The nitric oxide (NO) formation and intrinsic nitrosation may be involved in the possible mechanisms of liver fluke-associated carcinogenesis. We still do not know much about the responses of inducible NO synthase (iNOS) induced by Clonorchis sinensis infection. This study was conducted to explore the pathological lesions and iNOS expressions in the liver of mice with different infection intensity levels of C. sinensis. Extensive periductal inflammatory cell infiltration, bile duct hyperplasia, and fibrosis were commonly observed during the infection. The different pathological responses in liver tissues strongly correlated with the infection intensity of C. sinensis. Massive acute spotty necrosis occurred in the liver parenchyma after a severe infection. The iNOS activity in liver tissues increased, and iNOS-expressing cells with morphological differences were observed after a moderate or severe infection. The iNOS-expressing cells in liver tissues had multiple origins.

Background: This retrospective study aimed to validate the safety and effectiveness of hepatectomy for huge hepatocellular carcinoma (HCC). Materials and Methods: Data of patients who underwent hepatectomy for HCC between January 2006 and December 2012 were reviewed. The patients were divided into three groups: huge HCC(${\geq}10cm$ in diameter), large HCC(${\geq}5$ but<10 cm in diameter) and small HCC(<5cm in diameter). Results: Characteristics of pre-operative patients in all three groups were homogeneously distributed except for alpha fetal protein (AFP)(p<0.001).The 30, 60, 90-day post-operative mortality rates were not different among the three groups (p=0.785, p=0.560, and p=0.549). Laboratory data at 1, 3, and 7 days after surgery also did not vary. The 5-year overall survival (OS) and 5-year disease-free survival (DFS) rates in the huge and large HCC groups were lower than that of the small HCC group (OS: 32.5% vs 36.3% vs 71.2%, p=0.000; DFS: 20.0% vs 24.8% vs 40.7%, p=0.039), but there was no difference between the huge and large HCC groups (OS: 32.5% vs 36.3%, p=0.667; DFS: 20.0% vs 24.8%, p=0.540). In multivariate analysis, five independent poor prognostic factors that affected OS were significantly associated with worse survival (p<0.05), namely, AFP level, macrovascular invasion, Edmondsone Steiner grade, surgical margin and Ishak score. AFP level, macrovascular invasion, microvascular invasion, and surgical margin influenced disease-free survival independently (p<0.05). Conclusions: The safety of hepatectomy for huge HCC is similar to that for large and small HCC; and this approach for huge HCC may achieve similar long-term survival and disease-free survival as for large HCC.

