        Photocatalyst TiO2/WO3/GO nano-composite with high efficient photocatalytic performance for BPA degradation under visible light and solar light illumination

        A high efficient photocatalyst (TiO2/WO3/GO) was developed with tungsten trioxide doping and graphene oxide hybridization. XRD and SEM indicate the formation of crystalline anatase/rutile TiO2 and hexagonal and cubic WO3. UV–vis spectroscopy shows the excellent visible-light-responsive performance. The photocatalyst exhibits excellent photocatalytic performance for bisphenol A degradation under visible light and sunlight irradiations. Transformation by-products were identified and main degradation pathways were proposed. The photocatalytic mechanism is also proposed on the basis of the fact that charge separation is facilitated owing to photo-generated electrons transfer from TiO2 to WO3 to GO and photogenerated holes transfer from WO3 to TiO2.

        SsPsaH, a H subunit of the photosystem I reaction center of Suaeda salsa, confers the capacity of osmotic adjustment in tobacco

        Background: Abiotic stress effects agricultural production, so research on improving stress tolerance of crop is important. Suaeda salsa is a halophyte with high salt and drought tolerance and ability to desalinate saline soil and improve soil quality. Objective: To discover and utilize of salt and drought tolerance-related genes, we further investigated the mechanisms of salt and drought tolerance. Methods: Through screening a salt treated Suaeda salsa cDNA library and further cloning a H subunit of the photosystem I reaction center SsPsaH cDNA, and then the protein domain and phylogenetic analyses of PSI genes was conducted with the NCBI Blast, DNAMAN, and MotifScan programs. The S. salsa seedlings were subjected to various stress treatments and analyze expression of SsPsaH under these treatments by real-time RT-PCR. SsPsaH expression construct was introduced into S. pombe cells by electroporation and transformed into N. tabacum plants by the leaf disc transformation method. Results: A member of the H subunit of the Photosystem I reaction center (defined as SsPsaH) was obtained. The expression of SsPsaH was up-regulated by abscisic acid (ABA), salt, and drought stress treatments. Over-expressing SsPsaH in recombinant yeasts enhanced high salinity tolerance and increased tolerance to sorbitol during seed germination and seedling root development in tobacco, respectively. Some stress-related mark genes such as a LEA family gene of NtLEA, a binding protein of a drought response element of NtDREB, the ascorbate peroxidase gene (NtAPX) were also up-regulated in SsPsaH overexpressing transgenic tobacco lines. Conclusions: These results show that SsPsaH may contribute to the salt and osmotic stress response of plants.

        Effects of Astragalus membranaceus roots supplementation on growth performance, serum antioxidant and immune response in finishing lambs

        Objective: Astragalus membranaceus root is a well-known traditional Chinese herbal medicine with many biological active constituents. This study was conducted to examine the effects of Astragalus membranaceus root powder (AMP) on growth performance, serum antioxidant and immune response in finishing lambs. Methods: A total of thirty-six Guangling fat-tailed ram lambs (body weight = 19±2 kg, mean±standard deviation) were randomly assigned to one of six treatments for a 40 d feeding period, with the first 10 d for adaptation. Treatments consisted of the lambs' basal diets with addition of 0, 5, 10, 15, 20, and 30 g/kg of diet of AMP. Results: Response to supplementation level of AMP was quadratic (p≤0.032) for final weight and ADG with the greatest at 10 g/kg of diet, but dry matter intake was not affected (p≥0.227) by treatments. The increase of AMP supplementation resulted in a quadratic response in contents of triglyceride and creatinine (p<0.05), with the lowest values for 10 and 20 g/kg of diet, respectively. A linear and quadratic decrease was observed in activity of alkaline phosphatase in serum of lambs. As the AMP supplementation increased, the activities of total superoxide dismutase and total antioxidant capacity increased linearly (p≤0.018) and hydroxyl radical (OH<sup>-</sup>) decreased linearly (p = 0.002). For catalase (CAT) and malondialdehyde (MDA), quadratic (p≤0.001) effects were observed among treatments, with the greatest CAT and lowest MDA values at 10 g/kg AMP. Additionally, supplementing AMP up to a level of 10 or 15 g/kg of diet quadratically increased immunoglobulin and interleukin contents in the serum. Conclusion: The results indicated that AMP can be used as natural feed additive in the ration of lambs to improve ADG, antioxidant status, and immune functions, and the optimal dose was 10 g/kg of diet under the condition of this experiment.

