      • Purification, Characterization and Immunomodulating activity of a Water-soluble Polysaccharide Isolated from Korean Capsosiphon fulvescens

        Various seaweed polysaccharides are getting great attention for intensive research due to their diverse biological activities, including effects on the immune system and cancer. Capsosiphon fulvescens, a green algae is widely distributed in the Southern coastal area of Korea. Crude polysaccharides were obtained from this seaweed collected at a coastal area of Wando, Korea, mainly by dilute acid extraction, ethanol precipitation and CaCl2 precipitation, with an yield of approximately 2.49% in mass. It was further purified by DEAE-cellulose column chromatography. The purified polysaccharide was examined for its immunostimulating activity on Raw 264.7 cells and shown to stimulate the production of TNF-α in a dose-dependent manner. The primary structure of this polysaccharide was analyzed by High Performance Anion Exchange Chromatography and Fourier Transform Infrared (FT-IR) spectrometry. From the HPAEC-PAD analysis, the monosaccharide composition of this purified polysaccharide was shown to be mannose(58%), rhamnose(33%), galactose(3.8%), xylose(3.3%), fucose(0.76%), and arabinose(0.67%). The IR spectrum showed a strong absorption band at 1256 cm-1, indicating the presence of sulfate esters. Absorption bands at 845 cm-1and 1060 cm-1 characteristic for galactose-4-sulfate and Sulfur trioxide, respectively, were also observed. From HPLC analysis molecular mass of this polysaccharide was determined to be approximately 385 kDa.

        Isolation and Immunomodulating Activity of an Extracellular Polysaccharide Produced by Bacillus sp. PS-12

        토양에서 분리한 세포외 다당류 생산균주 PS-12는 형태학적, 생리학적, 화학적 분석에 의하여 Bacillus sp.에 속하는 균주로 동정되었으며, 분리균주 PS-12는 Bacillus sp. PS-12로 명명하였다. Bacillus sp. PS-12가 생산하는 세포외 다당류는 에탄올 침전, cetylpyridinium chloride (CPC) 침전과 gel permeation chromatography를 이용하여 정제하였으며, 정제된 다당류 PS-12의 당조성은 glucose, mannose, galactose와 fucose가 7:3.2:2:1의 몰비로 구성되어있었다. Bacillus sp. PS-12로부터 분리된 다당류 PS-12를 이용하여 면역증강효과를 확인하였다. TNF-α 및 IL-6 측정은 RAW264.7 대식세포주를 사용하였으며 cytokine정량을 위하여 ELISA kit를 이용하였다. RAW264.7 세포주에 대한 PS-12의 세포독성을 확인하기 위하여 세포독성이 10% 미만을 나타내는 농도인 2 ㎍/㎖을 PS-12의 최대농도로 측정하였다. PS-12는 2 ㎍/㎖에서 TNF-α를 정상세포보다 50배 이상 높은 수치로 생산하였다. 또한 IL-6의 생산을 농도 의존적으로 증가시켰다. 이러한 결과로부터 PS-12가 면역세포에 대해 세포독성을 거의 나타내지 않는 농도에서 대식세포로부터 TNF-α와 IL-6의 cytokine 생산을 함으로써 면역증강효과를 나타낸다는 것을 확인하였다. A bacterial strain producing highly viscous extracellular polysaccharide was isolated from soil. Through morphological, physiological and chemotaxonomical studies, it was identified as a Bacillus sp. and named Bacillus sp. PS-12. The extracellular polysaccharide, named PS-12 was purified by ethanol precipitation, cetylpyridinium chloride (CPC) precipitation and gel permeation chromatography. The purified polysaccharide was found to consist of glucose, mannose, galactose, and fucose, with a molar ratio of approximately 7:3.2:2:1, respectively. PS-12 was investigated for its immunostimulating activity on murine macrophage RAW264.7 cells using an ELISA assay. PS-12 stimulated the production of TNF-α to a level 50 times greater than the control and also induced IL-6 secretion in a dose-dependent manner. The cytotoxicity on RAW264.7 cells by PS-12 was relatively low with 10% cytotoxicity at 2 ㎍/㎖. These results indicate that PS-12 is less cytotoxic to immune cells and possess immunomodulating activity in which it can produce cytokines including TNF-α and IL-6 from macrophages.

      • Biological activities of a Water-soluble Polysaccharide(CPS) from Korean Capsosiphon fulvescens

        Seaweed extracts have recently been found to have biological activities including effects on the immune system, anticoagulant system, cancer, etc. Capsosiphon fulvescens is a green alga that is widely distributed in the Southern coastal area of Korea. Crude polysaccharides were obtained from this seaweed collected at a coastal area of Wando, Korea, mainly by dilute acid extraction, ethanol precipitation and CaCl2 precipitation. It was further purified by DEAE-cellulose column chromatography and its chemical composition, structural analysis, immunostimulating activity, anticoagulation activity, anticancer activity and osteogenesis was determined. From HPLC analysis molecular mass of this polysaccharide was determined to be approximately 385 kDa. Elemental analysis of CPS, The estimation of only four elements, that is carbon(31.9398% w/w), hydrogen(4.9022% w/w), nitrogen(0.6318% w/w) and sulfur(6.2166% w/w), was undertaken. CPS stimulated the production of TNF-α to a level 80 times greater than the normal. Also CPS induced IL-6 secretion from RAW264.7 cells in a dose-dependent manner. Moreover, Cf–PS treatment increased the expressions of cyclooxygenase-2 (COX-2) and the inducible form of nitric oxide (iNOS). The CPS could significantly increase anticoagulant activity, which was especially characterized by activated partial thrombloplastin time (APTT) and thrombloplatin time (TT). Two hundred ug/ml of CPS delayed the APTT up to 61 s. And TT of CPS reached up to 32 s. MTT assay showed that the CPS significantly inhibited the proliferation of cultured human MKN-45 gastric cancer cells in a dose-dependent manner. Also nitrite plays an important role in the formation of carcinogenic nitrosamine. One percentage of CPS eliminated the nitrite up to approximately 50%. CPS plays a role for osteoblastic bone metastasis by promoting both osteoblasts proliferation and apoptosis of osteoclasts. One mg/ml of CPS increased the osteoblast cell growth up to approximately 150% and decreased the osteoclast cell growth up to approximately 60% by induction of apoptosis.

