The Up-Regulation of miR-199b-5p in Erythroid Differentiation Is Associated with GATA-1 and NF-E2

        Li, Yuxia,Bai, Hua,Zhang, Zhongzu,li, Weihua,Dong, Lei,Wei, Xueju,Ma, Yanni,Zhang, Junwu,Yu, Jia,Sun, Guotao,Wang, Fang Korean Society for Molecular and Cellular Biology 2014 Molecules and cells Vol.37 No.3

MicroRNAs (miRNAs) represent a class of small non-coding regulatory RNAs that play important roles in normal hematopoiesis, including erythropoiesis. Although studies have identified several miRNAs that regulate erythroid commitment and differentiation, we do not understand the mechanism by which the crucial erythroid transcription factors, GATA-1and NF-E2 directly regulate and control differentiation via miRNA pathways. In this study, we identified miR-199b-5p as a key regulator of human erythropoiesis, and its expression was up-regulated during the erythroid differentiation of K562 cells. Furthermore, the increase of miR-199b-5p in erythroid cells occurred in a GATA-1- and NF-E2-dependent manner during erythrocyte maturation. Both GATA-1 and NF-E2 bound upstream of the miR-199b gene locus and activated its transcription. Forced expression of miRNA-199b-5p in K562 cells affected erythroid cell proliferation and maturation. Moreover, we identified c-Kit as a direct target of miR-199b-5p in erythroid cells. Taken together, our results establish a functional link among the erythroid transcription factors GATA-1/NF-E2, miR-199b-5p and c-Kit, and provide new insights into the coupling of transcription and post-transcription regulation in erythroid differentiation.

Effect of polymer and additive on the structure and property of porous stainless steel hollow fiber

        Xiao-Hua Ma,Zhen-Liang Xu,Yu Bai,Yue Cao 한국화학공학회 2014 Korean Journal of Chemical Engineering Vol.31 No.8

Porous stainless steel hollow fiber has been widely used due to its high mechanical strength, excellentthermal conductivity and good sealing properties compared with other porous supports. We successfully prepared porousstainless steel hollow fibers using polyacrylonitrile (PAN) as polymer via dry-wet spinning followed by sintering throughtemperature programming method. The PAN concentration had an obvious impact on the structure and property ofporous stainless steel hollow fiber even if it would be burned off during sintering. The results showed that the morphologycould be tuned by adjusting the concentration of PAN. With increasing PAN concentration in casting solutionfor spinning, the viscosity was increased dramatically, resulting in much compact structures with high pure water flux(higher than 3×105 L·m−2·h−1·Pa−1). A more dense structure could be obtained by adding additive polyvinylpyrrolidone(PVP) as viscosity enhancer.

Autophagy induced by H. pylori VacA regulated the survival mechanism of the SGC7901 human gastric cancer cell line

        Luo Juan,Bai Luyan,Tao Jun,Wen Yu,Li Mingke,Zhu Yunzhen,Luo Sufeng,Pu Guangyu,Ma Lanqing 한국유전학회 2021 Genes & Genomics Vol.43 No.10

Background Vacuolating cytotoxin (VacA) is an important virulence factor of Helicobacter pylori (H. pylori). It was previously believed that VacA can trigger the cascade of apoptosis on mitochondria to lead to cell apoptosis. Recently, it was found that VacA can induce autophagy. However, the molecular mechanism by which VacA induces autophagy is largely unknown. Objective We aimed to explore the molecular mechanism of autophagy induced by H. pylori in gastric cancer cells and the efect of autophagy on the survival of gastric cancer cells. Methods The autophagy of human gastric cancer cell line SGC7901 was detected by Western blot and RT-PCR in the treatment of VacA protein of H. pylori. The relationship between autophagy and reactive oxygen species (ROS) in the proliferation of gastric cancer cells were studied by gene expression silences (siRNA) and CM-H2DCFDA (DCF) staining. Results The results showed that VacA protein secreted by H. pylori in the supernatant stimulated autophagy in SGC7901 cells. After VacA protein treatment, the mRNA expressions of BECN1, ATG7 and PIK3C3, were up-regulated. ATG7 silencing by siRNA inhibited VacA-induced autophagy. Furthermore, our data demonstrated that VacA protein increased ROS levels. Addition of the antioxidant N-acetyl-l-cysteine (NAC) suppressed the levels of ROS, leading to inhibition of autophagy. Conclusions H. pylori VacA is a key toxin that induces autophagy by increased ROS levels. And our fndings demonstrated that VacA signifcantly inhibited proliferation in SGC7901 cells.

