Metabolic Flux Analysis of Beijerinckia indica for PS-7 Production

In order to investigate central metabolic changes in Beijerinckia indica, cells were grown on different carbon sources and intracellular flux distributions were studied under varying concentrations of nitrogen. Metabolic fluxes were estimated by combining material balances with extracellular substrate uptake rate, biomass formation rate, and exopolysaccharide (EPS) accumulation rate. Thirty-one metabolic reactions and 30 intracellular metabolites were considered for the flux analysis. The results revealed that most of the carbon source was directed into the Entner-Doudoroff pathway, followed by the recycling of triose-3-phosphate back to Hexose6-phosphate. The pentose phosphate pathway was operated at a minimal level to supply the precursors for biomass formation. The different metabolic behaviors under varying nitrogen concentrations were observed with flux analysis.

Polysialic acid (PSA) is a capsular polysaccharide obtained from aerobic fermentation with Escherichia coli. To enhance PSA production and eliminate the influence of phosphate on the PSA purification process, a lower level of initial phosphate was adopted with pH control. The resulting PSA yield reached 4.1 g/L in fed-batch fermentation with 2.5 g/L K2HPO4 and E. coli strain CCTCC M208088. In addition, an ammonia water (NH4OH) feeding strategy to control the pH at 6.4 was developed resulting in PSA production that reached as high as 5.2 g/L. NMR spectra confirmed the purified biopolymer as a α-2,8linked PSA, identical to the published NMR spectra, with a molecular weight in the range of 16 ~ 50 kDa.

Purpose: To investigate the prevalence of spina bifida occulta (SBO) and its relationship with the presence of overactive bladder (OAB) in middle-aged and elderly people in China. Methods: A cross-sectional community-based survey was carried out at 7 communities in Zhengzhou City, China from December 15, 2013 to June 10, 2014, where residents aged over 40 years were randomly selected to participate. All of the participants underwent lumbosacral radiographic analysis and relevant laboratory tests. A questionnaire including basic information, past medical history and present illness, and the OAB symptom score was filled out by all participants. Chi-square tests and logistic regression were used for data analysis with a P-value of <0.05 denoting statistical significance. Results: A total of 1,061 subjects were qualified for the final statistical analysis (58.8±11.7 years; male, 471 [44.4%]; female, 590 [55.6%]). The overall prevalence of SBO was 15.1% (160 of 1,061): 18.3% (86 of 471) in men and 12.5% (74 of 590) in women. Among these subjects, 13.7% (145 of 1,061) had OAB: 13.2% (62 of 471) in men and 14.1% (83 of 590) in women. The results of logistic regression showed that age, SBO, history of cerebral infarction (HCI), and constipation were risk factors for OAB (P<0.05), while sex, history of childhood enuresis (HCE), body mass index (BMI), and diabetes mellitus (DM) were not (P>0.05). In men, age, SBO, and constipation were risk factors for OAB (P<0.05), while HCE, BMI, DM, HCI, and benign prostate hyperplasia were not (P>0.05). In women, age, SBO, and HCI were risk factors for OAB (P<0.05), while HCE, BMI, DM, vaginal delivery, and constipation were not (P>0.05). Conclusions: The prevalence of SBO is high and it is related to OAB in middle-aged and elderly people in China.

We investigated the reduction leaching process of manganese dioxide ore using black locust as reductant in sulfuric acid solution. The effect of parameters on the leaching efficiency of manganese was the primary focus. Experimental results indicate that manganese leaching efficiency of 97.57% was achieved under the optimal conditions: weight ratio of black locust to manganese dioxide ore (WT) of 4:10, ore particle size of 63 μm, 1.7 mol·L-1 H2SO4, liquid to solid ratio (L/S) of 5:1, leaching time of 8 h, leaching temperature of 368 K and agitation rate of 400 r·min-1. The leaching rate of manganese, based on the shrinking core model, was found to be controlled by inner diffusion through the ash/inert layer composed of associated minerals. The activation energy of reductive leaching is 17.81 kJ·mol-1. To conclude the reaction mechanism, XRD analysis of leached ore residue indicates manganese compounds disappear; FTIR characterization of leached residue of black locust sawdust shows hemicellulose and cellulose disappear after the leaching process.
Application of RBF Neural Network in Fault Diagnosis of FOG SINS

Taking FOG SINS (fiber-optic gyroscope strapdown inertial system) as an object, a new fault diagnostic scheme based on RBF(radial basis function) neural network is proposed. Being capable of training and simulating data off-line, neural networks provide a solution to overcome some drawbacks of the quantitative fault diagnosis. The fault tree of FOG SINS is analyzed, which is the basis of the study of neural network fault diagnosis technology. The structure and inferential mechanism of RBF network used for elementary fault diagnosis are discussed in detail. Training simulation results of the neural network are given and an improved effect with real data is obtained, which show the feasibility of the proposed scheme.
Transcription Regulation Network Analysis of MCF7 Breast Cancer Cells Exposed to Estradiol

Background: In breast cancer, estrogen receptors have been demonstrated to interact with transcription factors to regulate target gene expression. However, high-throughput identification of the transcription regulation relationship between transcription factors and their target genes in response to estradiol is still in its infancy. Purpose: Thus, the objective of our study was to interpret the transcription regulation network of MCF7 breast cancer cells exposed to estradiol. Methods: In this work, GSE11352 microarray data were used to identify differentially expressed genes (DEGs). Results: Our results showed that the MYB (v-myb myeloblastosis viral oncogene homolog [avian]), PGR (progesterone receptor), and MYC (v-myc myelocytomatosis viral oncogene homolog [avian]) were hub nodes in our transcriptome network, which may interact with ER and, in turn, regulate target gene expression. MYB can up-regulate MCM3 (minichromosome maintenance 3) and MCM7 expression; PGR can suppress BCL2 (B-cell lymphoma 2) expression; MYC can inhibit TGFB2 (transforming growth factor, beta 2) expression. These genes are associated with breast cancer progression via cell cycling and the $TGF{\beta}$ signaling pathway. Conclusion: Analysis of transcriptional regulation may provide a better understanding of molecular mechanisms and clues to potential therapeutic targets in the treatment of breast cancer.
