        Identification of novel alleles induced by EMS-mutagenesis in key genes of kernel hardness and starch biosynthesis in wheat by TILLING

        To identify novel allelic variations in key genes of wheat quality, the present study used the targeting induced local lesions in genomes platform to detect point mutations in target genes. The wheat variety Longfumai 17 was treated by the mutagen ethyl methanesulfonate to produce a bulk M2 generation, and the population included 1122 plants. A total length of 3906.80 kb nucleotides was analyzed, and the average mutation density was 1/244.17 kb. The identified mutations included G>A substitutions (43.75%), C>T substitutions (31.25%), A insertions (12.50%), T insertions (6.25%), and deletions (6.25%). These point mutations led to changes in amino acids and thus the encoded protein sequences, ultimately producing 18.75% of missense mutations, 12.50% of frame shift mutations, 6.25% of nonsense mutations, 25.00% of silent mutations and 37.50% of non-coding region mutations. In the kernel hardness gene Pinb and 3 starch synthesis genes waxy, Agp2 and SSIIa-A, we detected 16 different point mutations in 25 mutant lines. The Pinb gene harbored two missense mutations and a nonsense mutation; the C>T missense mutation resulted in a novel allele, this novel allele and the nonsense mutation alerted protein 3D structure; the waxy gene presented missense and frame shift mutations; the Agp2 gene carried a missense mutation; the SSIIa-A incurred a missense mutation and a frame shift mutation that resulted in premature protein termination. All the frame shift mutations, nonsense mutations and the Pinb novel allele resulted in allelic variation of their corresponding genes, which in turn affected their gene functions. The identified mutant lines can be used as intermediate materials in wheat quality improvement schemes.

        Bacterial Community Structure and Function Shift in Rhizosphere Soil of Tobacco Plants Infected by Meloidogyne incognita

        Root-knot nematode disease is a widespread and catastrophic disease of tobacco. However, little is known about the relationship between rhizosphere bacterial community and root-knot nematode disease. This study used 16S rRNA gene sequencing and PICRUSt to assess bacterial community structure and function changes in rhizosphere soil from Meloidogyne incognita-infected tobacco plants. We studied the rhizosphere bacterial community structure of M. incognita-infected and uninfected tobacco plants through a paired comparison design in two regions of tobacco planting area, Yuxi and Jiuxiang of Yunnan Province, southwest China. According to the findings, M. incognita infection can alter the bacterial population in the soil. Uninfested soil has more operational taxonomic unit numbers and richness than infested soil. Principal Coordinate Analysis revealed clear separations between bacterial communities from infested and uninfested soil, indicating that different infection conditions resulted in significantly different bacterial community structures in soils. Firmicutes was prevalent in infested soil, but Chloroflexi and Acidobacteria were prevalent in uninfested soil. Sphingomonas, Streptomyces, and Bradyrhizobium were the dominant bacteria genera, and their abundance were higher in infested soil. By PICRUSt analysis, some metabolism-related functions and signal transduction functions of the rhizosphere bacterial community in the M. incognita infection-tobacco plants had a higher relative abundance than those uninfected. As a result, rhizosphere soils from tobacco plants infected with M. incognita showed considerable bacterial community structure and function alterations.

        Evaluation of antifatigue and antioxidant activities of the marine microalgae Isochrysis galbana in mice

        The present work aimed to estimate the possible anti-fatigue effect and potential mechanism of Isochrysis galbana (IG) in mice. The anti-fatigue activity of IG (100, 200, and 400 mg/kg) was elucidated by a weight-loaded forced swimming test, and the potential mechanism was explored by determination of fatigue-related biochemical parameters. The results showed that pretreatment with IG significantly extended the exhaustive swimming time and increased the levels of liver glycogen, muscle glycogen and blood glucose in a dose-dependent manner. Besides, the increased levels of alanine aminotransferase, aspartate aminotransferase, blood lactic acid, lactic dehydrogenase, creatine kinase, and blood urea nitrogen by exhausted swimming, were dramatically attenuated by pretreatment with IG. Furthermore, supplementation with IG significantly enhanced the glutathione peroxidase and superoxide dismutase levels, while attenuated the level of malonaldehyde. Taken together, IG possessed appreciable efficacy to alleviate fatigue, and the mechanism might be associated with favorably modulating the process of energy consumption, metabolism, and attenuating oxidative stress injury.

