        Does H3K27M Mutation Impact Survival Outcome of High-Grade Spinal Cord Astrocytoma?

        Objective: To evaluate the impact of H3K27M mutation in the prognosis of histological high-grade intramedullary astrocytoma. Methods: A total of 78 patients who were diagnosed with high-grade spinal cord astrocytoma were included. Clinical data consisting demographic, radiological, molecular features and treatment data were recorded. Univariate and multivariate Cox analysis were performed to investigate variables associated with survival outcome of histological high-grade spinal cord astrocytoma. Results: Median survival time was 21 months. Overall survival (OS) at 1 and 3 years was 65.7% and 40.7%, respectively. Sex, location, and tumor span did not present significant association with OS. Patients with H3K27M mutation showed significant shorter duration of symptom than patients with H3K27 wild-type. As respect to adjuvant treatment, adjuvant radiotherapy and chemotherapy were associated with favorable OS (both p = 0.01). Younger patients (age ≤ 18 years) had shorter OS (p = 0.008) than adult patients (age > 18 years). Of note, H3K27M mutation did not show significant impact on the survival outcome, regardless of histology grade 3 or grade 4 (p = 0.3). Conclusion: Histological high-grade spinal cord astrocytoma has dismal prognosis. Our study demonstrated that H3K27M mutation did not show significant impact on survival outcome of histological high-grade spinal cord astrocytoma.

      • Research of Acoustic Emission Testing Method with Application to Monitored for Wind Turbines

        Acoustic Emission method of nondestructive check is based on exertion wave radiation and their registration during fast local material structure reorganization. It is used as a means of analysis of materials, constructions, productions control and diagnosis during operating time. In the article, it is applied to structural health monitoring of Wind Turbines. Acoustic emission testing is based condition monitoring system uses data already collected at the wind turbine controller. It is an effective way to monitor wind turbines for early warning of failures and performance issues. We used a number of measurements to develop anomaly detection algorithms and investigated classification techniques using clustering algorithms and principal components analysis for capturing fault signatures. When registering signal amplitude it is required to consider its frequency distribution connecting each amplitude rate with the corresponding vibration rate. The correlation and signal to noise ratio were evaluated by the algorithm based on frequency spectrum. Classical method of frequency distortion influence exclusion consists of FRF calculation with subsequent adjustment of received signals spectral characteristics.

        A Joint Control Strategy for Automobile Active Grille Shutter and Cooling Fan

        As an emerging technology improving fuel consumption, active grille shutter (AGS) has been applied in engine cooling system. For the reason of unreasonable matching method of grille opening and cooling fan, the problem of engine overcooling is widespread. As components with similar function of adjusting cooling air flow, the AGS and the cooling fan are separated from each other in parameter matching and operation which reduce cooling efficiency and fuel economy. To achieve precise cooling and improve fuel economy, a joint control strategy for AGS and cooling fan was proposed by using computational fluid dynamics. In order to verify the feasibility of the strategy, the fuel economy tests were carried out at 0 km/h, 90 km/h and 120 km/h, respectively. The results showed that the fuel consumption under the three working conditions was reduced by 0.01 L/ 100 km, 0.07 L/ 100 km and 0.13 L/ 100 km, respectively, indicating that the proposed strategy can significantly reduce fuel consumption.

        Anticancer effects of metformin in experimental animal models of different types of cancer: a systematic review and meta-analysis

        To systematically evaluate the effects of metformin on tumors in experimental animal models of different types of cancer. Pubmed, Embase, Cochrane, and Web of Science databases were searched for studies on metformin used in various experimental animal tumor models from 2008 to 2022. Meta-analysis was performed using STATA 16.0 software after screening literature extraction data and methodological quality evaluation by inclusion and exclusion criteria. A total of 24 studies with 1108 model animals were included. Meta-analysis results showed that this study used meta-analysis for quantitative synthesis of study results and found that tumor model animals of different species showed different degrees of reduction in tumor volume, weight, and number after metformin intervention.

        Osteogenic and Angiogenic Potency of VEGF165-Transfected Canine Bone Marrow Mesenchymal Cells Combined with Coral Hydroxyapatite in Vitro

        BACKGROUND: To explore the osteogenic and angiogenic potential of human vascular endothelial growth factor 165 (hVEGF165) gene-transfected canine bone marrow mesenchymal stem cells (BMSCs) combined with coral hydroxyapatite (CHA) scaffold. METHODS: We constructed a lentiviral vector and transfected canine BMSCs with the best multiplicity of infection. Osteogenesis was induced in the transfected groups (GFP-BMSCs group and hVEGF-BMSCs group) and non-transfected group (BMSCs group), followed by the evaluation of alkaline phosphatase (ALP) activity and alizarin red S staining. Cells from the three groups were co-cultured with CHA granules, respectively to obtain the tissue-engineered bone. MTT assay and fluorescence microscopy were employed to assess cell proliferation and adhesion. The expression of osteogenic and angiogenic related genes and proteins were evaluated at 7, 14, 21, and 28 days post osteoinduction in cell culture alone and cell co-culture with CHA, respectively using RT-PCR and ELISA. RESULTS: The hVEGF165 gene was transfected into BMSCs successfully. Higher ALP activity and more calcified nodules were found in the hVEGF-BMSCs group than in the control groups (p < 0.001). Cells attached and proliferated in CHA particles. Both cells cultured alone and cells co-culture with CHA expressed more osteogenic and angiogenic related genes and proteins in the hVEGF-BMSCs group compared to the GFP-BMSCs and BMSCs groups (p < 0.05). CONCLUSION: High expression of hVEGF165 in BMSCs potentially promote the osteogenic potential of BMSCs, and synergically drive the expression of other osteogenic and angiogenic factors. hVEGF-BMSCs co-cultured with CHA expressed more osteogenic and angiogenic related factors, creating a favorable microenvironment for osteogenesis and angiogenesis. Also, the findings have allowed for the construction of a CHA-hVEGF-BMSCs tissue-engineered bone.         Eccentrically compressive behaviour of RC square short columns reinforced with a new composite method

