the effect of child-centered play therapy on a child with depression and withdrawl, due to depression of mother. The subject was a 8-year-old girl who had received child-centered play therapy during 45 sessions. The purpose of this therapy was to express negative emotion and to enhance autonomy and self-confidence, Observed outcomes of therapy reported decreased depression, heighten self-esteem and self-confidence and improved adjustment in home and school. This study supports child-centered play therapy as effective intervention for children who have depression and withdrawl problems.
제4형 가족성 고중성지방혈증 임산부에서 발생한 급성 췌장염 1례

Plasma levels of cholesterol (TC) and triglyceride(TG) physiologically increase during pregnancy. The lipid increment is 23-53% above the pregravidic level for TC and 2- to 3-fold of the prepregnancy level for TG. If the TC and TG are higher than normal values in pregnancy, the patient must be carefully monitored. Acute pancreatitis is the main consequence of hyperlipidemia and occurs either during pregnancy, in the third trimester, or in the puerperium. Mortality is high both for the mother (21%) and the fetus (20%). We report a case of 28 year-old pregnant woman at 29 weeks gestation with hypercholesterolemia (TC = 357 ㎎/dl) and severe hypertriglyceridemia (TG = 1300 ㎎/dl). The patient was admitted to the hospital because of severe epigastric pain, nausea and vomiting. Total serum cholesterol was increased and trigyleride was markedly increased. The electroporesis pattern of serum lipoprotein showed increase in pre-beta lipoprotein fraction, suggesting IV hypertriglyceridemia pattern. According to a review of the literature, the incidence of pancreatitis during pregnancy is 1 in 1000 to 3000 pregnancies. Severe hypertriglyceridemia in pregnancy should be treated with a careful restriction of calories and fat: for preventing acute pancreatitis, hospitalization for intravenous fluid therapy and plasma exchange must be required.
NMR Structural Analysis and 3D Homology Modelling of APG8a from Arabidopsis thaliana

The gene coding for APG8a (At4g21980), a protein from Arabidopsis thaliana, is involved in the autophagy process. The protein is an interesting candidate for structure determination by NMR spectroscopy. Toward this end, APG8a has been produced recombinantly in Escherichia coli and typical NMR experiments such as $^{15}N-HSQC$, HNCA, HN(CO)CA, CBCA(CO)NH, HCCH-TOCSY, HNCO were performed. The backbone resonances, HN, N, CA, CB, and C' were sequence-specifically assigned, and the secondary structures including 3 $\alpha$ helices and $4\beta$ strands were deduced based on the assignments. Due to the intrinsic flexibility or the effect of the denaturant, the backbone resonances were not fully observed. Since the structure calculation by NMR data was not possible, the 3-dimensional model was built based on the sequence homology, and compared with the NMR results. The overall structure of the model could explain and complement the NMR derived secondary structures.

Escherichia coli responds to ever-changing external and internal stresses by rapidly adjusting its physiology for better survival. This adjustment occurs at all levels including metabolites as well as mRNAs and proteins. Although there has been many reports describing E. coli's adaptation to various stresses regarding transcriptomics or proteomics, only a few investigations have been reported regarding this adaptation viewed from metabolites' perspective. We applied four different types of stresses at four different doses as imposed by NaCl, sorbitol, ethanol, and pH to investigate the similarities or differences among the stresses, and which stress causes the largest perturbation of the metabolite composition. We profiled the metabolites under such external stresses by using two-dimensional NMR spectroscopy and identified 39 metabolites including amino acids, sugars, organic acids, and nucleic acids. According to our statistical analysis, the osmotic stress caused by sorbitol differentiated itself from others, while NaCl showed the largest dose dependent metabolic perturbations. We hope this work will form a foundation on which an approach to a successful protein production is systematically provided by a favorable metabolic environment by imposing proper external stresses.

The strategy to incorporate [$^{15}$ N]-labeled amino acids were discussed. Instead of using specific auxotrophic strains for selective labeling, the prototrophic strain, BL2l(DE3), was used with a plasmid, pLysS, and found to be very effective for several amino acids including alanine, lysine, leucine, and threonine. Isoleucine, valine, glutamine, and tyrosine were also found to be effective despite some diffusion into other amino acids. Interesting result was obtained when [$^{15}$ N]-labeled glycine was tried: only glycines were labeled when amino acid mixture was added in the growth medium, and serines were co-labeled when amino acids were omitted. These results can be used as a guideline when selective labeling strategy is considered, and when the resulting data are interpreted.
