      • 당뇨병이 없는 성인에서의 당화 혈색소 농도와 관련된 인자

        연구배경 : 당화 혈색소는 최근 2∼3개월 동안의 혈당 농도를 반영하며, 당뇨병 환자들에서의 심혈관 질환의 발생과 미세혈관 합병증의 예측인자이기도 하다. 최근 연구들에서 당화 혈색소 농도가 당뇨병 환자들 뿐만 아니라 일반인들에 있어서 사망률을 예측하는 중요한 인자임이 밝혀졌다. 본 연구는 당뇨병이 없는 성인들에서 당화 혈색소 농도와 관련된 인자들을 심혈관 위험인자를 중심으로 알아보고자 한다. 방법 : 2001년 4월 1일에서부터 2001년 7월 31일까지 일개 대학 병원의 건강 증진 센터를 방문하여 진료를 받은 20세 이상의 수진자 중 당뇨병이 없고 혈중 당화 혈색소 농도가 본 건강 증진센터의 정상 상한수치인 7.1%이하인 1,778명을 대상으로 하였다. 모든 대상들은 성명, 연령, 신장, 체중, 혈압, 공복시 혈당, 총콜레스테롤, 중성지방, 고밀도콜레스테롤, 당화 혈색소가 측정되었고 자가 기입식 설문조사를 통하여 흡연력이 조사되었다. 당화 혈색소와 관련된 인자들에 대하여 성별로 t-test또는 ANOVA test로 평균값 비교를 한 후 유의한 차이를 보인 인자들에 대하여 당화 혈색소를 4분위수로 나눈 후 중회귀 로짓 분석(multiple logistic regression analysis)으로 가장 낮은 당화 혈색소의 제1구간에 대한 가장 높은 제 4간의 당화 혈색소의 교차비를 구하였다. 결과 : 성별로 각 인자들에 대한 당화 혈색소 농도의 평균값 분석시 남성과 여성 모두에서 연령과 체질량 지수가 증가함에 따라 당화 혈색소 농도의 평균의 유의하게 증가하였고(P<0.05), 남성에서는 총콜레스테롤 240㎎/㎗ 이상, 중성지방 200㎎/㎗ 이상, 고밀도콜레스테롤, 공복시 혈당 110㎎/㎗ 이상에서 평균값이 유의하게 높았으며, 고밀도 콜레스테롤 35㎎/㎗ 이상에서 평균값이 유의하게 낮았고(P<0.05), 여성에서는 고혈압, 총콜레스테롤 240㎎/㎗ 이상, 중성지방 200㎎/㎗ 이상, 공복시 혈당 110㎎/㎗ 이상에서 평균값이 유의하게 높았고(P<0.05). 남성에서의 흡연, 고혈압, 여성에서의 흡연, 고밀도콜레스테롤에 대한 당화 혈색소 농도의 평균값에서는 유의한 차이가 없었다(P<0.05). 로지스틱 다중회귀 분석으로 교차비를 구하였을 때, 남성과 여성에서 연령이 증가할수록 당화 혈색소 농도에 대한 교차비가 유의하게 증가하였고(P<0.05), 비만, 총콜레스테롤 240㎎/㎗ 이상, 공복시 혈당 110㎎/㎗ 이상의 변수들에서 당화 혈색소 농도에 대한 교차비가 유의하게 높았으며(P<0.05), 남서에서는 고밀도콜레스테롤 35㎎/㎗ 이상에 대한 교차비가 유의하게 낮았다(P<0.05). 결론 : 본 연구에서 당뇨병이 없는 성인에서 당화 혈색소 농도와 관련된 인자로 남성과 여성 모두에서는 연령의 증가, 비만, 고콜레스테롤혈증, 공복시 혈당능 장애이며, 남성에서의 저고밀도콜레스테롤혈증도 관련이 있었다. Backgroud : Glycated hemoglobin concentration(HbA1c) is an indicator of average blood glucose concentration over 2-3 months, and in diabetes patients, it is a predictor of cardiovascular disease and microvascular complication. In recent studies, Glycated hemoglobin concentration is predictor of mortality in diabetes patient and people without diabetes. Our study is designed to obtain factors associated glycated hemoglobin in adults without diabetes. Methods : We screened 1778 adults over 20 years who were visited the Health Promotion Center in a university hospital from April 2001 to July 2001, had no diabetes and had HbA1c below 7.1%. Every subjects were assessed with respect to gender, age, height, weight, blood pressure, fasting blood glucose(FBS), total cholesterol, high density lipoprotein(HDL)-cholesterol, triglyceride, HbA1c and smoking were assessed by self-administered questionnaires. factors associated HbA1c were analyzed by t-test and ANOVA test, multiple logistic regression. Results : In men and women, as age and BMI were increased. the means of HbA1c is increased and that of HbA1c was higher in higher total cholesterol(≥100㎎/㎗). In men, the means of HbA1c was lower in higher HDL cholesterol(<35㎎/㎗) and in women, that of HbA1c was higher in hypertension. By multiple logistic regression, in men and women as age is increased, the odd ratio of HbA1c is increased, and that of HbA1c was higher in obesity, higher total cholesterol(≥240㎎/㎗), higher FBS(<110㎎/㎗). In men, the odd ratio of HbA1c was lower in higher HDL cholesterol(<35㎎/㎗). Conclusion : factors associated glycated hemoglobin(HbA1c) in adults without diabetes were increasing age, obesity, hypercholesterolemia, impaired fasting glucose in both sex and in men, low HDL cholesterolemia is too associated.

