        Deflection prediction for reinforced concrete deep beams

        A simplified method, developed from the softened strut-and-tie model, for determining the mid-span deflection of deep beams at ultimate state is proposed. The mid-span deflection and shear strength predictions of the proposed model are compared with the experimental data collected from 70 simply supported reinforced concrete deep beams, loaded with concentrated loads located at a distance a from an end reaction. The comparison shows that the proposed model can accurately predict the mid-span deflection and shear strength of deep beams with different shear span-to-depth ratios, different concrete strengths, and different horizontal and vertical hoops.

        High-strength concrete deep beams with web openings strengthened by carbon fiber reinforced plastics

        The objective of this study is to examine the effect of carbon fiber reinforced polymer (CFRP) on the shear strengths of deep beams with web openings. A total of 18 high-strength concrete deep beams with web openings were tested. Twelve were externally wrapped with four layers of CFRP, six of them strengthened in the horizontal direction and the others in the vertical direction. The parameters of the configuration of CFRP, the sizes of the openings and the locations of the openings were covered in this study. The test results indicates the shear strengths of deep beams with openings sized 60×40 mm were about 16% higher than that with openings sized 68×68 mm. For deep beams with openings sized 60×40 mm, the lower the locations of openings the higher the shear strengths were. The test results also indicate the shear strengths of deep beams with web openings strengthened by CFRP wrapped in the vertical direction can be enhanced by about 10%. However, the shear strengths of deep beams with web openings strengthened by CFRP wrapped in the horizontal direction can only be enhanced by about 6%. The shear strengths of deep beam, with different size and location of web openings and strengthened by different configuration of CFRP can be reasonably predicted by the empirical formulas of Kong and Sharp.

        Behavior of reinforced concrete corbels

        Test results of thirteen reinforced concrete corbels with shear span-to-depth ratio greater than unity are reported. The main variables studied were compressive strength of concrete, shear span-todepth ratio and parameter of vertical stirrups. The test results indicate that the shear strengths of corbels increase with an increase in compressive strength of concrete and parameter of vertical stirrups. The shear strengths of corbels also increase with a decrease in shear span-to-depth ratio. The smaller the shear span-to-depth ratio of corbel, the larger the stiffness and the shear strength of corbel are. The higher the concrete strength of corbel, the higher the stiffness and the shear strength of corbel are. The larger the parameter of vertical stirrups, the larger the stiffness and the shear strength of corbel are. The softened strut-and-tie model for determining the shear strengths of reinforced concrete corbels is modified appropriately in this paper. The shear strengths predicted by the proposed model and the approach of ACI Code are compared with available test results. The comparison shows that the proposed model can predict more accurately the shear strengths of reinforced concrete corbels than the approach of ACI Code.

        Tests of reinforced concrete deep beams

        This study reports the test results of twelve reinforced concrete deep beams. The deep beams were tested with loads applied through and supported by columns. The main variables studied were the shear span-to-depth ratios, and the horizontal and vertical stirrups. The shear strengths can be effectively enhanced for deep beams reinforced with both horizontal and vertical stirrups. The test results indicate the shear strengths of deep beams increase with the decrease of the shear span-to-depth ratios. The normalized shear strengths of the deep beams did not increase proportionally with an increase in effective depth. An analytical method for predicting the shear strengths of deep beams is proposed in this study. The shear strengths predicted by the proposed method and the strut-and-tie model of the ACI Code are compared with available test results. The comparison shows the proposed method can predict the shear strengths of reinforced concrete deep beams more accurately than the strut-and-tie model of the ACI Code.

