Reduction Leaching of Manganese Dioxide Ore Using Black Locust as Reductant in Sulfuric Acid Solution

( Jian Rong Xue ),( Hong Zhong ),( Shuai Wang ),( Chang Xin Li ),( Jin Zhong Li ),( Fang Fang Wu ) 한국화학공학회 2015 Korean Chemical Engineering Research(HWAHAK KONGHA Vol.53 No.4

We investigated the reduction leaching process of manganese dioxide ore using black locust as reductant in sulfuric acid solution. The effect of parameters on the leaching efficiency of manganese was the primary focus. Experimental results indicate that manganese leaching efficiency of 97.57% was achieved under the optimal conditions: weight ratio of black locust to manganese dioxide ore (WT) of 4:10, ore particle size of 63 μm, 1.7 mol·L-1 H2SO4, liquid to solid ratio (L/S) of 5:1, leaching time of 8 h, leaching temperature of 368 K and agitation rate of 400 r·min-1. The leaching rate of manganese, based on the shrinking core model, was found to be controlled by inner diffusion through the ash/inert layer composed of associated minerals. The activation energy of reductive leaching is 17.81 kJ·mol-1. To conclude the reaction mechanism, XRD analysis of leached ore residue indicates manganese compounds disappear; FTIR characterization of leached residue of black locust sawdust shows hemicellulose and cellulose disappear after the leaching process.

      • Induction of Apoptosis by IGFBP3 Overexpression in Hepatocellular Carcinoma Cells

        Han, Jian-Jun,Xue, De-Wen,Han, Qiu-Rong,Liang, Xiao-Hong,Xie, Li,Li, Sheng,Wu, Hui-Yong,Song, Bao Asian Pacific Journal of Cancer Prevention 2014 Asian Pacific journal of cancer prevention Vol.15 No.23

Background: The insulin-like growth factor (IGF) system comprises a group of proteins that play key roles in regulating cell growth, differentiation, and apoptosis in a variety of cellular systems. The aim of this study was to investigate the role of insulin-like growth factor binding protein 3 (IGFBP3) in hepatocellular carcinoma. Materials and Methods: Expression of IGF2, IGFBP3, and PTEN was analyzed by qRT-PCR. Lentivirus vectors were used to overexpress IGFBP3 in hepatocellular carcinoma cell (HCC) lines. The effect of IGFBP3 on proliferation was investigated by MTT and colony formation assays. Results: Expression of IGF2, IGFBP3, and PTEN in several HCC cell lines was lower than in normal cell lines. After 5-aza-2'-deoxycytidine/trichostatin A treatment, significant demethylation of the promoter region of IGFBP3 was observed in HCC cells. Overexpression of IGFBP3 induced apoptosis and reduced colony formation in HUH7 cells. Conclusions: Expression of IGF2, IGFBP3, and PTEN in several HCC cell lines was lower than in normal cell lines. After 5-aza-2'-deoxycytidine/trichostatin A treatment, significant demethylation of the promoter region of IGFBP3 was observed in HCC cells. Overexpression of IGFBP3 induced apoptosis and reduced colony formation in HUH7 cells.

      • A New Cell Counting Method to Evaluate Anti-tumor Compound Activity

        Wang, Xue-Jian,Zhang, Xiu-Rong,Zhang, Lei,Li, Qing-Hua,Wang, Lin,Shi, Li-Hong,Fang, Chun-Yan Asian Pacific Journal of Cancer Prevention 2014 Asian Pacific journal of cancer prevention Vol.15 No.8

Determining cell quantity is a common problem in cytology research and anti-tumor drug development. A simple and low-cost method was developed to determine monolayer and adherent-growth cell quantities. The cell nucleus is located in the cytoplasm, and is independent. Thus, the nucleus cannot make contact even if the cell density is heavy. This phenomenon is the foundation of accurate cell-nucleus recognition. The cell nucleus is easily recognizable in images after fluorescent staining because it is independent. A one-to-one relationship exists between the nucleus and the cell; therefore, this method can be used to determine the quantity of proliferating cells. Results indicated that the activity of the histone deacetylase inhibitor Z1 was effective after this method was used. The nude-mouse xenograft model also revealed the potent anti-tumor activity of Z1. This research presents a new anti-tumor-drug evaluation method.