The land subsidence poses a nonnegligible threat to production and life, especially in dredger fill area. The essence of land subsidence is soil consolidation. Better evaluation of the degree of soil consolidation presents significant progress for predicting land subsidence. In this study, SBAS-InSAR was conducted to Chenjia town, the typical dredger fill area in Chongming East Shoal, Shanghai. SAR data and precise Orbit data were collected from the ESA satellite Sentinel-1 in recent 2 years with approximately one month intervals. the relationship curve of cumulative subsidence with time is obtained after processing. On this basis, the final settlement and soil consolidation can be estimated by applying the method proposed by Professor Huang Wenxi of China. The results show that: The subsidence rate is faster than 20 mm/a; The estimated final settlement is 26.6-52.9 mm, and the corresponding consolidation degree is 54.6 % on average; Soil mass has not yet been fully consolidated and it still takes a long time to achieve stability. In conclusion, the method proposed in this study can provide a clearer understanding of the consolidation degree, and a new reference for the evaluation and prevention of land subsidence. Future research will focus on longer monitoring time to enrich and improve the method. A simple, sensitive and reproducible high-performance liquid chromatography (HPLC) method has been validated for determining concentrations of glutamate, glycine, and alanine in human plasma. Proteins in plasma were precipitated with perchloric acid, followed by derivatization with 6-aminoquinolyl-N-hydroxysuccinimidyl carbamate (AQC). Simultaneous analysis of glutamate, glycine, and alanine is achieved using reversed-phase HPLC conditions and ultraviolet detection. Excellent linearity was observed for these three amino acids over their concentration ranges with correlation coefficients (r)>0.999. The intra- and inter-day precision were below 10%. This method utilizes quality control samples and demonstrates excellent plasma recovery and accuracy. The developed method has been successfully applied to measure plasma glutamate, glycine, and alanine in twenty volunteers.
Increased accumulation of anthocyanins in transgenic potato tubers by overexpressing the 3GT gene
Qing Wei,Qing Yang,Quan-Yi Wang,Zhi-Hang Feng,Bing Wang,Yun-Feng Zhang 한국식물생명공학회 2012 Plant biotechnology reports Vol.6 No.1
In order to explore a biotechnological method for improving potato tuber color and creating plants with increased anthocyanin contents, a potato UDP-glucose: flavonoid-3-O-glucosyltransferase (3GT) gene was inserted behind the GBSSI promoter of pBin19, and this construct was introduced into Solanum tuberosum L. cultivar De´sire´e plants by Agrobacterium-mediated transformation. Six independent transgenic lines overexpressing the 3GT gene were identified by PCR and Southern blot analysis from 18 kanamycin-resistant plants. Due to the expression of 3GT gene, the tuber color and the anthocyanin content were enhanced noticeably in the transgenic plants compared to the wild-type control plants. This result suggests that the 3GT gene can potentially be used to improve potato color and enhance levels of antioxidants in the diet.
Effect of Channel Curvature for Jet Impinging on Blade Leading Edge
Qing-Yang Zhao,Eui Yeop Jung,Seok Min Choi,Hyung Hee Cho 대한기계학회 2013 대한기계학회 춘추학술대회 Vol.2013 No.12
The characteristics of fluid flow and heat transfer affected by the curvature variation in a jet impinged leading edge channel have been investigated numerically using three-dimensional Reynolds-averaged Navier-Stokes analysis via shear stress transport turbulence model and γ - Reθ transitional turbulence model. A constant heat flux condition has been applied on the leading edge surface. Jet to leading edge surface distance is constant as three times of jet diameter. The leading edge curved surface curvature is set as variables by the reason of multiple shape of real turbine blade. Results presented in this study include contour of Nusselt number, velocity vector and local Nusselt number distribution along the central line on the leading edge surface. Based on the data above, the average Nusselt number, stagnation Nusselt number along the central line of leading edge and average pressure supplied on each jet are compared between each case. The varied leading edge curved surfaces change the cross flow intensity which influences the impinging jet flow direction. As a result of the phenomenon of jet flow direction change, the heat transfer on the leading edge is influenced significantly. Result indicates that with the decrease of leading edge curved surface diameter, the cross flow effect improves and heat transfer uniformity decreases. In the case of small diameter leading edge, the average heat transfer decrease and the Nusselt value decreases along the streamwise direction due to the crossflow from upstream jet flow. Besides the heat transfer uniformity, the thermal performance comparison is also investigated in this study.

Objective: This research aimed to study the effects of Artemisia argyi flavonoids (AAF) supplemented in diets on the growth performance and immune function of broiler chickens challenged with lipopolysaccharide (LPS). Methods: A total of one hundred and ninety-two 1-d-old broiler chicks were assigned into 4 treatment groups, which were, respectively, fed a basal diet (control), fed a diet with 750 mg/kg AAF, fed a basal diet, and challenged with LPS, fed a diet with 750 mg/kg AAF, and challenged with LPS. Each treatment had six pens with 8 chicks per pen. On days 14, 16, 18, 20 (stress phase I) and 28, 30, 32, 34 (stress phase II), broilers were injected with LPS (500 ㎍/kg body weight) or an equivalent amount of saline. Results: The results demonstrated that dietary AAF significantly improved the body weight (d 21) and alleviated the decrease of average daily gain in broilers challenged with LPS on d 21 and d 35 (p<0.05). Dietary AAF increased bursa fabricius index, and dramatically attenuated the elevation of spleen index caused by LPS on d 35 (p<0.05). Furthermore, serum interleukin-6 (IL-6) concentration decreased with AAF supplementation on d 21 (p<0.05). Diet treatment and LPS challenge exhibited a significant interaction for the concentration of IL-1β (d 21) and IL-6 (d 35) in serum (p<0.05). Additionally, AAF supplementation mitigated the increase of IL-1β, IL-6 in liver and spleen induced by LPS on d 21 and 35 (p<0.05). This study also showed that AAF supplementation significantly reduced the expression of IL-1β (d 21) and nuclear transcription factor kappa-B p65 (d 21 and 35) in liver (p<0.05), and dietary AAF and LPS treatment exhibited significant interaction for the gene expression of IL-6 (d 21), toll like receptor 4 (d 35) and myeloid differentiation factor 88 (d 35) in spleen (p<0.05). Conclusion: In conclusion, AAF could be used as a potential natural immunomodulator to improve growth performance and alleviate immune stress in broilers challenged with LPS.
