        Abnormal differentiation of regulatory T cells and Th17 cells induced by perinatal bisphenol A exposure in female offspring mice

        Background Bisphenol A (BPA) is an environmental estrogen widely exposed to human beings, and there are more studies on its reproductive toxicity, endocrine disruption and neurobehavioral disorders. Recent few studies have found that BPA has immunotoxicity, and its mechanism is not clear. Therefore, the effects of BPA on immune system have attracted extensive attention. The aim of this study was to investigate the effect and mechanism of perinatal exposure to BPA on regulatory T cells (Treg) and Th17 cells in female offspring mice. Methods Twenty-one pregnant C57BL/6 mice were randomly divided into three groups: a control group, low-dose BPA (0.2 μg/mL) and high-dose BPA (2.0 μg/mL) exposure group. All received BPA exposure via drinking water from gestational day 6 to the end of lactation. Female offspring were fed a normal diet and drinking water for 1 month. The percentages of Treg and Th17 cells, the levels of Foxp3 and RORγt protein and IL-17 and TGF-β from spleen tissue or blood were measured in female offspring. Results The percentage of Treg cells and levels of Foxp3 protein decreased, while the percentage of Th17 cells and levels of RORγt protein increased, which showed a dose–effect relationship. The levels of serum TGF-β were significantly lower and the levels of serum IL-17 were statistically higher in BPA-exposed female offspring compared with controls (P < 0.05 or P < 0.01). But there were no statistical difference in the levels of serum TGF-β and IL-17 between 0.2 μg/mL and 2.0 μg/ mL BPA groups (P > 0.05). Conclusion BPA exposure during pregnancy and lactation could cause abnormal differentiation and function of Treg and Th17 cells in female offspring mice, which was associated with down-regulated Foxp3 and up-regulated RORγt protein, respectively. Our findings indicated that BPA exposure during early development may play an important role in the development of autoimmune diseases later.

        Application of Intraoperative Ultrasonography for Guiding Microneurosurgical Resection of Small Subcortical Lesions

        Objective: We wanted to evaluate the clinical value of intraoperative ultrasonography for real-time guidance when performing microneurosurgical resection of small subcortical lesions. Materials and Methods: Fifty-two patients with small subcortical lesions were involved in this study. The pathological diagnoses were cavernous hemangioma in 25 cases, cerebral glioma in eight cases, abscess in eight cases, small inflammatory lesion in five cases, brain parasite infection in four cases and the presence of an intracranial foreign body in two cases. An ultrasonic probe was sterilized and lightly placed on the surface of the brain during the operation. The location, extent, characteristics and adjacent tissue of the lesion were observed by high frequency ultrasonography during the operation. Results: All the lesions were located in the cortex and their mean size was 1.3 ± 0.2 cm. Intraoperative ultrasonography accurately located all the small subcortical lesions, and so the neurosurgeon could provide appropriate treatment. Different lesion pathologies presented with different ultrasonic appearances. Cavernous hemangioma exhibited irregular shapes with distinct margins and it was mildly hyperechoic or hyperechoic. The majority of the cerebral gliomas displayed irregular shapes with indistinct margins, and they often showed cystic and solid mixed echoes. Postoperative imaging identified that the lesions had completely disappeared, and the original symptoms of all the patients were significantly alleviated. Conclusion: Intraoperative ultrasonography can help accurately locate small subcortical lesions and it is helpful for selecting the proper approach and guiding thorough resection of these lesions.         Optimum control system for earthquake-excited building structures with minimal number of actuators and sensors

        For vibration control of civil structures, especially large civil structures, one of the important issues is how to place a minimal number of actuators and sensors at their respective optimal locations to achieve the predetermined control performance. In this paper, a methodology is presented for the determination of the minimal number and optimal location of actuators and sensors for vibration control of building structures under earthquake excitation. In the proposed methodology, the number and location of the actuators are first determined in terms of the sequence of performance index increments and the predetermined control performance. A multi-scale response reconstruction method is then extended to the controlled building structure for the determination of the minimal number and optimal placement of sensors with the objective that the reconstructed structural responses can be used as feedbacks for the vibration control while the predetermined control performance can be maintained. The feasibility and accuracy of the proposed methodology are finally investigated numerically through a 20-story shear building structure under the El-Centro ground excitation and the Kobe ground excitation. The numerical results show that with the limited number of sensors and actuators at their optimal locations, the predetermined control performance of the building structure can be achieved.