      • Transmembrane Protein 166 Expression in Esophageal Squamous Cell Carcinoma in Xinjiang, China

        Sun, Wei,Ma, Xiu-Min,Bai, Jing-Ping,Zhang, Guo-Qing,Zhu, Yue-Jie,Ma, Hai-Mei,Guo, Hui,Chen, Ying-Yu,Ding, Jian-Bing Asian Pacific Journal of Cancer Prevention 2012 Asian Pacific journal of cancer prevention Vol.13 No.8

Objective: Transmembrane protein 166 (TMEM166) expression in esophageal squamous cell carcinoma (ESCC) and remote normal esophageal tissues was examined to assess any role in tumour biology. Methods: TMEM166 mRNA expression in 36 cases with ESCC (36 tumour samples, 36 remote normal esophageal tissue samples) was detected by RT-PCR. TMEM166 protein expression was analysed in paraffin-embedded tissue samples from the same cases by immunohistochemistry. Results: Semi-quantitative analysis showed TMEM166 mRNA expression in ESCCs to be significantly lower than in remote normal esophageal tissues ($0.759{\pm}0.713$ vs. $2.622{\pm}1.690$, P=0.014). TMEM166 protein expression was also significantly reduced (69.4% vs. 94.4%, P<0.01). Conclusion: TMEM166 mRNA and protein expression demonstrated significant reduction in ESCCs compared with remote esophageal tissues, albeit with no correlation with tumour size, differentiation, stage, and lymph node metastasis, suggesting a role in regulating autophagic and apoptotic processes in the ESCC.

Synthesis and Tribological Performance Study of Ultrahigh Molecular Weight Polyethylene Composites Reinforced with Graphene Oxide

        Xinjun Hu,Xiaojie Yan,Yu Ma,Yongxiao Bai 한국고분자학회 2017 폴리머 Vol.41 No.1

In order to improve the hardness and tribological properties of ultrahigh molecular weight polyethylene (UHMWPE), UHMWPE composites reinforced with graphene oxide (GO) nanosheets by ball milling is presented in this article. The results show that the content of 20 wt% GO composite exhibits the lowest friction coefficient and the best wear resistance among all specimens under distilled water lubrication conditions. The mechanism of GO content on tribological properties of the composites under dry conditions and distilled water were investigated. Super low friction coefficient value and the best wear resistance under distilled water is attributed to water provides passivation of interacting dangling bonds on the sliding surface of GO. This study provides a new path to synthesize the high wear resistance and mechanical performance GO/UHMWPE composite materials under water condition and may have huge potential applications in many related fields, such as biomedical and marine engineering.

Xylazole inhibits NO-cGMP pathway in fetal rat nerve cells

        Xinyu Wang,Yue Wu,Lin Liu,Hui Bai,Zhiheng Zhang,Mingchao Zhao,Tianwen Ma,Xiaopeng Song,Lina Jia,Liangyu Lv,Yue Yu,Xinyu Xu,Hong Chen,Li Gao 대한수의학회 2022 Journal of Veterinary Science Vol.23 No.1

Background: Xylazole (Xyl) is a veterinary anesthetic that is structurally and functionally similar to xylazine. However, the effects of Xyl in vitro remain unknown. Objectives: This study aimed to investigate the anesthetic mechanism of Xyl using fetal rat nerve cells treated with Xyl. Methods: Fetal rat nerve cells cultured for seven days were treated with 10, 20, 30, and 40 μg/ mL Xyl for 0, 5, 10, 15, 20, 25, 30, 45, 60, 90, and 120 min. Variations of amino acid neurotransmitters (AANTs), Nitric oxide-Cyclic GMP (NO-cGMP) signaling pathway, and ATPase were evaluated. Results: Xyl decreased the levels of cGMP and NO in nerve cells. Furthermore, Xyl affected the AANT content and Na+-K+-ATPase and Ca2+-Mg2+-ATPase activity in nerve cells. These findings suggested that Xyl inhibited the NO-cGMP signaling pathway in nerve cells in vitro. Conclusions: This study provided new evidence that the anesthetic and analgesic effects of Xyl are related to the inhibition of the NO-cGMP signaling pathway.

