본 연구에서는 도라지 종자의 기능성을 평가하기 위해 ELISA를 이용한 멜라토닌 함량 분석, 최적 추출용매 선정, 그리고 추출물에 대한 화학적 항산화능 및 3T3-L1 세포주를 이용한 세포 내 항산화능을 조사하였다. 종자로부터 멜라토닌 추출에는 75% 에탄올이 가장 우수하였으며 이 추출물의 멜라토닌 함량은 146.8 ng/g이었다. 해당 추출물의 페놀화합물 함량은 50.2 μg GAE/g이었으며 DPPH 라디칼 소거능, ABTS 라디칼 소거능, FRAP, ORAC법으로 분석한 항산화능이 각각 519.1, 645.1, 471.1, 1,438.7 μmol TE/g으로 나타났다. 이러한 종자 추출물의 멜라토닌 및 페놀화합물함량, 그리고 항산화능은 싹이나 뿌리보다 높았다. 3T3-L1 세포주에 40 μg/mL 추출물을 처리한 세포 내 항산화능 분석에서도 종자 추출물이 21.5 unit의 항산화능을 보인 반면에 싹이나 뿌리 추출물의 항산화능은 나타나지 않았다. 이로부터 도라지 종자는 싹이나 뿌리보다 멜라토닌 함량이 높고 항산화능이 우수한 새로운 기능성 식품자원으로서의 활용가능성을 확인하였다. The melatonin content and antioxidant activity of Platycodon grandiflorum (PG) seed extract were measured and compared to those of PG sprout and root extracts. Among the solvents used to extract melatonin from seeds, 75% ethanol was better than methanol or 25, 50 or 100% ethanol (v/v), resulting in 146.8 ng melatonin per g extract by ELISA assay. The polyphenol content of seed extract prepared using 75% ethanol was 50.2 μg gallic acid equivalent (GAE)/g. This extract also exhibited dose-dependent antioxidant activity in various assays, showing 519.1 μmol Trolox equivalent (TE)/g in 2,2-diphenyl-1-picrylhydrazyl radical scavenging, 645.1 μmol TE/g in 2,2’-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) radical scavenging, 471.1 μmol TE/g in ferric reducing antioxidant power, and 1,438.7 μmol TE/g in oxygen radical absorbance activity. Both the melatonin content and the antioxidant activity were highest in seed extract, followed by sprout and root extracts. Longer sprouting lowered the melatonin content and the antioxidant capacity. In a cellular antioxidant activity (CAA) test using 3T3-L1 cells, seed extract (40 μg/mL) showed 21.5 units CAA while neither sprout nor root extract exhibited any activity. These results suggest that PG seed is a novel functional foodstuff superior to its sprout and root in melatonin content and antioxidant activity.
Tae-Eum Type as an Independent Risk Factor for Obstructive Sleep Apnea

<P>Obstructive sleep apnea (OSA) is prevalent and associated with several kinds of chronic diseases. There has been evidence that a specific type of Sasang constitution is a risk factor for metabolic and cardiovascular diseases that can be found in patients with OSA, but there are no studies that address the association between the Sasang constitution type (SCT) and OSA. The purpose of this study was to investigate the association between the SCT and OSA. A total of 652 participants were included. All participants were examined for demographic information, medical history, and completed an interviewer-administered questionnaire on life style and sleep-related variables. Biochemical analyses were performed to determine the glucose and lipid profiles. An objective recording of OSA was done with an unattended home PSG using an Embla portable device. The apnea-hypopnea index (AHI) and oxygen desaturation index (ODI) were significantly higher in the Tae-eum (TE) type as compared to the So-eum (SE) and the So-yang (SY) types. Even after adjusting for confounding variables, the TE type still had a 2.34-fold (95% CI, 1.11–4.94; <I>P</I> = 0.0262) increased risk for OSA. This population-based cohort study found that the TE constitutional type is an independent risk factor for the development of OSA.</P>

<P>Objectives: It has been hypothesized that Sasang constitutional types (SCTs) have a specific hypoactive organ, which can account for vulnerability to related diseases or symptoms. This study examined the relationship between SCTs and irritable bowel syndrome (IBS). Design: Cross-sectional study in a population-based cohort study in Korea. Participants: 1362 individuals (705 men and 657 women) who participated in the Korean Genome and Epidemiology Study. Outcome measures: The participants were classified into SCTs by the integrated diagnostic model and asked about symptoms related to IBS using the Rome II criteria. Results: The prevalence of IBS differed significantly among the SCTs, with 33 (18.3%) of the So-eum (SE) type, 74 (9.9%) of the Tae-eum (TE) type, and 57 (13.2%) of the So-yang (SY) type having IBS. Even after adjustment for possible confounders, the SE type for both sexes continued to show 1.82-fold (95% confidence interval [CI], 1.05-3.16) excess odds of having IBS. Men with SE type had a 2.97 times (95% CI, 1.34-6.58) and a 2.50 times (95% CI, 1.15-5.47) significantly higher odds of having IBS than the TE and SY types, respectively. In analysis for the joint effect of SCT and psychological stress, the multivariate odds ratio of IBS was 3.21 (95% CI, 1.33-7.75) for the SE type and Psychological Well-Being Index-Short Form (PWI-SF) score (<27), and 5.83 (95% CI, 1.80-18.88) for the SE type and PWI-SF (>= 27) compared with the TE type and PWI-SF score (<27). Conclusions: The SE type of SCT is an independent risk factor for IBS. The findings support the hypothesis that persons with SE type are vulnerable to gastrointestinal diseases.</P>
Association of Sasang Constitutional Type with Bone Mineral Density, Osteopenia, and Osteoporosis

Object Although Taeeum and Soyang constitutional types have bigger body shapes and higher body mass index values than those with the Soeum, the relationship between the Sasang constitutional type and bone mass density is controversial and the association of osteopenia and osteoporosis remains unknown. Therefore, we investigated the relationship between bone mineral density, osteopenia, and osteoporosis according to Sasang constitutional type. Methods A total of 2,508 participants were included in this study. Among the study participants, 1,396 had Taeeum type, 276 had Soeum type, and 836 had Soyang type, respectively. The relationships to bone mass density, osteopenia, and osteoporosis in those with Sasang constitutional type were estimated using logistic and linear regression models. Results Bone mass density was significantly higher in the order of Taeeum, Soyang, and Soeum group (p < 0.01). Soeum group in comparison with Taeeum or Soyang group was significantly associated with a high odds ratio for osteopenia and osteoporosis except in the hip and femoral neck in the comparison of Taeeum and Soeum group (p < 0.01). Moreover, the bone mass density of Soeum group decreased more rapidly as the age increased when compared with Taeeum and Soyang group. Conclusions Our findings may contribute to the early prevention and management of high-risk individuals with poor bone mass density, osteopenia, and osteoporosis using Sasang constitution medicine.