        Flexural Properties and Electrical Conductivity of Epoxy Resin/Carbon Fiber Cloth/Metallic Powder Composites

        In this study, epoxy-based composites were prepared using diglycidyl ether of bisphenol A (DGEBA) as the matrix, carbon fiber cloth (CFC) as the reinforcing agent, and metallic powder as the conductive filler. The mechanical and electrical properties of DGEBA/CFC and DGEBA/CFC/metallic powder composites were investigated using several techniques. The morphology of the fracture surface was investigated using field-emission scanning electron microscopy. The flexural strength of the DGEBA/CFC composites was found to significantly increase with the addition of CFC. The electrical conductivity of the DGEBA/CFC composites significantly increased with the increasing amount of CFC and further increased after the addition of metallic powders. The electrical conductivity of the DGEBA/CFC composites significantly improved with the addition of a small amount of graphene.

      • A novel trifluoromethyl benzopyran induces G1 cell cycle arrest and apoptosis in HeLa human cervical carcinoma cells

        <P>In the present study, a biologically active 4-(trifluoromethyl)phenyl piperazin moiety was linked to a 2,2- dimethyl -2H-benzopyran template to generate (3R,4S)-2,2-dimethyl-6-nitro-4-(4-(3-(trifluoromethyl)phenyl)piperazin-1-yl) chroman -3-ol (C110g), and the cellular and molecular mechanisms by which C110g exerts cytotoxic effects on the HeLa human cervical cancer cell line were further investigated. C110g suppressed the viability of HeLa cells in both concentration- and time-dependent manner (IC50 of 17 ?M) by inducing DNA damage and G1 cell cycle arrest. Characteristic changes in nuclear morphology and Annexin V/PI staining pointed to apoptosis as the mode of cell death. The levels of p53 and p21 were increased in the C110g-treated cells, with a corresponding increase in Bax/Bcl-2 protein ratio. Subsequently, C110g induced the cytoplasmic release of cytochrome c from the mitochondria accompanied by a decreased mitochondrial membrane potential and activation of caspase-3 and -9. These results confirmed that the C110g transduced the apoptotic signal via the mitochondrial pathway. Caspase-8, typically associated with the initiation of the death receptor pathway, was activated, suggesting the extrinsic pathway might also be involved. However, C110g did not result in reactive oxygen species (ROS) generation. Taken together, these findings indicate that the DNA damage-dependent p53-regulated mitochondrial pathway as well as the extrinsic pathway play a crucial role in C110g-induced apoptosis, which provide a better understanding of the molecular mechanisms of trifluoromethyl benzopyrans in cervical cancer.</P>

        Effects of graded levels of cupric citrate on growth performance, antioxidant status, serum lipid metabolites and immunity, and tissue residues of trace elements in weaned pigs