        CRISPR/Cas9-mediated knockout of the Vanin-1 gene in the Leghorn Male Hepatoma cell line and its effects on lipid metabolism

        Objective: Vanin-1 (VNN1) is a pantetheinase that catalyses the hydrolysis of pantetheine to produce pantothenic acid and cysteamine. Our previous studies have shown that the VNN1 is specifically expressed in chicken liver which negatively regulated by microRNA-122. However, the functions of the VNN1 in lipid metabolism in chicken liver haven’t been elucidated. Methods: First, we detected the VNN1 mRNA expression in 4-week chickens which were fasted 24 hours. Next, knocked out VNN1 via CRISPR/Cas9 system in the chicken Leghorn Male Hepatoma cell line. Detected the lipid deposition via oil red staining and analysis the content of triglycerides (TG), low-density lipoprotein-C (LDL-C), and highdensity lipoprotein-C (HDL-C) after VNN1 knockout in Leghorn Male Hepatoma cell line. Then we captured various differentially expressed genes (DEGs) between VNN1- modified LMH cells and original LMH cells by RNA-seq. Results: Firstly, fasting-induced expression of VNN1. Meanwhile, we successfully used the CRISPR/Cas9 system to achieve targeted mutations of the VNN1 in the chicken LMH cell line. Moreover, the expression level of VNN1 mRNA in LMH-KO-VNN1 cells decreased compared with that in the wild-type LMH cells (p<0.0001). Compared with control, lipid deposition was decreased after knockout VNN1 via oil red staining, meanwhile, the contents of TG and LDL-C were significantly reduced, and the content of HDL-C was increased in LMH-KO-VNN1 cells. Transcriptome sequencing showed that there were 1,335 DEGs between LMH-KO-VNN1 cells and original LMH cells. Of these DEGs, 431 were upregulated, and 904 were downregulated. Gene ontology analyses of all DEGs showed that the lipid metabolism-related pathways, such as fatty acid biosynthesis and long-chain fatty acid biosynthesis, were enriched. KEGG pathway analyses showed that “lipid metabolism pathway”, “energy metabolism”, and “carbohydrate metabolism” were enriched. A total of 76 DEGs were involved in these pathways, of which 29 genes were upregulated (such as cytochrome P450 family 7 subfamily A member 1, ELOVL fatty acid elongase 2, and apolipoprotein A4) and 47 genes were downregulated (such as phosphoenolpyruvate carboxykinase 1) by VNN1 knockout in the LMH cells. Conclusion: These results suggest that VNN1 plays an important role in coordinating lipid metabolism in the chicken liver.

        Traditional Chinese medicine for COVID-19 pandemic and emerging challenges: An online cross-sectional survey in China

        Background We aimed to investigate use of infection control behaviours, preventative and therapeutic interventions, and outcomes among respondents to an online survey during the COVID-19 pandemic in China. Methods The survey was designed by an international team, translated and adapted to simplified Chinese, including 132 kinds of traditional Chinese medicine (TCM) preparation recommended by guidelines. It was distributed and collected from February to May 2021, with data analysed by WPS spreadsheet and wjx.cn. Descriptive statistics were used to describe demographics and clinical characteristics, diagnosis, treatments, preventative behaviours and interventions, and their associated outcomes. Results The survey was accessed 503 times with 341 (67.8%) completions covering 23 provinces and four municipalities in China. Most (282/341, 82.7%) respondents reported no symptoms during the pandemic and the majority (290/341, 85.0%) reported having a SARS-CoV-2 PCR test at some point. Forty-five (13.2%) reported having a respiratory infection, among which 19 (42.2%) took one or more categories of modern medicine, e.g. painkillers, antibiotics; 16 (35.6%) used TCM interventions(s); while seven respondents combined TCM with modern medicine. All respondents reported using at least one behavioural or medical approach to prevention, with 22.3% taking TCM and 5.3% taking modern medicines. No respondents reported having a critical condition related to COVID-19. Conclusion We found evidence of widespread use of infection control behaviours, modern medicines and TCM for treatment and prevention of COVID-19 and other respiratory symptoms. Larger scale studies are warranted, including a more representative sample exploring TCM preparations recommended in clinical guidelines.         Dynamic and static properties of sandwich-like graphene-reinforced composite plate

