양파 가수분해물 제조를 위해 celluclast 1.5L와 pectinex에 의한 양파 가수분해물 제조시 반응조건에 따른 영향과 조미액의 활용을 검토하였다. Celluclast 1.5L과 pectinex의 혼합비가 1 : 3일때 가장 높은 86%의 가수분해도를 보였으며, 총당의 생성량은 54㎎/ml으로 가장 높았다. 반응온도가 25~45℃에서 가수분해도는 83~86%로 커다란 차이가 없는 반면 65℃에서 가수분해시 66% 정도의 낮은 가수분해도를 보였으며, 총당의 생성량은 반응온도가 25℃에서 45℃로 증가할수록 총당의 생성량이 증가하는 경향을 보였으며, 45℃에서 이후의 반응온도에서는 총당 생성량의 변화가 미미하였다. pH에 따른 가수분해도와 총당의 변화를 측정한 결과, pH 2.5~5.5인 산성 조건에서는 비교적 높은 76~86%의 가수분해도를 보였으며, 총당의 생성량 역시 pH 4.5에서 54㎎/ml로 가장 높았다. 효소 첨가량이 증가할수록 가수분해도가 증가하는 경향을 보였으나, 0.5% 이상 첨가시 가수분해도의 증가는 미미하였다. 반응시간이 증가할수록 가수분해도가 증가하여 2시간 가수분해시 84%의 가수분해도를 보였으며, 6시간 가수분해시 88%의 가수분해도를 보였다. 총당 생성량은 4시간 반응시까지 증가하는 경향을 보였으며, 4시간 이상 가수분해시 총당의 변화량은 미미하였다. 이러한 조건에서 생산한 양파 가수분해물의 조미액으로의 활용 여부를 검토하고자 관능검사를 실시한 결과, 가수분해물과 양파를 직접 사용한 두군간에 큰 차이를 보이지 않았으며, 고기 양념으로 가수분해물 사용할 때 고기 중량에 대하여 5% 이하로 사용하는 것이 바람직하였다. To develop an onion sauce, reaction conditions of celluclast 1.5L and pectinex on onion were investigated and organoleptic evaluations were carried out. Degree of hydrolysis(D.H) of hydrolysate by a mixture of celluclast 1.5L and pectinex was a higher than that by each enzyme. Hydrolysate by a mixture of celluclast 1.5L and pectinex(1 : 3v/v) showed 86% of D.H. and total sugar content of the hydrolysate was 54㎎/ml. Hydrolysates showed 83~86% of D.H. at reaction temperature of 25℃ to 45℃. Total sugar content of the hydrolysate was increased with increasing temperature. D.H. and total sugar content of hydrolysate was 76~86% and 51~53㎎/ml, respectively, under acidic conditions. D.H. and total sugar content of hydrolysate were also increased with increasing time. Bitterness, sweetness and ordor of roasted pork prepared by adding onion and onion hydrolysate were significantly different(p<0.05), but color and preference between two groups were not significantly different(p<0.05) between two groups. There was no significant difference(p<0.05) in sweetness and bitterness of the roasted pork prepared by adding different amounts of onion hydrolysate, although ordor and preference of the roasted pork were significantly different(p<0.05).

The production of sorbose from sorbitol in resting cell system of Acetobacter suboxydans was studied. The conversion of sorbose from sorbitol was markedly influenced by several factors such as the substrate concentration, reaction time, temperature, pH. metal ions, growth factors and aeration in the resting cells. Sorbose production rapidly increased in the range of 6 ㎎/㎖ cells with the concentration of 5% sorbitol. For production of sorbose from sorbitol, optimal temperature and pH were 30℃ and 6.0. The production of sorbose from sorbitol was activated by 1 mM of Al^(+3) while inhibited by Ni^(+2). The conversion of sorbitol to sorbose was stimulated by the adding of 1 mM p-aminobenzoic acid and nicotinic acid, respectively. During incubation of 1.5 ㎖ of reaction mixture in 50 ㎖ of Erlenmeyer flask, 5% sorbitol was completely converted to sorbose after 20 hours.

The antioxidant and synergistic effect of methanol extracts of onion skin were investigated by measuring peroxide value. The inhibitory effects of the extracts against metal catalyzed oxidation were also studied. The antioxidant activity of methanol extract (0.02%) of onion skin was stronger than that of mixed tocopherol or ascorbic acid (0.02%), but weaker than that of BHT (0.02%). However, the methanol extract at the concentration of 0.04% exhibited strong antioxidant effect, comparable to that of BHT (0.02%). The methanol extract showed very strong synergistic effect with the mixed tocopherol. The methanol extracts at the concentration of 0.03% and 0.04% acted as metal deactivator in the presence of FeCl₃. As a metal deactivator, methanol extract of onion skin appeared to be more effective than citric acid.

Protein tyrosine phosphatases (PTPs) are pivotal regulators of key cellular functions, including cell growth, differentiation, and adhesion. Previously, we reported that leukocyte common antigen-related (LAR) tyrosine phosphatase promotes osteoblast differentiation in MC3T3-E1 preosteoblast cells. In the present study, the mechanism of the regulatory action of LAR on osteoblast differentiation was investigated. The mineralization of extracellular matrix and calcium accumulation in MC3T3-E1 cells were markedly enhanced by LAR overexpression, and these effects were further increased by treatment with an MEK inhibitor. In addition, LAR overexpression dramatically reduced extracellular signal-regulated kinase (Erk) activation during osteoblast differentiation. In contrast, a marginal effect of the inactive LAR mutant on Erk activation was detected. Expression of osteoblast-related genes such as ALP, BSP, DLX5, OCN, and RUNX2, was increased by LAR overexpression during osteoblast differentiation. On the basis of these results, we propose that LAR functions as a positive regulator of osteoblast differentiation by modulating ERK activation. Therefore, LAR phosphatase could be used as a novel regulatory target protein in many bone-associated diseases, including osteoporosis.
