IL-33–Induced Hematopoietic Stem and Progenitor Cell Mobilization Depends upon CCR2

<P>IL-33 has been implicated in the pathogenesis of asthma, atopic allergy, anaphylaxis, and other inflammatory diseases by promoting the production of proinflammatory cytokines and chemokines or Th2 immune responses. In this study, we analyzed the in vivo effect of IL-33 administration. IL-33 markedly promoted myelopoiesis in the bone marrow and myeloid cell emigration. Concomitantly, IL-33 induced hematopoietic stem and progenitor cell (HSPC) mobilization and extramedullary hematopoiesis. HSPC mobilization was mediated mainly through increased levels of CCL7 produced by vascular endothelial cells in response to IL-33. In vivo treatment of IL-33 rapidly induced phosphorylation of ERK, JNK, and p38, and inhibition of these signaling molecules completely blocked the production of CCL7 induced by IL-33. Consistently, inhibitor of CCR2 markedly reduced IL-33–mediated HSPC mobilization in vivo and migration of HSPCs in response to CCL7 in vitro. IL-33–mobilized HSPCs were capable of homing to, and of long-term reconstitution in, the bone marrow of irradiated recipients. Immune cells derived from these recipients had normal antifungal activity. The ability of IL-33 to promote migration of HSPCs and myeloid cells into the periphery and to regulate their antifungal activity represents a previously unrecognized role of IL-33 in innate immunity. These properties of IL-33 have clinical implications in hematopoietic stem cell transplantation.</P>

<P>A facile route to the fabrication of a broadband Al wire grid polarizer (WGP) was demonstrated using nanoimprint lithography (NIL) and oblique angle deposition (OAD) methods. An Al film with the desired shape for a 1D grating pattern was formed by OAD directly on the top of a polymer nanostructure, which was patterned by NIL as a substrate at a pitch of 120 nm, width of 50 nm and height of 160 nm. The results show that transmittance of the transverse magnetic wave for the fabricated WGP was an average of 67% and the average polarization extinction ratio was 22 dB in the visible and near infrared wavelength regime. The optical properties of the various shapes of Al patterns on the 1D polymer nanostructures were also simulated using a rigorous finite difference time domain simulation. This work can open up a cost-effective fabrication method for high- performance and large-area WGPs.</P>

Rapid increase of human population together with environmental changes to reduce crop yields are threatening human survival since these problems are expected to cause food shortage soon. Edible insects are getting attention as a new food resource that can solve forthcoming food shortages since they are particularly rich in protein and unsaturated fatty acids. Thus, when they are used as food, it is important to prepare the extract formulation with high yield of proteins and other nutritional components with minimal damage to them. In this study, 5 extraction methods were used, which were hot water (HW), autoclave (AT), high pressure treatment for 5 minutes (HP5), high pressure treatment for 10 minutes (HP10) and enzymatic hydrolysis (ENZ), and ten extracts were prepared by using one of these methods or combinations thereof. The extracts prepared were HW, AT, AT/ENZ, AT/ENZ/HW HP5/HW, HP5/ENZ, HP5/ENZ/HW, HP10/HW, HP10/ENZ, HP10/ENZ/HW, which were investigated for yields, molecular sizes of the components, amino acid (AA) composition and antioxidant activities. The extracts prepared using ENZ method were observed to contain high contents of total AA, high ratio of essential/non-essential AA and high antioxidant activity. Thus, ENZ method is recommended to prepare the high quality-extract of this insect.
A Comprehensive and Comparative Review of Global Gastric Cancer Treatment Guidelines

Countries differ in their treatment expertise and research results regarding gastric cancer; hence, treatment guidelines are diverse based on evidence and medical situations. A comprehensive and comparative review of each country’s guidelines is imperative to understand the similarities and differences among countries. We reviewed and compared five gastric cancer treatment guidelines in terms of endoscopic, surgical, perioperative, and palliative systemic treatment based on evidence levels and recommendation grades, as well as the postoperative follow-up strategies for each guideline. The Korean, Chinese, and European guidelines provided evidence and grading of the recommendations. The United States guidelines suggested categories for evidence and consensus. The Japanese guidelines suggested evidence and recommendations only for systemic treatment. The Korean and Japanese guidelines described endoscopic treatment, surgery, and lymphadenectomy in detail. The Chinese, United States, and European guidelines more intensively considered perioperative chemotherapy. In particular, the indications for chemotherapy and the regimens recommended by each guideline differed slightly. Considering their medical situations, each guideline had some diversity in terms of adopting evidence, which resulted in heterogeneous recommendations. This review will help medical personnel to comprehensively understand the diversity in gastric cancer treatment guidelines for each country in terms of evidence and recommendations.
Intratumorally Establishing Type 2 Innate Lymphoid Cells Blocks Tumor Growth

<P>A long-standing question in the field of tumor immunotherapy is how Th2 cytokines block tumor growth. Their antitumor effects are particularly prominent when they are secreted continuously in tumors, suggesting that Th2 cytokines may create a tumor micro-environment unfavorable for tumor growth independently of adaptive immunity. In this study, we show that local production of IL-33 establishes a high number of type 2 innate lymphoid cells (ILC2s) with potent antitumor activity. IL-33 promotes secretion of a massive amount of CXCR2 ligands from ILC2s but creates a tumor microenvironment where tumor cells express CXCR2 through a dysfunctional angiogenesis/hypoxia/reactive oxygen species axis. These two signaling events converge to reinforce tumor cell-specific apoptosis through CXCR2. Our results identify a previously unrecognized antitumor therapeutic pathway wherein ILC2s play a central role.</P>

<P>Diarylalkynones were synthesized from one-pot Pd-catalyzed carbonylative and noncarbonylative coupling reactions of propiolic acid with aryl iodides under a carbon monoxide atmosphere. Aryl iodide (2.0 equiv), propiolic acid (1.0 equiv), Pd(PPh<SUB>3</SUB>)<SUB>2</SUB>Cl<SUB>2</SUB> (5 mol %), CuCl (10 mol %), Et<SUB>3</SUB>N (6.0 equiv), and CO (8 atm) were reacted under optimized conditions in CH<SUB>3</SUB>CN at 80 °C for 1 h. <P><B>Abstract</B></P> <P>Optical properties of nucleobase thin films were studied by attenuated total reflection (ATR) and surface-enhanced Raman spectroscopy (SERS). Adenine and guanine films were deposited on fused silica and silver at room temperature by thermal evaporation, and the normal dispersion of refractive indices of transparent adenine and guanine films in the visible and near-infrared regions were analyzed. The measured ATR spectra of adenine (guanine) films and numerical simulations by optical transfer matrix formalism demonstrate that the shift of surface plasmon resonance (SPR) wavelength is approximately linearly proportional to the adenine (guanine) film thickness, indicating that SPR can be used for quantitative measurements of biomaterials. The Raman spectra indicated that the adenine (guanine) films can be deposited by thermal evaporation. The adenine (guanine) films on silver exhibited Raman intensity enhancement as compared to those on glass, which was attributed to the SPR effect of silver platform and might play a role as a hot plate for SERS detection of biomaterials.</P>