        Objective: The goal of this study was to investigate the effects of cupric citrate (CuCit) on growth performance, antioxidant indices, serum lipid metabolites, serum immune indices, and tissue residues of copper (Cu), zinc, and iron in weaned pigs. Methods: A total of 180 weaned pigs (Duroc×Landrace×Large White) with an average body weight of 8.98±1.21 kg were randomly assigned to a corn-soybean meal control ration, or 4 similar rations with 30, 60, 120, or 240 mg/kg Cu as CuCit. All diets contained 10 mg/kg Cu as cupric sulfate from the vitamin-mineral premix. The experiment was divided into two phases: 0 to 14 d (phase 1) and 15 to 28 d (phase 2). Results: Average daily gain (ADG; linearly, p<0.01) and average daily feed intake (ADFI; linearly and quadratically, p<0.05) were affected by an increase in CuCit during phase 2. Overall period, ADG (p<0.05) and ADFI (p<0.01) were linearly increased with increasing dietary levels of CuCit. Serum malondialdehyde concentrations (p<0.05) and glutathione peroxidase activity (p<0.01) linearly decreased and increased respectively with an increase in CuCit. Serum levels of Cu-Zn superoxide dismutase were linearly affected with an increase in CuCit (p<0.01). Hepatic malondialdehyde levels decreased with an increase in CuCit (linearly and quadratically, p<0.01). Serum total cholesterol concentrations were quadratically affected (p<0.05) and decreased in pigs fed Cu as CuCit at 60 and 120 mg/kg and increased in pigs fed 240 mg/kg Cu as CuCit. Serum high-density lipoprotein concentrations were linearly affected with an increase in CuCit (p<0.01). Serum IL-1β levels were quadratically affected (p<0.05) by dietary treatment. Compared with other treatments, 240 mg/kg Cu from CuCit quadratically increased hepatic (p<0.01) and renal (p<0.05) Cu concentrations, and quadratically decreased hepatic and renal iron concentrations (p<0.05). Conclusion: Cu administered in the form of CuCit at a dosage range of 30 to 60 mg/kg, effectively enhanced the growth performance and antioxidant status of weaned pigs.

        Building Asian climate change scenario by multi‐regional climate models ensemble. Part II: mean precipitation

        <P><B>ABSTRACT</B></P><P>Under the framework of an project ‘Building Asian Climate Change Scenarios by Multi‐Regional Climate Models Ensemble’, the ability of eight regional climate models and two fine‐resolution global climate models to reproduce late 20th century (1981–2000) precipitation climatology is assessed. Future precipitation change (2041–2060) under the A1B scenario is also quantified by applying four different ensemble methods: equal weighting, weighted mean (WM), reliability ensemble averaging (REA) and performance‐based ensemble averaging, after applying fourfold cross‐validation using observation and multi‐model‐simulated precipitation. The results indicate that the ensemble of simulated precipitation outperforms any single RCM in many aspects. Among the four ensemble approaches, the WM and REA methods show better skill in improving the simulation results, and are used for ensemble prediction of regional climate in Asia. Under the A1B scenario, the WM method estimates future precipitation change of approximately 0.2 mm day<SUP>−1</SUP> with less precipitation in northern and western China and northern India, and more precipitation in most other areas in Asia. The future annual precipitation will decrease by 0.1‐0.5 mm day<SUP>−1</SUP> in northern India, Pakistan and the central area of southern China. No significant change is found over eastern Kazakhstan, Mongolia, north‐central and western China.</P>

        Building Asian climate change scenario by multi‐regional climate models ensemble. Part I: surface air temperature

        <P><B>ABSTRACT</B></P><P>Under the Asia‐Pacific Network for Global Change (APN) project ‘Building Asian Climate Change Scenarios by Multi‐Regional Climate Models Ensemble’ (RMIP III, Regional Model Intercomparision Project), the simulation results of eight regional climate models (RCMs) and two fine‐resolution global climate models are validated for reproducibility of the current surface air temperature climatology (1981–2000), and are used to generate surface air future temperature projections (2041–2060) over the CORDEX‐EA (A Coordinated Regional climate Downscaling Experiment‐East Asia) domain. Four ensemble methods, namely, the equal weighting, the weighted mean, the reliability ensemble averaging, and the performance‐based ensemble averaging, are employed to generate the multi‐model projection of regional climate change over the region. The results show that the regional temperature ensembles of the present climate obtained from all four methods can outperform a single RCM result in aspects of the spatial distribution as well as the seasonal variation over East Asia. The four ensemble methods are then used to project the regional temperature climatology under the IPCC emission scenario of A1B for 2041–2060. Compared with the control climate of 1981–2000, the annual mean temperature of the future climate (2041–2060) increases 1–2 °C in low latitude areas and 2–3 °C in middle–high latitude areas over Asia.</P>