      • Preparation and Antitumor Activity of a Tamibarotene-Furoxan Derivative

        Wang, Xue-Jian,Duan, Yu,Li, Zong-Tao,Feng, Jin-Hong,Pan, Xiang-Po,Zhang, Xiu-Rong,Shi, Li-Hong,Zhang, Tao Asian Pacific Journal of Cancer Prevention 2014 Asian Pacific journal of cancer prevention Vol.15 No.15

Multi-target drug design, in which drugs are designed as single molecules to simultaneously modulate multiple physiological targets, is an important strategy in the field of drug discovery. QT-011, a tamibarotene-furoxan derivative, was here prepared and proposed to exert synergistic effects on antileukemia by releasing nitric oxide and tamibarotene. Compared with tamibarotene itself, QT-011 displayed stronger antiproliferative effects on U937 and HL-60 cells and was more effective evaluated in a nude mice U937 xenograft model in vivo. In addition, QT-011 could release nitric oxide which might contribute to the antiproliferative activity. Autodocking assays showed that QT-011 fits well with the hydrophobic pocket of retinoic acid receptors. Taken together, these results suggest that QT-011 might be a highly effective derivative of tamibarotene and a potential candidate compound as antileukemia agent.

      • Value of PAX1 Methylation Analysis by MS-HRM in the Triage of Atypical Squamous Cells of Undetermined Significance

        Li, Shi-Rong,Wang, Zhen-Ming,Wang, Yu-Hui,Wang, Xi-Bo,Zhao, Jian-Qiang,Xue, Hai-Bin,Jiang, Fu-Guo Asian Pacific Journal of Cancer Prevention 2015 Asian Pacific journal of cancer prevention Vol.16 No.14

Background: Detection of cervical high grade lesions in patients with atypical squamous cells of undetermined significance (ASCUS) is still a challenge. Our study tested the efficacy of the paired boxed gene 1 (PAX1) methylation analysis by methylation-sensitive high-resolution melting (MS-HRM) in the detection of high grade lesions in ASCUS and compared performance with the hybrid capture 2 (HC2) human papillomavirus (HPV) test. Materials and Methods: A total of 463 consecutive ASCUS women from primary screening were selected. Their cervical scrapings were collected and assessed by PAX1 methylation analysis (MS-HRM) and high-risk HPV-DNA test (HC2). All patients with ASCUS were admitted to colposcopy and cervical biopsies. The Chisquare test was used to test the differences of PAX1 methylation or HPV infection between groups. Results: The specificity, sensitivity, and accuracy for detecting CIN2 + lesions were: 95.6%, 82.4%, and 94.6%, respectively, for the PAX1 MS-HRM test; and 59.7%, 64.7%, and 60.0% for the HC2 HPV test. Conclusions: The PAX1 methylation analysis by MS-HRM demonstrated a better performance than the high-risk HPV-DNA test for the detection of high grade lesions (CIN2 +) in ASCUS cases. This approach could screen out the majority of low grade cases of ASCUS, and thus reduce the referral rate to colposcopy.

Genetic Overlap in the Quantitative Resistance of Rice at the Seedling and Adult Stages to Xanthomonas oryzae pv. oryzae

        Yong-Li Zhou,Xue-Wen Xie,Mei-Rong Xu,Jin-Ping Zang,Ling-Hua Zhu,Jian-Long Xu,Zhi-Kang Li 한국식물학회 2012 Journal of Plant Biology Vol.55 No.2

