        Endophytic fungi harbored in Panax notoginseng: diversity and potential as biological control agents against host plant pathogens of root-rot disease

        Background: Endophytic fungi play an important role in balancing the ecosystem and boosting host growth. In the present study, we investigated the endophytic fungal diversity of healthy Panax notoginseng and evaluated its potential antimicrobial activity against five major phytopathogens causing rootrot of P. notoginseng. Methods: A culture-dependent technique, combining morphological and molecular methods, was used to analyze endophytic fungal diversity. A double-layer agar technique was used to challenge the phytopathogens of P. notoginseng. Results: A total of 89 fungi were obtained from the roots, stems, leaves, and seeds of P. notoginseng, and 41 isolates representing different morphotypes were selected for taxonomic characterization. The fungal isolates belonged to Ascomycota (96.6%) and Zygomycota (3.4%). All isolates were classified to 23 genera and an unknown taxon belonging to Sordariomycetes. The number of isolates obtained from different tissues ranged from 12 to 42 for leaves and roots, respectively. The selected endophytic fungal isolates were challenged by the root-rot pathogens Alternaria panax, Fusarium oxysporum, Fusarium solani, Phoma herbarum, and Mycocentrospora acerina. Twenty-six of the 41 isolates (63.4%) exhibited activity against at least one of the pathogens tested. Conclusion: Our results suggested that P. notoginseng harbors diversified endophytic fungi that would provide a basis for the identification of new bioactive compounds, and for effective biocontrol of notoginseng root rot.

      • Prognostic Significance of Interactions Between ER Alpha and ER Beta and Lymph Node Status in Breast Cancer Cases

        Objective: Both estrogen receptors, ER alpha ($ER{\alpha}$) and ER beta ($ER{\beta}$), are expressed in 50-70% of breast cancer cases. The role of $ER{\alpha}$ as a prognostic marker in breast cancer has been well established as its expression is negative correlated with tumor size and lymph node metastasis. $ER{\beta}$ is also a favorable prognostic predictor although this is less well documented than for $ER{\alpha}$. Materials and Methods: To explore whether ERs independently or together might influence clinical outcome in breast cancer, the correlation between the ERs with the clinicopathological features was analyzed in 84 patients. Results: $ER{\alpha}$ expression negatively correlated with tumor stage (r=-0.246, p=0.028) and tended to be negatively correlated with lymph node status (r=-0.156, p=0.168) and tumor size (r=-0.246, p=0.099). Also, $ER{\beta}$ was negatively correlated with nodal status (r=-0.243, p=0.028), as was coexpression of $ER{\alpha}$ and $ER{\beta}$ (p=0.043, OR=0.194, 95% CI= 0.040-0.953). Conclusion: Coexpression of ERs might serve as an indicator of good prognosis in breast cancer patients.

        Calculation of eigenvalue and eigenvector derivatives with the improved Kron's substructuring method

        For large-scale structures, the calculation of the eigensolution and the eigensensitivity is usually very time-consuming. This paper develops the Kron's substructuring method to compute the first-order derivatives of the eigenvalues and eigenvectors with respect to the structural parameters. The global structure is divided into several substructures. The eigensensitivity of the substructures are calculated via the conventional manner, and then assembled into the eigensensitivity of the global structure by performing some constraints on the derivative matrices of the substructures. With the proposed substructuring method, the eigenvalue and eigenvector derivatives with respect to an elemental parameter are computed within the substructure solely which contains the element, while the derivative matrices of all other substructures with respect to the parameter are zero. Consequently this can reduce the computation cost significantly. The proposed substructuring method is applied to the GARTEUR AG-11 frame and a highway bridge, which is proved to be computationally efficient and accurate for calculation of the eigensensitivity. The influence of the master modes and the division formations are also discussed.

      • Down-regulation of Phosphoglucose Isomerase/Autocrine Motility Factor Enhances Gensenoside Rh2 Pharmacological Action on Leukemia KG1α Cells

        Aims and Background: Ginsenoside Rh2, which exerts the potent anticancer action both in vitro and in vivo, is one of the most well characterized ginsenosides extracted from ginseng. Although its effects on cancer are significant, the underlying mechanisms remain unknown. In this study, we sought to elucidate possible links between ginsenoside Rh2 and phosphoglucose isomerase/autocrine motility factor (PGI/AMF). Methods: $KG1{\alpha}$, a leukemia cell line highly expressing PGI/AMF was assessed by western blot analysis and reverse transcription- PCR (RT-PCR) assay after transfection of a small interfering (si)-RNA to silence PGI/AMF. The effect of PGI/AMF on proliferation was measured by typan blue assay and antibody array. A cell counting kit (CCK)-8 and flow cytometry (FCM) were adopted to investigate the effects of Rh2 on PGI/AMF. The relationships between PGI/AMF and Rh2 associated with Akt, mTOR, Raptor, Rag were detected by western blot analysis. Results: KG1${\alpha}$ cells expressed PGI/AMF and its down-regulation significantly inhibited proliferation. The antibody array indicated that the probable mechanism was reduced expression of PARP, State1, SAPK/JNK and Erk1/2, while those of PRAS40 and p38 were up-regulated. Silencing of PGI/AMF enhanced the sensibility of $KG1{\alpha}$ to Rh2 by suppressing the expression of mTOR, Raptor and Akt. Conclusion: These results suggested that ginsenoside Rh2 suppressed the proliferation of $KG1{\alpha}$, the same as down-regulation of PGI/AMF. Down-regulation of PGI/AMF enhanced the pharmacological effects of ginsenoside Rh2 on KG1${\alpha}$ by reducing Akt/mTOR signaling.

