        Genome-wide investigation of a Korean synthetic breed, Woori-Heukdon using the Illumina PorcineSNP60K BeadChip

        Background: Woori-Heukdon (KWH) is a Korean synthetic pig breed generated using Chookjin-Duroc (KCD), Chookjin-Chamdon (KCC), and their crossbreds. Currently, there is a severe lack of studies investigating the Korean breed populations including wild boars (KWB) throughout the genome. Objective: This study was performed to investigate the genetic characteristics of Korean pig populations at the genome-wide level. Methods: Using the SNP dataset derived from genotyped and downloaded datasets using the Illumina PorcineSNP60K BeadChip, we compared the genomes of 532 individuals derived from 23 pig breeds to assess the genetic diversity, inbreeding coefficient, genetic differentiation, and population structure. Results: KWB showed the lowest average expected heterozygosity (HE = 0.1904), while KWH showed the highest genetic diversity (HE = 0.02859) among Korean populations. We verified that the genetic composition of KWH, showing USD of 74.8% and KCC of 25.2% in ADMIXTURE analysis. In population structure analyses, KCC was consistently shown to be separated from other pig populations. In addition, we observed gene flow from Western pigs to a part of Chinese populations. Conclusion: This study showed that Korean native pigs, KCC have genetic differences in comparison with Chinese and Western pigs; despite some historical records and recent genetic studies, we could not find any clear evidence that KCC was significantly influenced by Chinese or Western breeds in this study. We also verified the theoretical genomic composition of KWH at the molecular level in structure analyses. To our knowledge, this is the first genomic study to investigate the genomic characteristics of KWH and KCC.

        N/A Most authorities agree the higher incidence of gallstones among diabetic patient. Although many reports on gallstones or diabetes mellitus have been published, and actual correlation between gallstones and diabetes mellitus remains obscure. The purpose of this reports is to know the incidence of gallstone in diabetics and of diabetics in the patients with gallstone. Authors studied prospectively and clinically the diabetics in a group of 173 patients with gallstones and gallstone in a group of 98 diabetic patients who were admitted to Yonsei University Yong-Dong Hospital for the same period of April 1983 to May 1984. The results obtained are summarized as follow: The incidence of gallstone to general admission patients is 1.5% and the prevalence of diabetes mellitus to total in-patients is 0.8% The gallstone was found in 11.0% of diabetics and were diabetic 19.4% of patients with gallstones. The age of peak incidence of gallstones is 5th decade and female was more prevalent than male with the ratio of 1.37:1. The age of peak incidence of diabetes mellitus is 6 th decade and female is less prevalent with the ratio of 0.72:l.3, The most common characteristic finding of patients of gallstones with diabetes mellitus is symptomless(42.1%)4, There is no significant difference in serum levels of cholesterol and triglyceride between diabetic and non-diabetic groups or with and without gallstones. 5. Comparative study on location of stones revealed, 72.8% in gallbladder only, 12.7% in common bile duct only, 4. 6% in intrahepatic duct only, and remainders in multiple site. There is no significant difference in terms of location of stone between diabetic and nondiabetic group.

      • Construction of recombinant Autographa californica nucleopolyhedrovirus containing genomic segments of Spodoptera litura granulovirus

        Recently, the genome of Spodoptera litura granulovirus (SlGV) which encodes 133 putative open reading frames (ORFs) was completely sequenced. In this study, to screen novel insecticidal genes of SlGV, we first constructed an advanced plasmid capture system, pPCS-TPI, which contains not only pUC19 ori and ampicillin resistance gene but also Autographa californica nucleopolyhedrovirus (AcMNPV) ORF603 and ORF1629 homologous region between Tn7L and Tn7R. In order to introduce genomic segments of SlGV into the genome of AcMNPV, genomic DNA of SlGV was digested with EcoRI and self-ligated. These self-ligated segments were in vitro transposed with the pPCS-TPI donor by the help of TnsABC* transposase. By this, 10 EcoRI-digested genomic segments of the SlGV were cloned, and these clones were co-transfected with the bApGOZA DNA into sf9 cells to generate corresponding recombinant virus, respectively. The resulting recombinant viruses harboring genomic segments of the SlGV could be used to investigate the insecticidal activity and/or other functions originated from the introduced genomic segments of the SlGV.