      • The XPD Lys751Gln Polymorphism has Predictive Value in Colorectal Cancer Patients Receiving Oxaliplatin-Based Chemotherapy: a Systemic Review and Meta-analysis

        Background: The predictive value of the xeroderma pigmentosum group D (XPD) Lys751Gln polymorphism regarding clinical outcomes of patients with colorectal cancer (CRC) receiving oxaliplatin-based chemotherapy has been evaluated in numerous published studies, but the results remain inconclusive. Therefore, we performed a meta-analysis to determine the precise role of the XPD Lys751Gln polymorphism in this clinical situation and optimize individual chemotherapy. Materials and Methods: A multiple search strategy was used to identify eligible studies. Pooled odds ratios (ORs), generalized odds ratio (ORG) and their 95% confidence intervals (CIs) were used to estimate the objective response, while hazard ratios (HRs) with 95%CIs were used for progression-free survival (PFS) and overall survival (OS). Results: A total of 17 studies including 2,286 patients met the inclusion criteria. Overall, the XPD 751Gln allele was associated with a non-significant reduced objective response to oxaliplatin-based chemotherapy in all patients or in the Asian and Caucasian subgroups. However, poor PFS and OS of CRC patients treated with oxaliplatin-based regimens were significantly related to the XPD 751Gln allele in the dominant model (PFS: HR=2.10, 95%CI: 1.65-2.67; OS: HR=3.18, 95%CI: 1.57-6.47). On stratified analysis by ethnicity, these relationships were more pronounced in Asians (PFS: HR=2.49, 95%CI: 1.79-3.47; OS: HR=5.25, 95%CI: 3.46-7.94) than in Caucasians (PFS: HR=1.73, 95%CI: 1.22-2.46; OS: HR=1.78, 95%CI: 1.06-2.99). Conclusions: The XPD Lys751Gln polymorphism may have prognostic value in patients with CRC undergoing oxaliplatin-based chemotherapy.

        Apatinib Combined with Local Irradiation Leads to Systemic Tumor Control via Reversal of Immunosuppressive Tumor Microenvironment in Lung Cancer

        Purpose This study aimed to investigate the potential systemic antitumor effects of stereotactic ablative radiotherapy (SABR) and apatinib (a novel vascular endothelial growth factor receptor 2 inhibitor) via reversing the immunosuppressive tumor microenvironment for lung carcinoma. Materials and Methods Lewis lung cancer cells were injected into C57BL/6 mice in the left hindlimb (primary tumor; irradiated) and in the right flank (secondary tumor; nonirradiated). When both tumors grew to the touchable size, mice were randomly divided into eight treatment groups. These groups received normal saline or three distinct doses of apatinib (50 mg/kg, 150 mg/kg, and 200 mg/kg) daily for 7 days, in combination with a single dose of 15 Gy radiotherapy or not to the primary tumor. The further tumor growth/regression of mice were followed and observed. Results For the single 15 Gy modality, tumor growth delay could only be observed at the primary tumor. When combining SABR and apatinib 200 mg/kg, significant retardation of both primary and secondary tumor growth could be observed, indicated an abscopal effect was induced. Mechanism analysis suggested that programmed death-ligand 1 expression increased with SABR was counteract by additional apatinib therapy. Furthermore, when apatinib was combined with SABR, the composition of immune cells could be changed. More importantly, this two-pronged approach evoked tumor antigen–specific immune responses and the mice were resistant to another tumor rechallenge, finally, long-term survival was improved. Conclusion Our results suggested that the tumor microenvironment could be managed with apatinib, which was effective in eliciting an abscopal effect induced by SABR.