The genetic components responsible for the qualitative and quantitative resistance of rice to three Chinese races (C2, C4, and C5) of Xanthomonas oryzae pv. oryzae (Xoo)were investigated at the seedling and adult stages in two successive years in set of Lemont/Teqing cross introgression lines (ILs) in a Teqing background, to create a complete linkage map using 160 well-distributed SSR markers. Teqing was resistant to C2 and C4, but moderately susceptible to C5,whereas Lemont was susceptible to all three races. Highly significant correlations were detected among the resistance to different races at different developmental stages. A major gene (Xa4), 14 main-effect QTLs (M-QTLs), and 18 epistatic QTLs were identified in the two developmental stages over 2 years, and were largely responsible for the segregation of resistance in the ILs. In 2007, the Lemont alleles at all loci in the seedling stage, except QBbr10 to C4, increased lesion length (LL) or decreased resistance. The Teqing allele at the Xa4 locus acted as a resistance gene against C2 and C4, but acted as a M-QTL when its resistance was overcome by the virulent race C5. MQTLs showed a degree of race specificity and had a cumulative effect on resistance. Most M-QTLs (94%) consistently expressed resistance to the same race at the seedling and adult stages, indicating that a high degree of genetic overlap exists between Xoo resistance at both developmental stages in rice. Among the digenic interactions, most co-introgressed Lemont alleles at the two epistatic loci lead to significantly smaller LL with all three races, compared to other types of interacting alleles at both development stages. The results indicate that a high level of resistance may be achieved by the cumulative effect of multiple M-QTLs, including the residual effects of “defeated” major resistance genes and the epistatic effects of co-introgression from diverse susceptible varieties.

Sentinel lymph Node mapping versus systematic pelvic lymphadenectomy on the prognosis for patients with intermediate-high-risk Endometrial Cancer confined to the uterus before surgery: trial protocol for a non-inferiority randomized controlled trial (SNE

        Jun Guan,Yu Xue,Rong-yu Zang,Ji-hong Liu,Jian-qing Zhu,Ying Zheng,Bo Wang,Hua-ying Wang,Xiao-jun Chen 대한부인종양학회 2021 Journal of Gynecologic Oncology Vol.32 No.4

Background: Sentinel lymph node (SLN) mapping has been recommended as an alternative staging approach to lymphadenectomy for apparent uterine-confined endometrial cancer (EC). However, the prognostic value of SLN mapping alone instead of systematic lymphadenectomy on EC patients remains unclear. Methods: A multi-center, open label, non-inferiority randomized controlled trial has been designed to identify if SLN mapping alone is not inferior to pelvic lymphadenectomy on prognosis of patients with intermediate-high-risk EC clinically confined to uterus. Eligible patients will be 1:1 randomly assigned to accept SLN mapping or pelvic lymphadenectomy. The primary endpoint is the 2-year progression-free survival (PFS). The second points are the 5-year PFS, 5-year overall survival, surgery-related adverse events and life quality. A total of 780 patients will be enrolled from 6 hospitals in China within 3-year period and followed up for 5 years. Trial Registration: ClinicalTrials.gov Identifier: NCT04276532

Characterization of SiC/C Nanocomposite Powders Synthesized by Arc-Discharge

        Zhou, Lei,Yu, Jie Yi,Gao, Jian,Wang, Dong Xing,Gan, Xiao Rong,Xue, Fang Hong,Huang, Hao,Dong, Xing Long Korean Society of Microscopy 2015 Applied microscopy Vol.45 No.4

In this paper, three carbon sources, i.e., solid graphite, gaseous CH4 and liquid ethanol, and one solid silicon source were employed to synthesize SiC/C nanocomposite powders by arc-discharge plasma. The processing conditions such as the component ratios of raw materials, atmospheric gases, etc. were adjusted for controllable synthesis of the nanopowders. It is indicated that both of solid graphite and silicon can be co-evaporated and reacted to form nanophases of cubic ${\beta}$-SiC with ~50 nm in mean size and a little free graphite; the carbon atoms decomposed from gaseous $CH_4$ favor to combine with the evaporated silicon atoms to form the dominant SiC nanophase; liquid carbon source of ethanol can also be used to harvest the main ${\beta}$-SiC and minor 6H-SiC phases in the assembly of nanoparticles. The as-prepared SiC/C nanocomposite powders were further purified by a heat-treatment in air and their photocatalytic performances were then greatly improved.

