Effect of Turbulent Fluctuation on Settling Behavior of Dredged Slurry in Sheared Condition

For the land shortage problem upon the city development in coastal areas, it is of great importance to improve the overall understanding of the settlement behavior of dredged slurry and determine the required site water storage capacity as well as the mechanical properties of the newly formed dredged slurry. This paper presents an experimental investigation on the effect of turbulent fluctuation on the settling behavior of dredged slurry in sheared condition via a self-developed testing apparatus namely double cylinder settling column. Test results showed that the settling velocity of the soil-water interface initially increased then decreased with the increasing rotation speed of the inner column. The interface would disappear and a stable suspended layer was observed when the rotation speed was greater than 20 rad/min, where water contents and grain composition were not changed over the depth. The particle content was closer to that of the original sample as the rotation speed kept increasing in the suspended layer. It was found that the stabilized flocs were gradually broken down by the turbulence effect existing in the sheared slurry. Moreover, the stabilized flocs were losing the stability gradually as the flowing velocity increased till 10 rad/min and the floc structure was broken down completely once the flowing velocity was greater than 20 rad/min.

A novel bright-green leaf mutant, bgl11, derived from Nipponbare (Oryza sativa L. ssp. japonica) treated by ethyl methanesulfonate (EMS), exhibited a distinct brightgreen leaf phenotype throughout development. Chlorophyll contents of bgl11 decreased significantly than that of its wild-type parent. Genetic analysis suggested that the brightgreen leaf trait was controlled by a single recessive nuclear gene, which was tentatively designed as BGL11(t). To isolate the BGL11(t) gene, a map-based cloning strategy was employed, and the gene was finally mapped in a 94.7 kb region between marker InDel11-5 and InDel11-9 on the long arm of chromosome 11, in which no gene leaded to leaf-color mutation had been mapped or cloned. Cloning and sequencing analysis revealed that, LOC_Os11g38040, which was predicted to encode an expressed protein, had a 9 bp segment deletion in the coding region of bgl11. Furthermore, the transgenic plants with wild-type gene LOC_Os11g38040were restored to normal phenotype. Accordingly, the gene (LOC_Os11g38040) was identified as the BGL11(t) gene. These results are very valuable for further study on BGL11(t)gene and illuminating the mechanism of chloroplast development in rice.

In this work, a highly sensitive method using a colorimetric probe coupled to dispersive liquid?liquid microextraction (DLLME) was developed for the quantitative determination of dopamine (DA) in serum. The DA in serum was concentrated by DLLME to increase the detection sensitivity and reduce the matrix effects. After the DLLME process, a colorimetric probe of silver triangular nanoparticles (AgTNPs) was used to detect DA, which was based on the plasma transformation of AgTNPs caused by strong interactions with melamine (MA). The results showed that DA could inhibit the aggregation of AgTNPs induced by MA, resulting in the recovery of the surface plasmon resonance (SPR) peak of AgTNPs. Thus, the DLLME method followed by colorimetric probe detection of DA can be achieved. The parameters affecting the proposed method were optimized, under the optimal conditions, a linear calibration curve was obtained over a concentration range of 5 to 250?nM with a recovery from 94.4 to 101.3%. The detection limit was 1.6 nM (at an S/N ratio of 3). The present method was successfully applied to determine DA in human serum.

Aim: To investigate the expression of protein kinase $CK2{\alpha}$ ($CK2{\alpha}$) in human thyroid disease and its relationship with thyroid cancer metastasis. Materials and Methods: Using immunohistochemistry we measured the expression of $CK2{\alpha}$ in 76 benign and malignant human thyroid cancer tissues, including 10 pairs of papillary carcinoma tissues with or without lymph node cancerous metastasis and similarly 10 pairs of lymph nodes. Results: The expression of $CK2{\alpha}$ was found to be higher in thyroid carcinoma cases (papillary carcinoma, follicular carcinoma, anaplastic carcinoma and medullary carcinoma) than in ones such as chronic lymphocytic thyroiditis, nodular goiter and adenoma. These findings were also confirmed by RT-PCR and Western blotting. More strikingly, elevated expression of $CK2{\alpha}$ in thyroid papillary carcinoma tissues was not only significantly associated with lymph node cancerous metastasis and clinical stage of thyroid cancers; but also correlated with epithelial-mesenchymal transition (EMT) and high tenascin C (TNC) expression. In addition, EMT and high TNC expression in thyroid carcinoma tissues was significantly associated with lymph node cancerous metastasis. Conclusions: Elevated expression of nuclear $CK2{\alpha}$ is a poor prognosis indicator in lymph node cancerous metastasis of human thyroid cancers.

