      • THE EFFECT OF CATHETER-DIRECTED ETHANOL SCLEROTHERAPY ON OVARIAN RESERVE IN PATIENTS WITH RECURRENT ENDOMETRIOSIS: COMPARATIVE ANALYSIS WITH PRIMARY ENDOMETRIOSIS

        Objective: Although the surgical treatment remains gold standared in endometrioma, ovarian reserve is injured after the hemostasis and excision of the cyst. Catheter-directed ethanol sclerotherpy is known to affect the ovarian function less, increase efficacy, and reduce complications. This study aims to investigate the changes in ovarian reserve after catheter-directed ethanol sclerotherpy in recurrent endometrioma. Methods: Electronic medical records and pictures in patients with endometrioma who underwent catheter-directed ethanol sclerotherpy were obtainted in single institution from August 2014 to April 2022. Patient with aged >18 years old and AMH level >1.0 and <5.0 were enrolled in the study and age, BMI, cyst diameter, unilaterality, number of loculation and AMH level and CA-125 level before and after 1month, 6month, 1year and 2year of sclerotherapy were obtained. Results: 139 patients were enrolled in the study under IRB. There was significant differnece in AMH level before sclerotherapy in primary and recurrent endometrioma(2.68 vs. 2.04, p-value=0.011). However, there was no statistically significant difference in delta value of AMH after sclerotherapy in primary and recurrent endometrioma in 1month, 6month, 1year and 2year after sclerotherpy(91.78% vs. 96.14%, 74.75% vs. 81.58%, 83.98% vs. 65.79%, 97.08% vs. 52.52%, with p-value=0.593, 0.695, 0.217, 0.069, respectively). Also, this results were consistent regardless of unilaterality and loculation and CA-125 level showed no differerence between primary and recurrent endometrioma on every 1month, 6month, 1year and 2year after sclerotherpy. Conclusion: The effect of Catheter-directed ethanol sclerotherpy on ovarian reserve is not inferior in recurrent endometrioma compared to primary endometrima. Since sclerotherapy is known to less deteriorate the ovarian function than surgical removal of endometrioma, clinician could consider this as the first-line therapy in patient with recurrent endometrioma.

        Multidrug and Toxic Compound Extrusion Protein-1 (MATE1/SLC47A1) Is a Novel Flavonoid Transporter

        <P>Dietary flavonoids have various biological functions. However, their cellular transport mechanisms are largely unknown. We have determined that the multidrug and toxic compound extrusion transporter-1 (MATE1) is a membrane transporter for flavonoids and has a high affinity for quercetin. HEK293T cells overexpressing MATE1 exhibited increased intracellular quercetin accumulation. This effect disappeared in the presence of a MATE1 inhibitor and after MATE1 gene knockdown. HepG2 cells expressed MATE1 significantly, with the uptake quercetin of which was dramatically reduced with MATE1 inhibition. On the basis of immunofluorescence analysis, MATE1 was highly expressed in peroxisomes and the endoplasmic reticulum (ER) as well as in plasma membranes in the liver and intestine, which suggests potential accumulation of quercetin in peroxisomes and the ER in these tissues. Fluorescent microscopic analysis confirmed selective accumulation of qurcetin in peroxisome. The effects of quercetin on cellular lipid reduction and glucose uptake were exaggerated with MATE1 overexpression. In conclusion, MATE1 is a membrane transporter for quercetin; its overexpression enhances the hypolipidemic activity of quercetin and cellular glucose transport.         A Study on the Proposal of the Behavioral Principles for the Training of Korea Coast Guard in the Pandemic Era10.22471/disaster.2022.7.2.52

