        Minus-C odorant binding protein TcasOBP7G contributes to reproduction and defense against phytochemical in the red flour beetle, Tribolium castaneum

        Insect odorant binding proteins (OBPs) paly an important role in the recognition and transmission signals. One odorant binding protein gene, TcasOBP7G, was significantly up-regulated after exposure to eucalyptol. Never theless, the causal role of TcasOBP7G in Tribolium castaneum is still unclear. Here, TcasOBP7G was cloned and characterized from T. castaneum, aligning with the typical characteristic of Minus-C OBPs. The analysis of gene expression profile showed that TcasOBP7G is highly expressed at late larval and adult stage, with particularly high expression levels in ovary and testis of adults, suggesting that TcasOBP7G might be associated with reproduction of T. castaneum. Furthermore, RNAi of TcasOBP7G led to a decrease in spawning, while the development of gonads was normal, suggesting that TcasOBP7G may medicate the reproduction of T. castaneum. Additionally, the mRNA level of TcasOBP7G was significantly increased after exposure to eucalyptol, and knockdown of TcasOBP7G resulted in an increased susceptibility of T. castaneum to eucalyptol, indicating that TcasOBP7G is associated with the tolerance of T. castaneum to eucalyptol. Altogether, these results indicate that TcasOBP7G is required for both reproduction and defense against phytochemical in T. castaneum. This study will provide a theoretical basis for the development of novel control strategies based on the olfaction of insects.

        A sigma class glutathione S-transferase gene regulated by the CncC pathway is required for phytochemical tolerance in the red flour beetle, Tribolium castaneum

        Insect glutathione S-transferases (GSTs) play a crucial role in the detoxification of exogenous compounds, especially insecticides and plant allelochemicals. A sigma class GST gene, TcGSTS7, mediates the response to eugenol in Tribolium castaneum. However, the mechanism underlying this effect remains largely unknown. In this study, TcGSTS7, which exhibits a structural motif and domain organization characteristic of GSTs, was cloned from the T. castaneum genome. Spatiotemporal expression analysis revealed that TcGSTS7 was most highly expressed at the late larva stage and was mainly expressed in the fat body and epidermis of larvae and adults, suggesting that TcGSTS7 may play a potential role in the protection against toxic xenobiotics in T. castaneum. Furthermore, the expression of TcGSTS7 was significantly induced after exposure to eugenol, while RNA inter ference (RNAi) targeting TcGSTS7 enhanced the sensitivity of the beetle to eugenol, indicating that TcGSTS7 is involved in the tolerance of T. castaneum to this insecticide. Interestingly, the depletion of TcCncC, which encodes a transcription factor of the CncC pathway that has been associated with the regulation of detoxification-related genes in insects, led to a reduction in the TcGSTS7 transcript level following exposure to eugenol, which suggests that TcGSTS7 acts downstream of the CncC pathway. Combined, these results indicated that TcGSTS7 partici pates in the tolerance of T. castaneum to phytochemicals in a CncC pathway-dependent manner. These findings have implications for the development of novel drugs for use in pest control.

        The Influence of Changing PV Array Interconnections under a Non-uniform Irradiance

        Usually, the output characteristics of a photovoltaic (PV) array are significantly affected by non-uniform irradiance which is caused by ambient obstacles, clouds, orientations, tilts, etc. Some local maximum power points (LMPP) in the current-voltage (I-V) curves of a PV array can result in power losses of the array. However, the output power at the global maximum power point (GMPP) is different in different interconnection schemes in a PV array. Therefore, based on the theoretical analysis and mathematical derivation of different topological structures of a PV array, this paper investigated the output characteristics of dual series PV arrays with different interconnections. The proposed mathematical models were also validated by experimental results. Finally, this paper also concluded that in terms of performance, the total cross tied (TCT) interconnection was not always the optimal structure, especially in a dual series PV array. When one of the PV modules was severely mismatched, the TCT worked worse than the series parallel (SP) structure. This research can provide guidance for switching the interconnection to gain the greatest energy yield in a changeable- structure PV system.

        Operational data-based adaptive improvement method of gas turbine component characteristics for performance simulation

        Accurate component maps are crucial for gas turbine performance simulation. However, generating component maps is challenging due to limited data availability and individual characteristic differences between gas turbines. Thus, a component characteristic adaptation method is proposed here. Initially, the original component analytical formulas (OCAF) are enhanced, and the analytical normalized characteristic parameters (ANCPs) are calculated. Subsequently, the real normalized characteristic parameters (RNCPs) are calculated reversely based on field-measured data. Next, the tuning factors are optimized to obtain optimal improved component analytical formulas (ICAF). Finally, the effectiveness of the proposed method is validated using LM2500+ gas turbine field data and compared with two previous adaptive methods. The results reveal that the proposed method offers high tunability and computational efficiency during the adaptation process, significantly improving the accuracy of the gas turbine performance simulation model. This study paves the way for more reliable gas turbine performance simulations and enhanced fault diagnosis in the field.

