콜롬비아 소가모스 지역에서 미세먼지 감축 시나리오를 TAPM 모델을 활용하여 도출하고 평가하였다. 시나리오 1의 제철소 및 금속 등 중공업 오염원을 대상으로 미세먼지 방지 기술을 이용하여 배출량을 80% 감축하는 방안에서는 도시 중심 평균 농도는 6.5 μg m-3로 1.5 % 감소하였고, 모델영역에서 최고 평균농도는 387 μg m-3 로 변동 없었다. 시나리오 2의 벽돌 및 석회공장 등을 대상으로 연료를 석탄에서 코크스로 교체하여 90 % 감축하는 방안에서는 도시 중심 평균 농도는 4.5 μg m-3로 31.8%, 모델영역에서 최고 평균농도는 111 μg m-3 로 71.6 % 감소하였다. 시나리오 1과 시나리오 2를 결합한 시나리오 3에서는 도시 중심에서 평균 농도는 4.3 μg m-3로 34.8 %, 모델영역에서 최고 평균농도는 110 μg m-3 로 71.7 % 감소하였다. 소가모스 지역에서 대기환경을 개선하는 로드맵 수립에는 수공업에서 주로 사용되는 석탄 연료를 청정연료인 코크스로 교체하는 방안이 최우선으로 고려되어야 하겠다. 이후 미세먼지 방지 시설을 굴뚝에 설치하는 방안도 차선책으로 고려하여 환경과 경제가 선 순환하는 녹색성장관점에서 장기적인 미세먼지 개선 마스터 플랜이 수립되어야 하겠다. Reduction scenarios for PM10 concentration in Sogamoso, Colombia were analyzed using the TAPM( The Air Pollution Model). The average concentration of PM10 in the downtown area was 6.5 μg m-3, a reduction of 1.5 %. The maximum average concentration in the model domain was 387 μg m-3 without any reduction in scenarios 1 which the PM10 emissions from heavy industries such as iron and metal was reduced by 80 % using the prevention technology. In scenario 2 which the PM10 emissions from the brick and lime industries was reduced by 90 % substituting coke (a cleaner fuel) for coal, the average concentration of PM10 in the downtown area was 4.5 μg m-3, a reduction of 31.8 % , the maximum average concentration in the model domain was 111 μg m-3 , a reduction of 71.6 %. In scenario 3 which the scenarios 1 and 2 were combined, the average concentration of PM10 in the downtown area was 4.3 μg m-3, a reduction of 34.8 % and the maximum average concentration was 110 μg m-3, a reduction of 71.7 %. To put forward a road map for the reduction of PM10 in the Sogamos region, substituting coke for coal in manual industries should have priority over reduction policies. Also, it is highly recommended that a longterm master plan for reducing PM10 concentrations should be established with a focus on environmentally-friendly development and future green growth policy with a win-win relationship between economy and environment.

<P>In this study, we investigated the relative expression of the Rous sarcoma virus (RSV) promoter-driven expression of enhanced green fluorescent protein (EGFP) in fibroblasts of transgenic quails. We analyzed the direct influence of CpG methylation of the RSV promoter on the transcriptional activity of delivered transgenes. Embryonic fibroblasts collected from homozygous transgenic quail (TQ2) were treated with 50?μM of DNA methyltransferase inhibitor followed by 5-aza-2'-deoxycytidine (5-azadC) for 48?h, and changes in expression were then analyzed by flow cytometry. The results show a significant increase of EGFP expression in TQ2 embryonic fibroblasts (QEFs) (2.64% to 79.84%). Subsequent methylation-specific amplification revealed that 5-azadC significantly reduced the CpG methylation status in the RSV promoters of the QEFs (86.42 to 48.41%); even after 5-azadC was withdrawn, CpG methylation remained decreased in expanded culture (16.28%). Further analysis showed that potential transcription factor binding sites existed in the CpG methylation site of the RSV promoter. These results may provide the basis for understanding the epigenetic mechanism responsible for transgenic animal production and genetic preservation.</P>

