        치주인대 세포의 교원질 생성에 대한 Substance P의 효과

        Substance P는 교정력이 가해진 치아의 치주인대 중 인장력을 받는 부위에 많이 분포하는 neuropeptide 중의 하나이며, 또한 여러 조직에서 neurogenic inflammation을 야기하는 neuropeptide 중의 하나로도 알려져 있다. 그러나 중요한 세포의 단백기질인 교원질의 생성에 대한 Substance P의 효과는 잘 알려져 있지 않다. 따라서 이 연구의 목적은 배양 치주인대 세포에서 교원질 생성에 대한 Substance P의 효과를 평가하는 것이었다. collagenase-digestion method로 교원질 생성을 평가하였고 mRNA 수준에서 작용효과를 평가하기 위하여 Northern blot hybridization을 시행하였다. 이 연구는 또한 교원질 생성에 대한 prostaglandin과 gelatinase 생성도 포함하였으며 변성된 교원질의 분해를 평가하기 위하여 Zymography를 이용하였다. 비교원성 단백질, 교원성 단백질, 상대교원질에 대한 dose-dependent effect를 보면 Substance P는 비교원성 단백질 합성을 증가시켰으나 교원성 단백질 합성은 감소시켰다. 그리하여 총 단백합성에 대한 상대적인 교원질 생성을 나타내는 상대교원질은 7%에서 3.6%로 감소시켰다. 세포를 indomethacin과 동시에 처리할 때 substance P의 교원질 합성 억제효과는 나타나지 않았다. 이것은 Substance P의 교원질 합성 억제효과가 prostaglandin의 생성 때문이라는 것을 의미한다. Substance P의 교원질 합성 억제효과가 procollagen mRNA의 정상(steady-state)수준에 부합하는가를 평가하기 위하여 northern blot hybridization을 시행한 결과 Substance P는 α1(1) procollagen mRNA의 양적 변동을 일으키지 않았다. Substance P의 교원질 생성 억제효과는 전사이후의 어떤 단계에서 이루어지는 현상임을 나타낸다. 치주인대세포에서 gelatinase 생성에 대한 Substance P의 효과를 알아보기 위하여 zymography를 이용하였다. zymogram을 보면 Substance P는 치주인대세포에서 gelatinase 생성에는 아무 효과도 나타내지 않음을 알 수 있다. Substance P의 교원질 생성 억제효과가 치주인대세포에 대해 선택적인가 아닌가를 알아보기 위하여 MC3T3-E1세포를 이용하였는데 Substance P는 MC3T3-E1세포의 교원질 합성에는 영향을 미치지 않았다. 이상에서 Substance P는 인간의 치주인대세포에서 교원질 합성을 억제하였다. 이 효과는 procollagen mRNA와 gelatinase 생성의 정상(steady-state) 수준의 변화 때문이 아니라 prostaglandin 생성과 연관이 있음을 알았다. Substance P is one of the neuropeptide which presents highly in tension site of periodontal ligament during the orthodontic tooth movement. It has been also known as one of the neuropeptides which cause neurogenic inflammation in various tissues and organs. However, there is no report about the effect of substance P on major extracellualar matrix protein, collagen production. The purpose of this study was to evaluate the collagen production by substance P in human periodontal ligament cell, The collagenase-digestion method was used to evaluate collagen production and also used Northern blot hybridization for the evaluation of collagen mRNA level. This study also included in terms of prostanglandins and gelatinase production with respect to collagen production. For the collagen degradation, zymography was used to estimate denatured collagen degradation. Dose-dependent effect of substance P on noncollagen protein, collagen, and percent collagen was that substance P increased noncollagen protein synthesis, but decreased collagen systnisis. So the percent collagen, which determined by relative collagen production against total protein production, was decreased from 7% to 3.6%. This inhibitory effect of substance P on collagen production was disappeared when cells were treated concomitantly with indomethacin. It means that substance P-induced inhibitory effect on collagen production was due at least in part to the production of prostaglandins. To evaluate whether substance P-induced inhibitory effect on collagen production is correspond to the steady-state levels of procollagen mRNA, Northern blot hybridizartion was performed and it showed that substance P has no effect on the steady-state level of α1(1) procollagen mRNA. It means that the inhibitory effect of substance P on collagen production was due to the change of a certain mechanism after posttranscription. In this context, gelatinase production by substance P in periodontal ligament cells was evaluated by zymography. Zymogram showed that substance P has no effect on gelatinase production in periodontal ligament cells. To explore wheter substance P-induced inhibitory effect on collagen production is selevtive in periodontal ligament cells or not, MC3T3-31 cells which originated from mouse calvaria was used. It showed that substance P has no effect on collagen production in MCDTD-E1 cells. Taken together, substance P inhibits collagen production in human periodontal ligament cells. This effect was not due to the change of the steady-state level of procollagen mRNA and gelatinase production, but due at least in part to the change of prostaglandins production.

