소목(Caesalpinia sappan) 추출물의 Streptococcus mutans에 대한 성장, 산생성, 부착 및 비수용성 글루칸 합성 억제에 미치는 영향

Streptococcus mutans is known as the causative bacterial playing the most important role in forming plaque and it is being noticed as major causative bacteria of dental caries. Therefore, development of more effective, substantial and safe preventive agent against dental caries and periodontal disease is honestly required. The present study was designed to investigate the effect of chloroform extract(CE), n-butanol extract(BE),methanol extract(ME) and water extract(WE) of Caesalpinia on the growth, acid production, adhesion, and insoluble glucan synthesis of Streptococcus mutans. The CE, BE, ME and WE decreased the growth and acid production of S.mutans than that of control, especially, the ME was more effective than other extracts. The effect of CE, BE, ME and WE on the adherences to saliva-coated hydroxyapatite bead(S-HA) were evaluated. At the concentration of 2,000 ㎍/㎖, the CE showed 32% rate of bacterial adherence to S-HA, whereas, other extracts of the same concentration producedgreater than 50% rate. The CE and BE were significantly decreased the reduction rates of water insoluble glucan formation from sucrose by crude glucosyltransferase of S.mutans. Thus, the application of C. sappan can be considered a useful and a practical method for the prevention of dental caries.

This paper is about establishment of authentic text and annotation on Lee Sang's literature. Frist, we pointed out the fallacies of the first printing of Lee Sang's Japanese Poems and scrutinized the fallacies of translation and compiling process of collection in many aspects. The fallacies of translation were grouped 10category such as opposite translation, absurd translation, colloquial expression, chinese writing style expression and so on. Some of the results have already been pointed out but many of them are new We believe that this paper ascertained more authentic text version but the establishment of authentic text has not been finished. Still, we can not identify initial name 'R' presented in the prefaces of 〈ChoSeon and Architecture(朝鮮と建築)〉. We reserve this part of work unfinished for another time and another researcher. Secondly, this paper also focused on 〈Raven's eye view(烏瞰圖)〉 which is the most annotated poem of Lee Sang. After we studied former researcher's opinions about the title 〈Raven's eye view〉, We bring up the possibility that the title 〈bird's eye view(烏瞰圖)〉is more original one than 〈Raven's eye view〉. In addition to that, we studied annotation of 〈poem number 1〉the most discussed poem of Lee Sang's and studied 〈poem number 1(詩第1號)〉. Annotation work about poems of Lee Sang first developed by Lee Ur-rhyueng and studied more seriously by Lee Seoung-hun. Based on their studies. This paper presented more particular and detailed annotation work after fully surveying almost all papers, criticisms, and books, which are related to Lee Sang. Specifically we supplemented annotation that Lee Seoung-hun omitted annotation amongst papers published by 1989 and added new research results. We still feel something wanting that we can not bring up everything. It is because of the shortage of research period and different style of this paper from general paper. The remainder of works are annotation of full 〈Raven's eye view〉and the whole works of Lee' Sang. Then, we can avoid duplicated study and will present new results based of authentic text version. If this is completed, we can enhance the level of study about Lee Sang.

The purpose of this study is to find out the optimal mixing ratios of amounts of Mal-Cha for preparation of Jeung-Pyun through Sensory, Mechanical characteristics test. The proximate composition of Mal-Cha were a 5.46±0.15% of moisture, 4.43±0.11% of total nitrogen, 7.52±0.21% of crude lipid, 8.74% of crude fiber, 8.51±0.09% of ash. Overall quality in 1.5% Jeung-Pyun showed the worst of 0, 0.5, 1.0% Jeung-Pyuns(p<0.05), especially 1.0% Jeung-Pyun had relatively the best overall quality. But the sweetness, sourness, flavor, hardness, moistness were not significantly different among all Jeung- Pyuns. 0, 0.5, 1.0% Jeung-Pyuns showed the best quality of Jeung-Pyun through sensory, mechanical property. Total color difference was increased as amount of Mal-Cha(p<0.001).