      • 7β-Hydroxycholesterol이 사람 백혈암에 대한 세포 독성 효과

        The cytotoxic activity of 7β-hydroxycholesterol (7β-OHC) has been evaluated using various leukemia cell lines by MTT test. Among the tested cell lines, 7β-OHC showed to be more cytotoxic to THP-1, human monocytic leukemia cell line. IC50 value of 7β-OHC on THP-1 was 10μM. Kinetic of on THP-1 showed that growth of THP-1 began to decrease at 36 h after treatment 7β-OHC. 7β-OHC induced apoptosis shown by DNA fragmentation by gel electrophoresis. 7β-OHC induced apoptosis through down-regulation of Bcl-2 expression. Taken together, these results suggested that treatment with 7β-OHC inhibits the proliferation of THP-1 cells through apoptosis.

      • Expression of constitutive endothelial and inducible nitric oxide synthase in the sciatic nerve of Lewis rats with experimental autoimmune neuritis

        This study examined the expression of constitutive endothelial nitric oxide synthase (eNOS) and inducible NOS (iNOS) in the sciatic nerve of Lewis rats with experimental autoimmune neuritis(EAN). Western blot analysis showed that both eNOS and iNOS expressions in the sciatic nerves of rats increased significantly during the peak stage of EAN, but declined thereafter. Only minimal amounts of these enzymes were identified in normal rat sciatic nerves. Immunohistochemical studies showed that eNOS was increased in vascular endothelial cells and Schwann cells, but not in inflammatory cells, during the peak stage of EAN. However, iNOS was found mainly in inflammatory macrophages in sciatic nerve EAN lesions. These findings suggest that, depending on the stage of peripheral nervous system autoimmune disease, the increased expressions of both eNOS and iNOS might be involved in either the production of detrimental effects during the induction stage of EAN or in the recovery from EAN paralysis. ⓒ 2002 Elsevier Science B.V.       • HL-60 세포의 성장에 대한 제주자생 해조류의 효과

        The purpose of the present study is to examine the inhibitory effect of extracts of marine algae growing in Jeju on the growth of HL-60 leukemia cells. The effect was observed by measurement of metabolic activity and counting of viable cells. The methanol extracts on 36 species of marine algae growing in Jeju were screened for cytotoxic effect at the concentration of 100㎍/㎖. In results, the inhibitory effect on the growth was observed in the treatment of crude extracts on 7 species of marine algae. In particular, Dictyota dichotoma, Myelophycus simplex, Laurencia okamurae, Plocamium telfairiae, Petalonia binghamiae, Codium minus marketdly inhibited the growth of HL-60 cells.

      • 당유자와 하귤의 숙성에 따른 이화학적 성분 변화

        Dangyooja and Hageul were picked in two week’s interval from September, and physicochemical properties were measured. Average weight, and ratios of flesh and juice increased, while ratio of peel decreased with the ripening of Dangyooja and Hageul. Average weight of Dangyooja was higher than that of Hageul. Ratios of the juice from Dangyooja increased greatly compared with that of Hageul with the ripening. Total acidity of the juice from Dangyooja increased upto 3.61% and then decreased, and soluble solid was 7.9˚Brix. Total acidity and soluble solid of the juice from Hageul showed the same trend as that of Dangyooja except the higher values. Pectinesterase activity of the juice from Hageul was 1.76 times lower than that from Dangyooja. The juice from Hageul was more turbid than that from Dangyooja and showed green and yellow color. The citric and malic acids were the major organic acids, while the ascrbic, oxalic and fumaric acids were the minor in the juice. Citric and malic acids increased greatly and then decreased slightly with the ripening. Total organic acids of the juice from Dangyooja and Hageul were 3.25∼5.51% and 4.35∼6.04%, respectively