      • Personality consistency analysis in cloned quarantine dog candidates

        <P>In recent research, personality consistency has become an important characteristic. Diverse traits and human-animal interactions, in particular, are studied in the field of personality consistency in dogs. Here, we investigated the consistency of dominant behaviours in cloned and control groups followed by the modified Puppy Aptitude Test, which consists of ten subtests to ascertain the influence of genetic identity. In this test, puppies are exposed to stranger, restraint, prey-like object, noise, startling object, etc. Six cloned and four control puppies participated and the consistency of responses at ages 7-10 and 16 weeks in the two groups was compared. The two groups showed different consistencies in the subtests. While the average scores of the cloned group were consistent (<I>P</I> = 0.7991), those of the control group were not (<I>P</I> = 0.0089). Scores of Pack Drive and Fight or Flight Drive were consistent in the cloned group, however, those of the control group were not. Scores of Prey Drive were not consistent in either the cloned or the control group. Therefore, it is suggested that consistency of dominant behaviour is affected by genetic identity and some behaviours can be influenced more than others. Our results suggest that cloned dogs could show more consistent traits than non-cloned. This study implies that personality consistency could be one of the ways to analyse traits of puppies.</P>

      • Chrysin attenuates atopic dermatitis by suppressing inflammation of keratinocytes

        <P><B>Abstract</B></P> <P>We previously reported the inhibitory effect of chrysin, a natural flavonoid plentifully contained in propolis, vegetables and fruits, on the mast cell-mediated allergic reaction. In this study, we evaluated the effect of chrysin on atopic dermatitis (AD) and defined underlying mechanisms of action. We used an AD model in BALB/c mice by the repeated local exposure of 2,4-dinitrochlorobenzene (DNCB) and house dust mite (<I>Dermatophagoides farinae</I> extract, DFE) to the ears. Repeated alternative treatment of DNCB/DFE caused AD-like skin lesions. Oral administration of chrysin diminished AD symptoms such as ear thickness and histopathological analysis, in addition to serum IgE and IgG2a levels. Chrysin decreased infiltration of mast cells, and reduced serum histamine level. Chrysin also suppressed AD by inhibiting the inflammatory responses of Th1, Th2, and Th17 cells in mouse lymph node and ear. Interestingly, chrysin significantly inhibited the production of cytokines, Th2 chemokines, CCL17 and CCL22 by the down-regulation of p38 MAPK, NF-κB, and STAT1 in tumor necrosis factor (TNF)-α/interferon (IFN)-γ-stimulated human keratinocytes (HaCaT). Chrysin also inhibited TNF-α/IFN-γ-stimulated IL-33 expression in HaCaT cells and mouse primary keratinocytes. Taken together, the results indicate that chrysin suppressed AD symptoms, suggesting that chrysin might be a candidate for the treatment of AD and skin allergic diseases.</P> <P><B>Highlights</B></P> <P> <UL> <LI> Chrysin suppresses AD symptoms by inhibiting T helper cell immune reaction. </LI> <LI> Chrysin downregulates CCL17 and CCL22 expression via p38 MAPK, STAT1, and NF-κB in human keratinocytes. </LI> <LI> Chrysin and tacrolimus have overlapping effects in inhibiting AD immune responses. </LI> <LI> Chrysin might be a candidate for the treatment of skin allergic diseases. </LI> </UL> </P>