        The relationship between odd- and branched-chain fatty acids and microbial nucleic acid bases in rumen

        Objective: This study aims to identify the relationship between odd- and branched-chain fatty acids (OBCFAs) and microbial nucleic acid bases in the rumen, and to establish a model to accurately predict microbial protein flow by using OBCFA. Methods: To develop the regression equations, data on the rumen contents of individual cows were obtained from 2 feeding experiments. In the first experiment, 3 rumen-fistulated dry dairy cows arranged in a $3{\times}3$ Latin square were fed diets of differing forage to concentration ratios (F:C). The second experiment consisted of 9 lactating Holstein dairy cows of similar body weights at the same stage of pregnancy. For each lactation stage, 3 cows with similar milk production were selected. The rumen contents were sampled at 4 time points of every two hours after morning feeding 6 h, and then to analyse the concentrations of OBCFA and microbial nucleic acid bases in the rumen samples. Results: The ruminal bacteria nucleic acid bases were significantly influenced by feeding diets of differing forge to concentration ratios and lactation stages of dairy cows (p<0.05). The concentrations of OBCFAs, especially odd-chain fatty acids and C15:0 isomers, strongly correlated with the microbial nucleic acid bases in the rumen (p<0.05). The equations of ruminal microbial nucleic acid bases established by ruminal OBCFAs contents showed a good predictive capacity, as indicated by reasonably low standard errors and high R-squared values. Conclusion: This finding suggests that the rumen OBCFA composition could be used as an internal marker of rumen microbial matter.

        vfr, A Global Regulatory Gene, is Required for Pyrrolnitrin but not for Phenazine-1-carboxylic Acid Biosynthesis in Pseudomonas chlororaphis G05

        In our previous study, pyrrolnitrin produced in Pseudomonas chlororaphis G05 plays more critical role in suppression of mycelial growth of some fungal pathogens that cause plant diseases in agriculture. Although some regulators for pyrrolnitrin biosynthesis were identified, the pyrrolnitrin regulation pathway was not fully constructed. During our screening novel regulator candidates, we obtained a white conjugant G05W02 while transposon mutagenesis was carried out between a fusion mutant G05ΔphzΔprn::lacZ and E. coli S17- 1 (pUT/mini-Tn5Kan). By cloning and sequencing of the transposon-flanking DNA fragment, we found that a vfr gene in the conjugant G05W02 was disrupted with mini-Tn5Kan. In one other previous study on P. fluorescens, however, it was reported that the deletion of the vfr caused increased production of pyrrolnitrin and other antifungal metabolites. To confirm its regulatory function, we constructed the vfr-knockout mutant G05Δvfr and G05ΔphzΔprn::lacZΔvfr. By quantifying β-galactosidase activities, we found that deletion of the vfr decreased the prn operon expression dramatically. Meanwhile, by quantifying pyrrolnitrin production in the mutant G05Δvfr, we found that deficiency of the Vfr caused decreased pyrrolnitrin production. However, production of phenazine-1-carboxylic acid was same to that in the wild-type strain G05. Taken together, Vfr is required for pyrrolnitrin but not for phenazine-1-carboxylic acid biosynthesis in P. chlororaphis G05.

        Numerical Investigations of Turbulent Flows around Hydrofoil by Using Implicit Large Eddy Simulation

        Based on Implicit Large Eddy Simulation (ILES), the Adaptive Local Deconvolution Method (ALDM) was proposed to develop an implicit Sub-Grid Scale (SGS) model for three-dimensional (3D) turbulent unsteady flow around the NACA0012 Hydrofoil at a high Reynolds number of 2400,000. With the help of User Define Function (UDF) of the Fluent CFD software, the 3D turbulent unsteady flows were carried out by using ILES and LES respectively. The lift coefficients predicted by both models were in good agreement with the experimental data and the relative error of lift coefficient predicted by both models decreases firstly and then increases with the increase of the incident angle, but the numerical results by ILES were more close to the experimental, which verified the feasibility and accuracy of the ILES model. In addition, the predicted pressure contour and streamline distributions by ILES and LES at different incident angles in a wide range were also investigated further. The larger of the incident angle, the production and shedding of the vortex are stronger. The predicted results by both the models have the similar tendency, and the ILES could more finely capture the flow patterns. For a large incident angle of 20°, the internal flow analyses by ILES could well reproduce the whole process of the vortex from generation to development and shedding around the hydrofoil.
