Over-expression of SlWRKY39 leads to enhanced resistance to multiple stress factors in tomato

        Xiao-chun Sun,Yong-feng Gao,Hui-rong Li,Shu-zhang Yang,Yong-sheng Liu 한국식물학회 2015 Journal of Plant Biology Vol.58 No.1

The WRKY transcription factors are one of the well-characterized classes of plant transcription factors, which participated in various biotic and abiotc stress responses. Previous study showed that there are 81 WRKY genes in tomato, wherein a number of SlWRKY genes including SlWRKY39 were significantly up-regulated under salt, drought stress and PstDC3000 infection. However little is known about their physiological role in tomato. In this study, by using a forward genetic approach, we demonstrated transgenic plants over-expressing SlWRKY39 showing enhanced resistance to multiple stress factors including PstDC3000, salt and drought. Transgenic plants accumulated higher level of proline and lower level of malonic dialdehyde. Compared with wild type, the expression of pathogenesis-related genes SlPR1, SlPR1a1 and environmental stress related genes SlRD22, SlDREB2A were up-regulated in the transgenic plants. These results indicated that SlWRKY39 is a positive regulatory component of tomato against biotic and abiotic stress probably via activating the expression of both pathogenesis-related genes and stress related genes.

Effects of Oxygen Content on the Simultaneous Microbial Removal of SO2 and NOx in Biotrickling Towers

        Xing Chun Wang,Xiao Yi Bi,Pei Shi Sun,Jin Quan Chen,Ping Zou,Xiao Ming Ma,Jing Zhang,Hai Yu Wang,Xiao Yi Xu 한국생물공학회 2015 Biotechnology and Bioprocess Engineering Vol.20 No.5

This study uses microbial methods to research the influence of oxygen (O2) content on the removal efficiency of sulfur dioxide (SO2) and nitrogen oxides (NOx) in a tandem twin-towers desulfurization and denitrification process system. Oxygen can play a significant role in biotrickling towers. Other important factors had already been optimized prior to the study, including inlet concentration, gas flow rate, and temperature. SO2 and NOx were prepared by a chemical method. A gas flow meter was used to regulate nitrogen (N2) that had been stored in steel cylinders. In this way, the O2 content was adjusted in the biotrickling towers by controlling the N2 flow rate. Five gradients of O2 content were selected for investigation, namely 4, 8, 12, 16, and 20%. Results indicated that the SO2 removal efficiency from mixed gas (SO2 and NOx) can reach 100%, from all of the five O2 gradients, in biotrickling towers. In a tandem twin-towers desulfurization and denitrification process system, the NOx removal efficiency and the inlet concentration of nitrogen dioxide (NO2) gradually increased as the O2 content increased. Specifically, the average removal efficiency of NOx increased from 49.28 to 80.85% as the O2 content changed from 4 to 20%. The oxygen levels influenced the removal of NOx but the SO2 removal efficiency in mixed gas was always stable.

Resistance Function of Rice Lipid Transfer Protein LTP110

( Xiao Chun Ge ),( Ji Chao Chen ),( Ning Li ),( Yi Lin ),( Chong Rong Sun ),( Kai Ming Cao ) 생화학분자생물학회 2003 BMB Reports Vol.36 No.6

Plant lipid transfer proteins (LTPs) are a class of proteins whose functions are still unknown. Some are proposed to have antimicrobial activities. To understand whether LTP110, a rice LTP that we previously identified from rice leaves, plays a role in the protection function against some serious rice pathogens, we investigated the antifungal and antibacterial properties of LTP110. A cDNA sequence, encoding the mature peptide of LTP110, was cloned into the Impact-CN prokaryotic expression system. The purified protein was used for an in vitro inhibition test against rice pathogens, Pyricularia oryzae and Xanthomonas oryzae. The results showed that LTP110 inhibited the germination of Pyricularia oryzae spores, and its inhibitory activity decreased in the presence of a divalent cation. This suggests that the antifungal activity is affected by ions in the media; LTP110 only slightly inhibited the growth of Xanthomonas oryzae. However, the addition of LTP110 to cultured Chinese hamster ovarian cells did not retard growth, suggesting that the toxicity of LTP110 is only restricted to some cell types. Its antimicrobial activity is potentially due to interactions between LTP and microbe-specific structures.