        지역공동체를 위한 수문/수질 평가 의사결정지원시스템

        의사결장지원시스템은 다양한 분야에 적용되어 왔으며, 그 중 수자원 및 수질 관련 분야에도 다각적으로 적용되어 왔다. 본 연구에서는 미 농무성의 자연자원보전국(NRCS, Natural Resources Conservation Service)에서 개발한 유출곡선법(Curve Number Method)과 EMC(Event Mean Concentration)을 사용한 L-THIA(Long-Term Hydrologic Impact Assessment) 수문/수질 모형을 강우자료 데이터베이스, 웹기반 지리정보시스템, 웹 사용자 편의 시스템과 통합한 수문/수질 L-THIA web 의사결정지원시스템을 개발하였다. L-THIA web은 도시계획가나 지방자치단체, 또는 지방의 공동체가 사용할 수 있도록 쉽고 단순한 사용자 편의 시스템을 제공하고 있으며, 미국의 50개 본토의 주와 카운티(County) 이름으로 기상자료와 수문토양분류(Hydrologic Soil Group)을 인터넷 지리정보시스템을 이용하여 제공하고 있다. 본 연구는 지방자치단체 및 지역공동체의 실무자를 사용자로 수문/수질 평가 및 관리를 위한 시스템으로 유용하게 활용될 수 있을 것으로 사료된다.

        Synthesis of a Novel Series of Imidazo[1,2-α]pyridines as Acyl-CoA: Cholesterol Acyltransferase (ACAT) Inhibitors

        A novel series of imidazo[1,2-$\alpha$]pyridines was designed, synthesized, and tested for their ability to inhibit acyl- CoA:cholesterol acyltransferase. Preliminary lead optimization efforts resulted in the identification of ACAT inhibitors represented by analogues 5b, 5c, 6a, 6c, 7b, and 7c. The ACAT inhibitory activity of these compounds was further established by potent inhibition of cholesteryl ester formation in HepG2 cells by a representative analogue 7b.

        Synchronous second primary cancers in patients with squamous esophageal cancer: clinical features and survival outcome

        Background/Aims: Unexpected diagnosis of synchronous second primary cancers (SPC) complicates physicians’ decision-making because clinical details of squamous esophageal cancer (EC) patients with SPC have been limited. We evaluated clinical features and treatment outcomes of patients with synchronous SPC detected during the initial staging of squamous EC. Methods: We identified a total of 317 consecutive patients diagnosed with squamous EC. Relevant clinical and cancer-specific information were reviewed retrospectively. Results: EC patients with synchronous SPC were identified in 21 patients (6.6%). There were significant differences in median age (70 years vs. 63 years, p = 0.01), serum albumin level (3.3 g/dL vs. 3.9 g/dL, p < 0.01) and body mass index (20.4 kg/m2 vs. 22.8 kg/m2, p = 0.01) between EC patients with and without SPC. Head and neck, lung and gastric cancers accounted for 18.2%, 22.7%, and 18.2% of SPC, respectively. Positron emission tomography-computed tomography (PET-CT) detected four cases (18.2%) of SPC that were missed on CT. Management plans were altered in 13 of 21 patients (61.9%) with detected SPC. Curative esophagectomy was attempted in 28.6% of EC patients with SPC (vs. 59.1% of patients without SPC; p = 0.006). EC patients with SPC had significantly lower 5-year survival than patients without SPC (10.6% vs. 36.7%, p = 0.008). Conclusions: Synchronous SPC were found in 6.6% of squamous EC patients, and PET-CT contributed substantially to the detection of synchronous SPC. EC patients with SPC had poor survival due to challenges of providing stage-appropriate treatment.

      • Use of Microsatellite Markers to Identify Commercial Melon (Cucumis melo L.) Varieties

        Microsatellite is one of the most suitable markers for variety identification as it has great discrimination power for varieties with narrow genetic variation. The relationship between marker genotypes and 58 melon varieties was analyzed. Of the 400 pairs of simple sequence repeat (SSR) primers screened against 11 melon varieties. 28 pairs showed highly polymorphism on the basis of PIC (Polymorphism Information Contents) value. These markers were applied to constructing DNA profile data base of 58 major melon varieties through multiplex PCR and fluorescence based automatic detection system. A total of 111 polymorphic amplified fragments were obtained by using 28 SSR markers. The average of PIC value was 0.643 ranging from 0.401 to 0.846. One hundred eleven SSR loci were used to calculate Jaccard’s distance coefficients for UPGMA cluster analysis. A clustering group of varieties, based on the results of SSR analysis, were categorized into plant shape and fruit type. Almost all of the varieties were discriminated by marker genotypes. This information may be used to select comparative variety through comparison of genetic relationship between existing variety and candidate varieties in distinctive tests and protection of plant breeders’ intellectual property rights through variety identification.
