        The Up-Regulation of miR-199b-5p in Erythroid Differentiation Is Associated with GATA-1 and NF-E2

        MicroRNAs (miRNAs) represent a class of small non-co-ding regulatory RNAs that play important roles in normal hematopoiesis, including erythropoiesis. Although studies have identified several miRNAs that regulate erythroid commitment and differentiation, we do not understand the mechanism by which the crucial erythroid transcription factors, GATA-1and NF-E2 directly regulate and control differentiation via miRNA pathways. In this study, we identified miR-199b-5p as a key regulator of human erythropoiesis, and its expression was up-regulated during the erythroid differentiation of K562 cells. Furthermore, the increase of miR-199b-5p in erythroid cells occurred in a GATA-1- and NF-E2-dependent manner during erythrocyte maturation. Both GATA-1 and NF-E2 bound upstream of the miR-199b gene locus and activated its transcription. Forced expression of miRNA-199b-5p in K562 cells affected erythroid cell proliferation and maturation. Moreover, we identified c-Kit as a direct target of miR-199b-5p in erythroid cells. Taken together, our results establish a functional link among the erythroid transcription factors GATA-1/NF-E2, miR-199b-5p and c-Kit, and provide new insights into the coupling of transcription and post-transcription regulation in erythroid differentiation.

        Adsorption of different molecular weight lignosulfonates on dimethomorph powder in an aqueous system

        Different molecular weight lignosulfonates were acquired by ultrafiltration and their adsorption behavior on dimethomorph powder were investigated. The results showed that as the molecular weight of lignosulfonates increased the adsorption amounts increased and the adsorption processes were driven by van der Waals forces and hydrophobic effects. The adsorption isotherms were fitted well by Langmuir model. Relations of adsorption amounts and zeta potentials with particle size of dimethomorph particles were established to disclose the dispersing mechanism of dimethomorph suspensions. The average particle sizes were determined by zeta potentials rather than adsorption amounts, which confirmed the dispersing mechanisms were electrostatic repulsive forces.

      • Optimization of Physical Education Teaching Mode in Colleges based on Data Mining Clustering Algorithm

        Deep neural networks (DNNs) and their learning algorithms have been widely used in large data analysis. With the practice of teaching network reform in higher education, many colleges began to use the network to change the traditional teaching methods, and made several achievements. In this paper, the authors first analyze the infinite depth neural network, and construct evaluation index system by using network data mining method. The result shows that 58.09% students think the evaluation from college physical education is formative assessment, 27.94% students think that the evaluation reflects the magnitude progress, and 30.15% students think that it reflects the students' self-evaluation. Physical teaching evaluation is an important part of physical education; however, the survey found that physical education students are lack of understanding of teaching evaluation. Therefore, colleges should make comprehensive evaluation of physical education, and formulate scientific and rational teaching evaluation concept.

        Identification of genes involved in inbreeding depression of reproduction in Langshan chickens

        Objective: Inbreeding depression of reproduction is a major concern in the conservation of native chicken genetic resources. Here, based on the successful development of strongly inbred (Sinb) and weakly inbred (Winb) Langshan chickens, we aimed to evaluate inbreeding effects on reproductive traits and identify candidate genes involved in inbreeding depression of reproduction in Langshan chickens. Methods: A two-sample t-test was performed to estimate the differences in phenotypic values of reproductive traits between Sinb and Winb chicken groups. Three healthy chickens with reproductive trait values around the group mean values were selected from each of the groups. Differences in ovarian and hypothalamus transcriptomes between the two groups of chickens were analyzed by RNA sequencing (RNA-Seq). Results: The Sinb chicken group showed an obvious inbreeding depression in reproduction, especially for traits of age at the first egg and egg number at 300 days (p<0.01). Furthermore, 68 and 618 differentially expressed genes (DEGs) were obtained in the hypothalamus and ovary between the two chicken groups, respectively. In the hypothalamus, DEGs were mainly enriched in the pathways related to vitamin metabolism, signal transduction and development of the reproductive system, such as the riboflavin metabolism, Wnt signaling pathway, extracellular matrix-receptor interaction and focal adhesion pathways, including stimulated by retinoic acid 6, serpin family F member 1, secreted frizzled related protein 2, Wnt family member 6, and frizzled class receptor 4 genes. In the ovary, DEGs were significantly enriched in pathways associated with basic metabolism, including amino acid metabolism, oxidative phosphorylation, and glycosaminoglycan degradation. A series of key DEGs involved in folate biosynthesis (gamma-glutamyl hydrolase, guanosine triphosphate cyclohydrolase 1), oocyte meiosis and ovarian function (cytoplasmic polyadenylation element binding protein 1, structural maintenance of chromosomes 1B, and speedy/RINGO cell cycle regulator family member A), spermatogenesis and male fertility (prostaglandin D2 synthase 21 kDa), Mov10 RISC complex RNA helicase like 1, and deuterosome assembly protein 1) were identified, and these may play important roles in inbreeding depression in reproduction. Conclusion: The results improve our understanding of the regulatory mechanisms underlying inbreeding depression in chicken reproduction and provide a theoretical basis for the conservation of species resources.         Carbon Source Affects Synthesis, Structures, and Activities of Mycelial Polysaccharides from Medicinal Fungus Inonotus obliquus