      • Anti-tumor Effects and Apoptosis Induction by Realgar Bioleaching Solution in Sarcoma-180 Cells in Vitro and Transplanted Tumors in Mice in Vivo

        Xie, Qin-Jian,Cao, Xin-Li,Bai, Lu,Wu, Zheng-Rong,Ma, Ying-Ping,Li, Hong-Yu Asian Pacific Journal of Cancer Prevention 2014 Asian Pacific journal of cancer prevention Vol.15 No.6

Background: Realgar which contains arsenic components has been used in traditional Chinese medicine (TCM) as an anticancer drug. However, neither Realgar nor its formula are soluble in water. As a result, high dose of Realgar has to be administered to achieve an effective blood medicine concentration, and this is associated with adverse side effects. The objective of the present study was to increase the solubility of a formula using hydrometallurgy technology as well as investigating its effects on in vitro and in vivo cell proliferation and apoptosis in Sarcoma-180 cell line. Materials and Methods: Antiproliferative activity of Realgar Bioleaching Solution (RBS) was evaluated by MTT assay. Further, effects of RBS on cell proliferation and apoptosis were studied using flow cytometry and transmission electron microscopy. Kunming mice were administered RBS in vivo, where arsenic specifically targeted solid tumors. Results: The results indicated that RBS extract potently inhibited the tumor growth of Sarcoma-180 cell line in a dose-dependent manner. Flow cytometry and transmission electron microscopy further indicated that RBS significantly induced cell apoptosis through the inhibition of cell cycle pathway in a dose-dependent manner. Further, on RBS administration to mice, arsenic was specifically targeted to solid tumor.s Conclusions: RBS could substitute for traditional Realgar or its formula to work as a potent tool in cancer treatment.

Effect of Different Cooling Rates in High Rheological Rate Forming Process on Mechanical Properties of Zr57Cu20Al10Ni8Ag5 Bulk Metallic Glass

        Xiao‑Bo Mei,Bao‑Chuan Liu,Wei Jiang,Quan Xu,Qi‑Dong Zhang,Yu‑Bai Ma,Fang‑Qiu Zu 대한금속·재료학회 2020 METALS AND MATERIALS International Vol.26 No.9

The influence of cooling rates on the mechanical properties of a Zr-based bulk metallic glass prepared with high rheologicalrate forming (HRRF) was investigated and compared with traditional suction cast methods. Amorphous samples of Zr57Cu-20Ni8Al10Ag5 were prepared in copper molds with different sizes in order to obtain different cooling rates for both HRRFand traditional cast methods. These specimens were subjected to compression experiments, including microhardness testing,X-ray diffraction testing and differential scanning calorimetry analysis. The results indicate that the plasticity of the samplesformed by HRRF are higher than that of the as-cast ones at the same cooling rates, while the microhardness manifests theopposite principle. As the cooling rate increases further, the difference in plasticity further increases between two methods, indicating that the plasticity of metallic glasses is more sensitive to cooling rates during the HRRF process. At the core ofthis phenomenon is the fact that HRRF methods can introduce more free volume into glasses than traditional cast methodswith an elevated cooling rate are able to.

Factors that Influence the Presciption of Antipsychotics for Patients with Schizophrenia in China

        Tian-Mei Si,Liang Shu,Ke-Qing Li,Xie-He Liu,Qi-Yi Mei,Gao-Hua Wang,Pei-Shen Bai,Li-Ping Ji,Xian-Sheng Chen,Cui Ma,Jian-Guo Shi,Hong-Yan Zhang,Hong Ma,Xin Yu 대한정신약물학회 2011 CLINICAL PSYCHOPHARMACOLOGY AND NEUROSCIENCE Vol.9 No.3

Objective: To investigate the patterns of antipsychotic use in China and to analyze the factors that influence antipsychotic prescriptions. Methods: A standardized survey was conducted from May 20 to 24 2002 in five different regions of China with varying economic levels. The patterns of antipsychotic medication use were analyzed in a sample of 4,779 patients with schizophrenia. The survey gathered information on demographic characteristics, clinical profiles, and antipsychotic medications prescribed. Multiple logistic regression was used to analyze factors related to patterns of antipsychotic medication use. Results: A plurality of patients with schizophrenia was treated with clozapine (39%); this was followed by risperidone, sulpride, chlorpromazine, perphenazine, and haloperidol. More than 56.3% of patients were treated with only one atypical antipsychotic. The mean daily dose of chlorpromazine was 365±253 mg (mean±standard deviation), and 6.5% of patients were treated with depot injections of typical antipsychotic medications. A total of 73.7% (n=3,523) of patients with schizophrenia received monotherapy,24.8% (n=1,183) received two antipsychotics, 1.1% (n=52) received three antipsychotics, and one received four different antipsychotics. Patients often simultaneously received other classes of medications including anticholinergic agents, benzodiazepines,β-blockers, antidepressants, and mood stabilizers. Economic status and clinical symptoms were the main factors that contributed to the patterns of antipsychotic prescription. Conclusion: The present study suggests that atypical antipsychotic medications, especially clozapine, are the primary psychiatric treatments of choice in the management of schizophrenia in China. Moreover, the economic status and clinical profile of the patient are the major factors affecting the prescription of antipsychotic medication.
