        The culture conditions for mycelial growth and sclerotial formation of Polyporus umbellatus

        Polyporus umbellatus (Syn. Grifola umbellata) is a sclerotium forming mushroom belongs to family Polyporaceae of Polyphorales, Basidiomycota. The sclerotia of P. umbellatus have long been used for traditional medicines in China, Korea and Japan. This study was initiated to obtain the basic data for artificial sclerotial production of P. umbellatus. Here, we investigated the favorable conditions for mycelial growth of P. umbellatus and its symbiotic fungus Armillaria mellea. We also evaluate the favorable carbon and nitrogen sources for sclerotial formation in dual culture between P. umbellatus and A. mellea. The favorable conditions for mycelial growth of P. umbellatus were 20oC and pH 4, while optimal conditions for mycelial growth of A. mellea were 25oC and pH 6. The carbon sources for optimal mycelial growth of P. umbellatus were fructose and glucose, while carbon sources for favorable mycelial growth of A. mellea were also fructose and glucose. The nitrogen sources for favorable mycelial growth P. umbellatus were peptone and yeast extract, while optimal mycelial growth of A. mellea were obtained in peptone and yeast extract. When P. umbellatus and A. mellea were dual cultured on carbon sources, sclerotia were induced on basal media supplemented with glucose, fructose and maltose at pH 4~6, while nitrogen sources inducing sclerotia were basal media supplemented with peptone and yeast extract for 60 days at 20oC under dark condition.

        Influence of particle distribution on filter coefficient in the initial stage of filtration

        Under the condition that the size of suspended particles is nonhomogeneous, we studied how filter grain and suspended particles affect filter coefficient in the early stage. A stochastic method was used to study the variation of the initial filter coefficient. Through physical experiment, the collected data include the initial inflow and outflow concentration,the size distributions of particles in suspension and so on. By introducing the standard capture probability function P(d) and the characteristic length of filter bed Lc, this separated the grains’ and particles’ influence on filter coefficient. An example showed that we could use P(d), Lc to ascertain the result of effluent distribution, ratio of Cout to Cin in any depths and equivalent filter coefficient λm. We also studied the filter coefficient λl during the experiment and the first-order derivative of λl.

        Deep Learning-based Target Masking Scheme for Understanding Meaning of Newly Coined Words

        최근 대량의 텍스트 분석을 위해 딥 러닝(Deep Learning)을 활용하는 연구들이 활발히 수행되고 있으며, 특히 대량의 텍스트에 대한 학습 결과를 특정 도메인 텍스트의 분석에 적용하는 사전 학습 언어 모델(Pre-trained Language Model)이 주목받고 있다. 다양한 사전 학습 언어 모델 중 BERT(Bidirectional Encoder Representations from Transformers) 기반 모델이 가장 널리 활용되고 있으며, 최근에는 BERT의 MLM(Masked Language Model)을 활용한 추가 사전 학습(Further Pre-training)을 통해 분석 성능을 향상시키기 위한 방안이 모색되고 있다. 하지만 전통적인 MLM 방식은 신조어와 같이 새로운 단어가 포함된 문장의 의미를 충분히 명확하게 파악하기 어렵다는 한계를 갖는다. 이에 본 연구에서는 기존의 MLM을 보완하여 신조어에 대해서만 집중적으로 마스킹을 수행하는 신조어 표적 마스킹(NTM: Newly Coined Words Target Masking)을 새롭게 제안한다. 제안 방법론을 적용하여 포털 ‘N’사의 영화 리뷰 약 70만 건을 분석한 결과, 제안하는 신조어 표적 마스킹이 기존의 무작위 마스킹에 비해 감성 분석의 정확도 측면에서 우수한 성능을 보였다. Recently, studies using deep learning to analyze a large amount of text are being actively conducted. In particular, a pre-trained language model that applies the learning results of a large amount of text to the analysis of a specific domain text is attracting attention. Among various pre-trained language models, BERT(Bidirectional Encoder Representations from Transformers)-based model is the most widely used. Recently, research to improve the performance of analysis is being conducted through further pre-training using BERT"s MLM(Masked Language Model). However, the traditional MLM has difficulties in clearly understands the meaning of sentences containing new words such as newly coined words. Therefore, in this study, we newly propose NTM(Newly coined words Target Masking), which performs masking only on new words. As a result of analyzing about 700,000 movie reviews of portal "N" by applying the proposed methodology, it was confirmed that the proposed NTM showed superior performance in terms of accuracy of sensitivity analysis compared to the existing random masking.