        Constrained Relay Node Deployment using an improved multi-objective Artificial Bee Colony in Wireless Sensor Networks

        Wireless sensor networks (WSNs) have attracted lots of attention in recent years due to their potential for various applications. In this paper, we seek how to efficiently deploy relay nodes into traditional static WSNs with constrained locations, aiming to satisfy specific requirements of the industry, such as average energy consumption and average network reliability. This constrained relay node deployment problem (CRNDP) is known as NP-hard optimization problem in the literature. We consider addressing this multi-objective (MO) optimization problem with an improved Artificial Bee Colony (ABC) algorithm with a linear local search (MOABCLLS), which is an extension of an improved ABC and applies two strategies of MO optimization. In order to verify the effectiveness of the MOABCLLS, two versions of MO ABC, two additional standard genetic algorithms, NSGA-II and SPEA2, and two different MO trajectory algorithms are included for comparison. We employ these metaheuristics on a test data set obtained from the literature. For an in-depth analysis of the behavior of the MOABCLLS compared to traditional methodologies, a statistical procedure is utilized to analyze the results. After studying the results, it is concluded that constrained relay node deployment using the MOABCLLS outperforms the performance of the other algorithms, based on two MO quality metrics: hypervolume and coverage of two sets.

        Design of HUST-PTF beamline control system for fast energy changing

        A proton therapy facility is under development at Huazhong University of Science and Technology (HUST). To meet the need for fast energy changes during treatments, a beamline control system (BCS) has been designed and implemented. The BCS coordinates and controls various beamline devices by adopting a distributed architecture divided into three layers: the client, server, and device layers. Among these, the design of the server layer is the key to realize fast energy changes. The server layer adopts the submodule programming paradigm and optimizes the data interface among modules, allowing the main workflow to be separated from the device workflow and data. Furthermore, this layer uses asynchronous, multithreaded, and thread-locking methods to improve the system's ability to operation efficiently and securely. Notably, to evaluate the changing energy status over time, a dynamic node update method is adopted, which can dynamically adjust the update frequency of variable nodes. This method not only meets the demand for fast updates on energy changes but also reduces the server's communication load in the steady state. This method is tested on a virtual platform, and the results are as expected.

      • Design and Implementation of iOS-based Mobile Application about Awakening by CNR

        With the growing popularity of the iPhone, there is a constantly increasing demand of users for mobile applications of iPhone, in which the alarm clock application is contained. But existing alarm clock application is not favored by many users because of functional singleness. This paper presents an iOS-based mobile application on alarm clock with online radio support, and designs and implements its framework by virtue of Objective-C and SQLite in the Xcode 4.5.2 development environment, based on the popular three-tier MVC software design structure. After the application is installed and deployed on iPhone, it is available to users by providing some functions such as auto-playing radio, setting radio alarm clock, turning off the alarm by shaking iPhone, sleeping timer by radio, reserving radio program, binding and sharing microblog. This proposed iOS-based mobile application provides users with more choices and allows the user to enjoy a more colorful leisure time. The practical test from some people shows that the proposed application is very popular with users, which also illustrates its practicability and effectiveness.

