        Preparation of activated carbon spheres and their electrochemical properties as supercapacitor electrode

        The carbon spheres (CSs) synthesized by an ultrasonic-spray pyrolysis method were activated for supercapacitor electrode. There are plenty of cracks on the surface of the activated carbon spheres (ACSs), which expend with increasing the activation temperature and activator dosage. The specifc capacitance of ACSs increases with the activation temperature and activator dosage and reach to maximal value at certain conditions. Importantly, the ACS sample activated at relatively low activation temperature (600 °C) and 7 of mass ratio of KOH to CSs has the highest specifc capacitance (about 209 F g−1 at 50 mA g−1 of current density) and indicates the excellent cycling stability after 1000 consecutive charge–discharge cycles. Furthermore, the graphene sheets could be found in the samples that were activated at 1000 °C. And the electrode prepared by the sample has the very low series resistance because of the excellent conductivity of the formed graphene sheets.

      • BRCA1 Gene Exon 11 Mutations in Uighur and Han Women with Early-onset Sporadic Breast Cancer in the Northwest Region of China

        The prevalence of BRCA1 gene mutations in breast cancer differs between diverse ethnic groups. Relatively little information is known about patterns of BRCA1 mutations in early-onset breast cancer in women of Uighur or Han descent, the major ethnic populations of the Xinjiang region in China. The aim of this study was to identify BRCA1 mutations in Uighur and Han patients with early-onset (age <35 years), and sporadic breast cancer for genetic predisposition to breast cancer. For detection of BRCA1 mutations, we used a polymerase chain reaction single-stranded conformation polymorphism approach, followed by direct DNA sequencing in 22 Uighur and 13 Han women with early-onset sporadic breast cancer, and 32 women with benign breast diseases. The prevalence of BRCA1 mutations in this population was 22.9% (8/35) among early-onset sporadic breast cancer cases. Of these, 31.8% (7/22) of Uighur patients and 7.69% (1/13) of Han patients were found to have BRCA1 mutations. In 7 Uighur patients with BRCA1 mutations, there were 11 unique sequence alterations in the BRCA1 gene, including 4 clearly disease-associated mutations on exon 11 and 3 variants of uncertain clinical significance on exon 11, meanwhile 4 neutral variants on intron 20 or 2. None of the 11 BRCA1 mutations identified have been previously reported in the Breast Cancer Information Core database. These findings reflect the prevalence of BRCA1 mutations in Uighur women with early-onset and sporadic breast cancer, which will allow for provision of appropriate genetic counseling and treatment for Uighur patients in the Xinjiang region.

      • Association of Family History with Cancer Recurrence and Survival in Patients with Hepatitis B-Related Hepatocellular Carcinoma (A Propensity Score Matching Analysis)

        Aims: A family history of liver cancer is regarded as a risk factor for hepatocellular carcinoma (HCC) development. We investigated the association between family history and cancer recurrence and survival in patients with hepatitis B virus (HBV)- related HCC. Methods: Patients who underwent curative resection of HBV-related HCC between 2003 and 2013 from a tertiary hepatobiliary center in China were enrolled in this study. A family history was defined as a self-reported history of primary liver cancer in the first-degree relatives. Propensity score matching (PSM) and multivariable Cox-regression analyses were performed to compare the overall survival (OS) and recurrence-free survival (RFS) between patients with and without a family history of liver cancer. Results: Of 1,112 patients, 183 patients (16.5%) had a family history of liver cancer. A family history was not associated with OS and RFS (P=0.994 and 0.428) in the entire cohort. Using PSM, 179 pairs of patients with and without a family history but with comparable baseline characteristics and operative variables were created. A family history was associated with decreased OS and RFS (P=0.042 and 0.006) in the PSM cohort. On multivariable Cox-regression analyses, a family history was significantly associated with decreased OS (HR: 1.574, 95% CI: 1.171-2.116, P=0.003) and RFS (HR: 1.534, 95% CI: 1.176-2.002, P=0.002) after adjusting for other prognostic factors. Conclusions: A family history of liver cancer was associated with decreased OS and RFS rates after curative resection in patients with HBV-related HCC.

        Regulatory Effects of Resveratrol on Antioxidant Enzymes: a Mechanism of Growth Inhibition and Apoptosis Induction in Cancer Cells

        Resveratrol (RSV) is a natural polyphenol that is known as a powerful chemopreventive and chemotherapeutic anticancer molecule. This study focused on the effects of RSV on the activities and expression levels of antioxidant enzymes in the cancer cells. Prostate cancer PC-3 cells, hepatic cancer HepG2 cells, breast cancer MCF-7 cells and the non-cancerous HEK293T kidney epithelial cells were treated with a wide range of RSV concentrations (10-100 M) for 24-72 h. Cell growth was estimated by trypan blue staining, activities of the antioxidant enzymes were measured spectrophotometrically, expression levels of the anti-oxidant enzymes were quantified by digitalizing the protein band intensities on Western blots, and the percentage of apoptotic cells was determined by flow cytometry. Treatment with a low concentration of RSV (25 M) significantly increased superoxide dismutase (SOD) activity in PC-3, HepG2 and MCF-7 cells, but not in HEK293T cells. Catalase (CAT) activity was increased in HepG2 cells, but no effect was found on glutathione peroxidase (GPX) upon RSV treatment. RSV-induced SOD2 expression was observed in cancer cells, although the expression of SOD1, CAT and GPX1 was unaffected. Apoptosis increased upon RSV treat- ment of cancer cells, especially in PC-3 and HepG2 cells. Together, our data demonstrated that RSV inhibits cancer cell growth with minimal effects on non-cancerous cells. We postulate that the disproportional up-regulation of SOD, CAT and GPX expression and enzymatic activity in cancer cells results in the mitochondrial accumulation of H2O2, which in turn induces cancer cell apoptosis.