        옥돔과 옥두어 판별을 위한 PCR 검사법 개발과 검증

        본 연구에서는 형태학적으로 판별이 어려운 옥돔과 옥두어의 종 특이 primer를 개발, 검증한 후 모니터링을 통해 위변조 된 옥돔의 유통을 예방하고자 하였다. 옥돔과 옥두어 염기서열은 clustal omega 프로그램을 이용하여 정리하였고, primer3 프로그램을 이용하여 primer를 설계하였다. Multiplex PCR 결과는 옥돔과 옥두어에 대한 종 특이적 증폭이 확인되었고, PCR 반응을 확인하기 위한 공통 유전자에 대한 증폭이 확인되었다. 옥돔을 288 bp, 옥두어를 159 bp, 공통 유전자를 502 bp로 증폭되어 각각 PCR product 사이에 100 bp 이상 차이가 나타나 정확하게 판별이 가능하였다. Multiplex PCR 민감도 실험결과 옥돔 primer가 1 ng, 옥두어 primer가 1 ng, 공통 유전자 primer가 1 ng까지 밴드가 확인되었다. 모니터링 실험결과, 옥돔 38건, 옥두어 13건으로 판정되어 시료의 어종과 실험결과가 100% 일치함을 확인하였고, 위변조 사례는 나타나지 않았다. 본 실험에서 개발된 multiplex PCR 방법은 특이도와 민감도가 확보되었고 모니터링을 통해 유통, 판매되고 있는 옥돔과 옥두어의 판별에 적합함을 확인하였다. We developed and validated species-specific primers for Branchiostegus japonicus and Branchiostegus albus to prevent the sale of B. albus as B. japonicus. Primers for B. japonicus and B. albus were designed against the cytochrome b gene. Multiplex PCR showed a 288 bp amplicon for B. japonicus, a 159 bp amplicon for B. albus, and a 502 bp amplicon for the internal control. The PCR product bands for B. japonicus, B. albus, and the internal control were present at 1 ng each. The specificity and sensitivity of the primers developed in this study were validated by testing 38 B. japonicus strains and 13 B. albus strains. Using this monitoring method, fake fish did not appear due to the agreement between the experimental results and the species. Therefore, the developed multiplex PCR method was suitable for differentiating B. japonicus and B. albus.

        Development and Characterization of 10 Polymorphic Microsatellite Loci in the Korean Endemic Freshwater Fish Iksookimia koreensis, and Their Cross-species Amplification in the Endemic I. longicorpa

        The genus Iksookimia (Actinopterygii: Cypriniformes: Cobitidae) is a bottom-dwelling freshwater loaches, which are well-known as their endemism and high geographic variation. However, population genetic relationships among Iksookimia spp. have remained unclear due to a shortage of genetic markers that can be applied generally in the genus. Here, we developed high-resolving microsatellite markers using I. koreensis and I. longicorpa as representatives of Iksookimia species because of their wide distribution range and phylogenetic position. Ten of polymorphic microsatellite loci were isolated from Iksookimia koreensis and were successfully cross-amplified in I. longicorpa. The mean number of observed alleles per locus was about 10.4 (range, 2-17) for I. koreensis and about 13.2 (range, 2-24) for I. longicorpa. The loci, IK03 and IK08, deviated from the Hardy-Weinberg equilibrium in I. koreensis, after applying the Bonferroni correction. The microsatellite markers obtained in the present study will be useful to evaluate population genetic structure and to establish conservation strategies for I. koreensis and related Iksookimia species.

      • Enhanced CO Oxidation and Cyclic Activities in Three-Dimensional Platinum/Indium Tin Oxide/Carbon Black Electrocatalysts Processed by Cathodic Arc Deposition

        <P>There have been extensive efforts to develop competitive electrocatalysts using carbon black (CB) supports for high-performance proton-exchange membrane fuel cells with less usage of Pt. Herein, we propose a very promising electrocatalyst architecture based on the three-dimensional Pt/indium tin oxide (ITO)/CB support structure which was enabled by a nonconventional deposition process ensuring very uniform impregnation of Pt and ITO nanoparticles into the CB network. The unusual scales of the Pt (∼1.9 nm) and ITO (∼5.6 nm) nanoparticles were directly related to unexpectedly better performance of the electrocatalytic activities. As a highlight, the electrochemical surface area of the electrocatalyst was maintained very well after the 3000 cycle-accelerated durability evaluation by demonstrating an excellent retention of ∼74.9%. Particularly, the CO tolerance exhibited a low value of ∼0.68 V as the absorption current peak, compared to ∼0.79 V for a commercial Pt/CB catalyst containing twice more Pt.</P> [FIG OMISSION]</BR>