        Xanthomonas campestris pv. glycines 8ra produces bacteriocin, glycinecin, which inhibits the growth of bacteria specifically belong to Xanthomonas spp. Xanthomonas spp. includes many important plant pathogenic bacteria such as X. c. pv. vesicatoria which cause the bacterial leaf spot in pepper and tomato and X. oryzae pv. oryzae which causes bacterial leaf blight in rice. Glycinecin has many of good properites as a biological control agent. Since glycinecin inhibit the growth of phytopathogenic Xanthomonas sp, it can be used to control disease without any harm to beneficial bacteria for pant mostly belonged to Pseudomonas sp. However, it needs some modification to develop glycinecin as a effective bilogical control agent. The clone involved in the secretion of bacteriocin production were isolated and named glyAsec.

        Significant hearing loss is the most frequent birth defect than any other birth defect for which there is mandated screening. The goal of universal newborn screening program(UNHSP) is diagnosis and intervention of every hearing handicapped child by 6 months of gae. It requires universal screening of every newborn before hospital discharge. The usual modes for newborn hearing loss detection are otoacoustic emission (OAE) or automated auditory brainstem response(AABR) testing. Recently, combined OAE/ABR system which facilitaes UNHSP with markedly low referral rate is staring to offer a turning point. Various screening protocol has been developed and modified according to the hospital situations and personnels assigned to testing. These protocols and acquired data need to be assessed and verified by a community or nationwide tracking system for quality controle of UNHSP. Initiating a UNHSP in Jeju Island would be a great cornerstone for nationwide UNHSP in Korea. Continued vigilance on the point of the child care provider, otolaryngologist and audiologist is necessary to detect hearing loss in all well-child examination as well as in children with known risk factors.

        Polyorchidism, defined as the presence of more than two testes, is a rare anomaly with approximately 98 cases reported in literature. We report the case of a 73-year-old man who had trichorchidism, two normal testes and one perineal testis, but he had not subjective symptom. All testes were normal in size and in shape. We have reviewed the literature.

      • Increased expression of phospholipase D1 in the spinal cords of rats with experimental autoimmune encephalomyelitis

        Phospholipase D1 (PLD1) expression was studied in the central nervous system (CNS) under the condition of induced experimental autoimmune encephalomyelitis (EAE) in Lewis rats. After inducing EAE, the expression of PLD1 was analyzed by Western blot and immunohistochemistry. Western blot analysis showed that expression of the isozymes PLD1 significantly increased in the spinal cord at the peak stage of EAE, and declined thereafter. Immunohistochemistry showed that PLD1 ­ positive cells increased in number in EAE lesions, which consisted mainly of ED1 ­ positive macro ­ phages and glial fibrillary acidic protein ­ positive astrocytes. In contrast, PLD1 was only weakly expressed in some spinal cord astrocytes in control rats. These results suggest that PLD1 is increased in autoimmune CNS inflammation, and possibly involved in the activation of macrophages and astrocytes in EAE lesions. ⓒ 2001 Elsevier Science Ireland Ltd, All rights reserved.

      • 인슐린의 세포사멸억제 작용에서 신호전달경로의 특성

        Insulin has a variety of activities like as glucose uptake, inhibition of lipolysis, glycogen biosynthesis in a number of cells and tissues. Recently insulin has known to exert an antiapoptotic activity in different cell systems. However, it is not clear by which insulin can protect apoptosis cells from apoptosis. Thus, CHO cells expressing the human insulin receptors (CHO-IR) were used to evaluate the importances of different signaling steps activated by insulin in this process. The Ras/Raf-1/MEK/ERK cascade and the serine/threonine protein kinase Akt are two survival pathways that may be activated in response to insulin. We tested the hypothesis that inhibition of farnesylated Ras was casually related to manumycin-induced apoptosis and showed that the response to manumycin was found to be independent of K-Ras function. Moreover, blocking Ras/Raf-1/MEK/ERK cascade by the expression of a transdominant inhibitory mSOSI mutant in CHO-IR cells kept cells to sensitive to the antiapoptotic action of insulin. Insulin-dependent activation of Akt was blocked by 4th treatment with manumycin. This study suggest that the deplection of short-lived farnesylated proteins by manumycin suppresses the antiapoptotic action of insulin at least in part by disrupting Akt­activation but not of the K-Ras/Raf-1/MEK/ERK-dependent cascade.