        A graphene-reinforced composite plate (GRCP) with a sandwich-like structure is proposed. The nonlinear mechanical characteristics of the sandwich-like GRCP are investigated and compared to those of functionally graded and uniform GRCPs. The modified HalpinTsai model is employed to estimate the material parameters, and the Hamilton principle is used to derive the governing equation for the composite plates. The effects of the graphene nanoplatelet (GPL) distribution patterns, weight fraction, and the number of layers on the static and dynamic nonlinear responses of the plates are investigated. The results show that the various GPL distribution patterns have different enhancement effects on plates. Compared with the graded and uniform distribution patterns, the sandwich-like distribution pattern has significant advantages in terms of anti-vibration and anti-bending properties. Furthermore, it is found that the stresses of the sandwich-like GRCPs are discontinuous, there exist stress mutation phenomena between the surface layers and the pure matrix layer.

        Rapid Detection and Monitoring Therapeutic Efficacy of Mycobacterium tuberculosis Complex Using a Novel Real-Time Assay

        We combined real-time RT-PCR and real-time PCR (R/P) assays using a hydrolysis probe to detect Mycobacterium tuberculosis complex (MTBC)-specific 16S rRNA and its rRNA gene (rDNA). The assay was applied to 28 nonrespiratory and 207 respiratory specimens from 218 patients. Total nucleic acids (including RNA and DNA) were extracted from samples, and results were considered positive if the repeat RT-PCR threshold cycle was ≤35 and the ratio of real-time RT-PCR and real-time PCR load was ≥1.51. The results were compared with those from existing methods, including smear, culture, and real-time PCR. Following resolution of the discrepant results between R/P assay and culture, the overall sensitivity, specificity, positive predictive values (PPV), and negative predictive values (NPV) of all samples (including nonrespiratory and respiratory specimens) were 98.2%, 97.2%, 91.7%, and 99.4%, respectively, for R/P assay, and 83.9%, 89.9%, 72.3%, and 94.7%, respectively, for real-time PCR. Furthermore, the R/P assay of four patient samples showed a higher ratio before treatment than after several days of treatment. We conclude that the R/P assay is a rapid and accurate method for direct detection of MTBC, which can distinguish viable and nonviable MTBC, and thus may guide patient therapy and public health decisions.

        Comparative Genomics Profiling of Clinical Isolates of Helicobacter pylori in Chinese Populations Using DNA Microarray

        In order to search for specific genotypes related to this unique phenotype, we used whole genomic DNA microarray to characterize the genomic diversity of Helicobacter pylori (H. pylori) strains isolated from clinical patients in China.The open reading frame (ORF) fragments on our microarray were generated by PCR using gene-specific primers. Genomic DNA of H. pylori 26695 and J99 were used as templates. Thirty-four H. pylori isolates were obtained from patients in Shanghai. Results were judged based on ln(x) transformed and normalized Cy3/Cy5 ratios. Our microarray included 1882 DNA fragments corresponding to 1636 ORFs of both sequenced H. pylori strains. Cluster analysis, revealed two diverse regions in the H. pylori genome that were not present in other isolates. Among the 1636 genes, 1091 (66.7%) were common to all H. pylori strains, representing the functional core of the genome. Most of the genes found in the H. pylori functional core were responsible for metabolism, cellular processes, transcription and biosynthesis of amino acids, functions that are essential to H. pylori’s growth and colonization in its host. In contrast, 522(31.9%) genes were strain-specific genes that were missing from at least one strain of H. pylori. Strainspecific genes primarily included restriction modification system components, transposase genes, hypothetical proteins and outer membrane proteins. These strain-specific genes may aid the bacteria under specific circumstances during their long-term infection in genetically diverse hosts. Our results suggest 34 H. pylori clinical strains have extensive genomic diversity. Core genes and strain-specific genes both play essential roles in H. pylori propagation and pathogenesis. Our microarray experiment may help select relatively significant genes for further research on the pathogenicity of H. pylori and development of a vaccine for H. pylori.