        Position and Force Control of Bilateral Teleoperation Systems With Time-varying Delays Based on Force Estimation

        A position and force tracking control based on force estimation is proposed for bilateral teleoperation systems with time-varying delays. A time-delay state observer is employed to estimate the system state variables affected by the delays. To estimate the interaction forces effectively, a force estimation algorithm with adaptive law is designed. Based on the estimated states and forces, a P+D controller is designed to simultaneously guarantee the position and force tracking of the system. The stability and tracking performance of the closed-loop system are proved via Lyapunov functions, and the feasibility of the proposed control is verified by both simulations and experiments. The proposed control can improve the position and force tracking performance of bilateral teleoperation systems under time-varying delays. Meanwhile, it neither requires force measurement nor the bound of the derivative of the time-varying delays to be within one.

        In Vivo Protein Transduction: Delivery of PEP-1-SOD1 Fusion Protein into Myocardium Efficiently Protects against Ischemic Insult

        Myocardial ischemia-reperfusion injury is a medical problem occurring as damage to the myocardium following blood flow restoration after a critical period of coronary occlusion. Oxygen free radicals (OFR) are implicated in reperfusion injury after myocardial ischemia. The antioxidant enzyme, Cu, Zn-superoxide dismutase (Cu, Zn-SOD, also called SOD1) is one of the major means by which cells counteract the deleterious effects of OFR after ischemia. Recently, we reported that a PEP-1-SOD1 fusion protein was efficiently delivered into cultured cells and isolated rat hearts with ischemia-reperfusion injury. In the present study, we investigated the protective effects of the PEP-1-SOD1 fusion protein after ischemic insult. Immunofluorescecnce analysis revealed that the expressed and purified PEP-1-SOD1 fusion protein injected into rat tail veins was efficiently transduced into the myocardium with its native protein structure intact. When injected into Sprague-Dawley rat tail veins, the PEP-1-SOD1 fusion protein significantly attenuated myocardial ischemia-reperfusion damage; characterized by improving cardiac function of the left ventricle, decreasing infarct size, reducing the level of malondialdehyde (MDA), decreasing the release of creatine kinase (CK) and lactate dehydrogenase (LDH), and relieving cardiomyocyte apoptosis. These results suggest that the biologically active intact forms of PEP-1-SOD1 fusion protein will provide an efficient strategy for therapeutic delivery in various diseases related to SOD1 or to OFR.

        Measurement of rivulet movement and thickness on inclined cable using videogrammetry

        Stay cables in some cable-stayed bridges suffer large amplitude vibrations under the simultaneous occurrence of rain and wind. This phenomenon is called rain–wind-induced vibration (RWIV). The upper rivulet oscillating circumferentially on the inclined cable surface plays an important role in this phenomenon. However, its small size and high sensitivity to wind flow make measuring rivulet size and its movement challenging. Moreover, the distribution of the rivulet along the entire cable has not been measured. This paper applies the videogrammetric technique to measure the movement and geometry dimension of the upper rivulet along the entire cable during RWIV. A cable model is tested in an open-jet wind tunnel with artificial rain. RWIV is successfully reproduced. Only one digital video camera is employed and installed on the cable during the experiment. The camera records video clips of the upper rivulet and cable movements. The video clips are then transferred into a series of images, from which the positions of the cable and the upper rivulet at each time instant are identified by image processing. The thickness of the upper rivulet is also estimated. The oscillation amplitude, equilibrium position, and dominant frequency of the rivulet are presented. The relationship between cable and rivulet variations is also investigated. Results demonstrate that this non-contact, non-intrusive measurement method has good resolution and is cost effective.

        Baimantuoluosides D-G, Four New Withanolide Glucosides from the Flower of Datura metel L.

        In our search for bioactive anti-psoriasis compounds from the flower of Datura metel L, we isolated four new withanolide glucosides, baimantuoluosides D, E, F and G (1-4). The structures of the new compounds are (5α,6α,7β,22R)-5,6,7,27-tetrahydroxy-1-oxowitha-2,24-dien-27-O-β-D-glucopyranoside (1), (5α,6β,7α,22R)-5,6,7,27-tetrahydroxy-1-oxowitha-2,24-dien-27-O-β-D-glucopyranoside (2), (5α,6β,7α,12β,22R)-5,6,7,12,27-pentahydroxy-1-oxowitha-2,24-dien-27-O-β-D-glucopyranoside (3), and (5α,6β,22R)-5,6,27-trihydroxy-1-oxowitha-2,24-dien-27-O-β-D-glucopyranoside (4) on the basis of chemical and physicochemical evidence.
