        Characterization of a novel variant HMW‐glutenin gene from Elymus canadensis

        High molecular weight (HMW) glutenin subunits (GS) play a key role in the determination of end‐use quality of wheat and other cereal crops. In this study, we report the isolation and characterization of both promoter region and ORF of novel HMW‐GS allele 1St1.3 from a perennial Triticeae species,Elymus canadensis. The amino acid (AA) sequences of E. canadensis 1St1.3 were deduced as 434 aa. Its protein primary structure comprises a signal peptide with a conserved N‐terminal domain, a central repetitive domain and a C‐terminal domain. E. canadensis 1St 1.3 possesses several distinct characteristics which are different from those of wheat HMW‐GSs. The N‐terminal domains of E. canadensis 1St 1.3 resemble that of y‐type subunits, while their C‐terminal domains are more similar to x‐type subunits. The deletion of 85 bp fragment has been observed in promoter region of 1St 1.3, however which has not interrupted the expression of this gene. Our results indicate that 1St 1.3 is novel HMW‐GS variants which will be valuable for enhancing our understanding of structural differentiation and the evolutionary relationship among HMW‐GSs in Triticeae species.

        High Temperature Deformation Behavior and Processing Map for a Nickel-Titanium Shape Memory Alloy

        The hot deformation behavior of 49.2Ti-50.8Ni shape memory alloy was studied using hot compressive deformationtesting in the temperature range of 1023-1323 K and at strain rates of 0.01-10 s-1. The work-hardening rate wasinduced to analyze the stress-strain curves, and the critical stress σc and the dynamic recovery saturation stress σsatwere measured which can be specified approximately by the expressions: σsat-1.12σp and σc-0.86σp. An Arrheniusmodel was calculated to describe the relationship between peak stress and the Z parameter. The relationshipbetween deformation activation energy, the deformation conditions and the effect of Ni component in a binaryTiNi alloy on the activation energy were discussed in this work. With the help of electron backscattering diffraction,a connected mode dynamic recrystallization microstructure was confirmed in peak efficiency regimes(850 °C & 0.01 s-1 and 1050 °C & 10 s-1) of the processing map.

        Almost Sure Stability of Dual Switching Continuous-time Nonlinear System With Deterministic and Stochastic Subsystems

        In this paper, the almost surely globally asymptotical stability and the almost surely exponential stability for dual switching continuous-time nonlinear system are investigated by using the probability analysis method and stochastic Multi-Lyapunov function, respectively. Different from the previous research results, it is the first time that dual switching continuous-time nonlinear system is used as a study object to investigate its switching stability. Then, the probability analysis method is used to overcome the deficiency that the ergodicity no longer holds due to the variable transition rate of Markov process. Some sufficient conditions for the globally asymptomatic stability almost surely and the almost surely exponential stability of dual switching continuous-time nonlinear system are given under the pre-designed deterministic switching strategy. Finally, two numerical examples are provided to verify the effectiveness of the proposed approach.

        The Nrf2 antioxidant defense system in intervertebral disc degeneration: Molecular insights

        Intervertebral disc degeneration (IDD) is a common degenerative musculoskeletal disorder and is recognized as a major contributor to discogenic lower back pain. However, the molecular mechanisms underlying IDD remain unclear, and therapeutic strategies for IDD are currently limited. Oxidative stress plays pivotal roles in the pathogenesis and progression of many age-related diseases in humans, including IDD. Nuclear factor E2-related factor 2 (Nrf2) is a master antioxidant transcription factor that protects cells against oxidative stress damage. Nrf2 is negatively modulated by Kelch-like ECH-associated protein 1 (Keap1) and exerts important effects on IDD progression. Accumulating evidence has revealed that Nrf2 can facilitate the transcription of downstream antioxidant genes in disc cells by binding to antioxidant response elements (AREs) in promoter regions, including heme oxygenase-1 (HO-1), glutathione (GSH), superoxide dismutase (SOD), catalase (CAT), and NADPH quinone dehydrogenase 1 (NQO1). The Nrf2 antioxidant defense system regulates cell apoptosis, senescence, extracellular matrix (ECM) metabolism, the inflammatory response of the nucleus pulposus (NP), and calcification of the cartilaginous endplates (EP) in IDD. In this review, we aim to discuss the current knowledge on the roles of Nrf2 in IDD systematically.

        A New Lupane-Triterpene Glycoside from the Leaves of Acanthopanax gracilistylus

        A new and two known lupane-triterpene glycosides were isolated from the hot MeOH fraction of the leaves of Acanthopanax gracilistylus W. W. Smith. Based on the physical properties and spectroscopic data, their chemical structures were determined as acankoreoside A (1), acankoreoside D (2), and $3{\alpha}-hydroxy-lup-23-al-20(29)-en-28-oic$ acid $28-O-{\alpha}-L-rhamnopyranosyl-(1{\rightarrow}4)-{$beta}-D-glucopyranosyl-(1{\rightarrow}6)-{\beta}-D-glucopyranosyl$ ester (3), respectively. To our best knowledge, compand 3 appears to be novel, which was named as wujiapioside A.