        Purpose: This study proposes the Korea Coast Guard trainee behavior principle based on the value of the Korea Coast Guard in response to the Pandemic era. Through the proposed principle of action, we will provide philo-sophical standards for value judgment to members of the organization in various dilemma situations that Korea Coast Guard will face in the future. Recently, as a large number of young employees in various fields have been recruited in the Korea Coast Guard organizations, controversy has arisen due to the difference between the ex-isting policies and the expectations of the younger generation. This means that the organization should make more efforts to establish and share the priority of organizational value to its members. Method: The Korea Coast Guard organizations have unique cultural characteristics that have been formed with unique history. This characteristic of the members is to examine the result of the formation of philosophical value of the Korea Coast Guard organization, along with the value policy of the Korea Coast Guard focusing on the educational regulations applied to the current students by the Korea Coast Guard Education Academy. As a research method of this paper, I would like to examine the behavior style required by members according to the unique characteristics of the Korea Coast Guard organizations in phenomenological aspects. The media press releases were used to maintain objective grounds among various problems caused by the characteristics of the Korea Coast Guard, and the internal voice of the organization was referred to the internal bulletin board of the Korea Coast Guard Agency. Results: As a conclusion of this study, based on the review of the philosophy and educational goals of the Korea Coast Guard organization. The principle of action is proposed 'Five Promises of Korea Coast Guard Stu-dents'. The proposed principle of student behavior is organized so that it can be applied to actual education center life from the perspective of the student. The first regulation of the prize and penalty for the students can be seen as a behavior that is supported and trusted by the people through 'honorable life' that helps others. Second, it emphasizes that marine police will move toward a steady learning and effort as a guardian of the sea, along with the educational goal of 'trying to become an expert in the sea every moment' and prioritizing the expertise and practical skills. Third, the Coast Guard respect the human rights of the people who always keep their promises and do not follow injustice. Fourth, considering the spirit of public service value for the nation and the people that the result suitable for the public interest is more precious, overcoming the individual desire and discerning what is more suitable for the public interest is the value to be cultivated as a public official who serves the people afterward, and above all, it can be wary of the situation that falls into the individualistic tendency, forgetting that he is a 'serviceman of the people'. Fifth, the direction of the challenge spirit shown in the 'expert in the ocean' that 'I am willing to challenge and overcome for a better future' and the future-oriented image that pursues the world's best pursued by educational goals can be confirmed. Conclusion: As such, this research suggests that the fundamental cause for this dilemmatic situation is on the absence of the values based on the organizational philosophies, and it is necessary for the organization to set up the operational methods based on its philosophical values taking into account the principles of action which are appropriate to the task of KCG(Korea Coast Guard). Also, this research asserts that the operation based on philosophical grounds contributes to the sequential positivity such as ‘psychological stability of the members’ and ‘creating hard working environments’ this will also help its members’ learning process and enhance the orga

        Purpose : Smoking adversely affects the health of smokers and passive smoking threatens the health of people around smokers. The purpose of this review was to summarize the effect of smoking on sleep. Methods : We reviewed 430 articles on smoking and the effect of medication for smoking on sleep. These 430 articles consisted of 78 articles on the physiological effects of nicotine, 20 articles on the effects of nicotine on sleep, 56 articles on smoking related diseases provoking a sleep disorder, 183 articles on other substances (except nicotine) provoking a sleep disorder, and 93 articles on the effects of stopping smoking and medication for stopping smoking on sleep. Results : We summarized the physiological effects of nicotine on sleep, the effects of other substances such as acetaldehyde on sleep, smoking related illnesses including obstructive sleep apnea, chronic obstructive pulmonary disease, chronic rhinosinusitis, cardiovascular and cerebrovascular disease, depression, sleep bruxism, and restless legs syndrome, and the effects of smoking cessation on sleep. Conclusion : Research on the effect of smoking on sleep is necessary to understand the mechanism of smoking. More research on smoking, sleep, and brain mechanisms will give us a closer insight into the relationship between smoking and sleep.

        Association between Fractures and Low Muscle Mass in Korean Menopausal Women: Data from Korean National Health and Nutrition Survey (2010–2011)