      • Efficacy of Prophylactic Entecavir for Hepatitis B Virus-Related Hepatocellular Carcinoma Receiving Transcatheter Arterial Chemoembolization

        Background and Aims: Hepatitis B virus (HBV) reactivation was reported to be induced by transcatheter arterial chemoembolization (TACE) in HBV-related hepatocellular carcinonma (HCC) patients with a high incidence. The effective strategy to reduce hepatitis flares due to HBV reactivation in this specific group of patients was limited to lamivudine. This retrospective study was aimed to investigate the efficacy of prophylactic entecavir in HCC patients receiving TACE. Methods: A consecutive series of 191 HBV-related HCC patients receiving TACE were analyzed including 44 patients received prophylactic entecavir. Virologic events, defined as an increase in serum HBV DNA level to more than 1 log10 copies/ml higher than nadir the level, and hepatitis flares due to HBV reactivation were the main endpoints. Results: Patients with or without prophylactic were similar in host factors and the majorities of characteristics regarding to tumor factors, HBV status, liver function and LMR. Notably, cycles of TACE were parallel between the groups. Ten (22.7%) patients receiving prophylactic entecavir reached virologic response. The patients receiving prophylactic entecavir presented significantly reduced virologic events (6.8% vs 54.4%, p=0.000) and hepatitis flares due to HBV reactivation (0.0% vs 11.6%, p=0.039) compared with patients without prophylaxis. Kaplan-Meier analysis illustrated that the patients in the entecavir group presented significantly improved virologic events free survival (p=0.000) and hepatitis flare free survival (p=0.017). Female and Eastern Cooperative Oncology Group (ECOG) performance status 2 was the only significant predictors for virological events in patients without prophylactic antiviral. Rescue antiviral therapy did not reduce the incidence of hepatitis flares due to HBV reactivation. Conclusion: Prophylactic entecavir presented promising efficacy in HBV-related cancer patients receiving TACE. Lower performance status and female gender might be the predictors for HBV reactivation in these patients.

      • Hepatitis B Virus DNA Negativity Acts as a Favorable Prognostic Factor in Hepatocellular Carcinoma Patients

        Background: This retrospective study was aimed to investigate the efficacy of prophylactic agents in hepatocellular carcinoma (HCC) patients receiving TACE and compare the difference between lamivudine and entecavir. Materials and Methods: A consecutive series of 203 HBV-related HCC patients receiving TACE were analyzed including 91 patients given prophylactic agents. Virologic events, defined as an increase in serum HBV DNA level to more than 1 log10 IU/ml higher than the nadir level, hepatitis flares due to HBV reactivation and progression free survival (PFS) were the main endpoints. Results: Some 48 (69.6%) reached virologic response. Prophylaxis significantly reduced virologic events (8.8% vs 58.0%, p=0.000) and hepatitis flares (1.1% vs 13.4%, p=0.001). Patients presenting undetectable HBV DNA levels displayed a significantly improved PFS as compared to those who never achieved undetectable HBV DNA. Prophylaxis and e-antigen positivity were the only significant variables associated with virologic events. In addition, prophylaxis was the only independent protective factor for hepatitis flares. Liver cirrhosis, more cycles of TACE, HBV DNA negativity, a lower Cancer of the Liver Italian Program score, non-metastasis and no hepatitis flares were protective factors for PFS. Prophylactic lamivudine demonstrated similar efficacy as entecavir. Conclusions: Prophylactic agents are efficacious for prevention of HBV reactivation in HCC patients receiving TACE. Achievement of undetectable HBV DNA levels displayed a significant capability in improving PFS. Moreover, persistent tumor residual lesions, positive HBV DNA and hepatitis B flares might be causes of tumor progression in these patients.

      • Staged Improvement in Awareness of Disease for Elderly Cancer Patients in Southern China

        Background: In mainland China, awareness of disease of elderly cancer patients largely relies on the patients' families. We developed a staged procedure to improve their awareness of disease. Materials and Methods: Participants were 224 elderly cancer patients from 9 leading hospitals across Southern China. A questionnaire was given to the oncologists in charge of each patient to evaluate the interaction between family and patients, patient awareness of their disease and participation in medical decision-making. After first cycles of treatment, increased information of disease was given to patients with cooperation of the family. Then patient awareness of their disease and participation in medical decision-making was documented. Results: Among the 224 cancer elderly patients, 26 (11.6%) made decisions by themselves and 125 (55.8%) delegated their rights of decision-making to their family. Subordinate family members tended to play a passive role in decision-making significantly. Patients participating more in medical decision-making tended to know more about their disease. However, in contrast to the awareness of disease, patient awareness of violation of medical recommendations was reversely associated with their participation in medical decision-making. Improvement in awareness of diagnosis, stages and prognosis was achieved in about 20% elderly cancer patients. About 5% participated more actively in medical decision-making. Conclusions: Chinese elderly cancer patient awareness of disease and participation in medical decision-making is limited and relies on their family status. The staged procedure we developed to improve patient awareness of disease proved effective.

