      • A Multi-dimensional Taylor Network (MTN)-Based Approach for Nonlinear Stochastic Systems Tracking Control

        The problem of tracking control for stochastic nonlinear systems is investigated in this paper. Because of the randomness and nonlinearity of stochastic nonlinear systems, the existing methods are sometimes difficult to achieve the desired tracking performance. In this paper, a new network controller (multi-dimensional Taylor network) is proposed, which only relies on the output of system. Firstly we give the structure of multi-dimensional Taylor network (MTN), and then prove the MTN has a good approximation performance. Secondly, Design a MTN control strategy relying on the system output, which will guarantee the tracking of system output to desired output. An example is given to illustrate the effectiveness of the proposed design approach.

        Adaptive Tracking Control of a Class of Nonlinear Systems with Input Delay and Dynamic Uncertainties Using Multi-dimensional Taylor Network

        This paper concerns on the control problem of a class of nonlinear systems with input delay and dynamic uncertainties using multi-dimensional Taylor Network (MTN) control method. Firstly, a new variable is introduced to eliminate the effect of input delay by combining Padé approximation with Laplace transformation. Secondly, a MTN-backstepping-based control strategy is constructively designed by introducing a new coordinate transformation, and the proposed controller has the advantages of simple structure and good real-time performance. Finally, the effectiveness of the proposed MTN-based control approach is demonstrated by three examples.

        A kind of NiTi-wire shape memory alloy damper to simultaneously damp tension, compression and torsion

        NiTi-wire shape memory alloy (SMA) dampers, that utilize NiTi SMA wires to simultaneously damp tension, compression and torsion, was developed for structural control implementation in this study. First, eight reduced-scale NiTi-wire SMA dampers were constructed. Then tension, compression and torsion experiments using the eight reduced-scale NiTi-wire SMA dampers of different specification were done. The experimental results revealed all of the eight reduced-scale NiTi-wire SMA dampers had the ability to simultaneously supply tension-compression damping and torsion damping. Finally, mechanics analysis of the NiTi-wire SMA dampers was done based on a model of the SMA-wire restoring force and on tension-compression and torsion damping analysis. The damping analytical results were found to be similar to the damping experimental results.

        Methylation Status of Transcriptional Modulatory Genes Associated with Colorectal Cancer in Northeast China

        Background/Aims: Methylation status plays a causal role in carcinogenesis in targeted tissues. However, the relationship between the DNA methylation status of multiple genes in blood leukocytes and colorectal cancer (CRC) susceptibility as well as interactions between dietary factors and CRC risks are unclear. Methods: We performed a case-control study with 466 CRC patients and 507 cancer-free controls to investigate the association among the methylation status of individual genes, multiple CpG site methylation (MCSM), multiple CpG site heterogeneous methylation and CRC susceptibility. Peripheral blood DNA methylation levels were detected by performing methylation-sensitive high-resolution melting. Results: Total heterogeneous methylation of CA10 and WT1 conferred a significantly higher risk of CRC (adjusted odds ratio [ORadjusted], 5.445; 95% confidence interval [CI], 3.075 to 9.643; ORadjusted, 1.831; 95% CI, 1.100 to 3.047; respectively). Subjects with high-level MCSM (MCSM-H) status demonstrated a higher risk of CRC (ORadjusted, 4.318; 95% CI, 1.529 to 12.197). Additionally, interactions between the high-level intake of fruit and CRH, WT1, and MCSM on CRC were statistically significant. Conclusions: The gene methylation status of blood leukocytes may be associated with CRC risk. MCSM-H of blood leukocytes was associated with CRC, especially in younger people. Some dietary factors may affect hypermethylation status and influence susceptibility to CRC.

      • Targeting SHCBP1 Inhibits Cell Proliferation in Human Hepatocellular Carcinoma Cells

        Src homology 2 domain containing (SHC) is a proto-oncogene which mediates cell proliferation and carcinogenesis in human carcinomas. Here, the SHC SH2-domain binding protein 1 (SHCBP1) was first established to be up-regulated in human hepatocellular carcinoma (HCC) tissues by array-base comparative genome hybridization (aCGH). Meanwhile, we examine and verify it by quantitative real-time PCR and western blot. Our current data show that SHCBP1 was up-regulated in HCC tissues. Overexpression of SHCBP1 could significantly promote HCC cell proliferation, survival and colony formation in HCC cell lines. Furthermore, knockdown of SHCBP1 induced cell cycle delay and suppressed cell proliferation. Furthermore, SHCBP1 could regulate the expression of activate extracellular signal-regulated kinase 1/2 (ERK1/2) and cyclin D1. Together, our findings indicate that SHCBP1 may contribute to human hepatocellular carcinoma by promoting cell proliferation and may serve as a molecular target of cancer therapy.

        Control Design for Stochastic Nonlinear Systems with Full-state Constraints and Input Delay: A New Adaptive Approximation Method

        In this paper, the full state constraints and input delay of stochastic nonlinear systems are studied. A new adaptive control algorithm is proposed using backstepping approach and multi-dimensional Taylor network (MTN) method. Firstly, the input delay problem is dealt with by introducing a new variable using the Padé approximation with Laplace transform. Secondly, MTNs are employed to approximate unknown nonlinear functions, and the barrier Lyapunov functions (BLFs) are constructed to deal with the state constraints. Based on this, a new approximation-based adaptive controller is proposed. Thirdly, it is proved that the proposed control method can ensure that all signals in the closed-loop system are semi-global ultimately uniformly bounded (SGUUB) in probability and the tracking error converges to a small neighborhood of the origin. Finally, two simulation examples are given to illustrate the effectiveness of the proposed design method.

