        MicroRNA analysis reveals the role of miR-214 in duck adipocyte differentiation

        Objective: Fat deposition in poultry is an important factor in production performance and meat quality research. miRNAs also play important roles in regulating adipocyte differentiation process. This study was to investigate the expression patterns of miRNAs in duck adipocytes after differentiation and explore the role of miR-214 in regulating carnitine palmitoyltransferases 2 (CPT2) gene expression during duck adipocyte differentiation. Methods: Successful systems for the isolation, culture, and induction of duck primary fat cells was developed in the experiment. Using Illumina next-generation sequencing, the miRNAs libraries of duck adipocytes were established. miRanda was used to predict differentially expressed (DE) miRNAs and their target genes. The expression patterns of miR-214 and CPT2 during the differentiation were verified by quantitative real-time polymerase chain reaction and western blot. Luciferase reporter assays were used to explore the specific regions of CPT2 targeted by miR-214. We used a miR-214 over-expression strategy in vitro to further investigate its effect on differentiation process and CPT2 gene transcription. Results: There were 481 miRNAs identified in duck adipocytes, included 57 DE miRNA candidates. And the 1,046 targets genes of DE miRNAs were mainly involved in p53 signaling, FoxO signaling, and fatty acid metabolism pathways. miR-214 and CPT2 showed contrasting expression patterns before and after differentiation, and they were selected for further research. The expression of miR-214 was decreased during the first 3 days of duck adipocytes differentiation, and then increased, while the expression of CPT2 increased both in the transcriptional and protein level. The luciferase assay suggested that miR-214 targets the 3’untranslated region of CPT2. Overexpression of miR-214 not only promoted the formation of lipid droplets but also decreased the protein abundance of CPT2. Conclusion: Current study reports the expression profile of miRNAs in duck adipocytes differentiated for 4 days. And miR-214 has been proved to have the regulator potential for fat deposition in duck.         Effect of Lateral Boundary Scheme on the Simulation of Tropical Cyclone Track in Regional Climate Model RegCM3

        The effect of the lateral boundary scheme in regional climate model (RCM) on the track simulation of tropical cyclone (TC) was investigated using RegCM3, for the case of Winnie (1997), which formed in the Western Pacific and landed on China in August 1997. The results show that there is an inevitable simulation error in the track of Winnie, and the narrower buffer zone size (BZS) will make a great error. However, it was demonstrated that a much broader BZS does not allow a better track simulation of Winnie, and the optimal BZS does not reduce the track error substantially. Moreover, the configuration scheme of nudging parameters plays an important role in the track simulation, and different nudging parameter configuration scheme could make the root mean square errors (RMSEs) of simulated track by more than two times. Nevertheless, the optimal configuration scheme can reduce the track error effectively by maintaining the equilibrium between the two additional nudging terms in the prognostic equations in the buffer zone, whereas both the strong nudging scheme and the weak nudging scheme distort the track simulation of the Winnie. It is also found that the simulated weaker west Pacific subtropical high (WPSH), which leads to the turning of the TC ahead of time, is the reason for the track simulation error. A possible approach for reducing track simulation error of TCs is also discussed.

        Effects of ultrasonic treatment on the gel properties of microbial transglutaminase crosslinked soy, whey and soy–whey proteins

        This paper studied the influences of diverse ultrasonic power treatments on the physico-chemical properties of soy–whey mixed protein induced by microbial transglutaminase. Two groups of 15% (m/v) of protein solution-sole protein (as control group) and mixed protein were prepared and processed under different ultrasonic powers for 30 min. After ultrasonic power treatments, gel properties were significantly increased: under 300 W, the gel hardness of mixed protein reached a maximum of 998.9 g, with its water binding capacity scoring a maximum of 87%. According to the analysis of fluorescence emission spectrum, the fluorescence intensity and maximum absorption peak had changed, for different ultrasonic power treatments had exposed more groups. The Fourier Transform Infrared Spectroscopy also suggested that ultrasonic power treatments could change the secondary structure of gel samples. The scanning electron microscope demonstrated that the network structure of mixed protein gel displayed more regular and uniform after ultrasonic treatments.

      • Application of Smart Technology in the Integrated Environmental Management of Urban Wetland Park

        Wetland Park is one of the important ecological infrastructures. Besides providing the ecosystem service function and value, it is also an important place for leisure and entertainment. Efficient environmental management is an important measure to promote the sustainable development of the wetland park. By combining the smart technology with routine management method, the efficiency of the environmental management for the urban wetland park can be improved. The application of smart technology set the users' requirements as the basic goal, which is divided into four layers: perception layer, transport layer, support layer and application layer. The perception layer consists of sensor, camera, RFID, mobile terminal device and so on; the transport layer is composed of optical fiber, WIFI and 3G network; the support layer is composed of data management system, information open platform and service support system; and the application layer is composed of four modules including panoramic-view park, environmental maintenance, smart tourism and public participation. A wetland park located in Zhangzhou city, Fujian, China, was taken as a practice case.

