        Evaluation of Renal Pathophysiological Processes Induced by an Iodinated Contrast Agent in a Diabetic Rabbit Model Using Intravoxel Incoherent Motion and Blood Oxygenation Level-Dependent Magnetic Resonance Imaging

        Objective: To examine the potential of intravoxel incoherent motion (IVIM) and blood oxygen level-dependent (BOLD) magnetic resonance imaging for detecting renal changes after iodinated contrast-induced acute kidney injury (CI-AKI) development in a diabetic rabbit model. Materials and Methods: Sixty-two rabbits were randomized into 2 groups: diabetic rabbits with the contrast agent (DCA) and healthy rabbits with the contrast agent (NCA). In each group, 6 rabbits underwent IVIM and BOLD imaging at 1 hour, 1 day, 2 days, 3 days, and 4 days after an iohexol injection while 5 rabbits were selected to undergo blood and histological examinations at these specific time points. Iohexol was administrated at a dose of 2.5 g I/kg of body weight. Further, the apparent transverse relaxation rate (R2*), average pure molecular diffusion coefficient (D), pseudo-diffusion coefficient (D*), and perfusion fraction (f) were calculated. Results: The D and f values of the renal cortex (CO) and outer medulla (OM) were significantly decreased compared to baseline values in the 2 groups 1 day after the iohexol injection (p < 0.05). A marked reduction in the D* values for both the CO and OM was also observed after 1 hour in each group (p < 0.05). In the OM, a persistent elevation of the R2* was detected for 4 days in the DCA group (p < 0.05). Histopathological changes were prominent, and the pathological features of CI-AKI aggravated in the DCA group until day 4. The D, f, and R2* values significantly correlated with the histological damage scores, hypoxiainducible transcription factor-1α expression scores, and serum creatinine levels. Conclusion: A combination of IVIM and BOLD imaging may serve as a noninvasive method for detecting and monitoring CIAKI in the early stages in the diabetic kidney.

      • Energy factor of high-strength-steel frames with energy dissipation bays under repeated near-field earthquakes

        This investigation contributes to quantification of the inelastic seismic demands for high strength steel moment resisting frames equipped with energy dissipation bays (HSSF-EDBs) subjected to seismic sequences composed of repeated near-field ground motions. The emphasis is placed on the energy factor demand. A statistical examination of a database with more than eighty million energy factor demands of single-degree-of-freedom (SDOF) oscillators representing HSSF-EDBs responding in different yielding stages is conducted. The research findings show that in the damage-control stage, the energy factor which quantifies the peak seismic demand of a HSSF-EDB structure is insensitive to the repeated near-field earthquake motions. In contrast, a remarkable elevation of the energy factor is observed when oscillators characterising HSSF-EDBs progress into the ultimate stage. In addition, an increasing post-yielding stiffness ratio of the nonlinear force-displacement response in the damage-control stage may produce a detrimental effect on HSSF-EDBs progressing into the ultimate stage under repeated near-field earthquakes due to the corresponding evident increase of seismic demands. A nonlinear regression model quantifying the mean energy factor demand of the system under repeated near-field earthquake motions is proposed to facilitate performance-based seismic design of HSSF-EDBs.

      • Graphene transistor based on tunable Dirac fermion optics

        <P><B>Significance</B></P><P>We report an electrically tunable graphene quantum switch based on Dirac fermion optics (DFO), with electrostatically defined analogies of mirror and collimators utilizing angle-dependent Klein tunneling. The device design allows a previously unreported quantitative characterization of the net DFO contribution and leads to improved device performance resilient to abrupt change in temperature, bias, doping, and electrostatic environment. The electrically tunable collimator and reflector demonstrated in this work, and the capability of accurate in situ characterization of their performance, provide the building blocks toward more complicated functional quantum device architecture such as highly integrated electron-optical circuits.</P><P>We present a quantum switch based on analogous Dirac fermion optics (DFO), in which the angle dependence of Klein tunneling is explicitly utilized to build tunable collimators and reflectors for the quantum wave function of Dirac fermions. We employ a dual-source design with a single flat reflector, which minimizes diffusive edge scattering and suppresses the background incoherent transmission. Our gate-tunable collimator–reflector device design enables the quantitative measurement of the net DFO contribution in the switching device operation. We obtain a full set of transmission coefficients between multiple leads of the device, separating the classical contribution from the coherent transport contribution. The DFO behavior demonstrated in this work requires no explicit energy gap. We demonstrate its robustness against thermal fluctuations up to 230 K and large bias current density up to 10<SUP>2</SUP> A/m, over a wide range of carrier densities. The characterizable and tunable optical components (collimator–reflector) coupled with the conjugated source electrodes developed in this work provide essential building blocks toward more advanced DFO circuits such as quantum interferometers. The capability of building optical circuit analogies at a microscopic scale with highly tunable electron wavelength paves a path toward highly integrated and electrically tunable electron-optical components and circuits.</P>