      • Effects of Gastric Cancer Cells on the Differentiation of Treg Cells

Hu, Jing-Lan,Yang, Zhen,Tang, Jian-Rong,Fu, Xue-Qin,Yao, Lan-Jie Asian Pacific Journal of Cancer Prevention 2013 Asian Pacific journal of cancer prevention Vol.14 No.8

The aim of this study was evaluated the prevalence of Treg cells in peripheral blood in patients with gastric cancer, and investigate the effect of gastric cancer cells on their differentiation. ELISA was employed to assess the concentrations of TGF-${\beta}$ and IL-10 in gastric cancer patients' serum. Then, mouse gastric cancer cells were co-cultured with T lymphocytes or T lymphocytes + anti-TGF-${\beta}$. Flow cytometric analysis and RT-PCR were then performed to detect Treg cells and TGF-${\beta}$ and IL-10 expression in gastric cancer cells. Our data showed that the expression of TGF-${\beta}$ and IL-10 in the patients with gastric cancer was increased compared to the case with healthy donors. The population of Treg cells and the expression levels of TGF-${\beta}$ and IL-10 in the co-culture group were much higher than in the control group (18.6% vs 9.5%) (P<0.05). Moreover, the population of Treg cells and the expression levels of TGF-${\beta}$ and IL-10 in the co-culture systerm were clearly decreased after addition of anti-TGF-${\beta}$ (7.7% vs 19.6%) (P<0.01). In conclusion, gastric cancer cells may induce Treg cell differentiation through TGF-${\beta}$, and further promote immunosuppression.

      • Clinical Study of Hepatectomy Combined with Jianpi Huayu Therapy for Hepatocellular Carcinoma

        Zhong, Chong,Li, Hui-Dong,Liu, Dong-Yang,Xu, Fa-Bin,Wu, Jian,Lin, Xue-Mei,Guo, Rong-Ping Asian Pacific Journal of Cancer Prevention 2014 Asian Pacific journal of cancer prevention Vol.15 No.14

Background: Traditional Chinese Medicine (TCM) possesses several advantages for treating patients with hepatocellular carcinoma (HCC). The theory of 'Jianpi Huayu Therapy' rooted from 'Jin Kui Yao Lue'is one of the most important therapies in this respect. This study was conducted to investigate the clinical effect and safety of hepatectomy combining with 'Jianpi Huayu Therapy' in the treatment of HCC. Materials and Methods: One hundred and twenty patients with HCC were randomized allocated into hepatectomy combined with 'Jianpi Huayu Therapy' group (treatment group, n=60) and hepatectomy alone group (control group, n=60). Disease- free survival (DFS) and overall survival (OS) were the primary end-points. Liver function at the end of one week after surgery, complications, average days of hospitalization as well as performance status (PS) at the end of one month post operation were also compared. Results: No significant differences existed between two groups on baseline analysis (p>0.05). No treatment related mortality occurred in either group. Post-operative complications were detected among 14 patients (23.3%) in the treatment group, and 12 (20.0%) in the control group (p=0.658). Alanine aminotransferase (ALT) at the end of one week after operation was lower in the treatment than control groups (p=0.042). No significant differences in other indexes of liver function were discovered between two groups. Average days of hospitalization reduced by 0.9 day in treatment group than in control (p=0.034). During follow-up, 104 patients (86.6%) developed recurrence. The rates of 1-, 3-, and 5-year DFS and median DFS for all patients were 77.4%, 26.3%, 9.0% and 25.6 months (range, 6.0~68.0), respectively (78.2%, 29.2%, 14.3% and 28.7 months for the 48 patients in the treatment group and 75.0%, 23.3%, 6.4%, and 22.6 months for the 56 patients in the control group (p=0.045)). 101 patients had died at the time of censor, with 1-, 3-, and 5-year overall survival rates and median survival for all patients of 97.5%, 76.4%, 40.5% and 51.2 months (range, 10.0~72.0), respectively (98.3%, 78.0%, 43.6% and 52.6 months, for treatment and 96.7%, 74.7%, 37.4%, and 49.8 months, for controls, respectively (p=0.048)). Conclusions: Hepatectomy combined with 'Jianpi Huayu therapy'was effective in the treatment of HCC, and reduced post-operative recurrence and metastasis and improved DFS and OS of HCC patients.