        Isolation and enzymatic characterization of the first reported hyaluronidase from Yak (Bos grunniens) testis

        A novel hyaluronidase (BgHya1) from Yak testis was isolated and shown to have compara-tively high activity on sodium hyaluronate. However, surveys on BgHya1 are still limited. The enzyme was purifiedthrough gel filtration on Sephacryl S-100 and cation-exchange on SP Sepharose fast flow; the purity was confirmedby a reverse phase FPLC Shodex C4 column. The specific activity of the purified BgHya1 was 20.4 U/mg assayed bythe colorimetric method against 0.85 U/mg for the crude enzyme, representing a 24-fold purification. It was a monomericprotein of 55 kDa estimated by sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE) and SephacrylS-200. It exhibited maximum activity in the presence of 0.15 M NaCl at 37 oC, pH 3.8, and a specificity to sodiumhyaluronate higher than that of chondroitin-4-sulfate, chondroitin-6-sulfate, and dermatan. The Km value for BgHya1,using sodium hyaluronate as substrate, was 0.106 mg/mL. Activity of BgHya1 was inhibited mildly by Ca2+and Fe2+,and significantly by Fe3+, Mg2+, EDTA, urea, heparin, and 0.5 M NaCl. It was not affected by Cu2+,Zn2+,Co2+, ascorbicacid, PMSF, DTT, glutathione (reduced), or L-cysteine. BgHya1 was shown to be heat unstable in the range of 4-45 oC. In terms of storage stability, 92% of the activity was retained after four weeks at 4 oC, and 58% at room temperature. In addition, adding BSA (1.0 mg/mL) to the enzyme sample prior to freezing resulted in complete retention of enzymeactivity. This work yielded a high purity hyaluronidase, the first one isolated from by-product.

        Nutritional Characteristics and Active Components in Liver from Wagyu×Qinchuan Cattle

        We investigated nutritional characteristics and active components in the liver of Wagyu${\times}$Qinchuan cattle and Qinchuan cattle produced in Shaanxi (China). We observed significant differences (p<0.05) in the proximate composition of protein, fat, carbohydrate, total energy, and glycogen. Wagyu${\times}$Qinchuan cattle liver showed higher (p<0.05) sodium, iron, zinc, and selenium concentrations than Qinchuan cattle liver. The amino acid composition of Wagyu${\times}$Qinchuan cattle liver was richer (p<0.05) in 13 types of amino acids, with the exception of Asp (10.06%), Val (5.86%), and Met (1.72%). Total essential amino acids accounted for almost half the composition (39.69%) in Wagyu${\times}$Qinchuan cattle liver. Wagyu${\times}$Qinchuan cattle liver had lower (p<0.05) levels of monounsaturated fatty acids (18.2%), but higher (p<0.05) levels of polyunsaturated fatty acids (35.11%), compared with Qinchuan cattle liver (23.29% and 28.11%, respectively). The thrombogenic index was higher in Qinchuan cattle liver (0.86) than in Wagyu${\times}$Qinchuan cattle liver (0.70), and the glutathione (38.0 mg/100g) and L-carnitine (2.12 ${\mu}M/g$) content was higher (p<0.05) in Wagyu${\times}$Qinchuan cattle liver than in Qinchuan cattle liver (29.8 mg/100g and 1.41 ${\mu}M/g$, respectively). According to the results obtained, the liver of Wagyu${\times}$Qinchuan cattle, which is insufficiently used, should be increasingly utilized to improve its commercial value.

        Mesenchymal-to-epithelial transition of osteoblasts induced by Fam20c knockout

        Background: Fam20c is intimately related to tissue development and diseases. At present, it has been reported that Fam20c regulates the mineralization of osteoblasts, but there are few reports on other effects. Objective: To study the effect of Fam20c on osteoblasts by knocking out the Fam20c gene. Methods: Fam20c knockout osteoblasts were constructed by transfecting mouse osteoblasts with lentivirus. The proliferation, migration and mineralization of Fam20c knockout cells were detected by CCK-8, scratch test and alizarin red staining assays. The subcellular structure was observed by transmission electron microscopy. RT-PCR was used to detect the differential expression of mesenchymal-to-epithelial transition (MET)-related marker genes and core transcription factors. The differential expression of MET-related proteins was detected by immunofluorescence or Western blot. Transcriptome analysis of Fam20c knockout osteoblasts was performed, and real-time PCR was used to verify transcriptome analysis related to MET. Results: The proliferation ability of osteoblasts was not significantly changed after Fam20c deletion, but the migration ability and mineralization ability were significantly weakened. There were tight junctions between Fam20c knockout cells. The expression of mesenchymal cell marker genes and core transcription factors was significantly decreased, and the expression of epithelial cell marker genes was significantly increased. The expression of mesenchymal cell marker proteins was significantly decreased, and the expression of epithelial cell marker proteins was significantly increased. Multiple signalling molecules and pathways involved in MET have changed. Conclusions: Knockdown of Fam20c resulted in MET. Fam20c affects the transcription of key factors in osteoblast MET.