        Zero-Current Phenomena Analysis of the Single IGBT Open Circuit Faults in Two-Level and Three-Level SVGs

        The fact that the reliability of IGBTs has become a more and more significant aspect of power converters has resulted in an increase in the research on the open circuit (OC) fault location of IGBTs. When an OC fault occurs, a zero-current phenomena exists and frequently appears, which can be found in a lot of the existing literature. In fact, fault variables have a very high correlation with the zero-current interval. In some cases, zero-current interval actually decides the most significant fault feature. However, very few of the previous studies really explain or prove the zero-current phenomena of the fault current. In this paper, the zero-current phenomena is explained and verified through mathematical derivation, based on two-level and three-level NPC static var generators (SVGs). Mathematical models of single OC fault are deduced and it is concluded that a zero-current interval with a certain length follows the OC faults for both two-level and NPC three-level SVGs. Both inductive and capacitive reactive power situations are considered. The unbalanced load situation is discussed. In addition, simulation and experimental results are presented to verify the correctness of the theoretical analysis.

      • Stress-induced expression of the sweetpotato gene <i>IbLEA14</i> in poplar confers enhanced tolerance to multiple abiotic stresses

        <P><B>Abstract</B></P> <P>Late embryogenesis abundant (LEA) proteins are small, highly hydrophilic proteins that act as protectors of macromolecules and increase abiotic stress tolerance in plants. We previously reported that overexpressing sweetpotato <I>IbLEA14</I> under the control of the <I>CaMV 35S</I> promoter increased osmotic and salt stress tolerance in transgenic sweetpotato calli. In this study, we generated transgenic poplar plants (<I>Populus alba × P. glandulosa</I>) expressing <I>IbLEA14</I> under the control of the oxidative stress-inducible <I>SWPA2</I> promoter (referred to as SL plants). Among the 15 SL plants obtained, three lines (SL2, SL7, and SL12) were established based on <I>IbLEA14</I> transcript levels, tolerance to salt stress and Southern blot analysis. The SL plants exhibited less damage in response to methyl viologen-mediated oxidative stress than non-transgenic (NT) plants. SL plants also showed enhanced tolerance to drought, salt, and heat stress, which was associated with higher photosystem II efficiency and lower malondialdehyde levels compared with NT plants. Furthermore, SL plants had higher levels of monolignol biosynthesis-related gene transcripts under drought stress compared with NT plants. Finally, SL plants exhibited increased tolerance to heat stress, which is associated with the high thermostability of IbLEA14 protein. SL plants might be useful for reforestation on global marginal lands, including desertification and reclaimed areas.</P> <P><B>Highlights</B></P> <P> <UL> <LI> <I>IbLEA14</I> gene was transformed into poplar plants. </LI> <LI> Transgenic poplars exhibit increased tolerance to MV-mediated oxidative, drought, salt and heat stress. </LI> <LI> Overproduction of IbLEA14 affects lignification and thermostability of transgenic poplars. </LI> </UL> </P>

        Open source board based acoustofluidic transwells for reversible disruption of the blood–brain barrier for therapeutic delivery

        Background Blood–brain barrier (BBB) is a crucial but dynamic structure that functions as a gatekeeper for the central nervous system (CNS). Managing sufficient substances across the BBB is a major challenge, especially in the development of therapeutics for CNS disorders. Methods To achieve an efficient, fast and safe strategy for BBB opening, an acoustofluidic transwell (AFT) was developed for reversible disruption of the BBB. The proposed AFT was consisted of a transwell insert where the BBB model was established, and a surface acoustic wave (SAW) transducer realized using open-source electronics based on printed circuit board techniques. Results In the AFT device, the SAW produced acousto-mechanical stimulations to the BBB model resulting in decreased transendothelial electrical resistance in a dose dependent manner, indicating the disruption of the BBB. Moreover, SAW stimulation enhanced transendothelial permeability to sodium fluorescein and FITC-dextran with various molecular weight in the AFT device. Further study indicated BBB opening was mainly attributed to the apparent stretching of intercellular spaces. An in vivo study using a zebrafish model demonstrated SAW exposure promoted penetration of sodium fluorescein to the CNS. Conclusions In summary, AFT effectively disrupts the BBB under the SAW stimulation, which is promising as a new drug delivery methodology for neurodegenerative diseases.

