From 3D to 4D printing: approaches and typical applications

With the additional dimension, 4D printing is emerging as a novel technique to enable configuration switching in 3D printed items. Inthis paper, four major approaches, namely self-assembly of elements, deformation mismatch, bi-stability, and the Shape memory effect(SME), are identified as the generic approaches to achieve 4D printing. The main features of these approaches are briefly discussed. Utilizingthese approaches either individually or in a combined manner, the potential of 4D printing to reshape product design is demonstratedby a few example applications.

Insulin-like growth factor-binding protein-3 (IGFBP3) has been identified as a putative tumor suppressor with multifunctional roles in the IGF axis. Recently, there have been a growing body of studies investigating the relation between the IGFBP3 A-202C polymorphism, circulating IGFBP3 and prostate cancer risk, but their outcomes varied leading to controversy. Hence, it is necessary to perform a meta-analysis covering all eligible studies to shed a light on the association of IGFBP3 A-202C and cancer risk. Finally, we included a total of 11 relevant articles between 2003 and 2010 covering 14 case-control studies including 9,238 cases and 8,741 controls for our analysis. Our results showed that A-202C was a marginal risk factor of prostate cancer (allele contrast: OR=1.08, 95% CI :1.01-1.16; dominant model: OR=1.11, 95% CI :1.01-1.22; heterozygote codominant model: OR=1.11, 95% CI :1.03-1.18; homozygote contrast: OR=1.19, 95% CI :1.03-1.37). Stratification analysis revealed that sample size and control source were two major heterogeneous meta-factors especially in the recessive model (source: Population-based control group :p=0.30,I2=16.7%, Hospital-based control group: p=0.20, I2=30.3%; sample size: Small: p=0.22,I2= 32.8%, Medium: p=0.09,I2=48%, Large p=0.60,I2=0.0%); However, contrary to previous findings, no significance was found in racial subgroups. No significant publication bias was found in our analysis. Considering the robustness of the results and the discrepancy among some studies, there might be some unsolved confounding factors, and further more critical large studies are needed for confirmation.

This paper proposes a new 2R1T PRS-PRU-PUR overconstrained parallel mechanism (PM). Firstly, the kinematics and stiffness are studied based on screw theory and strain energy. Compared with a typical 3-PRS PM, the main advantage of the proposed PRSPRU-PUR PM is that the orientational range β is improved by 50 %. Secondly, the original PM’s kinematic and stiffness performance are evaluated by using the global transmission index and stiffness index, which are 0.675 and 594 respectively, which causes the decrease of the efficiency of some configurations. Finally, the design parameters are optimized to enhance the PM’s kinematic and stiffness performance. The corresponding optimized indices were increased to 0.915 and 2171, respectively, confirming the validity of the optimal design method.

GST pull-down assays were used to characterize the SARS-CoV membrane (M) and nucleocapsid (N) interaction, and it was found that the amino acids 211-254 of N protein were essential for this interaction. When tetrad glutamines (Q) were replaced with glutamic acids (E) at positions of 240-243 of the N protein, the interaction was disrupted.
Gene Expression Profiles of HeLa Cells Impacted by Hepatitis C Virus Non-structural Protein NS4B

By a cDNA array representing 2308 signal transduction related genes, we studied the expression profiles of HeLa cells stably transfected by Hepatitis C virus nonstructural protein 4B (HCV-NS4B). The alterations of the expression of four genes were confirmed by real-time quantitative RT-PCR; and the aldo-keto reductase family 1, member C1 (AKR1C1) enzyme activity was detected in HCV-NS4B transiently transfected HeLa cells and Huh-7, a human hepatoma cell line. Of the 2,308 genes we examined, 34 were up-regulated and 56 were down-regulated. These 90 genes involved oncogenes, tumor suppressors, cell receptors, complements, adhesions, transcription and translation, cytoskeletion and cellular stress. The expression profiling suggested that multiple regulatory pathways were affected by HCV-NS4B directly or indirectly. And since these genes are related to carcinogenesis, host defense system and cell homeostatic mechanism, we can conclude that HCV-NS4B could play some important roles in the pathogenesis mechanism of HCV.
Spillover Effects of US Monetary Policy on Asian Countries

Ever since the global financial crises of 1997 and 2008, the recurrence of crises ledus evaluate the spillover effects of US monetary policy on Asian economies (Jan. 2005 - June 2017). This study makes precise use of various monetary policies (interest rate, money supply, exchange rate, and capital account) from the United States to different blocs (ASEAN, South Asian economies, and North Asia) and their spillover effects on the corresponding variables. Panel Vector Autoregression (PVAR) was applied in the cases. The findings are as follows. First, USD exchange rate volatility affects all Asian economies, ASEAN in particular; however, it doesn’t hold for the North Asian economies. For the exchange rate via SDR standard, only the South Asian countries are subjected to dollar volatility. The Fed money supply is ranked second in spillover effects, especially for the South Asian economies. The Fed’s policy interest exerts an impact on ASEAN, Northeast Asia, and South Asian economies. However, the US capital account movement only affects the latter two. The reasons behind the various intensities of spillover effects are interpreted accordingly. Regional location and size of economy might serve as an explanation for the spillover.

$N^1$-(2, 5-dimethoxyphenyl)-$N^8$-hydroxyoctanediamide (N25) is a novel SAHA cap derivative of HDACi, with a patent (No. CN 103159646). This invention is a hydroxamic acid compound with a structural formula of $RNHCO(CH_2)6CONHOH$ (wherein R=2, 5dimethoxyaniline), a pharmaceutically acceptable salt which is soluble. In the present study, we investigated the effects of N25 with regard to drug distribution and molecular docking, and anti-proliferation, apoptosis, cell cycling, and $LD_{50}$. First, we designed a molecular approach for modeling selected SAHA derivatives based on available structural information regarding human HDAC8 in complex with SAHA (PDB code 1T69). N25 was found to be stabilized by direct interaction with the HDAC8. Anti-proliferative activity was observed in human glioma U251, U87, T98G cells and human lung cancer H460, A549, H1299 cells at moderate concentrations ($0.5-30{\mu}M$). Compared with SAHA, N25 displayed an increased antitumor activity in U251 and H460 cells. We further analyzed cell death mechanisms activated by N25 in U251 and H460 cells. N25 significantly increased acetylation of Histone 3 and inhibited HDAC4. On RT-PCR analysis, N25 increased the mRNA levels of p21, however, decreased the levels of p53. These resulted in promotion of apoptosis, inducing G0/G1 arrest in U251 cells and G2/M arrest in H460 cells in a time-dependent and dose-dependent manner. In addition, N25 was able to distribute to brain tissue through the blood-brain barrier of mice ($LD_{50}$: 240.840mg/kg). In conclusion, our findings demonstrate that N25 will provide an invaluable tool to investigate the molecular mechanism with potential chemotherapeutic value in several malignancies, especially human glioma.
