        To construct expression vector for the human HPRT gene, pRSVneo plasmid conferring resistance to neomycin-kanamycin(Tn5) antibiotics and pHPT 31 containing human HPRT cDNA were used to subclone human HPRT cDNA. For construction of recombinant pRSVneo carrying human HPRT, pRSVneo-HPRT, the human HPRT cDNA fragment from pHPT31 plasmid was inserted into polyadenylation site (BamHI site) behind the neo gene fragment of pRSVneo plasmid. For the other expression vector, pRSV-HPRT, the neo gene and small portion of the untranslated 5′franking region were removed from the pRSVneo-HPRT. The HPRT gene of the two vectors were constructed in right orientation. Each recombinant vector was introduced into cultured HPRT-deficient mouse cell line, Sp2/0 by calcium mediated DNA transfection and the transfected cells were selected under HAT selection condition. The HPRT activity of the lysates from the selected cell was higher than that of the lysates from spleen cells of mouse. The HPRT activity of pRSVneo-HPRT vector was higher than that of pRSV-HPRT one. To find out whether the HPRT activity of the transfected cells selected under HAT medium was expressed by transfected vectors or by spontaneous mutations, it was cultured to reselect in G418 medium. It showed that the nature of the HPRT activity was resulted from the transfected vectors.

      • 동절기 신관리 석실고분 내부의 온·습도 분포특성

        개방된 고분의 장기보존은 조상의 얼이 담긴 문화재를 길이 후손에 물려주어야 하는 우리들의 시대적 사명이라 할 수 있다. 그런데 고분 내부의 공기조화를 필요로 하는 경우 설비측면에서 고분내부의 공기조화 문제를 검토한 사례는 그 동안 많지 않았다. 특히 고분 내부환경에 관한 온·습도 자료는 매우 제한적으로 알려져 있는 상태이다. 본 연구는 고분의 공기조화와 관련하여 부하산정 등의 기초자료로 활용하기 위한 고분내부의 온·습도자료를 원격측정 기술을 적용하여 획득한 결과를 소개하며, 이를 통하여 고분내부의 실내환경 조건에 대한 일반의 이해를 돕고, 더 나아가 공조기 설치에 필요한 기초자료를 제공하고자 수행하였다. 측정 연구대상은 충남 공주시 신관동에 소재한 신관리석실고분(충청남도 기념물 제 7호)으로서 백제후기의 횡혈식 석실고분으로서 현실의 크기는 폭 1.25m, 깊이 2.73m 그리고 높이 1.50m이며, 현실입구는 개방된 상태에서 실내 5채널 및 실외공기에 대하여 각각 온도와 습도를 측정하였으며, 본 본문에서는 동절기(12월부터 다음 해 3월까지)의 자료를 소개한다. In recent years the importance of the preservation of cultural artefacts like ancient tombs has been widely accepted domestically and internationally as the quality of life improves. However not much attention has been paid for the facilities and systems to preserve those artefacts. Even the general understanding of the preservatory environment of the underground space as tombs is poor. In the present study, the temperature and relative humidity inside a selected artefact, Shinkwan-ri tomb, are monitored to improve the understanding of the indoor thermal environment, which may possibly be used when one consider air-conditioning for preservation. Total of 12 channel data are collected by using remote data acquisition system based on the cdma(Code Division Multiple Access) technology. Three-month-data representing winter season is reported with discussions.

      • 벗나무속 수종간의 핵 리보솜 DNA의 ITS Ⅰ 염기서열 비교

        Comparison of four species (Prunus yedoensis, P. sargentii, P. pendula for. ascendens, P. serrulata var. quelpaertensis) and one cultivar of P. yedoensis were carried out by comparing their nucleotide sequences from the internal transcribed spacers (ITS) I of nuclear ribosomal DNA (rDNA). The length of the ITS I region was ranged from 229 to 236 base pairs (bp). The G+C contents of ITS I ranged from 58.5 to 60.2%. The value of nucleotide divergence across for four Prunus species and one cultivar ranged from 0.4 to 6%. The pairwise distance between P. pendula for. ascendens and P. yedoensis was 0.004 showing the lowest value among any other pairs. On the other hand, the pairwise distance betwen P. yedoensis and cultivated P. yedoensis was 0.026. The nucleotide analysis based on ITS I sequence was considerably similar to previous studies on the basis of random amplified polymorphic DNA (RAPD).

      • 고분보존용 항온ㆍ항습 System의 동절기 운전특성

        본 연구는 여러 가지 문화재 보존영역 중 개방된 고분의 장기보존을 위한 최적 공기 조화 시스템의 개발을 목표로 하여 수행되었다. 고분의 장기보존을 위해서는 고분 내 공기조성, 온ㆍ습도 및 기류환경을 모니터하여 고분내 자연 실내환경의 기초자료를 확보한 수 고분의 주 구성요소인 석재 및 전재의 보조측면에서 문제점을 검토하여 최적 유지환경조건을 정의하고, 이상의 요구조건을 실현할 수 있는 공조시스템의 적용이 필요하다. 본 논문은 이 중 모형고분에 적용된 공조시스템의 동계운전성능에 대하여 소개한다.