        Molecular and functional characterization of the adiponectin (AdipoQ) gene in goat skeletal muscle satellite cells

        Objective: It is commonly accepted that adiponectin binds to its two receptors to regulate fatty acid metabolism in adipocytes. To better understand their functions in the regulation of intramuscular adipogenesis in goats, we cloned the three genes (adiponectin [AdipoQ], adiponectin receptor 1 [AdipoR1], and AdipoR2) encoding these proteins and detected their mRNA distribution in different tissues. We also determined the role of AdipoQ in the adipogenic differentiation of goat skeletal muscle satellite cells (SMSCs). Methods: SMSCs were isolated using 1 mg/mL Pronase E from the longissimus dorsi muscles of 3-day-old female Nanjiang brown goats. Adipogenic differentiation was induced in satellite cells by transferring the cells to Dulbecco's modified Eagle's medium supplemented with an isobutylmethylxanthine, dexamethasone and insulin cocktail. The pEGFP-N1-AD plasmid was transfected into SMSCs using Lipofectamine 2000. Expression of adiponectin in tissues and SMSCs was detected by quantitative polymerase chain reaction and immunocytochemical staining. Results: The three genes were predominantly expressed in adipose and skeletal muscle tissues. According to fluorescence and immunocytochemical analyses, adiponectin protein expression was only observed in the cytoplasm, suggesting that adiponectin is localized to the cytoplasm of goat SMSCs. In SMSCs overexpressing the AdipoQ gene, adiponectin promoted SMSC differentiation into adipocytes and significantly (p<0.05) up-regulated expression of AdipoR2, acetyl-CoA carboxylase, fatty-acid synthase, and sterol regulatory element-binding protein-1, though expression of CCAAT/enhancer-binding $protein-{\alpha}$, peroxisome proliferator-activated receptor ${\gamma}$, and AdipoR1 did not change significantly. Conclusion: Adiponectin induced SMSC differentiation into adipocytes, indicating that adiponectin may promote intramuscular adipogenesis in goat SMSC.

        Production of Polyhydroxyalkanoates (PHA) by Haloferax mediterranei from Food Waste Derived Nutrients for Biodegradable Plastic Applications

        Polyhydroxyalkanoates (PHA) are a family of microbial polyesters that are used as biodegradable plastics in replacement of conventional plastics for various applications. However, the high production cost is the barrier for PHA market expansion. This study aimed to utilize food waste as low-cost feedstock to produce poly(3-hydroxybutyrate-co-3-hydroxyvalerate) (PHBV) by Haloferax mediterranei. The effects of acetate (Ac), propionate (Pr), butyrate (Bu), and the short-chain carboxylates derived from food waste were examined on the microbial growth and PHBV production. Results showed that a mixture of carboxylates provided a 55% higher PHBV yield than glucose. The food-waste-derived nutrients achieved the yields of 0.41 to 0.54 g PHBV/g Ac from initial loadings of 450 mg/l to 1,800 mg/l Ac of total carboxylates. And the consumption of individual carboxylate varied between different compositions of the carbon source. The present study demonstrates the potential of using food waste as feedstock to produce PHBV by Haloferax mediterranei, which can provide economic benefits to the current PHA industry. Meanwhile, it will also help promote organic waste reduction in landfills and waste management in general.

      • Gate-tunable current partition in graphene-based topological zero lines

        <P>We demonstrate new mechanisms for gate-tunable current partition at topological zero-line intersections in a graphene-based current splitter. Based on numerical calculations of the nonequilibrium Green's functions and Landauer-Buttiker formula, we show that the presence of a perpendicular magnetic field on the order of a few Teslas allows for carrier sign dependent current routing. In the zero-field limit the control on current routing and partition can be achieved within a range of 10-90% of the total incoming current by tuning the carrier density at tilted intersections or by modifying the relative magnitude of the bulk band gaps via gate voltage. We discuss the implications of our findings in the design of topological zero-line networks where finite orbital magnetic moments are expected when the current partition is asymmetric.</P>