      • ANXA2 Regulates the Behavior of SGC-7901 Cells

        Sun, Meng-Yao,Xing, Rui-Huan,Gao, Xiao-Jie,Yu, Xiang,He, Hui-Min,Gao, Ning,Shi, Hong-Yan,Hu, Yan-Yan,Wang, Qi-Xuan,Xu, Jin-Hui,Hou, Ying-Chun Asian Pacific Journal of Cancer Prevention 2013 Asian Pacific journal of cancer prevention Vol.14 No.10

ANXA2, a member of the annexin family, is overexpressed and plays important roles in tumor development. However, the significance of ANXA2 expression in gastric carcinoma has not been clarified. To elucidate its roles in growth of gastric cancer, ANXA2 expression in SGC-7901 cells was inhibited with a designated siRNA, then cell proliferation, cell cycling, apoptosis and motility were determined by MTT assay, flow cytometry, Hoechst 33342 staining and wound healing assay, respectively. To further assess the behavior of ANXA2 deleted SGC-7901 cells, changes of microstructures were observed under fluorescence microscopy, laser scanning confocal microscopy and electron microscopy. We found that inhibition of ANXA2 expression caused cell proliferation to decrease significantly with G1 arrest, motility to be reduced with changes in pseudopodia/filopodia structure and F-actin and ${\beta}$-tubulin expression, and apoptosis to be enhanced albeit without significance. At the same time, ANXA2 deletion resulted in fewer pseudopodia/filopodia, non-stained areas were increased, contact inhibition among cells reappeared, and expression of F-actin and ${\beta}$-tubulin was decreased, with induction of polymerized disassembled forms. Taken together, these data suggest that ANXA2 overexpression is important to maintain the malignancy of cancer cells, and this member of the annexin family has potential to be considered as a target for the gene therapy of gastric carcinoma.

      • Association Between XRCC1 Gene Polymorphisms and Risk of Glioma Development: A Meta-analysis

        Sun, Jian-Ying,Zhang, Chun-Yang,Zhang, Zhen-Jun,Dong, Yan-Fang,Zhang, An-Long,Wang, Zhi-Wei,Mei, Xiao-Long Asian Pacific Journal of Cancer Prevention 2012 Asian Pacific journal of cancer prevention Vol.13 No.9

Objective: Previous studies of the association between X-ray cross-complementing group 1 (XRCC1) gene polymorphisms and the gliomas risk have yielded conflicting results, and thus a meta-analysis was performed to provide a more accurate estimation. Methods: A computerized literature search of 5 electronic databases was conducted to identify the relevant studies. Fixed or random effect models were selected based on the heterogeneity test. Publication bias was estimated using Begg's funnel plots and Egger's regression test. Results: A total of 11 studies (3,810 cases and 6,079 controls), 7 studies (2,928 cases and 5,048 controls), and 4 studies (1,461 cases and 2,593 controls) were finally included in the analyses of the association between XRCC1 Arg399Gln, Arg194Trp, and Arg280His polymorphisms and glioma risk, respectively. The pooled results showed that GlnGln carriage was associated with moderately increased risk of gliomas in Asians (GlnGln vs. ArgArg, OR=1.490, 95%CI 1.031-2.153; GlnGln/ArgGln vs. ArgArg, OR=1.321, 95%CI 1.037-1.684), whereas a marginal association was revealed in Caucasians. For the Arg194Trp polymorphism, although a significant association was shown in the homozygous genotype comparisons (TrpTrp vs. ArgArg, OR = 2.209, 95%CI 1.398-2.945), no significant link was found on subgroup analysis stratified by ethnicity. With regard to the Arg280His polymorphism, no significant association was found in each comparison. No particular study was found to significantly influence the pooled results, and no potential publication bias was detected. Conclusions: This meta-analysis suggested that the XRCC1 Arg399Gln polymorphism is moderately associated with increased risk of gliomas in Asians, while Arg194Trp and Arg280His polymorphisms demonstrated no significant influence. Due to the limited studies and the potential confounders, further studies are needed to confirm these results.