Antibiotic-resistant Staphylococcus aureus (S. aureus) emerged in many parts of the world and increased complex clinical problems. Therefore, new agents are needed to treat the antibiotic-resistant S. aureus. Some natural products are candidates of new antibiotic substances. Previous reports showed that the extract of Coptidis Rhizoma and Phellodendri Cortex have antimicrobial effects. The main antibacterial substance of Coptidis Rhizoma and Phellodendri Cortex is berberine. In this study, we examined the antimicrobial activity of berberine to antibiotic-resistant S. aureus. We isolated the 13 strains of antibiotic-resistant S. aureus from the oral cavity of healthy children in a preschool, and investigated the antimicrobial activity of berberine. We examined the effects of berberine on the level of resistance to antibiotic-resistant S. aureus. We observed that the invasion of S. aureus into human gingival fibroblasts was blocked by berberine. Berberine inhibited the growth of antibiotic-resistant S. aureus as a dosw dependent maner. Minimum inhibitory concentration (MIC) of berberine against antibiotic-resistant S. aureus was 50∼100㎍/㎖. The inhibition of S. aureus invasion was depended on the concentration of berberine in human gingival fibroblasts. The MIC of ampicillin against antibiotic-resistant S. aureus was 32㎍/㎖. However, the MIC of ampicillin against antibiotic-resistant S. aureus in the presence of 50㎍/㎖ berberine was 1㎍/㎖. These data indicate that the combination with berberine and β-lactam antibiotics resulted in the synergistic reaction. These results suggest that the berberine may have the antimicrobial activities to antibiotic-resistnat S. aureus, and the mechanism may be the inhibition of growth and intracellular invasion, and alteration of antibiotic-resistance level of antibiotic-resistant S. aureus.

Geneally, rice seeds regardless indica or japonica are showing low germination ratio or completely lost germination ability together with lost of good eating quality under high temperature and humidity conditions. Thus, this study was designed to evaluate a longevity for conservation of good eating quality during long term storage in rice. For the longevity evaluation, germination ability was studied after 5 days of high temperature and humidity stress (50℃/RH 95%). Dharial, originated from Bangladesh and showing weedy type with red pericarp, was selected as a good donor for longevity genes. A mutant was developed from Dharial through EMS mutagenesis and two populations of Dharial/4*Ilmibyeo and Dharial/4*Gopumbyeo were also developed for genetic study. In the 2-DE analysis followed by MALDI-TOF MS with wild and mutant lines, several candidate genes were identified. In the longevity test of two populations, a few lines showing good germination ability after high temperature and humidity stress were selected and subjected to confirm the relationships between longevity and conservation of good eating quality under long term storage.
Enhancing Fatty Acid Production of <i>Saccharomyces cerevisiae</i> as an Animal Feed Supplement

<P><I>Saccharomyces cerevisiae</I> is used for edible purposes, such as human food or as an animal feed supplement. Fatty acids are also beneficial as feed supplements, but <I>S. cerevisiae</I> produces small amounts of fatty acids. In this study, we enhanced fatty acid production of <I>S. cerevisiae</I> by overexpressing acetyl-CoA carboxylase, thioesterase, and malic enzyme associated with fatty acid metabolism. The enhanced strain pAMT showed 2.4-fold higher fatty acids than the wild-type strain. To further increase the fatty acids, various nitrogen sources were analyzed and calcium nitrate was selected as an optimal nitrogen source for fatty acid production. By concentration optimization, 672 mg/L of fatty acids was produced, which was 4.7-fold higher than wild-type strain. Epizootic HPAIV, H5N6, and H5N8 infections produced severe loss in poultry and wild birds in the Republic of Korea from 2016 to 2017. But pathological lesions and antigen distribution of the novel HPAIV H5N6 clade 2.3.4.4 in natural cases have been rarely reported. Herein, we describe the pathological lesions and antigen localization in chickens (layer and Korean native), ducks, and Japanese quail naturally infected by HPAIV H5N6. Grossly, severe reddening, swelling, and some necrotic foci, which were similar to septicemia or viremia, were observed in skin and many visceral organs including trachea, lung, liver, spleen, and pancreas. Histopathologically, pulmonary congestion and edema, as well as necrotizing hepatitis, splenitis, pancreatitis, myocarditis, and encephalitis were observed. Immunohistochemically, numerous HPAIV antigens were detected in necrotic parenchymal cells and in blood vessels of the respiratory, lymphoid, digestive, urinary, nervous, and cardiovascular systems. The results indicate that HPAIV H5N6 spread to the entire body via blood and caused severe damage throughout the entire body. The HPAI H5N6 clade 2.3.4.4 virus was isolated from samples of all four cases.