        Uncertainty quantifi cation of the power control system of a small PWR with coolant temperature perturbation

        The coolant temperature feedback coefficient is an important parameter of reactor core power controlsystem. To study the coolant temperature feedback coefficient influence on the core power controlsystem of small PWR, the core power control system is built with the nonlinear model and fuzzy controltheory. Then, the uncertainty quantification method of reactor core parameters is established based onthe Latin hypercube sampling method and the Bootstrap method. Finally, under the conditions ofreactivity step perturbation and coolant inlet temperature step perturbation, uncertainty analysis for twocases is carried out. The result shows that with fuzzy controller and fuzzy PID controller, the uncertaintyof the coolant temperature feedback coefficient affects the core power control system, and the maximumuncertainties of core relative power, coolant temperature deviation, fuel temperature deviation and totalreactivity are acceptable

        Changes in plant anthocyanin levels in response to abiotic stresses: a meta-analysis

        Anthocyanins are small molecule antioxidants that play important roles in plant response and resistance to abiotic stresses. Their levels increase when plants are exposed to abiotic stress. However, the general response patterns and magnitude of anthocyanin increase, and how they confer resistance to abiotic stresses, are difficult to evaluate because of the influence of experimental variables. In this study, changes in plant anthocyanin content under different abiotic stresses and the effect of anthocyanin overproduction on various physiological indicators were investigated through meta-analysis derived from 1039 datasets of 102 studies. Results showed that among the different stress types, heavy metals, especially copper (Cu) and mercury (Hg), induced the most significant synthesis of anthocyanins. Among the different types of drought treatments, mannitol caused anthocyanin content in plants to increase by 2.4-fold, which was more significant than that of polyethylene glycol and natural drought. Furthermore, UV stress led plant anthocyanins to increase 1.3-fold in C4 plants, which was higher than that in C3 plants. In addition, anthocyanins increased the most when the applied stresses were low and moderate, and of short duration. Moreover, plants overexpressing genes encoding MYB transcription factors increased anthocyanin content by 5.8-fold and significantly improved plant stress tolerance. Among the 21 physiological indicators, glutathione and proline levels increased the most in transgenic plants overproducing anthocyanins when exposed to abiotic stress. Taken together, this meta-analysis indicates that anthocyanins enhance stress tolerance by improving the antioxidant, metal-chelating, and osmoregulatory abilities of the plant. The results presented here can help guide future applications of anthocyanins as plant growth regulators in dryland agriculture and breeding for plant stress tolerance.

        Improved Ethanol Production in Jerusalem Artichoke Tubers by Overexpression of Inulinase Gene in Kluyveromyces marxianus

        Ethanol production from Jerusalem artichoke tubers through a consolidated bioprocessing (CBP) strategy using the inulinase-producing yeast Kluyveromyces marxianus is an economical and competitive than that from a grainbased feedstock. However, poor inulinase production under ethanol fermentation conditions significantly prolongs the fermentation time and compromises ethanol productivity. Improvement of inulinase activity appears to be promising for increasing ethanol production from Jerusalem artichoke tubers by CBP. In the present study, expression of the inulinase gene INU with its own promoter in K. marxianus (K/INU2) was explored using the integrative cassette. Overexpression of INU was explored using chromosome integration via the HO locus of the yeast. Inulinase activity and ethanol were determined from inulin and Jerusalem artichoke tubers under fed-batch operation. Inulinase activity was 114.9 U/mL under aerobic conditions for K/INU2, compared with 52.3 U/mL produced by the wild type strain. Importantly, inulinase production was enhanced in K/INU2 under ethanol fermentation conditions. When using 230 g/L inulin and 220 g/L Jerusalem artichoke tubers as substrates, inulinase activities of 3.7 and 6.8 U/mL,respectively, were measured using K/INU2, comparing favorably with 2.4 and 3.1 U/mL, respectively, using the wide type strain. Ethanol concentration and productivity for inulin were improved by the recombinant yeast to 96.2 g/L and 1.34 g/L/h, respectively, vs 93.7 g/L and 1.12 g/L/h,respectively, by the wild type strain. Ethanol concentration and productivity improvements for Jerusalem artichoke tubers were 69 g/L and 1.44 g/L/h, respectively, from the recombinant strain vs 62 g/L and 1.29 g/L/h, respectively,from the wild type strain.
