麥酒麥의 (1-3,1-4)-β-glucanase 遺傳子의 構造와 發現 樣狀

禾穀類 種子 發芽時 호분층과 ??組織의 細胞壁에 多量 含有되어 있는 (1-3, 1-4)-β-glucan을 分解하여 種子의 發芽에 必要한 加水分解酵素의 分泌 및 擴散 浸透를 돕는다. 本 硏究에서는 國內 麥酒麥의 (1-3, 1-4)-β-glucanase 遺傳子의 構造 및 發現 에 대한 基礎 情報를 얻기 위하여 生化學的 및 分子生物學的 方法을 利用하여 發芽種子 및 어린 잎에서의 酵素活性과 遺傳子의 發芽樣狀 그리고 遺傳子의 構造 等을 調査하였다. 麥酒麥 發芽種子中 (1-3, 1-4)-β-glucanase 活性은 發芽 7日째에 제일 높았다. (1-3, 1-4)-β-glucanase의 活性測定에 使用한 (1-3, 1-4)-β-glucan이 (1-4)-β-glucanase에 의해 分解될 可能性이 있으므로 (1-4)-β-glucanase의 活性도 測定하였는데 (1-4)-β-glucanase의 活性이 매우 낮게 나타나서(1-4)-β-glucanase가 (1-3, 1-4)-β-glucanase을 分解하여 (1-3, 1-4)-β-glucanse의 活性에 影響을 미쳤을 可能性이 매우 낮았다. 麥酒麥의 (1-3, 1-4)-β-glucanase 遺傳子 構造를 PCR을 利用하여 調査하였던바 國內 麥酒麥의 (1-3, 1-4)-β-glucanase 遺傳子 構造는 外國의 다른 品種들이나 귀리의 遺傳子 構造와 대단히 類似하였다. 酵素活性 測定과 귀리의 (1-3, 1-4)-β-glucanase cDNA clone을 ?識子로 利用한 보리의 mRNA 分析에 의해서 發芽傳子 및 어린 잎에서 (1-3, 1-4)-β-glucanase가 種子發芽 以外에도 生育中인 組織細胞의 伸張과 發育 및 β-glucan 再利用에 泌要한 β-glucan 代謝에 關與하고 있음을 示唆하는 結果라 생각된다. (1-3, 1-4)-β-glucans hydrolysis of cell wall (1-3, 1-4)-β-glucanas accompanies degradation of endosperm cell walls during cereal seed germination. To investigate (1-3, 1-4)-β-glucanase expression pattern in Korean malting barley, (1-3, 1-4)-β-glucanase activity, gene structure and mRNA expression patterns werre examined. (1-3, 1-4)-β-glucanase activity increased as germination proceeded and the highest enzyme activity was detected in seeds germinated for 7 days. Because (1-4)-β-glucanase have a potential to degrade (1-3, 1-4)-β-glucans, (1-4)-β-glucanase activity was also determined. (1-4)-β-glucanase activity, however, was very low minimizing possibility of (1-4)-β-glucanase action on (1-3, 1-4)-β-glucan substrate. PCR technique was employed to investigate (1-3, 1-4)-β-glucanase gene structure. The amplification product of the expected size from the targeted sites of Dusan #29 indicated that (1-3, 1-4)-β-glucanase systems were conserved in different barley varieties and even in oats. (1-3, 1-4)-β-glucanase activity and mRNAs were detected in young leaves as well as in germinating seeds. This may suggest that (1-3, 1-4)-β-glucanase are also involved in β-glucan metabolism required for cell elongation and β-glucan reutilization during leaf growth and developmetn.
백두대간 관리범위 설정에 관한 연구(Ⅱ) : 준산악형 구간을 대상으로 In the Case of Semi-mountainous District

백두대간은 국토의 근간이자 중요한 생태축이다. 그러나 최근 백두대간 지역을 대상으로 한 각종 개발요구가 증가함에 따라 이에 적절히 대응하기 위한 보전 전략 수립과 체계적인 관리의 필요성이 부각되고 있다. 이 연구는 백두대간 지역을 효율적으로 보호하기 위한 관리구역 설정을 제안하고자 수행하였다. 접근 방법은 백두대간의 적절한 관리범위 설정을 위해 수계 및 유역을 바탕으로 인간 간섭의 영향정도를 파악하는 것으로, 수치지도, Landsat TM 영상 등이 사용되었다. 준산악지형의 특성을 갖는 남덕유산~소사고개 구간을 대상으로 유역확장방식을 사용하여 유역차수에 따른 경작지, 도시, 나지, 수계, 산림 등의 토지이용상태의 변화를 추적하였다. 그 결과 유역확장방식은 백두대간 관리범위 설정기법으로 효율적이었으며, 관리범위는 마루금을 중심으로 무주(8차)와 거창 (5차) 방향이 상이하게 설정되었다. Baekdu-daegan is the greatest mountain chain as well as the major ecological axis of the Korean Peninsula. In recent year, however, this area is faced with the various kinds of developmental urge. To cope adequately with these problems, this study was executed to prepare synthetic and systematic management with conservation-oriented strategy for Baekdu-daegan and to suggest spatially definite zoning for the managerial area. This study is to take into consideration the traditional concepts of stream and watershed as well as the actual disturbance on Baekdu-daegan area. The study area is selected with semi-mountainous type, from Namdeokyusan to Sosagogae. To propose the process for reasonably establishing the managerial boundary adjacent to the ridges, the analysis was carried out that ArcGIS was mainly used for its analysis with digital maps, Landsat TM image and ArcGIS Hydro Model. Landsat TM image was classified by 5 land use types such as cultivated land, urban area, barren area, water body and forest. Based on these analyses results, the managerial boundaries as alternatives from the Ridges were produced by watershed expansion process, and used for tracing the changes of areal ratio of various land use types to the relevant watersheds to search out the adequate managerial boundary. The results show that watershed expansion process could be effective tool for establishing the managerial boundary, and eighth expanded watershed toward Muju-Gun(west) and fifth expanded watershed toward Geochang-Gun (east) might be included for the adequate managerial boundary of the case site.
3-Cyano 및 3-Carbomethoxy Chromone과 Allenylstannane과의 반응

