        Production of a Recombinant Laccase from Pichia pastoris and Biodegradation of Chlorpyrifos in a Laccase/Vanillin System

        The recombinant strain P. pastoris GS115-lccC was used to produce laccase with high activity. Factors influencing laccase expression, such as pH, methanol concentration, copper concentration, peptone concentration, shaker rotate speed, and medium volume were investigated. Under the optimal conditions, laccase activity reached 12,344 U/L on day 15. The recombinant enzyme was purified by precipitating and dialyzing to electrophoretic homogeneity, and was estimated to have a molecular mass of about 58 kDa. When guaiacol was the substrate, the laccase showed the highest activity at pH 5.0 and was stable when the pH was 4.5~6.0. The optimal temperature for the laccase to oxidize guaiacol was 60℃, but it was not stable at high temperature. The enzyme could remain stable at 30℃ for 5 days. The recombinant laccase was used to degrade chlorpyrifos in several laccase/mediator systems. Among three synthetic mediators (ABTS, HBT, VA) and three natural mediators (vanillin, 2,6-DMP, and guaiacol), vanillin showed the most enhancement on degradation of chlorpyrifos. Both laccase and vanillin were responsible for the degradation of chlorpyrifos. A higher dosage of vanillin may promote a higher level of degradation of chlorpyrifos, and the 2-step addition of vanillin led to 98% chlorpyrifos degradation. The degradation of chlorpyrifos was faster in the L/V system (kobs = 0.151) than that in the buffer solution (kobs = 0.028).

        Raman Lidar for the Measurement of Temperature, Water Vapor, and Aerosol in Beijing in the Winter of 2014

        To measure atmospheric temperature, water vapor, and aerosol simultaneously, an efficient multi-function Raman lidar using an ultraviolet-wavelength laser has been developed. A high-performance spectroscopic box that utilizes multicavity interference filters, mounted sequentially at small angles of incidence, is used to separate the lidar return signals at different wavelengths, and to extract the signals with high efficiency. The external experiments are carried out for simultaneous detection of atmospheric temperature, water vapor, and aerosol extinction coefficient in Beijing, under clear and hazy weather conditions. The vertical profiles of temperature, water vapor, and aerosol extinction coefficient are analyzed. The results show that for an integration time of 5 min and laser energy of 200 mJ, the mean deviation between measurements obtained by lidar and radiosonde is small, and the overall trend is similar. The statistical temperature error for nighttime is below 1 K up to a height of 6.2 km under clear weather conditions, and up to a height of 2.5 km under slightly hazy weather conditions, with 5 min of observation time. An effective range for simultaneous detection of temperature and water vapor of up to 10 km is achieved. The temperature-inversion layer is found in the low troposphere. Continuous observations verify the reliability of Raman lidar to achieve real-time measurement of atmospheric parameters in the troposphere.

      • Construction enterprise cost management via value chain analysis

        Intense competition, lower tender and bid price, the owner"s nonstandard behaviors etc. have lowered the economic benefit of construction enterprises nowadays. Therefore, construction enterprises have to reinforce cost management and improve competitiveness to survivie and develop. With the overall improvement of level of management of safety, quality and technology of construction industry, cost management has become the most important factor assessing the competitiveness, viability and development potential of construction enterprises. Cost management by value chain anaysis focuses on the essence of cost and provides a new concept for solving problems of cost management of construction enterprises. Value chain of construction industry and internal value chain, longitudinal value chain and transverse value chain of construction enterprises were analyzed by value chain theories and methods and based on the review of the status quo of cost management of construction enterprises in China. Value chain cost management methods were proposed for construction enterprises. Acquirement of cost advantage by value chain analysis and its significance were discussed, which is helpful for construction enterprises to realize strategic cost management and enhance competitiveness.

        Thermo-triggerable self-assembly comprising cinnamoyl polymeric β cyclodextrin and cinnamoyl Pluronic F127

        <P><B>Abstract</B></P> <P>Thermo-triggerable self-assembly was prepared by co-dissolving cinnamoyl Pluronic F127 (CinPlu) and cinnamoyl polymeric β cyclodextrin (CinPβCD) in an aqueous phase. On TEM photo, the CinPlu/CinPβCD self-assembly was 100–200nm in diameter. The specific loading of Nile red (NR) in the assembly was calculated to be 5.5% (wt NR/wt polymer), and the molar ratio of NR to βCD residue in the assembly was about 0.89:1. No significant release of NR from the assembly was observed at 10°C and 20°C. However, when the temperature was raised to 30°C, 40°C, 50°C, and 60°C, the cumulative release amount in 5min was 17%, 25%, 32%, and 52%, respectively. The specific loading of doxorubicin (DOX) in the assembly was about 6.8% (wt DOX/wt polymer) (corresponding to the molar ratio of DOX to βCD residue was about 0.41:1). The DOX release from the assembly was proportional to the temperature of release medium. NR and DOX were likely to be expelled out of the cavity of βCD residue by the interaction of the thermally hydrophobicized Pluronic F127 chain (molecular piston) and the cavity of βCD residue (cylinder). After 4h-incubation with KB cell, DOX loaded in CinPlu/CinPβCD self-assembly was found to be internalized into the cancer cell more than free DOX, observed on a confocal laser scanning microscope and a fluorescence activated cell sorter. CinPlu/CinPβCD self-assembly enhanced the in vitro anti-cancer activity of DOX against KB cell without increasing significantly the in vitro toxicity of DOX against Raw264.7 cell.</P> <P><B>Highlights</B></P> <P> <UL> <LI> Cinnamoyl Pluronic F127 and cinnamoyl polymeric β cyclodextrin are self-assembled. </LI> <LI> The release amount of doxorubicin from the assembly is temperature-dependent. </LI> <LI> Doxorubicin seems to release by the interaction of β cyclodextrin and Pluronic F127. </LI> <LI> The assembly enhances the in vitro anti-cancer activity of doxorubicin. </LI> </UL> </P> <P><B>Graphical abstract</B></P> <P>[DISPLAY OMISSION]</P>