      • Study on Corner Reflectors Identification in Highway Deformation Monitoring

        CRInSAR is a newly developed technique to monitor ground deformation. In CRInSAR algorithm, the identification of Corner Reflectors in SAR images is necessary. Due to the uncertainty of traditional identification method, a new method based on the intensity and correlation coefficient of each CR candidates in SAR images is presented. The method has been successfully used to determine the locations of 11 CRs installed along a highway in six SAR images over the study area. The results show that the identification accuracy of the new method is about 1 pixel. It is effective and reliable especially in the area with lots of lightspots around the CR points. The method proposed can play important role in the highway deformation monitoring within CRInSAR algorithm.

        Overexpression of a Lotus corniculatus AP2/ERF transcription factor gene, LcERF080, enhances tolerance to salt stress in transgenic Arabidopsis

        The APETALA2/ethylene-responsive elementbinding factors (AP2/ERF) play central roles in the stressresponse in plants. In this study, we identified and isolateda novel salt stress-related gene, LcERF080, that encodes anAP2/ERF protein in Lotus corniculatus cultivar Leo. LcERF080 was classified into the B-4 group of the ERFsubfamily based on multiple sequence alignment andphylogenetic characterization. Expression of LcERF080was strongly induced by salt, abscisic acid, 1-aminocyclopropane-1-carboxylic acid, methyl jasmonate, and salicylicacid stresses. Subcellular localization assay confirmedthat LcERF080 is a nuclear protein. LcERF080 overexpressionin Arabidopsis resulted in pleiotropic phenotypeswith a higher seed germination rate and transgenic plantswith enhanced tolerance to salt stress. Further, under stressconditions, the transgenic lines exhibited elevated levels ofsoluble sugars and proline as well as relative moisturecontents but a lower malondialdehyde content than incontrol plants. The expression levels of hyperosmoticsalinity response genes COR15A, RD22, and P5CS1 werefound to be elevated in the LcERF080-overexpressingArabidopsis plants compared to the wild-type plants. Theseresults reveal that LcERF080 is involved in the responsesof plants to salt stress.

        ORiginal Article : Pyrrolidine Dithiocarbamate Inhibits Nuclear Factor κB and Toll-Like Receptor 4 Expression in Rats with Acute Necrotizing Pancreatitis

        Background/Aims: To investigate the expression of Tolllike receptor 4 (TLR4) in the pancreases of rats with acute necrotizing pancreatitis (ANP) and any changes upon treatment with pyrrolidine dithiocarbamate (PDTC), an inhibitor of nuclear factor κB (NF-κB), as well as to determine the relationship between TLR4 and NF-κB in ANP pathogenesis. Methods: A total of 72 SD rats were randomly divided into three groups, namely, the control (sham-operation), ANP, and ANP with PDTC pretreatment groups. The PDTC-pretreated group was intraperitoneally injected with PDTC at a dose of 100 mg/kg 1 hour before the induction of ANP. The expressions of TLR4 and NF-κB in pancreatic tissue were evaluated by immunohistochemistry and Western blot analysis. The mRNA levels of cytokines tumor necrosis factor α, interleukin (IL)-1β, and IL-6 were measured by reverse transcription polymerase chain reaction. Results: The expressions of TLR4, NF-κB, and cytokine (NF-κB target) genes in the pancreatic tissue increased more significantly in the ANP groups than in the sham-operation group at 3, 6, and 12 hours. Pretreatment with PDTC alleviated the inflammatory activation in the pancreas with ANP, causing a significant decrease in the expressions of TLR4, NF-κB, and cytokine genes in the pancreatic tissue. Conclusions: The expressions of TLR4 and NF- κB were increased in the pancreases of rats with ANP. PDTC not only inhibits NF-κB but also suppresses the expression of TLR4 and downregulates the expression of the related cytokine genes. (Gut Liver 2015;9:411-416)
