      • Enzymatic Extract from Mandarin Peel Inhibits Adipogenesis through Down-regulating of PPARγ and C/EBP α in 3T3-L1 adipocytes

        Mandarin peel (MP) is a by-product occurred during citrus juice or other products processing. The purpose of the current study was to investigate the potential anti-obesity activity of an enzymatic extract of MP in inhibiting adipogenesis in 3T3-L1 adipocytes. The enzymatic extract (MPCE) was prepared using the commercial food grade carbohydrase, Celluclast. Levels of lipid accumulation and triglyceride were significantly lower in MPCE treated cells than those in untreated cells. In addition, MPCE treatment reduced protein expression levels of peroxisome proliferator-activated receptor-γ (PPARγ), CCAAT/enhancer-binding proteins α (C/EBPα), sterol regulatory element-binding protein 1 (SREBP 1), and fatty acid binding protein 4 (FABP 4). These results suggest that MPCE possesses inhibitory effect on adipogenesis through down-regulating expression levels of adipogenesis-related proteins. Therefore, current findings demonstrate that MPCE possesses potent anti-obesity properties and could be a potential ingredient used in functional food industries.

        감귤피 효소적 추출물의 지방세포에서의 항비만 효과

        본 연구는 감귤 가공공정 중에 발생하는 가공부산물인 감귤피의 산업적 이용성 및 유용성을 증대시키고자 효소적 추출법을 적용하여 추출물을 제조하고 항비만 활성을 측정하였다. 감귤피 효소적 추출물(MPCE)의 폴리페놀 함량은 이전의 연구에서 보고한 유기용매 추출물보다 높은 함량을 나타내었다. MPCE의 항비만활성을 알아보기 위해 지방세포 분화억제에 미치는 영향을 확인한 결과 MPCE는 농도 의존적으로 지방세포의 분화 및 지방축적을 감소시켰으며 이는 세포독성에 의한 것이 아님을 세포생존율 측정을 통해 확인하였다. 또한 MPCE는 adipogenesis 관련 전사인자인 C/EBPα, SREBP-1, PPARγ, FABP4의 단백질 발현을 감소시켰다. 이러한 결과들을 통해 MPCE는 adipogenesis 관련 전사인자의 단백질 발현 억제하여 지방세포의 분화 및 지방축적을 억제하였으며, 이를 통해 항비만 효과를 나타낸다는 것을 확인하였다. 따라서 MPCE는 비만 예방 효능을 가진 기능성 식품 소재로서의 활용 가치가 있는 것으로 판단되며, 보다 명확한 항비만효능을 확인하기 위해 추후 동물 실험 등에 대한 연구가 필요할 것으로 생각된다. Mandarin peel (MP) is a by-product of the processing of citrus juice or other products. This study aimed to investigate the potential anti-obesity effect of an enzymatic extract of MP on the inhibition of adipogenesis in 3T3-L1 adipocytes.         Psychological Aspects of Child Maltreatment

        In this review, we provide information about the etiology, risk factors, and clinical presentations of maltreatment to help clinicians better understand child abuse and neglect. Child maltreatment is a major global health challenge that can result in severe consequences. Abused and neglected children are likely to develop psychiatric disorders, such as major depressive disorder, anxiety disorder, and posttraumatic stress disorder. Understanding child maltreatment is expected to prevent and reduce victimization in children, adolescents, and their families.

        호텔종사원의 감정노동이 직무만족과 서비스 사보타주에 미치는 영향 : 개인직무적합성의 조절효과를 중심으로

        본 연구는 호텔 종사원의 감정노동이 직무만족과 부정적 일탈 행동인 서비스 사보타주에 미치는 영향관계를 밝히고자 하였으며, 개인직무적합성의 조절효과를 규명하여 호텔 종사원의 감정관리와 경영자의 효과적인 인적자원관리 방안을 제시하고자 하였다. 연구를 수행하기 위해 서울, 부산, 제주에 위치한 5성급 호텔 종사원을 대상으로 설문조사를 실시하였으며, SPSS를 활용하여 분석을 실시하였다. 분석결과 첫째, 감정행동 중 표면행동은 직무만족에 부정적인 영향을 미치고, 서비스 사보타주 행동을 유발하는 반면, 감정행동 중 내면행동은 직무만족에 긍정적인 영향을 미치고, 서비스 사보타주 행동을 할 가능성을 낮추는 것으로 나타났다. 따라서 자신의 감정과 행해야 하는 행동이 부조화를 이루면 만족이 떨어지고, 부정적인 행동을 하게 될 확률이 높아지는 반면, 자신의 감정을 행해야 하는 행동에 맞추어 수정하면 그 감정으로 인해 만족이 높아지고, 부정적 일탈 행동을 할 확률이 낮아진다. 둘째, 직무만족은 서비스 사보타주에 부정적인 영향을 미친다. 셋째, 개인직무적합성은 표면행동이 직무만족에 미치는 영향 관계와 직무만족이 서비스 사보타주에 미치는 영향 관계에서 조절효과가 있는 것으로 나타났다. 하지만 내면행동이 직무만족에 미치는 영향 관계에서 개인직무적합성은 조절효과가 없는 것으로 나타났다. 연구 결과를 바탕으로 학문적 시사점과 실무적 시사점을 제시하였다. The purpose of this study was to clarify the effect of hotel employee s emotional labor on job satisfaction and service sabotage and to investigated the moderating effect of person job fit to establish an effective human resource management plan for hotel employees and managers. To conduct the study, a questionnaire survey was conducted on employees of 5-star hotels located in Seoul, Busan, and Jeju, and analysis was conducted using SPSS. As a result of analysis, first, it was found that surface acting negatively affects job satisfaction and induces service sabotage behavior, while deep acting positively affects job satisfaction and lowers service sabotage behavior. In other words, if one s emotions and behaviors are inconsistent, satisfaction decreases and the probability of negative behavior increases, whereas if one s emotions are modified according to behavior, satisfaction increases due to the emotion, and the probability of deviant behavior decreases. Second, job satisfaction negatively affects service sabotage. Third, it was found that person job fit had a moderating effect in the relationship between the influence of surface acting on job satisfaction and the effect of job satisfaction on service sabotage. However, in the relationship of the influence of deep acting on job satisfaction, it was found that person job fit had no moderating effect. Based on the research results, academic and practical implications were presented.

