      • Optimal design of bio-inspired isolation systems using performance and fragility objectives

        This study aims to propose a performance-based design method of a novel passive base isolation system, BIO isolation system, which is inspired by an energy dissipation mechanism called 'sacrificial bonds and hidden length'. Fragility functions utilized in this study are derived, indicating the probability that a component, element, or system will be damaged as a function of a single predictive demand parameter. Based on PEER framework methodology for Performance-Based Earthquake Engineering (PBEE), a systematic design procedure using performance and fragility objectives is presented. Base displacement, superstructure absolute acceleration and story drift ratio are selected as engineering demand parameters. The new design method is then performed on a general two degree-of-freedom (2DOF) structure model and the optimal design under different seismic intensities is obtained through numerical analysis. Seismic performances of the biologically inspired (BIO) isolation system are compared with that of the linear isolation system. To further demonstrate the feasibility and effectiveness of this method, the BIO isolation system of a 4-storey reinforced concrete building is designed and investigated. The newly designed BIO isolators effectively decrease the superstructure responses and base displacement under selected earthquake excitations, showing good seismic performance.

        Assessment Method of Slope Excavation Quality based on Point Cloud Data

        Slope excavation quality assessment is essential in water conservancy engineering. This study presents a quality assessment method to estimate the excavation quality of a slope based on point cloud data obtained from a 3D Terrestrial Laser Scanner (TLS). A data processing method, which includes coordinate registration, data partitioning, and noise removal, is proposed to organize raw data from the TLS and ensure accurate and available point cloud data of a slope surface. An excavation quality assessment method with seven indicators, namely, slope ratio, slope toe elevation, back break, volume of back break, remained semi-void rate, unevenness, and offset degree, is proposed to analyze the difference between a slope’s as-design excavation appearance and actual construction appearance. The as-design excavation appearance is expressed by the Building Information Modeling (BIM) model of a slope-excavation face, and it is a reference to assess the actual construction appearance. The slope excavation quality of a water conservancy project in southwest China is evaluated with the proposed method and the traditional method. The results demonstratethat the proposed method can estimate the slope excavation quality effectively, rapidly, and accuately, and can provide theoretical data support for subsequent work.

      • Analysis of Differential-expressed Proteins of Acidithiobacillus ferrooxidans Grown under Phosphate Starvation

        Acidithiobacillus ferrooxidans is one of the most important bacterium used in bioleaching, and can utilize $Fe^{2+}$ or sulphide as energy source. Growth curves for Acidithiobacillus ferrooxidans under phosphate starvation and normal condition have been tested, showing lag, logarithmic, stationary and aging phases as seen in other bacteria. The logarithmic phases were from 10 to 32 hours for Acidithiobacillus ferrooxidans cultivated with normal cultivating condition and from 20 to 60 hrs for Acidithiobacillus ferrooxidans cultivated phosphate starvation. Differences of protein patterns of Acidithiobacillus ferrooxidans growing in case of normal or phosphate starvation were separately investigated after cultivation at $30^{\circ}C$ by the analysis of two-dimensional gel electrophoresis (2-DE), matrix-assisted laser desorption/ionization (MALDI)-Mass spectrometry. There were total 6 protein spots identified, which were Recombination protein recA, RNA helicase, AP2 domain-containing transcription factor, NADH dehydrogenase I chain D, Hyothetical protein PF1669, and Transaldolase STY3758. From the 6 identified protein spots, 3 proteins were found to be decreased in expression at the cultivating condition of phosphate starvation, while another three upregulated.

        Anti-fouling nanofiltration membranes based on macromolecule crosslinked polyvinyl alcohol

        Nanofiltration membranes with excellent anti-fouling properties were fabricated through a green surfacecrosslink method. Poly (styrene-maleic anhydride) (SMA) blended polyethersulfone (PES) membrane wasused as a support. The SMA in the PES support served as a macromolecular crosslinker. After coating ofpolyvinyl alcohol (PVA) on the surface of the SMA-PES support, a PVA selective layer with gradient crosslinkedstructure was formed, through the esterification between the anhydride groups in SMA and thehydroxyl groups in PVA. This process adopts a full-water fabrication strategy without secondary pollution. The gradiently cross-linked PVA/SMA-PES membrane, with molecular weight cut-off (MWCO) about600 Da and permeance around 10-12 L/(m2hbar), exhibited good dye/salts separation performance. Inthe filtration of a mixed solution of Congo Red (CR) and calcium chloride (CaCl2), the membrane rejected98% CR and only had 10% rejection of CaCl2. In addition, this membrane processed an excellent antifoulingproperty. In the BSA fouling test, the pure water flux recovery ratio (FRR) was 99%. It is expectedthat the PVA/SMA-PES membrane can be used in organic desalination.