        The effects of various carbon sources on mycelial growth and polysaccharide synthesis of the medicinal fungus Inonotus obliquus in liquid fermentation were investigated. After 12-d fermentation, mycelial biomass, polysaccharide yield, and polysaccharide content were significantly higher in Glc+Lac group (glucose and lactose used as combined carbon source) than in other groups. Crude polysaccharides (CIOPs) and the derivative neutral polysaccharides (NIOPs) were obtained from mycelia fermented using Glc, fructose (Fru), Lac, or Glc+Lac as carbon source. Molecular weights of four NIOPs (termed as NIOPG, NIOPF, NIOPL, and NIOPGL) were respectively 780.90, 1105.00, 25.32, and 10.28 kDa. Monosaccharide composition analyses revealed that NIOPs were composed of Glc, Man, and Gal at different molar ratios. The NIOPs were classified as α-type heteropolysaccharides with 1→2, 1→3, 1→4, 1→6 linkages in differing proportions. In in vitro cell proliferation assays, viability of RAW264.7 macrophages was more strongly enhanced by NIOPL or NIOPGL than by NIOPG or NIOPF, and proliferation of HeLa or S180 tumor cells was more strongly inhibited by NIOPG or NIOPGL than by NIOPF or NIOPL, indicating that immune-enhancing and antitumor activities of NIOPs were substantially affected by carbon source. qRT-PCR analysis revealed that expression levels of phosphoglucose isomerase (PGI) and UDP-Glc 4-epimerase (UGE), two key genes involved in polysaccharide synthesis, varied depending on carbon source. Our findings, taken together, clearly demonstrate that carbon source plays an essential role in determining structure and activities of I. obliquus polysaccharides by regulating expression of key genes in polysaccharide biosynthetic pathway.

        Green and mild production of 5-aminolevulinic acid from algal biomass

        Algal biomass was converted into 5-aminolevulinic acid (5-ALA) in five chemical steps: conversion to 5- (chloromethyl)furfural (5-CMF), ammoniation, ring-opening (photo-oxidation), reduction, and hydrolyzation. Among them, we mainly focused on the 5-CMF production and the following ammoniation. To our knowledge, the mixed solvent catalytic system of deep eutectic solvent (DES) and low concentration hydrochloric acid is the first reported for the synthesis of 5-CMF from algal biomass, providing a 24.6% 5-CMF yield at 120 oC for 5 h. Potassium phthalimide (KPI) was employed as an ammoniation reagent with superb selectivity and activity instead of conventional sodium azide (NaN3). Optimizing the experimental design, a 23.7% 5-ALA yield along with high purity (>96%) was achieved from 5-CMF, and the total 5-ALA yield was 5.8% from algal biomass. This work provides a green and mild pathway for 5- ALA production from algal biomass.

        Nonlinear Adaptive Backstepping with ESO for the Quadrotor Trajectory Tracking Control in the Multiple Disturbances

        In this paper, we present a nonlinear adaptive backstepping with extended state observer (ESO) trajectory tracking controller for a quadrotor unmanned aerial vehicle (UAV) subject to the multiple disturbances, which include the parametric uncertainties, actuator faults and external disturbance. First, a six-degrees of freedom quadrotor UAV model with the multiple disturbances function is built. Second, the adaptive backstepping controller is designed to track the desired trajectory command aim at internal disturbance. And the adaptive backstepping controller with ESO is designed to track the desired trajectory command aim at external disturbance. Third, the stability of the system is proved by the circle criterion. Finally, under different flight scenarios, simulation results are given to demonstrate the effectiveness of the proposed method.

        Insect cytokine growth-blocking peptide may regulate density-dependent phase trait of cuticular melanization in the larval armyworm, Mythimna separata

        Growth-blocking peptide (GBP) is a 25 amino acid insect cytokine found in lepidopteran insects that has diverse biological activities such as larval growth regulation, paralysis induction, cell proliferation and stimulation of immune cells. Density-dependent phase polyphenism is a phenomenon of phenotypic plasticity in which the expression of a variety of traits can be affected by local population density. In the present study, the armyworm Mythimna separata larvae with four rearing densities (1 larva/vial, 2 larvae/vial, 4 larvae/vial and 6 larvae/vial) were tested for cuticular melanization and body weight throughout the third-fifth instar, and the functional role of GBP in regulating the changes was investigated. The results indicated that when reared at high densities, the larvae exhibited less body weight and more degree of cuticular melanization than larvae reared at low densities. The gene expression of GBP in armyworm larvae showed an initial rise and then decline trend with increased rearing densities in the third to fifth instar. Compared with control, more degree of cuticular melanization was observed in GBP-injected larvae (500 ng/larva in volume 50 μL) than that in Ringer’s solution-injected counterparts. Furthermore, the gene expression level of dopa decarboxylase and prophenoloxidase increased significantly in GBP-injected fifth instar larvae from 6 h to 12 h after injection, suggesting the role of GBP in modulating density-dependent phase trait of armyworm cuticular melanization.

        A New Closed-loop Control Allocation Method with Application to Direct Force Control

        A new closed-loop control allocation method is proposed to distribute the force and moment increment commands among multiple actuators at the same time. An innovative feedback configuration for the force and moment increment produced by the actuators deflections is added into the trajectory and attitude control loop, respectively. In addition, the trajectory and attitude controllers designed by nonlinear dynamic inversion are used to derive the desired force and moment increment commands. Then, the stability of the closed-loop system is guaranteed theoretically when the actuators are healthy and fail, where there is no a fault detection method to identify the failure. Finally, the proposed method is used to achieve direct force control for decoupling the attitude and flight trajectory in the longitudinal flight. The nonlinear simulation using Canard Rotor/Wing (CRW) aircraft longitudinal model in fixed-wing mode demonstrates that this method can achieve two direct force control modes: pitch pointing and direct lift, whether the actuators are healthy or fail.
