        Characterization of Ecdysteroid UDP-Glucosyltransferase Gene Promoter from Bombyx mori Nucleopolyhedrovirus

        Bombyx mori nucleopolyhedrovirus(BmNPV) ecdysteroid UDP-glucosyltransferase gene (egt) promoter fragments of different lengths were amplified from BmNPV ZJ-8 genomic DNA by PCR. Reporter plasmids pBmegt542-luc, pBmegt309-luc and pBmegtl59-luc with luciferase (lue) driven by egt promoters were constructed. Both in vitro and in vivo expressions showed that BmNPV egt promoter activity requires the transactivation of viral factor(s), and expression of luc was detected earliest at 24 hrs post infection (pi). BmNPV ZJ-8 homologous region 3 (hr3) increased the expression of luc by over 1,600-fold. Molting hormone of 1.0 - 2.0 $\mu\textrm{g}$/$m\ell$ can dramatically down regulate expression of luc. Juvenile hormone analogue of 0.5-2.0 ${\mu}g$/$m\ell$ increased expression of luc by 145.8% to 75.7%. Deletion assay revealed that the promoter fragment of 159 bp contains the basal promoter structure; Promoter fragments of 309 bp and 542 bp showed similar but much higher transcriptional activities than that of 159 bp, suggesting that nucleotide from -159 to -309 nt upstream the translation initiation site harbors the main cis-acting elements.

        Effects of Insect Hormones on the Replication of Nucleopolyhedrovirus

        An experimental study was undertaken to quantify the effects of insect hormones on the replication of nucleopolyhedrovirus (NPV). The results demonstrated that TCID/ sub 50/ at 72 h post-infection (hpi) rose systematically from 0.55$\times$10$^{8}$ /m1, for untreated cells, up to 1.67$\times$10$^{8}$ / ml at 3$\mu$g/ml, then dropped down to 1.45$\times$10$^{8}$ /m1 at 4 $\mu$g/ml, by adding ecdysone to the culture medium for Bm-N cells infected with a wild-type Bambyx mori. nucleopolyhedrovirus (BmNPV). The optimum enhancement of about 3 times on budded virus (BV) titer at 72 hpi was given at 3 $\mu$g/ml of ecdysone. While the polyhedra number had no obvious variation within the range of concentrations from 0 to 4 $\mu$g/ml. By addition of juvenile hormone analogue (JHA) into the media with this concentration range, the BmNPV TCID/ sub 50/ and polyhedra number at 72 hpi did not show significant changes. Also, the addition of either 3 $\mu$g/ml of ecdysone or 3 $\mu$g/ml of JHA to the culture media did not appear to affect the TCID/ sub 50/ and polyhedra number significantly in infected Sf-21 cells with the autographa californica nucleopolyhedrovirus (AcMNPV).

        Identification of candidate odorant‐degrading enzyme genes in the antennal transcriptome of Aphidius gifuensis

        Odorant‐degrading enzymes (ODEs) have been found in insect antennae and play a critical role in signal chemical degradation once the message is conveyed. Significant progress has been made in characterizing ODEs in a variety of pests but very little is known in their natural enemies. We have carried out an antennae‐ and sex‐specific transcriptome of Aphidius gifuensis, a natural enemy of aphid, to identify the candidate ODEs. Based on the antennae‐ and sex‐specific transcriptome, a total of 100 putative ODEs were identified including one aldehyde oxidase (AOX), four alcohol dehydrogenases (ADs), eight UDP‐glucuronosyltransferases (UGTs), 45 cytochrome P450 (P450s), nine glutathione S‐transferases (GSTs) and 40 carboxylesterases (CCEs or CXEs). Additionally, we used RT‐qPCR to determine the expression profiles of these genes in tissues of both sexes. Based on the phylogenic analysis and tissue‐expression patterns, AgifEstE4, AgifCXE3, AgifCCE4, AgifCCE7, and AgifCCE18 were suggested as key ODEs in A. gifuensis. In addition, the female or male specifically enriched genes, such as AgifCCE17, AgifEstB1, AgifCYP18a1, AgifUGT2C2, were also considered to involve in the chemosensory processing in A. gifuensis. This study not only identified the candidate ODEs in A. gifuensis but also provided source for further exploration of the molecular mechanisms of chemical signal transductions in A. gifuensis, as well as other hymenopteran species.