      • Vesicles composed of fatty acid and N-[3-(dimethylamino)propyl]-octadecanamide: effect of fatty acid chain length on physicochemical properties of vesicles

        <P><I>Background and objectives</I>: Vesicles, recently claimed as drug delivery carriers, were prepared by taking advantage of an electrostatic interaction between the carboxylic groups of fatty acids (FAs) and the amino groups of N-[3-(Dimethylamino)propyl]-Octadecanamide (DMAPODA). The study is to find out the effect of FAs' chain length on physicochemical properties of vesicles.</P><P><I>Methods</I>: Decanoic acid (DA), myristic acid (MA), stearic acid (SA) and behenic acid (BA) were used as FAs. Vesicles composed of them and DMAPODA were studied about formation on microscope, calorimetric analysis, size and zeta potential.</P><P><I>Results</I>: On microphotographs, all of FAs could form vesicles when mixed with DMAPODA in an equi-molar ratio. However, DA/DMAPODA vesicles were disintegrated during the homogenization. Due to the asymmetry of DA/DMAPODA associate, it seems to hardly form a stable and well-packed bilayer. On thermograms, the vesicles exhibited one strong peak, indicating that FAs can be homogeneously mixed with the cationic amphiphile. The sizes of the four kinds of vesicles suspended in an aqueous solution (final pH 7.5) were in the same order (hundreds of nanometer) on microphotographs. But, on a light scattering machine, the mean size of MA/DMAPODA vesicle was measured to be much greater than those of the other vesicles, possibly because of the low absolute value of the zeta potential. In addition, the medium pH value had a significant effect on the size of BA/DMAPODA vesicle possibly because the zeta potential was strongly dependent on the pH value.</P><P><I>Conclusion</I>: FAs' chain length would affect the physicochemical properties of vesicles composed of them and DMAPODA.</P>

        Risk Factors for Anxiety in Major Depressive Disorder Patients

        Objective: To analyze the sociodemographic and clinical factors related to anxiety in patients with major depressive disorder (MDD). Methods: This study involved a secondary analysis of data obtained from the Diagnostic Assessment Service for People with Bipolar Disorders in China (DASP), which was initiated by the Chinese Society of Psychiatry (CSP) and conducted from September 1, 2010 to February 28, 2011. Based on the presence or absence of anxiety-related characteristics, 1,178 MDD patients were classified as suffering from anxious depression (n=915) or non-anxious depression (n=263), respectively. Results: Compared with the non-anxious group, the anxious-depression group had an older age at onset (t=−4.39, p<0.001), were older (t=−4.69, p<0.001), reported more lifetime depressive episodes (z=−3.24, p=0.001), were more likely to experience seasonal depressive episodes (χ2=6.896, p=0.009) and depressive episodes following stressful life events (χ2=59.350, p <0.001), and were more likely to have a family history of psychiatric disorders (χ2=6.091, p=0.014). Their positive and total scores on the Mood Disorder Questionnaire (MDQ) and the 32-item Hypomania Checklist (HCL-32) (p<0.05) were also lower. The logistic regression analysis indicated that age (odds ratio [OR]=1.03, p<0.001), a lower total MDQ score (OR=0.94, p=0.011), depressive episodes following stressful life events (OR=3.04, p<0.001), and seasonal depressive episodes (OR=1.75, p=0.039) were significantly associated with anxious depression. Conclusion: These findings indicate that older age, fewer subclinical bipolar features, an increased number of depressive episodes following stressful life events, and seasonal depressive episodes may be risk factors for anxiety-related characteristics in patients with MDD.

        본 연구에서 유도결합형 플라즈마 발생 장치를 사용하였으며, Ar gas를 사용하여 플라즈마를 발생시켰다. Langmuir probe, cut-off probe를 이용해 인가 파워 및 압력 변화에 따른 플라즈마 특성을 관찰하였다. 압력이 증가할수록 플라즈마 전위와 전자 온도는 감소하며, 플라즈마의 밀도는 증가함을 알 수 있었다.