        Objectives: This study used the Korean National Health and Nutrition Examination Survey (KNHANES) to determine the association between fractures and low muscle mass.Methods: This cross-sectional study used the 2010–2011 KNHANES data. Low muscle mass was defined as (appendicular skeletal muscle mass [kg]/Height2 [m2]) < 5.45 kg/m2, which is < 2 SD below the sex-specific mean of a young reference group. Patients with T-scores between –1.0 and –2.5 indicated osteopenia, whereas those with T-scores lower than –2.5 indicated osteoporosis.Results: Out of 1,306 women enrolled in the study, 330 were diagnosed with low muscle mass according to the abovementioned diagnostic criterion. The prevalence of fractures at various sites was significantly higher in postmenopausal women with low muscle mass than in those without low muscle mass (relative risk [RR], 1.64; odds ratio [OR], 1.62; 95% confidence interval [CI], 1.06–2.48; P = 0.027). Furthermore, the prevalence of fractures was increased by the presence of osteopenia or osteoporosis in addition to low muscle mass (RR, 1.59; OR, 1.60; 95% CI, 1.02–2.49; P = 0.039) and by osteoporosis only (RR, 2.12; OR, 2.29; 95% CI, 1.11–4.70; P = 0.025).Conclusions: Fracture was more prevalent in postmenopausal women with low muscle mass than in those without low muscle mass. This finding is consistent in a subgroup analysis that included women who had osteoporosis or osteopenia. Moreover, the risk of fractures increased as low muscle mass worsened.

        Selective cytotoxicity of a novel mitochondrial complex I inhibitor, YK-135, against EMT-subtype gastric cancer cell lines due to impaired glycolytic capacity

        Epithelial-to-mesenchymal transition (EMT)-subtype gastric cancershave the worst prognosis due to their higher recurrence rate,higher probability of developing metastases and higher chemoresistancecompared to those of other molecular subtypes. Pharmacologicallyactionable somatic mutations are rarely found inEMT-subtype gastric cancers, limiting the utility of targeted therapies. Here, we conducted a high-throughput chemical screenusing 37 gastric cancer cell lines and 48,467 synthetic smallmoleculecompounds. We identified YK-135, a small-moleculecompound that showed higher cytotoxicity toward EMT-subtypegastric cancer cell lines than toward non-EMT-subtype gastriccancer cell lines. YK-135 exerts its cytotoxic effects by inhibitingmitochondrial complex I activity and inducing AMP-activatedprotein kinase (AMPK)-mediated apoptosis. We found that thelower glycolytic capacity of the EMT-subtype gastric cancer cellsconfers synthetic lethality to the inhibition of mitochondrialcomplex I, possibly by failing to maintain energy homeostasis. Other well-known mitochondrial complex I inhibitors (e.g., rotenoneand phenformin) mimic the efficacy of YK-135, supportingour results. These findings highlight mitochondrial complex Iinhibitors as promising therapeutic agents for EMT-subtype gastriccancers and YK-135 as a novel chemical scaffold for further drugdevelopment.

        SNAREs support atlastin-mediated homotypic ER fusion in <i>Saccharomyces cerevisiae</i>

        <P>Dynamin-like GTPases of the atlastin family are thought to mediate homotypic endoplasmic reticulum (ER) membrane fusion; however, the underlying mechanism remains largely unclear. Here, we developed a simple and quantitative in vitro assay using isolated yeast microsomes for measuring yeast atlastin Sey1p-dependent ER fusion. Using this assay, we found that the ER SNAREs Sec22p and Sec20p were required for Sey1p-mediated ER fusion. Consistently, ER fusion was significantly reduced by inhibition of Sec18p and Sec17p, which regulate SNARE-mediated membrane fusion. The involvement of SNAREs in Sey1p-dependent ER fusion was further supported by the physical interaction of Sey1p with Sec22p and Ufe1p, another ER SNARE. Furthermore, our estimation of the concentration of Sey1p on isolated microsomes, together with the lack of fusion between Sey1p proteoliposomes even with a 25-fold excess of the physiological concentration of Sey1p, suggests that Sey1p requires additional factors to support ER fusion in vivo. Collectively, our data strongly suggest that SNARE-mediated membrane fusion is involved in atlastin-initiated homotypic ER fusion.</P>

      • Catalytic Pyrolysis of Cork Oak Over Supercritical Hydrothermal Synthesized Nanosized ZrO<sub>2</sub>

        <P>The change in acidity of pyrolysis bio-oil was investigated using the catalytic pyrolysis of cork oak over zirconia synthesized by supercritical hydrothermal synthesis using a fixed bed reactor. Catalytic pyrolysis over zirconia reduced the content of acetic acid considerably and showed high selectivity to ketones and high-value-added aromatic compounds.</P>