        A Microscopic Cohesive Zone Model and Effects of Interface on the Transverse Mechanical Properties for Composites

        The carbon fiber/epoxy matrix interface plays an important role in the behavior and damage onset of unidirectionalfiber reinforced epoxy matrix composites and accurate modeling techniques are needed to study the effects of this complexregion on the composite response. In this work, a microscopic cohesive zone model (MCZM) based on atomic potentialenergy is established for the interface. A multiscale analysis method is proposed for predicting the transverse mechanicalproperties of unidirectional fiber composites: (1) At the microscale, MCZM is established for the interface; (2) At themesoscale, a unit cell model is established for the fiber, matrix and interface; and (3) At the macroscale, the homogenizationmethod, failure criteria and damage degradation models are used for predicting the transverse mechanical properties. Subsequently, the transverse mechanical properties and the damage evolution process are simulated with the multiscaleanalysis method. A comparison between the simulations and experiments shows that: (1) The maximum errors of thepredicted transverse modulus and transverse strength are -4.45 % and -1.28 %, respectively; and (2) MCZM can reflect thenonlinearity of epoxy matrix composite materials. Moreover, the effects of the interfacial strength on the macroscopictransverse mechanical properties and the damage onset are analysed. The simulation results show that: (1) The interfacialstrength has a more significant effect on the transverse strength and ultimate strain than on the transverse modulus; (2)Decreasing the interfacial strength has a greater effect on the transverse modulus, strength and ultimate strain than increasingthe interfacial strength; and (3) The interfacial strength can change the damage onset.

        Systemic Inflammatory Biomarkers, Especially Fibrinogen to Albumin Ratio, Predict Prognosis in Patients with Pancreatic Cancer

        Purpose Systemic inflammatory response is a critical factor that promotes the initiation and metastasis of malignancies including pancreatic cancer (PC). This study was designed to determine and compare the prognostic value of neutrophil-to-lymphocyte ratio (NLR), platelet-to-lymphocyte ratio (PLR), monocyte-to-lymphocyte ratio (MLR), and fibrinogen-to-albumin ratio (FAR) in resectable PC and locally advanced or metastatic PC. Materials and Methods Three hundred fifty-three patients with resectable PC and 807 patients with locally advan-ced or metastatic PC were recruited in this study. These patients were classified into a training set (n=758) and a validation set (n=402). Kaplan-Meier survival plots and Cox proportional hazards regression models were used to analyze prognosis. Results Overall survival (OS) was significantly better for patients with resectable PC with low preoperative PLR (p=0.048) and MLR (p=0.027). Low FAR, MLR, NLR (p < 0.001), and PLR (p=0.003) were significantly associated with decreased risk of death for locally advanced or metastatic PC patients. FAR (hazard ratio [HR], 1.522; 95% confidential interval [CI], 1.261 to 1.837; p < 0.001) and MLR (HR, 1.248; 95% CI, 1.017 to 1.532; p=0.034) were independent prognostic factors for locally advanced or metastatic PC. Conclusion The prognostic roles of FAR, MLR, NLR, and PLR in resectable PC and locally advanced or metastatic PC were different. FAR showed the most prognostic power in locally advanced or metastatic PC. Low FAR was positively correlated with OS in locally advanced or metastatic PC, which could be used to predict the prognosis.

        Effects of Length and Oxidation of Multi-walled Carbon Nanotubes on the Mechanical and Electrical Properties for Epoxy Matrix Composites

        This work investigated the effects of length and oxidation of multiwalled carbon nanotubes (CNTs) on theinterfacial strength, fracture toughness enhancement and conductivity for epoxy matrix composites by experiment. Firstly, aseven-step processing scheme was proposed to obtain highly dispersed CNT-epoxy composites. Subsequently, two feasiblemethods were presented to characterize the interfacial strength and used to study the effects of length and oxidation of CNTson the interfacial strength. Thirdly, based on the experimental results of the fracture toughness of CNT-epoxy composites, weproposed a new fracture theory for CNT-based composites which is contrary to the conventional fracture theory for fiberbasedcomposites. The experimental results show that the fracture toughness enhancement reaches maximum at the criticaloxidation time, when the interfacial strength equals CNT strength. At last, to obtain CNT-epoxy composites with both highmechanical and electrical properties, a feasible solution was put forward.

        A Multiscale Analysis Method for Predicting the Transverse Mechanical Properties of Unidirectional Fibre-reinforced Composites

        Mechanical property prediction methods for composites are very important as theoretical tools for engineeringstructural design. To more accurately capture the transverse mechanical properties of composites, a multiscale analysismethod is developed in this paper. Finally, the effects of the interfacial strength on the macroscopic transversemechanical properties and the damage onset are analysed. The following conclusions are drawn from the simulation results:(1) The interfacial strength has a more significant effect on the transverse strength and ultimate strain than on the transversemodulus; (2) Decreasing the interfacial strength has a greater effect on the transverse modulus, strength and ultimate strainthan increasing the interfacial strength; and (3) The interfacial strength can change the damage onset.