      • L-lactic Acid Production Using Adaptively Evolved Saccharomyces cerevisiae

        Lactic acid (LA) is one of the organic acids applied in various industries such as food additives, cosmetics, medical use, packaging, and pharmaceuticals. For this reason, the demand for LA is rapidly increasing in worldwide. Lactic acid bacteria is used as LA producing microbes but ironically it doesn’t tolerate high concentration of LA. But Saccharomyces cerevisiae yeast has high acid tolerance and its genetic engineering is not difficult. Therefore, S. cerevisiae already used as a platform to produce various valuable chemicals. In our previous study, we developed yeast strain that can produce LA (SR8LDH). In this study, the SR8LDH was adaptively evolved in 8% of LA condition and evolved strain was isolated (BK01). The BK01 strain was able to survive even at 8% LA condition or more. LA production of BK01 was up to 119 g/L and pH was even under 3 at that fermentation. In fermentation of lignocellulosic biomass, BK01 not only produced 12.5 g/L of LA, but also showed tolerance to 4 g/L of acetic acid, one of the fermentation inhibitors of lignocellulosic biomass. This suggests that BK01 could be presented as novel industrial LA producing yeast strains.

        괭생이모자반의 동시당화발효를 통한 Lactic Acid의 생산

        Sargassum horneri is inedible and invasive species of brown macroalgae that expands and drifts in Spring and Summer, causing damages to the ecosystem of the south coast of Korea, especially Jeju island. Korean government is preventively collecting large amounts of S. horneri biomass before reaching to the coast, but there is no other way to dispose or utilize the biomass but landfill and incineration. In the present study, S. honeri biomass was tested as a raw material for biological production of lactic acid, a type of commodity chemicals for polymer synthesis. Using commercial cellulases and lactic acid-producing engineered yeast, simultaneous saccharification and fermentation (SSF) conditions were optimized and lactic acid production was demonstrated. Finally, lactic acid was produced at a yield of 0.24 g lactic acid/g dried biomass and at a productivity of 0.73 g lactic acid/L-h, suggesting an industrial potential to biologically utilize S. honeri coastal waste.

        Sargassum horneri is inedible and invasive species of brown macroalgae that expands and drifts in Spring and Summer, causing damages to the ecosystem of the south coast of Korea, especially Jeju island. Simultaneous saccharification and fermentation (SSF) with lactic acid-producing engineered yeast was optimized in various conditions including type of commercial cellulases, solid loading, and scale-up fermentation. Finally, lactic acid was produced at a yield of 0.24 g lactic acid/g dried biomass and productivity of 0.73 g lactic acid/L-h, suggesting an industrial potential to biologically utilize S. honeri coastal waste.

        In vitro screening of extracts from 38 marine animal resources for novel cosmeceutical activities

        Marine resources have various biological activities and their constituents are more novel than those of land organisms. Several biologically active constituents have been found in marine organisms. Recently, many studies have reported that marine animals (MAs) can be used as functional ingredients in functional foods or nutraceutical due to their health benefits. However, no studies have extensively investigated the cosmeceutical activities of MAs extracts. Here, 70% ethanol extracts of 38 MAs were investigated for their activities of whitening and anti-aging properties for use as materials in novel cosmeceuticals. Anti-aging activities were determined by skin aging-related enzyme activities (anti-collagenase, anti-elastase, anti-hyaluronidase) and whitening activities (anti-tyrosinase, anti-3,4-dihydroxyl-L-phenylalanine [DOPA] oxidation) evaluated by colorimetric method. Among the 38 MAs, we found that Urechis unicinctus and Petrosia corticata extracts showed the strongest inhibitory effects against tyrosinase and DOPA oxidation, respectively. Our results additionally showed that Protankyra bidentata extract might provide a major source of anti-hyaluronidase and anti-elastase; meanwhile, anti-collagenase effects were similar in most MAs. Overall, these results suggest that extracts of marine animals have potential as a tyrosinase, collagenase, elastase, and hyaluronidase inhibitors. Taken together, MA resources could be considered as a novel cosmeceutical agent to be applied in cosmetic industry.