      • Ultra-sensitive graphene based mid-infrared plasmonic bio-chemical sensing using dielectric beads as a medium

        Liu, Xiao,Zhang, Duan,Wu, Ye-Cun,Yang, Mei,Wang, Qian,Coileá,in, Cormac Ó,.,Xu, Hongjun,Yang, Chen,Abid, Mohamed,Abid, Mourad,Liu, Huajun,Chun, Byong Sun,Shi, Qingfan,Wu, Han-Chun Elsevier 2017 Carbon Vol.122 No.-

<P>Graphene is moving beyond the realm of simple electronic devices toward areas such as advanced biochemical sensing. The infrared (IR) response of graphene, characterized by collective long-lived charge-carrier oscillations, could be applied in IR-absorption spectroscopy, typically used for bio-chemical analysis. However, direct light absorption by propagating plasmons in graphene is forbidden due to the large momentum mismatch. Proposed methods to overcome this bottleneck come at a cost, the use of noble metal particles on graphene reduces the spectral bandwidth and nano-structuring graphene is expensive. Here, we propose a simple and cheap method to fabricate large scale ultra-sensitive graphene based mid-IR biosensors, by introducing dielectric beads to excite mid-IR range plasmons. Interference from waves scattered by the beads excite surface plasmon polaritons, which propagate several micrometers in graphene and enhance the interaction between the molecules and mid-IR light. This method opens an interesting window for the application of graphene in bio-chemical sensing. (C) 2017 Elsevier Ltd. All rights reserved.</P>

Selection signature reveals genes associated with susceptibility loci affecting respiratory disease due to pleiotropic and hitchhiking effect in Chinese indigenous pigs

        Xu, Zhong,Sun, Hao,Zhang, Zhe,Zhang, Cheng-Yue,Zhao, Qing-bo,Xiao, Qian,Olasege, Babatunde Shittu,Ma, Pei-Pei,Zhang, Xiang-Zhe,Wang, Qi-Shan,Pan, Yu-Chun Asian Australasian Association of Animal Productio 2020 Animal Bioscience Vol.33 No.2

Objective: Porcine respiratory disease is one of the most important health problems causing significant economic losses. To understand the genetic basis for susceptibility to swine enzootic pneumonia (EP) in pigs, we detected 102,809 single nucleotide polymorphisms in a total of 249 individuals based on genome-wide sequencing data. Methods: Genome comparison of susceptibility to swine EP in three pig breeds (Jinhua, Erhualian, and Meishan) with two western lines that are considered more resistant (Duroc and Landrace) using cross-population extended haplotype homozygosity and F-statistic (FST) statistical approaches identified 691 positively selected genes. Based on quantitative trait loci, gene ontology terms and literature search, we selected 14 candidate genes that have convincible biological functions associated with swine EP or human asthma. Results: Most of these genes were tested by several methods including transcription analysis and candidate genes association study. Among these genes: cytochrome P450 1A1 and catenin beta 1 (CTNNB1) are involved in fertility; transforming growth factor beta receptor 3 plays a role in meat quality traits; Wnt family member 2, CTNNB1 and transcription factor 7 take part in adipogenesis and fat deposition simultaneously; plasminogen activator, urokinase receptor (completely linked to AXL receptor tyrosine kinase, r2 = 1) plays an essential role in the successful ovulation of matured oocytes in pigs; colipase like 2 (strongly linked to SAM pointed domain containing ETS transcription factor, r2 = 0.848) is involved in male fertility. Conclusion: These adverse genes susceptible to swine EP may be selected while selecting for economic traits (especially reproduction traits) due to pleiotropic and hitchhiking effect of linked genes. Our study provided a completely new point of view to understand the genetic basis for susceptibility or resistance to swine EP in pigs thereby, provides insight for designing sustainable breed selection programs. Finally, the candidate genes are crucial due to their potential roles in respiratory diseases in a large number of species, including human.

Conductivity relaxation in NiTiO3 at high temperatures

        Teng Li,Chun Chang Wang,Chang Mei Lei,Xiao Hong Sun,Guo Jing Wang,Liu Na Liu 한국물리학회 2013 Current Applied Physics Vol.13 No.8

NiTiO3 ceramics were prepared via the traditional solid-state reaction route. The dielectric properties of NiTiO3 ceramics have been systematically investigated in the temperature range from room temperature to 1073 K NiTiO3 ceramics exhibit intrinsic dielectric response in the temperature range below 400 K. Two relaxations were observed in the temperature range higher than 400 K. The relaxation activation energy is 0.95 eV and 1.17 eV for the low- and high-temperature relaxations, respectively. Our results strongly indicate that the two relaxations are related to conductivity relaxation associated with the singly and doubly ionized oxygen vacancies.