        Ruminal pH pattern, fermentation characteristics and related bacteria in response to dietary live yeast (Saccharomyces cerevisiae) supplementation in beef cattle

        Objective: In this study we aimed to evaluate the effect of dietary live yeast supplementation on ruminal pH pattern, fermentation characteristics and associated bacteria in beef cattle. Methods: This work comprised of in vitro and in vivo experiments. In vitro fermentation was conducted by incubating 0%, 0.05%, 0.075%, 0.1%, 0.125%, and 0.15% active dried yeast (Saccharomyces cerevisiae, ADY) with total mixed ration substrate to determine its dose effect. According to in vitro results, 0.1% ADY inclusion level was assigned in in vivo study for continuously monitoring ruminal fermentation characteristics and microbes. Six ruminally cannulated steers were randomly assigned to 2 treatments (Control and ADY supplementation) as two-period crossover design (30-day). Blood samples were harvested before-feeding and rumen fluid was sampled at 0, 3, 6, 9, and 12 h post-feeding on 30 d. Results: After 24 h in vitro fermentation, pH and gas production were increased at 0.1% ADY where ammonia nitrogen and microbial crude protein also displayed lowest and peak values, respectively. Acetate, butyrate and total volatile fatty acids concentrations heightened with increasing ADY doses and plateaued at high levels, while acetate to propionate ratio was decreased accordingly. In in vivo study, ruminal pH was increased with ADY supplementation that also elevated acetate and propionate. Conversely, ADY reduced lactate level by dampening Streptococcus bovis and inducing greater Selenomonas ruminantium and Megasphaera elsdenii populations involved in lactate utilization. The serum urea nitrogen decreased, whereas glucose, albumin and total protein concentrations were increased with ADY supplementation. Conclusion: The results demonstrated dietary ADY improved ruminal fermentation dosedependently. The ruminal lactate reduction through modification of lactate metabolic bacteria could be an important reason for rumen pH stabilization induced by ADY. ADY supplementation offered a complementary probiotics strategy in improving gluconeogenesis and nitrogen metabolism of beef cattle, potentially resulted from optimized rumen pH and fermentation.         Interval Observer-based Robust Trajectory Tracking Control for Quadrotor Unmanned Aerial Vehicle

        In this work, a robust trajectory tracking control strategy is developed for the quadrotor unmanned aerial vehicle (QUAV) with unknown external disturbance. The nonlinear dynamic model of QUAV is established. Different from the traditional disturbance observer and adaptive estimation method, this paper adopts the interval observer technique to approximate the upper and lower bounds of the unknown disturbance. In this way, the proposed interval observer can cover its all possible values and show its scope in real time, especially before the disturbance error estimation converges. Combining with the backstepping approach, a robust tracking control approach is proposed to ensure that the desired trajectory can be successfully followed. Finally, comparative numerical simulation results are provided to evaluate the presented robust tracking controller.

      • An Effective Framework for Contented-Based Image Retrieval with Multi-Instance Learning Techniques

        Multi-Instance Learning(MIL) performs well to deal with inherently ambiguity of images in multimedia retrieval. In this paper, an effective framework for Contented-Based Image Retrieval(CBIR) with MIL techniques is proposed, the effective mechanism is based on the image segmentation employing improved Mean Shift algorithm, and processes the segmentation results utilizing mathematical morphology, where the goal is to detect the semantic concepts contained in the query. Every sub-image detected is represented as a multiple features vector which is regarded as an instance. Each image is produced to a bag comprised of a flexible number of instances. And we apply a few number of MIL algorithms in this framework to perform the retrieval. Extensive experimental results illustrate the excellent performance in comparison with the existing methods of CBIR with MIL.

      • Impact of Caspase-8 (CASP8) -652 6N Del and D302H Polymorphisms on Prostate Cancer in Different Ethnic Groups

        Background: Despite evidence suggesting roles for caspase-8 (CASP8) -652 6N del and D302H polymorphisms in prostate cancer (PCa), the association of these polymorphisms with PCa risk remains inconclusive. Therefore, a meta-analysis was performed to more precisely estimate the association of CASP8 -652 6N del and D302H polymorphisms with PCa susceptibility. Materials and Methods: A comprehensive literature search was conducted to identify all case-control studies of CASP8 D302H and -652 6N del polymorphisms and PCa risk. Odds ratios (ORs) and 95% confidence intervals (CIs) were used to assess the strength of the association and the precision of the estimate, respectively. Results: Nine -625 6N del studies and 4 D302H studies were included. CASP8 -652 6N del and D302H polymorphisms were not significantly associated with PCa risk in the overall analyses. However, in the subgroup analysis stratified by ethnicity, -625 6N del was significantly associated with PCa risk in the East Asian and Indian populations under the recessive model. Furthermore, the subgroup analysis strongly suggested that D302H was associated with lower PCa risk in the Non-Indian population under the dominant model. Conclusions: In our meta-analysis, ethnic-specific differences were evident in the association of CASP8-625 6N del and D302H polymorphisms with PCa risk.