        We have already made the concentic cylindrical ESA. and investigated its characteristics. From those experimental results, we detected unexpected background peaks together with the peak corresponding to the extracting voltage, ??, on our energy spectrum of charged particle beams. Therefore, to find out the source of occurrence of those background peaks, we measured the voltage between extractor and slit of ESA conferring that the origin of occurrence of those peaks would be unexpected electric field between extractor and slit. As the results of our experiment, we discovered the existence of time-varying voltage similar to square wave form in this region when the extractor floated from ground and found out these background peaks are caused by floated extractor.

        정신보건관련시설 입원(소)자의 주요 정신장애 유병률 : 전국 20개 시설의 입원(소)자를 대상으로

        Objectives : One of the objectives of this study is to estimate the prevalence rates of psychiatric diagnoses in the combined populations of psychiatric hospitals, psychiatric nursing facilities and homeless asylums using the Korean version of the Composite International Diagnostic Interview (K-CIDI), which is a comprehensive and fully standardized interview schedule to assess psychiatric disorders for diagnosis. The Other objective is to compare with previously studied prevalence rates of Psy-chiatric diagnoses using the results of this study. Methods : The study subjects, aged from 181o 64 years, were randomly selected from 64,582 institutionalized population of psychiatric hospitals, Psychiatric nursing facilities and homeless asylums as of 30, June, 2001. Twelve trained interviewers administered the K-CIDI to the selected respondents. A total of 1,875 respondents (male 1,194, female 681) completed the interview. Results : The lifetime and one year prevalences of any diagnosis excluding nicotine dependence · withdrawal, anxiety disorder, eating disorder, somatoform disorder were 88.2% (male 88.1%, female 88.3%), and 65.8% (male 62.7%, female 73.7%), respectively. The lifetime and one year prevalences of schizophrenia and other psychotic disorders were 78.5% (male 75.3%, female 83.7%), and 56.8% (male 51.9%, female 64.8%), respectively. The lifetime and one year prevalences of alcohol use disorder (dependence/abuse) were 26.7% (male 37.9%, female 8.3%), and 8.7% (male 12.8%, female 2.1%), respectively. The lifetime and one year prevalences of mood disorder were 18.1% (male 13.5%, female 25.8%), and 10.4% (male 7.2%,female 16.9%), respectively. The lifetime and one year prevalences of schizophrenia and other psychotic disorders were comparable with the 2001 community survey by 1.19% to 1.32%, and 0.51% to 0.61%, respectively. The lifetime and one year prevalences of schizophrenia were also comparable by 0.16% to 0.28%, and 0.16% to 0.25%, respectively. There were no significant changes of prevalence rates when correction were applied to other psychiatric diagnosis. Conclusion : The results of this study could be used for evaluating the distribution of psychiatric diagnoses in mental health related facilities and for planning mental health policies.

        A past monthly data is not faithful so much for a short term. But, the stochastic generation technique was provide of a long-term data. Thus this study is used a data which generated a monthly inflow amounts data by Thomas-Fiering model. This model is needed a certain process which determination of distribution, decision of continuous durability, etc. It was generated a inflow data every one month as Thomas-Fiering method. The generated inflow data was used input data for a monthly cumulative analysis. This analysis obtained a storage capacities which would be required during droughts having various return periods. It was presented a equation of fitting regression that carried out regression analysis of 5,10, 20, 50 years period.