Objective: Self-sampled human papillomavirus (HPV) testing is a potential option forcer vical cancer screening, but research is scarce in Cambodia. We evaluated the feasibility,accuracy, and acceptability of self-sampled HPV testing using careHPV. Methods: A cross-sectional study including women aged 20–49 years attending 2 nationalhospitals in the capital city was conducted. Women under went both self-sampling andclinician-sampling of specimens, and were then asked to complete an acceptabilityquestionnaire. The paired samples were analyzed for high-risk HPV by careHPV andgenotyped by polymerase chain reaction (PCR). Results: A total of 375 women were eligible for inclusion. Based on PCR, 78.9% were negativefor HPV in both self and clinician-samples, 9.9% had a complete HPV type match, and 6.1%had all HPV types in clinician-samples also detected in self-samples. In 5.1%, one or more HPVtypes identified in the clinician-samples were missed in self-samples. When using careHPV,the overall agreement between the 2 sampling methods was 95.7% (95% confidence inter val[CI]=95.8–95.6) with good concordance (κ=0.66, 95% CI=0.56–0.76). Nearly 90% of the womenpreferred clinician-sampling over self-sampling, citing greater comfort, ease, and speed. Conclusion: Self-sampled HPV testing using careHPV could be an option for cer vical cancerscreening in Cambodia; however, it requires periodic quality control of handling procedures. In addition, women’s health education regarding the accuracy of self-sampled HPV testingand the importance of follow-up in cases of positive results is needed.

Objective: To identif y the maximum tolerated dose (MTD) of docetaxel combined with afixed dose of cisplatin (75 mg/m2) delivered as hyperthermic intraperitoneal chemotherapy(HIPEC) in patients with ovarian cancer. Methods: In this phase I trial, a time-to-event Bayesian optimal inter val design was used. Docetaxel was given at a starting dose of 60 mg/m2 and was increased in 5 mg/m2 incrementsuntil the MTD was determined or the maximum dose level of 75 mg/m2 was reached. The dose-limiting toxicity (DLT) rate was set at 25%, with a total sample size of 30 patients. HIPEC wasdelivered immediately following debulking surger y at a target temperature of 43°C for 90 minutes. Results: From August 2022 to November 2022, 30 patients were enrolled. Among thepatients who received a dose of docetaxel ≤65 mg/m2, no DLT was reported. DLTs wereobser ved in one patient who received 70 mg/m2 docetaxel (grade 3 anaemia) and in threepatients who received 75 mg/m2 docetaxel (one case of grade 3 anaemia, one case of grade3 hepatic impairment and one case of grade 4 thrombocytopenia). Patients treated withdocetaxel 75 mg/m2 in combination with cisplatin 75 mg/m2 had an estimated DLT rate of25%, which was the closest to the target DLT rate and was therefore chosen as the MTD. Conclusion: Docetaxel, in combination with a fixed dose of cisplatin (75 mg/m2), can be usedsafely at intraperitoneal doses of 75 mg/m2 in ovarian cancer patients who received HIPEC(43°C, 90 minutes) following debulking surger y. Objective: To analyze the detection rate of the inferior pyloric artery (IPA) in patients with gastric cancer by computed tomography arteriography (CTA). Materials and Methods: Fifty-four patients (48 males and 6 females; mean age, 59.0 ± 1.5 years) who had undergone radical gastrectomy for gastric cancer from September 2016 to July 2017 at our institution were recruited prospectively. Patients underwent abdominal contrast-enhanced CT scans and CTA imaging reconstruction before the operation. The origin of the IPA in all cases was determined by a radiologist based on CTA images and verified by the surgeon. The accuracy of CTA in diagnosing the origin of the IPA was calculated. Dominant vessels of the origin were analyzed. Results: IPAs were detected by CTA in 51 patients (94.4%). Among these, IPAs originated from the right gastroepiploic artery (RGEA) (24 cases), the gastroduodenal artery (GDA) (4 cases), and the anterior superior pancreaticoduodenal artery (ASPDA) (20 cases). In the remaining 3 cases, the IPAs contained two branches originating from the RGEA and ASPDA, respectively. During surgery, in 2 (3.7%) of the 54 cases of gastric cancer, IPAs could not be detected; the IPAs originated from the RGEA (22 cases), GDA (5 cases), and ASPDA (24 cases). One case had an IPA originating from both the RGEA and the GDA. Finally, the accuracy of CTA in diagnosing the origin artery of the IPA was 85.2% (46/54). Conclusion: CTA can detect the origin of the IPA accurately, which can aid surgeons while performing pylorus-preserving operations.

Apoptotic cell death plays a predominant role in histone deacetylase (HDAC) inhibitor-induced cytotoxicity. Nuclear morphological changes and activation of apoptotic executors are involved in CTS203-induced cell death. However, emerging issues of HDAC inhibitor-resistance have been observed in patients. Herein, MCF-7 cells were continuously exposed to CTS203 until the derived cells could proliferate normally in its presence. The newly obtained CTS203-resistant cells were nominated as MCF-7/203R. Compared to MCF-7 original cells, the MCF-7/203R cells were less sensitive to CTS203-induced apoptosis, with a minimal 6-fold higher $IC_{50}$ value. In contrast, the expression of Beclin-1 was dramatically up-regulated, positively correlated to the acquisition of CTS203-resistance. Our results revealed the participation of autophagy in acquired HDAC inhibitor-resistance and further identified Beclin-1 as a promising target for anti-drug resistance.