      • Phase II Trial of Loubo<sup>®</sup> (Lobaplatin) and Pemetrexed for Patients with Metastatic Breast Cancer not Responding to Anthracycline or Taxanes

        Purpose: This phase II study was undertaken to determine the efficacy and safety of Loubo$^{(R)}$ (Lobaplatin) in combination with pemetrexed in treating patients with metastatic breast cancer who failed to respond to anthracycline or taxanes. Patients and Methods: Metastatic breast cancer cases who had previously received an anthracycline and a taxane in either adjuvant or metastatic settings, were enrolled. All patients were recruited from Jiangsu Cancer Hospital and Research Institute, and were treated with Loubo$^{(R)}$ (Lobaplatin) 35 $mg/m^2$ (intravenous; on day 1) and pemetrexed 500 $mg/m^2$ (intravenous; on day 1) every 21 days. Efficacy and side effects were evaluated after at least two cycles of chemotherapy. Results: All eligible 19 patients completed at least 2 cycles of chemotherapy with pemetrexed and lobaplatin, and were evaluable. Overall, 3 (15.8%) patients achieved partial response, 11 (57.9%) stable disease, 5 (26.3%) progression of disease, with no complete remission. Response rate was 15.8%, disease control rate was 42.1%. The median survival time was 10.3 months. Neutrophil suppression occurred in 36.8% of patients who had grade 2 toxicity, and 26.3% had grade 3, 26.4% had grade 4. Thrombocytopenia was encountered as follows: 21.1% grade 2, 15.8% grade 3 and 5.5% grade 4. Incidences of anemia were 10.5% in grade 2, 5.3% grade 3 and 0% grade 4. Only 5.3% of patients required packed red blood cell transfusion. Grade 3 digestive tract toxicity occurred in 5.5% of patients. Other toxicities included elevated transaminase,oral mucositis and skin rashes. Conclusions: The regimen of lobaplatin and pemetrexed is modestly active in metastatic breast cancer patients who failed anthracycline or taxanes, and the toxicity profile suggesting that the doses of chemotherapy should be further modified.

      • A Novel Mutant of Human Papillomavirus Type 18 E6E7 Fusion Gene and its Transforming Activity

        Background: Persistent human papillomavirus (HPV) infection, especially with high-risk types such as HPV16 and HPV18, has been identified as the primary cause of cervical cancer. E6 and E7 are the major onco-proteins of high-risk HPVs, which are consistently expressed in HPV infected tissues but absent in normal tissues and represent ideal therapeutic targets for immunotherapy of cervical cancer. Materials and Methods: In this study, the optimized fusion gene HPV18 E6E7 (HPV18 ofE6E7) was constructed according to genetic codon usage for human genes. At the same time, for safety future clinical application, a mutant of HPV18 ofE6E7 fusion gene was generated by site-directed mutagenesis at L52G for the E6 protein and C98G for the E7 protein. Results: HPV18-E6E7 mutant (HPV18 ofmE6E7) constructed in this work not only lost the transformation capability for NIH 3T3 cells and tumorigenicity in BALB/c nude mice, but also maintained very good stability and antigenicity. Conclusion: These results suggest that the mutant should undergo further study for application as a safe antigenspecific therapeutic vaccine for HPV18-associated tumors.

        Diversity and Distribution of Bulb-associated fungi of Fritillaria Cirrhosae Bulbus Source Plants used in Traditional Chinese Medicine

        Diversity and community composition of bulb-associated fungi of Fritillaria Cirrhosae Bulbus source plants, which are used in traditional chinese medicine, in the eastern Himalaya- Hengduan Mountains, southwestern China, were estimated based on the internal transcribed spacer rDNA sequence analysis, using host plant species, geographic area, and plant phenology as variables. A total of 1,486 fungal sequences assigned to 251 operational taxonomical units (OTUs) were obtained from the bulbs. Fungal OTUs comprised 96.41% Ascomycotina, 3.52% Basidiomycotina, and 0.07% Zygomycotina. Sordariomycetes, Hypocreales, and Nectriaceae were the most frequent fungal lineages at each taxonomic rank. Fusarium, Ilyonectria, Tetracladium, Leptodontidium, and Tomentella were the top OTU-rich genera. Fusarium sp. 03, Ilyonectria rufa, Fusarium sp. 08, Ilyonectria sp. 03, and Leptodontidium orchidicola 03 represented the most frequent OTUs. Fusarium spp. were the most frequent general taxa. The distribution of fungal community exhibited preferences for host plant species, geographic area, and plant phenology. These findings are the foundation of our research on culturing and active metabolites of bulb-associated fungi of Fritillaria Cirrhosae Bulbus source plants.

        Effects of Cavity Size and Density on Polymer Micro Hot Embossing

        We designed nine micro molds with various cavity geometric parameters to investigate their effects on micropillars replication of the amorphous polymer via micro hot embossing. Picoseconds laser was used to fabricate the micro holes array on the stainless steel plates with 200 μm thickness as micro molds. The mechanical behavior of PMMA above glass transition temperature was investigated by uniaxial compression tests at various temperatures and strain rates, the results were used in simulation as constitutive relation. Confocal microscope and optical microscope showed that the replication height has a complicate nonlinear relationship with cavity size and density. DEFORM 3D was used to model and simulate the embossing process, and it is a good approach to investigate the micropallars replication heights, front angles and filling ratios.