        북한 벼 품종의 지대별 출수반응

        북한벼의 출수 반응을 검토하기 위하여, 작물시험장 인공기상실과 표고가 다른 수원, 철원, 진부 등 3 지역에서 이앙기를 달리하여 1998년과 1999년에 시험 수행한 결과를 요약하면 다음과 같다. 가. 지대별 이앙기 변동에 다른 출수일수 변화는 표고가 낮은 지역일수록, 이앙기가 늦어질수록 출수일수가 짧아졌으며, 지역과 이앙기 이동에 따른 경향을 살펴보면, 표고가 높은 진부지역에서 이앙기가 늦어질수록 출수일수가 단축되는 정도가 뚜렷하게 나타났다. 2. 북한 벼의 기본영양생장성은 조생종은 대조 품종인 오대벼보다 짧은 편이었고, 중생종은 오대벼에 비하여 긴 편이었다. 다. 감온성은 대비품종인 오대벼보다 대체로 예민한 것으로 나타났으며 극조생종인 올벼 2호는 감온성이 매우 예민한 반응을 보여 ′98년에는 42일, ′99년에는 39일이었다. The experiment was carried out to determine heading response of North Korean rice cultivars at NCES(national crop experiment station). RDA. South Korea in 1998 and 1999. This experiment was tested at three different locations which were Suwon, Cheolwon and Jinbu with 5 transplanting times with 10 cultivars. Duration of heading stage of the rice plant was shortened at lower altitude and in later transplanting date, respectively. Bases on the heading days, it was classified as the basic vegetative phase(BVP) and thermo-sensitive phase(TSP) in tested cultivars. BVPs and TSPs among treated varieties were grouped with four groups. BVPs of North Korean rice cultivars for early maturity were shoter than that of Odaebyeo of Korean rice cultivars, however, those for medium maturity were longer than that of the control cultivar, Odaebyeo. Meanwhile, TSPs of North Korean rice cultivars were more sensitive than that of the check cultivar of Odaebyeo, Correlation between 1998 and 1999 was more significant in basic vegetative phase than in thermo-sensitive stage. Therefore, the heading time of the rice plant primarily depended on the period of basic vegetative phase growth.

      • RNF20 Functions as a Transcriptional Coactivator for PPARγ by Promoting NCoR1 Degradation in Adipocytes

        <P>Adipose tissue is the key organ coordinating whole-body energy homeostasis. Although it has been reported that ring finger protein 20 (RNF20) regulates lipid metabolism in the liver and kidney, the roles of RNF20 in adipose tissue have not been explored. Here, we demonstrate that RNF20 promotes adipogenesis by potentiating the transcriptional activity of peroxisome proliferator–activated receptor-γ (PPARγ). Under normal chow diet feeding, <I>Rnf20</I> defective (<I>Rnf20</I><SUP><I>+/−</I></SUP>) mice exhibited reduced fat mass with smaller adipocytes compared with wild-type littermates. In addition, high-fat diet–fed <I>Rnf20</I><SUP><I>+/−</I></SUP> mice alleviated systemic insulin resistance accompanied by a reduced expansion of fat tissue. Quantitative proteomic analyses revealed significantly decreased levels of PPARγ target proteins in adipose tissue of <I>Rnf20</I><SUP><I>+/−</I></SUP> mice. Mechanistically, RNF20 promoted proteasomal degradation of nuclear corepressor 1 (NCoR1), which led to stimulation of the transcriptional activity of PPARγ. Collectively, these data suggest that RNF20-NCoR1 is a novel axis in adipocyte biology through fine-tuning the transcriptional activity of PPARγ.</P>

      • Image Sticking Property in the In-Plane Switching Liquid Crystal Display by Residual DC Voltage Measurements

        The residual DC phenomena in the in-plane switching(IPS)-liquid crystal display(LCD) by the voltage-transmittance (V-T) and capacitance-voltage (C-V) hysteresis method on rubbed polyimide (PI) surfaces were studied. We found that the residual DC voltage in the IPS-LCD was decreasing with the increasing concentration of cyano LCs. The residual DC voltage of the IPS-LCD can be improved by the high polarity of cyano LCs.