        A new composite reinforced method, namely self-compacting concrete filled circular CFRP-steel jacketing, was proposed in this paper. Experimental tests on eight RC square short columns reinforced with the new composite reinforced method and four RC square short columns reinforced with CFS jackets were conducted to investigate their eccentrically compressive behaviour. Nine reinforced columns were subjected to eccentrically compressive loading, while three reinforced columns were subjected to axial compressive loading as reference. The parameters investigated herein were the eccentricity of the compressive loading and the layer of CFRP. Subsequently, the failure mode, ultimate load, deformation and strain of these reinforced columns were discussed. Their failure modes included the excessive bending deformation, serious buckling of steel jackets, crush of concrete and fracture of CFRP. Moreover, these reinforced columns exhibited a ductile failure globally. Both the eccentricity of the compressive loading and the layer of CFRP had a significant effect on the eccentrically compressive behaviour of reinforced columns. Finally, formulae for the evaluation of the ultimate load of reinforced columns were proposed. The theoretical formulae based on the ultimate equilibrium theory provided an effective, acceptable and safe method for designers to calculate the ultimate load of reinforced columns under eccentrically compressive loading.

        Analysis of Factors Impacting Atmospheric Pressure Plasma Polishing

        Atmospheric pressure plasma polishing (APPP) is a noncontact precision machining technology that uses low temperature plasma chemical reactions to perform atom-scale material removal. APPP is a complicated process, which is affected by many factors. Through a preliminary theoretical analysis and simulation, we confirmed that some of the key factors are the radio frequency (RF) power, the working distance, and the gas ratio. We studied the influence of the RF power and gas ratio on the removal rate using atomic emission spectroscopy, and determined the removal profiles in actual operation using a commercial form talysurf. The experimental results agreed closely with the theoretical simulations and confirmed the effect of the working distance. Finally, we determined the element compositions of the machined surfaces under different gas ratios using X-ray photoelectron spectroscopy to study the influence of the gas ratio in more detail. We achieved a surface roughness of Ra 0.6 nm on silicon wafers with a peak removal rate of approximately 32 $mm^{3}$/min.

      • Association of Six Susceptibility Loci with Prostate Cancer in Northern Chinese Men

        Background/Aim: Six prostate cancer (PCa) susceptibility loci were identified in a genome-wide association study (GWAS) in populations of European decent. However, the associations of these 6 single-nucleotide polymorphisms (SNPs) with PCa has remained tobe clarified in men in Northern China. This study aimed to explore the loci associated with PCa risk in a Northern Chinese population. Methods: Blood samples and clinical information of 289 PCa patients and 288 controls from Beijing and Tianjin were collected. All risk SNPs were genotyped using polymerase chain reaction (PCR)-high resolution melting curve technology and gene sequencing. Associations between PCa and clinical covariates (age at diagnosis, prostate-specific antigen [PSA], Gleason score, tumor stage, and level of aggressiveness) and frequencies of alleles and genotypes of these SNPs were analyzed using genetic statistics. Results: Among the candidate SNPs, 11p15 (rs7127900, A) was associated with PCa risk (P = 0.02, odds ratio [OR] = 1.64, 95% confidence interval [CI] = 1.09-2.46). Genotypes showed differences between cases and controls on 11p15 (rs7127900, A), 11q13 (rs7931342, T), and HNF1B (rs4430796, A) (P = 0.03, P = 0.01, and P = 0.04, respectively). The genotype TG on 11q13 (rs7931342, T) was positively associated with an increased Gleason score (P = 0.04, OR = 2.15, 95% CI = 1.02-4.55). Patients carrying TG on 17q24 (rs1859962, G) were negatively associated with an increased body mass index (BMI) (P = 0.03, OR = 0.44, 95% CI = 0.21-0.92) while those with AG on HNF1B (rs4430796, A) were more likely to have PSA increase (P = 0.002). Conclusion: Our study suggests that 11p15 (rs7127900, A) could be a susceptibility locus associated with PCa in Northern Chinese. Genotype TG on 11q13 (rs7931342, T) could be related to an increased Gleason score, AG on HNF1B (rs4430796, A) could be associated with PSA increase, and TG on 17q24 (rs1859962, G) could be negatively associated with an increased BMI in Chinese men with PCa.