t-Butyldimethylsilyl triflate 존재에서 3-Cyano 및 3-Carbomethoxy Chromone(1a,b)과 Allenylstannane(2) 과의 반응에서 Chromone의 C-2위치에 propargyl기가 도입된 유도체(3a,b)를 합성하였다. 합성한 유도체의 상대적 구조는 ¹H-NMR을 통해 분석하였으며, T.S구조의 모델을 통하여 가능한 메카니즘을 제시하였다. Reaction of 3-Cyano and 3-Carbomethoxy Chromone(1a,b) with Allenylstannane(2) in the presence of tert-butyldimethylsilyl triflate afforded the corresponding the propargylation product(3a,b) in the moderate yield. The relative stereochemistry of 3a,b are obtained on the basis of ¹H-NMR analysis. And the possible reaction mechanism are suggested by the molecular models in the transition state.
고속가공에서 2중 신경망을 이용한 표면거칠기 예측과 가공DB 구축 효율화 방안

In this paper, a double artificial neural network (ANN) approach and the efficient machining database building scheme are presented for the prediction of surface roughness in high-speed machining. In this approach, 4 machining parameters and used for the prediction of cutting force components, and the combinations of 4 parameters and the predicted cutting force components are finally used for the prediction of surface roughness. The experimental results comparing the these results with the predicted values using simple 4 input nodes have been also investigated to verify the effectiveness of the proposed approach.

Purpose: Studies about the effect of radiation therapy (RT) on immune cells are usually limited to a high-grade glioma mostly exposed to chemotherapy and a high dose of steroid which also could affect immune cells. The purpose of this retrospective analysis of low-grade brain tumor patients treated by RT alone is to determine significant factors influencing neutrophil-to-lymphocyte ratio (NLR), absolute neutrophil counts (ANC), and absolute lymphocyte counts (ALC). Materials and Methods: A total of 41 patients who received RT between 2007 and 2020 were analyzed. Patients who received chemotherapy and high-dose of steroid were excluded. ANC and ALC were collected before starting RT (baseline) and within one-week before ending RT (post-treatment). Changes of ANC, ALC, and NLR between baseline and post-treatment were calculated. Results: ALC decreased in 32 patients (78.1%). NLR increased in 31 patients (75.6%). No patients developed grade 2 or higher hematologic toxicities. The decrease of ALC was significantly correlated with the dose to brain V15 in a simple and multiple linear regression (p = 0.043). Brain V10 and V20 adjacent to V15 were also marginally significant factors determining the reduction of lymphocytes (p = 0.050 and p = 0.059, respectively). However, it was difficult to find predictive factors affecting changes of ANC and NLR. Conclusion: In low-grade brain tumor patients who are treated by RT alone, ALC decreased and NLR increased in three-fourth of patients, although the magnitude was minimal. The decrease of ALC was mainly affected by low dose to the brain. However, RT dose was not correlated with changes of ANC or NLR.

<P>Dynamin 2 is known as a protein involved in cell migration and endocytosis. We aimed to investigate the association between dynamin 2 expressions and tumor progression in early cervical carcinoma (IB1-IIA). Dynamin 2 expression was evaluated at protein level in thirty seven paraffin-embedded, formalin-fixed tissues including four normal cervix tissues and compared with pathologic risk factors for recurrence after surgery in thirty three patients with squamous cell carcinoma of the cervix. The expression of dynamin 2 was not different according to clinical stage and lympho-vascular space invasion. However, there were inverse correlations between dynamin 2 expression and the depth of invasion in cervix (p = 0.003) and lymph node (LN) metastasis (p = 0.001). To evaluate the mechanism of dynamin 2 in tumor invasion and metastasis, we performed an in vitro experiment with dynamin 2 siRNA using several cervical carcinoma cell lines such as HeLa, MS751 and SiHa cells. We found the inhibition of dynamin 2 using specific siRNA enhanced the expression of matrix metalloproteinase-2. These results suggested that dynamin 2 might be involved in preventing tumor invasion and LN metastasis, possibly in relation with extracellular matrix degradation, and may be a prognostic marker for these risk factors in early squamous cell carcinoma of the cervix.</P>