        28-Day Oral Toxicity of Cadmium Selenide in Sprague-Dawley Rats

        This study was performed to evaluate the toxicity of cadmium selenide for a period of 28 days in Sprague-Dawley rats. Each of 10 healthy male and females rats per group received daily oral administration for 28-day period at dosage levels 30, 300 and 1,000 ㎎/㎏ of body weight. Mortality and clinical signs were checked, and body weight, water intake and food consumption were also recorded weekly. There were no dose-related changes in food consumption or urine volume. All animals survived to the end of study with no clinical signs or differences in body weight gain observed when compared with the control group. At the end of study, all animals including control group, were subjected to necropsy. Blood samples were collected for hematology tests including coagulation time and biochemistry analysis. Blood coagulation time and relative organ weight were unaffected by all received doses. White Blood Cell (WBC) counts significantly increased in the 300 ㎎/㎏ administered male animal group when compared to the control. Monocyte (MO) value were also increased significantly in both 300 and 1,000 ㎎/㎏ male animal group. However, Mean Corpuscular Volume (MCV) were significantly decreased compared with the control in the 1,000 ㎎/㎏ dose groups for male and female animals. Mean Corpuscular Hemoglobin (MCH) decreased significantly for female in the 300 and 1,000 ㎎/㎏ group compared to the control. Blood biochemical values of Inorganic phosphorus (IP) were significantly increased in both the 300 and 1,000 ㎎/㎏ dose groups in male animals when compared to the control. Creatinine (CRE) levels indicated significant increase in kidney function for the female, 30 ㎎/㎏ dose group when compared with control. There was a significant decrease in thymus absolute organ weight in the female, 1,000 ㎎/㎏ dose group when compared with control. Histopathological findings revealed no evidence of injury related to cadmium selenide except for one case of focal hepatic inflammation in the high dose (1,000 ㎎/㎏) group. One case of lung inflammation was also seen in the control group. Basis on these result, the No Observable Adverse Effect Level (NOAEL) of cadmium selenide was determined to be more than 1,000 mg/kg/day for male and female rats under conditions in this study.

      • Subgap Density-of-States-Based Amorphous Oxide Thin Film Transistor Simulator (DeAOTS)

        <P>The amorphous oxide thin-film transistor (TFT)-oriented simulator [subgap Density of states (DOS)-based Amorphous Oxide TFT Simulator (DeAOTS)] is proposed, implemented, and demonstrated for amorphous indium-gallium-zinc-oxide (a-IGZO) TFTs. It only consists of parameters having their physical meanings and is supplied with concrete techniques for parameter extraction. Among the physical parameters, the acceptor-like DOS <I>gA</I>(<I>E</I>) was experimentally extracted using the multifrequency <I>C</I>-<I>V</I> technique, whereas the donor-like DOS <I>gD</I>(<I>E</I>) and the doping concentration <I>ND</I> were extracted using numerical iterations. The simulation result reproduces the DOS and thin-film-thickness-dependence of dc <I>I</I>-<I>V</I> characteristics very well. Compared with the previously reported a-Si TFT models, the proposed DeAOTS model not only reflects the strong <I>V</I><SUB>GS</SUB> dependence of the effective mobility (μ<SUB>eff</SUB>) but also clarifies the relations between process-controlled DOS parameters and dc <I>I</I>- <I>V</I> characteristics based on experimentally extracted DOS parameters. Also, it sufficiently takes into account the peculiar situation of amorphous oxide TFTs where the free-carrier charge can be larger than the localized one out of the total induced charge. Moreover, it reproduces the measured electrical characteristics within the wide range of <I>V</I><SUB>GS</SUB>/<I>V</I><SUB>DS</SUB> with a single equation, not distinguishing the operation regions such as the subthreshold, linear, and saturation regimes.</P>