단백질-단백질 결합은 수용체 단백질, 효소, 세포 골격 단백질의 세포내 위치 결정 및 기능 조절에 중요한 역할을 한다. Postsynaptic density-95/disks large/zonula occludens-1 (PDZ) 도메인을 가진 단백질들은 시냅스 가소성, 신경세포 성장과 분화뿐만 아니라 많은 질병의 병태생리에 중요하게 관여하는 scaffold 단백질로 작용한다. Multi-PDZ domain protein 1 (MUPP1)은 13개 PDZ 도메인을 가지는 단백질로서 세포막 수용체 군집화, 신호전달 복합체 구성, 세포 골격 조정에 대한 매개 역할을 하는 것으로 알려지고 있지만 MUPP1의 세포 내 기능은 아직 명확히 밝혀지지 않았다. 본 연구에서 MUPP1의 아미노 말단 PDZ 도메인과 결합하는 새로운 단백질을 규명하기 위하여 효모 two-hybrid 방법을 이용하였고 Wdpcp (전에 Fritz로 알려짐)이 MUPP1과 결합하는 것을 확인하였다. Wdpcp는 planar cell polarity (PCP) effector로서 세포 이동과 섬모형성에 관여하는 것으로 알려져 있다. Wdpcp는 MUPP1의 첫 번째 PDZ 도메인과 결합하지만, 다른 PDZ 도메인과는 결합하지 않았다. 또한 MUPP1와 Wdpcp의 결합에서 Wdpcp의 C-말단부위가 결합에 필수적임을 효모 two-hybrid 방법으로 확인하였다. 이러한 단백질간 결합은 glutathione S-transferase (GST) pull-down assay, 공동면역침강, HEK-293T 세포에서의 발현위치를 통하여 추가적으로 확인하였다. 이러한 결과들은, MUPP1과 Wdpcp 결합은 세포내 액틴 다이내믹스(dynamics)와 세포이동 조절에 역할을 할 가능성을 시사한다. Protein-protein interactions regulate the subcellular localization and function of receptors, enzymes, and cytoskeletal proteins. Proteins containing the postsynaptic density-95/disks large/zonula occludens-1 (PDZ) domain have potential to act as scaffolding proteins and play a pivotal role in various processes, such as synaptic plasticity, neural guidance, and development, as well as in the pathophysiology of many diseases. Multi-PDZ domain protein 1 (MUPP1), which has 13 PDZ domains, has a scaffolding function in the clustering of surface receptors, organization of signaling complexes, and coordination of cytoskeletal dynamics. However, the cellular function of MUPP1 has not been fully elucidated. In the present study, a yeast two-hybrid system was used to identify proteins that interacted with the N-terminal PDZ domain of MUPP1. The results revealed an interaction between MUPP1 and Wdpcp (formerly known as Fritz). Wdpcp was identified as a planar cell polarity (PCP) effector, which is known to have a role in collective cell migration and cilia formation. Wdpcp bound to the PDZ1 domain but not to other PDZ domains of MUPP1. The C-terminal end of Wdpcp was essential for the interaction with MUPP1 in the yeast two-hybrid assay. This interaction was further confirmed in a glutathione S-transferase (GST) pull-down assay. When coexpressed in HEK-293T cells, Wdpcp was coimmunoprecipitated with MUPP1. In addition, MUPP1 colocalized with Wdpcp at the same subcellular region in cells. Collectively, these results suggest that the MUPP1-Wdpcp interaction could modulate actin cytoskeleton dynamics and polarized cell migration.

<P>Existing studies examining the control of mesenchymal stem cell (MSC) differentiation into desired cell types have used a variety of biochemical reagents such as growth factors despite possible side effects. Recently, the roles of biomimetic microphysical environments have drawn much attention in this field. We studied MSC differentiation and changes in gene expression in relation to osteoblast-like cell and smooth muscle-like cell type resulting from various microphysical environments, including differing magnitudes of tensile strain and substrate geometries for 8 days. In addition, we also investigated the residual effects of those selected microphysical environment factors on the differentiation by ceasing those factors for 3 days. The results of this study showed the effects of the strain magnitudes and surface geometries. However, the genes which are related to the same cell type showed different responses depending on the changes in strain magnitude and surface geometry. Also, different responses were observed three days after the straining was stopped. These data confirm that controlling microenvironments so that they mimic those in vivo contributes to the differentiation of MSCs into specific cell types. And duration of straining engagement was also found to play important roles along with surface geometry.</P>