        High expression of RAD51 promotes DNA damage repair and survival in KRAS-mutant lung cancer cells

        RAD51 recombinase plays a critical role in homologous recombination and DNA damage repair. Here we showed that expression of RAD51 is frequently upregulated in lung cancer tumors compared with normal tissues and is associated with poor survival (hazard ratio (HR) = 2, P = 0.0009). Systematic investigation of lung cancer cell lines revealed higher expression of RAD51 in KRAS mutant (MT) cells compared to wildtype (WT) cells. We further showed that MT KRAS, but not WT KRAS, played a critical role in RAD51 overexpression via MYC. Moreover, our results revealed that KRAS MT cells are highly dependent on RAD51 for survival and depletion of RAD51 resulted in enhanced DNA double strand breaks, defective colony formation and cell death. Together, our results suggest that mutant KRAS promotes RAD51 expression to enhance DNA damage repair and lung cancer cell survival, suggesting that RAD51 may be an effective therapeutic target to overcome chemo/radioresistance in KRAS mutant cancers.         Key Technologies of Supercapacitor Energy Storage System of IP Transmitter

        A practical application of a supercapacitor energy storage system in a polarization instrument is proposed on the basis of the energy storage requirements of an induced polarization (IP) transmitter for geophysical exploration. We focused on the energy storage system of a supercapacitor, the topology of the power converter, and the system control strategy as key technologies, and we performed theoretical research and experimental tests on the system and developed an experimental platform. The experiments validated the theoretical research on the key technologies of the supercapacitor energy storage system and demonstrated the effectiveness of the innovation. Results showed that the storage system is efficient and satisfies the energy storage needs of the IP transmitter.

        Observer-based Stabilization of Uncertain Switched Linear Systems with Admissible Edge-dependent Switching Signals

        This paper focuses on the stabilization of a class of uncertain switched linear systems with admissible edge-dependent (AED) switching signals, where disturbances exist in not only the state equation but also output equation. The performance of switchings from the current system mode to the other is shown by the concept of admissible transition edge-dependent weights (ATEWs). A special output transformation is adopted to construct a new system with the same states as the original system but different output, where the disturbance in measurement equation is decoupled. Then, a multiple-mode unknown input observer (UIO) with AED switching is proposed to simultaneously estimate the state and disturbance. Next, an observer-based controller is developed to stabilize the uncertain switched system, then the conditions ensuring the stability of the closed-loop system are provided. Finally, the validity of the proposed method is shown by an example.

        A Hybrid Link Protection Scheme for Ensuring Network Service Availability in Link-state Routing Networks

        The internet is playing an increasingly crucial role inboth personal and business activities. In addition, with the emergenceof real-time, delay sensitive and mission-critical applications,stringent network availability requirement is put forward for internetservice providers (ISPs). However, commonly deployed intradomainlink-state routing protocols react to link failures by globallyexchanging link state advertisements and recalculating routingtable, inevitably causing significant forwarding discontinuityafter a failure. Therefore, the loop-free criterion (LFC) approachhas been widely deployed by many ISPs for coping with the singlenetwork component failure scenario in large internet backbones. The success of LFC lies in its inherent simplicity, but this comesat the expense of letting certain failure scenarios go unprotected. To achieve full failure coverage with LFC without incurring significantextra overhead, we propose a novel link protection scheme,hybrid link protection (HLP), to achieve failure resilient routing. Compared to previous schemes, HLP ensures high network availabilityin a more efficient way. HLP is implemented in two stages. Stage one provides an efficient LFC based method (MNP-e). Thecomplexity of the algorithm is less than that of Dijkstra’s algorithmand can provide the similar network availability with LFC. Stage two provides backup path protection (BPP) based on MNP-e,where only a minimum number of links need to be protected, usingspecial paths and packet headers, to meet the network availabilityrequirement. We evaluate these algorithms in a wide spread ofrelevant topologies, both real and synthetic, and the results revealthat HLP can achieve high network availability without introducingconspicuous overhead. HLP not only needs around 10% time ofthat of full protection, but also provides full protection capabilitiesthat full protection provide.
