Background: It is well-established that serum testosterone in men decreases with age, yet the underlying mechanism of this changeremains elusive. Methods: The expression patterns of Fancd2 opposite-strand (Fancd2os) in BALB/c male mice and testicular tissue derived celllines (GC-1, GC-2, TM3, and TM4) were assessed using real-time polymerase chain reaction (RT-PCR), Western blot and immunofluorescence. The Fancd2os-overexpressing or knockdown TM3 cells were constructed by infecting them with lentivirus particlesand were used to evaluated the function of Fancd2os. The testosterone production was measured using enzyme linked immunosorbent assay (ELISA) and the steroidogenic enzymes such as steroidogenic acute regulatory protein (StAR), P450 cholesterol sidechain cleavage (P450scc), and 3β-hydroxysteroid dehydrogenase (3β-HSD) were analysed using RT-PCR. The apoptosis of TM3cells induced by ultraviolet light or testicular tissues was detected using flow cytometry, Western blot or dUTP-biotin nick end labeling (TUNEL) assays. Pearson correlation analysis was used to assess the correlation between the Fancd2os expression and TUNELpositive staining in mouse testicular Leydig cells. Results: The Fancd2os protein was predominantly expressed in mouse testicular Leydig cells and its expression increased with age. Fancd2os overexpression inhibited testosterone levels in TM3 Leydig cells, whereas knockdown of Fancd2os elevated testosteroneproduction. Fancd2os overexpression downregulated the levels of StAR, P450scc and 3β-HSD, while Fancd2os knockdown reversed this effect. Fancd2os overexpression promoted ultraviolet light-induced apoptosis of TM3 cells. In contrast, Fancd2os knockdown restrained apoptosis in TM3 cells. In vivo assays revealed that higher Fancd2os levels and mouse age were associated with increased apoptosis in Leydig cells and decreased serum testosterone levels. Pearson correlation analysis exhibited a strong positivecorrelation between the expression of Fancd2os and TUNEL-positive staining in mouse testicular Leydig cells. Conclusion: Our findings suggest that Fancd2os regulates testosterone synthesis via both steroidogenic enzymes and the apoptoticpathway.

Key events in football videos are fetched on the basis of accurate detection of shots. A quick and effective algorithm for detecting football video shots has very important meanings. Here it focuses on features of football videos by combining with a few basic shot detection methods to propose one shot detection method applicable for football videos. The experiment shows the feasibility of the proposed algorithm, and the experimental data shows that the algorithm has a good effect for soccer video shot detection and key frame extraction

Football is one of the most popular sports in the world. It is deeply loved by massive ball fans. Generally one complete football game lasts around two hours, but only a very little part of it has wonderful views which attract audiences. Although in sport matches, episodes which people care about are of high subjectivity, for football games, they share some in common. . So how to fetch out concerned events from tremendous football videos has become a very important and typical problem. Here we propose a new algorithm for extracting effectively key events from football videos, which is named key event extraction method based on attributes of shots. The method proves higher recall and precision rate.

A series of dimethine cyanine dyes were synthesized in a fast, efficient and high yield by the condensation of quaternary salts with 1H-indole-3-carbaldehyde in the presence of piperidine under solvent-free microwave irradiation. The products were identified by 1H NMR, IR, UV-Vis spectra and elemental analysis. The absorption and fluorescence properties of these dyes were investigated both experimentally and theoretically. Calculations performed at a combination of time-dependent density functional theory (TD-DFT) and the polarizable continuum model (PCM) reproduced the p-p* type absorption bands of the dyes. Regression analysis was used for studying theoretical results of the absorption maxima in different solvents. Compared with experimental counterparts, estimated overall uncertainties in the absorption maxima were about ?2%.

In the present study, reverse microemulsion was prepared to regulate the mineralization of calcium phosphate. Calcium chlorideand sodium hydrogen phosphate aqueous solution were, respectively, dropped into the mixture of Span 80, Tween 80, andn-heptaneto get two kinds of emulsions.n-Buranol was used to adjust the emulsions to transparent state. Calcium phosphate was prepared byadding phosphate microemulsion to Calcium microemulsion. Heating method was used to induce the phase separation. After beingwashed with acetone and ethanol, the mineralized deposition were studied with scanning electron microscopy (SEM), X-ray dirac-tion (XRD) and transmission electron microscopy (TEM). The results indicated that the minerals in the deposition were mainlymonetite (DCPA) and the nanosized needle-like DCPA exhibited the preferential orientation along 200 direction..

To investigate the antioxidant effect of vitexin-4"-O-glucoside, a flavone glycoside, isolated from the leaves of Crataegus pinnatifida Bge. var. major, we developed a simple and sensitive high-performance liquid chromatography (HPLC) method to determine levels of malondialdehyde (MDA) in ECV304 cell culture medium after induction by tert-butyl-hydroperoxide (TBHP). The preparation of analyzed samples involved a one-step derivatization with thiobarbituric acid (TBA). HPLC analysis was performed on a SynergiTM Hydro-RP, a polar end-capped C18 column (250×4.6 mm, 4 μm), using an acetonitrile-ammonium acetate aqueous solution (10 mM, pH 6.8) as the mobile phase under linear gradient conditions with UV detection at 532 nm. The calibration curve was linear over 0.0125-1.25 μM MDA (r = 0.9951). Relative standard deviations (RSDs) of intra-day and inter-day precision were less than 6.1% and 5.0%, respectively. The mean recovery was 96.9 ± 1.6%. The lower limit of quantification (LLOQ) of MDA was 0.0125 μM. This chromatographic method was successfully applied to investigating the in vitro antioxidant effect of vitexin-4"-O-glucoside. Vitexin-4"-O-glucoside (120 M) protected ECV304 cells from peroxidation induced by TBHP.

Stable and Recyclable SERS Substrates Based on Au-Loaded PET Nanocomposite Superhydrophobic Surfaces

Novel surface-enhanced Raman scattering (SERS) substrates with stable and recyclable properties have been prepared by assembling gold nanoparticles-loaded PET (AuNPs/PET) nanocomposite superhydrophobic surfaces. After a physical vapor deposition process, the AuNPs/PET surfaces with vast plasmonic "hot spots" showed superhydrophobic properties, and it can hold target molecules droplets for rapid SERS detection. From blown off droplets and rinsed substrates with water after detection, we found that no probe molecules remained on the surfaces from Raman spectra. The prepared substrates were not contaminated in the detection process. Furthermore, the new SERS substrates were used for rapidly detecting droplets of crystal violet (CV) and the lowest detection concentration was about 1 x 10 -9 M. The as-prepared AuNPs/PET substrates also have good performance in terms of reproducibility and recyclability.