      • 제주도 지하수의 전기비전도도와 염소이온의 지역별 변화 양상

        The specific electrical conductivity and the chloride contents were monitored at 98 sits in Cheju Island during two years (1994.2∼1995.11) to the purpose of characterized to regional variations of its elements. The areas of east and west were characterized by both higher elements contain than south and north area. Chloride and specific electrical conductivity values from Kimyoung to Shinhung areas showed more than 100㎎/ℓand 400 μmohs/㎝, respectively. Also west areas relatively high values of both elements were derived from agricultural practice and feeding cattle. However, the difference of difference of these areas indicated that the groundwater environments were different.

      • Various Digital Memory Behaviors of Functional Aromatic Polyimides Based on Electron Donor and Acceptor Substituted Triphenylamines

        <P>A series of aromatic polyimides (PIs) were synthesized via the polymerization of 3,3′,4,4′-diphenylsulfonyltetracarboxylic dianhydride with 4,4′-diaminotriphenylamine derivatives containing hydrogen, cyano, methoxy, or dimethylamine substituents. These PIs were thermally and dimensionally stable and produced high-quality thin films when applied in a conventional spin-coating process. Their structure and properties were characterized. Nanoscale thin films of the PIs demonstrated excellent electrical memory performance, with high stabilities and ON/OFF current ratios. The memory characteristics were found to be tunable by varying the substituents; nonvolatile write-once–read-many-times memory behavior, nonvolatile ON/OFF switching type memory behavior, and volatile dynamic random access memory behavior were observed. The memory characteristics were substantially influenced by the electron-accepting cyano- and electron-donating dimethylamine substituents but were apparently not affected by the electron-donating methoxy substituent. In addition, the film density was a significant factor influencing the observed memory behaviors, with larger film densities causing lower OFF-current levels. However, the critical switching-on voltage varied very little as the substituents were changed and was measured to be approximately ±2 V. All of the memory behaviors were found to be governed by a mechanism involving trap-limited space-charge-limited conduction and local filament formation. Overall, all of the PIs assessed in the present work were found to be suitable active materials for the low-cost mass production of high-performance, programmable unipolar memory devices that can be operated with very low power consumption, high ON/OFF current ratios, and high thermal and dimensional stability.</P><P><B>Graphic Abstract</B> <IMG SRC='http://pubs.acs.org/appl/literatum/publisher/achs/journals/content/mamobx/2012/mamobx.2012.45.issue-9/ma300311d/production/images/medium/ma-2012-00311d_0004.gif'></P>

      • Digital Memory Versatility of Fully π-Conjugated Donor–Acceptor Hybrid Polymers

        <P>The fully π-conjugated donor–acceptor hybrid polymers Fl-TPA, Fl-TPA-TCNE, and Fl-TPA-TCNQ, which are composed of fluorene (Fl), triphenylamine (TPA), dimethylphenylamine, alkyne, alkyne-tetracyanoethylene (TCNE) adduct, and alkyne-7,7,8,8-tetracyanoquinodimethane (TCNQ) adduct, were synthesized. These polymers are completely amorphous in the solid film state and thermally stable up to 291–409 °C. Their molecular orbital levels and band gaps vary with their compositions. The TCNE and TCNQ units, despite their electron-acceptor characteristics, were found to enhance the π-conjugation lengths of Fl-TPA-TCNE and Fl-TPA-TCNQ (i.e., to produce red shifts in their absorption spectra and significant reductions in their band gaps). These changes are reflected in the electrical digital memory behavior of the polymers. Moreover, the TCNE and TCNQ units were found to diversify the digital memory modes and to widen the active polymer layer thickness window. In devices with aluminum top and bottom electrodes, the Fl-TPA polymer exhibits stable unipolar permanent memory behavior with high reliability. The Fl-TPA-TCNE and Fl-TPA-TCNQ devices exhibit stable unipolar permanent memory behavior as well as dynamic random access memory behavior with excellent reliability. These polymer devices were found to operate by either hole injection or hole injection along with electron injection, depending on the polymer composition. Overall, this study demonstrated that the incorporation of π-conjugated cyano moieties, which control both the π-conjugation length and electron-accepting power, is a sound approach for the design and synthesis of high-performance digital memory polymers. The TCNE and TCNQ polymers synthesized in this study are highly suitable active materials for the low-cost mass production of high-performance, polarity-free, programmable, volatile, and permanent memory devices that can be operated with very low power consumption, high ON/OFF current ratios, and high reliability.</P><P><B>Graphic Abstract</B> <IMG SRC='http://pubs.acs.org/appl/literatum/publisher/achs/journals/content/aamick/2014/aamick.2014.6.issue-11/am5013134/production/images/medium/am-2014-013134_0012.gif'></P>

      • New Fullerene-Based Polymers and Their Electrical Memory Characteristics

        <P>Covalent incorporations into polymers of fullerene were achieved via the Cu(I)-catalyzed azide–alkyne click polymerizations of a fullerene derivative monomer functionalized with 5-(trimethylsilyl)pent-4-yn-1-yl groups and a comonomer functionalized with azidomethyl groups, producing the novel fullerene polymers P1-<I>C</I><SUB>60</SUB> and P2-<I>C</I><SUB>60</SUB>. Despite their extremely high fullerene loading levels, the polymers were soluble in common organic solvents and exhibited no aggregation of fullerene units in films. Moreover, devices containing these fullerene polymers were easily fabricated with common coating processes that exhibit excellent unipolar and bipolar flash memory characteristics as well as unipolar permanent memory characteristics, with high ON/OFF current ratios, long retention times, and low power consumption. These electrical switching behaviors were favorably operated by electron injection.