        MiR-1297 and MiR-26a-5p Inhibit Cell Progression of Keratinocytes in Cholesteatoma Depending on the Regulation of BMI1

        Cholesteatoma is a pathologically benign but clinically destructive middle ear disease characterized by hyperproliferative keratinocytes. B-cell-specific Moloney murine leukemia virus insertion site 1 (BMI1) has been reported to be upregulated in cholesteatoma tissues. This study aimed to explore the biological role and underlying mechanisms of BMI1 in the progression of cholesteatoma. The expression levels of microRNA (miR)-1297, miR-26a- 5p, and BMI1 in cholesteatoma tissues and cells were examined by quantitative real-time polymerase chain reaction (qRT-PCR) or western blot. Functional experiments were performed by CCK-8 assay for cell proliferation viability, 5-ethynyl-2'deoxyuridine (EdU) incorporation assay for DNA biosynthesis, colony formation assay for cloning forming ability analysis, transwell assay and wound healing assay for cell metastasis, flow cytometry for cell cycle distribution and cell apoptosis. The protein expression of apoptosis-associated proteins was investigated by western blot. Dual-luciferase reporter assay was conducted to verify the interaction between miR-1297 or miR-26a-5p and BMI1. BMI1 was highly expressed in cholesteatoma tumor tissues. Functional analyses showed that BMI1 knockdown could inhibit the proliferation, colony formation, migration, invasion, cell cycle progression and promoted the apoptosis of keratinocytes. Mechanically, BMI1 was a target of miR-1297 and miR-26a-5p. Moreover, the rescue experiments presented that BMI1 addition could abolish the suppressive effects of miR-1297 or miR-26a-5p overexpression on cell malignant behaviors in keratinocytes. BMI1 could exert an oncogenic role in the malignant development of cholesteatoma through serving as the targets of miR-1297 and miR-26a-5p, which might provide novel strategies for cholesteatoma treatment.

        Correlation of NUF2 Overexpression with Poorer Patient Survival in Multiple Cancers

        Purpose NUF2 has been implicated in multiple cancers recently, suggesting NUF2 may play a role in the common tumorigenesis process. In this study, we aim to perform comprehensive meta-analysis of NUF2 expression in the cancer types included in the Cancer Genome Atlas (TCGA). Materials and Methods RNA-sequencing data in 31 cancer types in the TCGA data and 11 independent datasets were used to examine NUF2 expression. Silencing NUF2 using targeting shRNAs in hepatocellular carcinoma (HCC) cell lines was used to evaluate NUF2’s role in HCC in vitro and in vivo. Results NUF2 up-regulation is significantly observed in 23 out of the 31 cancer types in the TCGA datasets and validated in 13 major cancer types using 11 independent datasets. NUF2 overexpression was clinically important as high NUF2 was significantly associated with tumor stages in eight different cancers. High NUF2 was also associated with significantly poorer patient overall survival and disease-free survival in eight and six cancers, respectively. We proceeded to validate NUF2 overexpression and its negative association with overall survival at the protein level in an independent cohort of 40 HCC patients. Compared to the non-targeting controls, NUF2 knockdown cells showed significantly reduced ability to grow, migrate into a scratch wound and invade the 8 μm porous membrane in vitro. Moreover, NUF2 knockdown cells also formed significantly smaller tumors than control cells in mouse xenograft assays in vivo. Conclusion NUF2 up-regulation is a common feature of many cancers. The prognostic potential and functional impact of NUF2 up-regulation warrant further studies.

        <P>Soft and conformable wearable electronics require stretchable semiconductors, but existing ones typically sacrifice charge transport mobility to achieve stretchability. We explore a concept based on the nanoconfinement of polymers to substantially improve the stretchability of polymer semiconductors, without affecting charge transport mobility. The increased polymer chain dynamics under nanoconfinement significantly reduces the modulus of the conjugated polymer and largely delays the onset of crack formation under strain. As a result, our fabricated semiconducting film can be stretched up to 100% strain without affecting mobility, retaining values comparable to that of amorphous silicon. The fully stretchable transistors exhibit high biaxial stretchability with minimal change in on current even when poked with a sharp object. We demonstrate a skinlike finger-wearable driver for a light-emitting diode.</P>

        An electrochemical sensor based on [Ru(bpy)2dpp]2+/SMWCNTs/Au modified glassy carbon electrode for the detection of 5ʹ‑GMP

        A sensitive electrochemical sensor for the selective detection of 5ʹ-guanylic acid (5ʹ-GMP) was prepared by combining sulfonated-multiwalled carbon nanotubes (SMWCNTs) and [Ru(bpy)2dpp]Cl2, which were dripped on the surface of a glass carbon electrode (GCE) immobilized with gold nanoparticles. The 5ʹ-GMP electrochemical biosensor was fabricated using [Ru(bpy)2dpp]2+/SMWCNTs/Au/GCE as working, Ag/AgCl as reference and Pt as auxiliary electrode connected by an electrochemical workstation. The modified electrode was characterized by cyclic voltammetry (CV) and electrochemical impedance spectroscopy (EIS). The results showed the sensor’s response current had the best peak shape and maximum peak when the pH of electrolyte was 3, scan speed of CV was in the range of 100 to 180 mV/s, and the enrichment time was in the range of 200 to 300 s. Under the optimum conditions, a linear analytical curve was obtained for 5ʹ-GMP concentrations in the range of 0.01 to 0.5 mmol L− 1, with a detection limit of 0.0014 mmol L− 1. The analytical results of the 5ʹ-GMP sensor were exhibited good consistent with the data from liquid chromatography. The sensor has good reproducibility, long-term stability and strong immunity to interference, and may be a powerful device for 5ʹ-GMP detection, with great advantages such as simple preparation and operation, low equipment cost.