        The K+/H+ antiporter AhNHX1 improved tobacco tolerance to NaCl stress by enhancing K+ retention

        High salinity is the one of important factors limiting plant growth and crop production. Many NHX-type antiporters have been reported to catalyze K+/H+ exchange to mediate salt stress. This study shows that an NHX gene from Arachis hypogaea L. has an important role in K+ uptake and transport, which affects K+ accumulation and plant salt tolerance. When overexpressing AhNHX1, the growth of tobacco seedlings is improved with longer roots and a higher fresh weight than the wild type (WT) under NaCl treatment. Meanwhile, when exposed to NaCl stress, the transgenic seedlings had higher K+/H+ antiporter activity and their roots got more K+ uptake. NaCl stress could induce higher K+ accumulation in the roots, stems, and leaves of transgenic tobacco seedlings but not Na+ accumulation, thus, leading to a higher K+/Na+ ratio in the transgenic seedlings. Additionally, the AKT1, HAK1, SKOR, and KEA genes, which are involved in K+ uptake or transport, were induced by NaCl stress and kept higher expression levels in transgenic seedlings than in WT seedlings. The H+-ATPase and H+-PPase activities were also higher in transgenic seedlings than in the WT seedlings under NaCl stress. Simultaneously, overexpression of AhNHX1 increased the relative distribution of K+ in the aerial parts of the seedlings under NaCl stress. These results showed that AhNHX1 catalyzed the K+/H+ antiporter and enhanced tobacco tolerance to salt stress by increasing K+ uptake and transport.

      • Risk Factors, Patterns and Long-Term Prognosis of Early and Late Recurrence in Patients with Hepatitis B Virus-Associated Hepatocellular Carcinoma

        Aims: Survival after liver resection of hepatocellular carcinoma (HCC) remains poor due to a high incidence of recurrence. We sought to investigate risk factors, patterns, and long-term prognosis among patients with early and late recurrence after liver resection for hepatitis B virus (HBV)-associated HCC. Methods: Data of consecutive patients undergoing curative resection for HBV-associated HCC were analyzed. According to the time to recurrence after surgery, recurrence was divided into early (≤ 2 years) and late recurrence (> 2 years). Characteristics, patterns of initial recurrence and post-recurrence survival (PRS) were compared between patients with early and late recurrence. Risk factors of early and late recurrence, and predictors of PRS were identified by univariable and multivariable Cox-regression analyses. Results: mong 894 patients, 322 (36.0%) and 282 (31.5%) developed early and late recurrence, respectively. On multivariable analyses preoperative HBV-DNA>104 copies/ml was associated with both early and late recurrence, while postoperative no/ irregular antiviral therapy was associated with late recurrence. Compared with patients with late recurrence, patients with early recurrence had a lower proportion of intrahepatic only recurrence (72.0% vs. 91.1%, P<0.001), as well as a lower chance of receiving potentially-curative treatments for recurrence (33.9% vs. 50.7%, P<0.001) and a worse median PRS (19.1 vs. 37.5 months, P<0.001). Multivariable analysis demonstrated that early recurrence was independently associated with worse PRS (HR 1.361, 95%CI 1.094-1.692, P=0.006). Conclusions: Although risk factors associated with early recurrence and late recurrence were different, a high preoperative HBV-DNA load was an independent hepatitis-related risk for both early and late recurrence.       • Polymorphisms in Genes of the De Novo Lipogenesis Pathway and Overall Survival of Hepatocellular Carcinoma Patients Undergoing Transarterial Chemoembolization

        Aberrant expression of genes in de novo lipogenesis (DNL) pathway were associated with various cancers, including hepatocellular carcinoma (HCC). Single nucleotide polymorphisms (SNPs) of DNL genes have been reported to be associated with prognosis of some malignancies. However, the effects of SNPs in DNL genes on overall survival of HCC patients receiving transarterial chemoembolization (TACE) treatment are still unknown. In present study, nine SNPs in three genes (ACLY, ACACA and FASN) in DNL pathway were genotyped using the Sequenom iPLEX genotyping system in a hospital-based cohort with 419 HCC patients treated with TACE, and their associations with HCC overall survival were evaluated by Cox proportional hazard regression analysis under three genetic models (additive, dominant and recessive). Although we did not find any significant results in total analysis (all p>0.05), our stratified data showed that SNP rs9912300 in ACLY gene was significantly associated with overall survival of HCC patients with lower AFP level and SNP rs11871275 in ACACA gene was significantly associated with overall survival of HCC patients with higher AFP level. We further identified the significant interactions between AFP level and SNP rs9912300 or rs11871275 in the joint analysis. Conclusively, our data suggest that genetic variations in genes of DNL pathway may be a potential biomarker for predicting clinical outcome of HCC patients treated with TACE.
