Reactive oxygen species(ROS) have been investigated to have pivotal roles on amyloidogenecity of $\beta-amyloidpeptide(A\beta)$, the major component of senile plaques in Alzheimer's disease(AD) brain. Addition of radical scavengers is one of the on-going strategies for therapeutic treatment for AD patients. Hsp104 protein including two ATP binding sites from Saccharomyces cerevisiae, as a molecular chaperone, was known to function as a protector of ROS generation when exposed to oxidative stress in our previous study. This observation has led us to investigate Hsp104 protein as a molecular mediator of $A{\beta}$ aggregation in this study. We have developed a new way of expression for Hsp104 protein using GST-fusion tag. As we expected, formation of $A{\beta}$ aggregate was protected by wild type Hsp104 protein, but not by the two ATP-binding site mutant, based on Thioflavin-T fluorescence. Interestingly, Hsp104 protein was observed to keep $A{\beta}$ from forming aggregates independent of ATP binding. On the other hand, disaggregation of $A{\beta}$ aggregates by wild type Hsp104 was totally dependent on the presence of ATP. On the other hand, mutant Hsp104 with two ATP binding sites altered exhibited no inhibition. Another effective antioxidant, hydrazine analogs of curcumin were also effective in $A{\beta}$ fibrilization as protectors against oxidative stress. Based on these observations we conclude that Hsp104 and curcumin derivatives, as protectors of oxidative stress, inhibit $A{\beta}$ aggregation in virto and can be candidates for therapeutic approaches in cure of some neurodegenerative disease.

N-Hydroxy diketopiperazine derivatives from L- and D-forms of Ser and Cys were synthesized for the first time and the ring conformation of both analogues in water was determined to be close to flat via NMR and ab initio calculations. However, the side chain of the Ser analogue was oriented toward a pseudo-equatorial position while that of the Cys analogue was slightly oriented toward a pseudo-axial direction, with a slightly distorted boat-shaped ring conformation. Among them, the D-Cys analogue was found to be a weak inhibitor of a-glucosidase.

This paper describes the spherical ammonium diuranate gel particles which are the intermediated material of the $UO_2$ microsphere for an VHTR(very high temperature reactor) nuclear fuel. The characteristics of the intermediate-ADU gel particles prepared by AWD(ageing, washing, and drying) and FB(fluidized-bed) apparatus were examined and compared in a sol-gel fabrication process. The electrical conductivity of washing filtrate from the FB treating and the surface area of dried-ADU gel particles were higher than those of AWD treating. Also, an internal pore volume in dried-ADU gel particles showed a more decrease in AWD treatment than FB treatment because of decomposition of PVA affected by the washing time. However, the internal microstructures of ADU gel particles were similar regardless of the process variation.

Background/Aims : Wilson's disease in an autosomal recessive genetic disorder which is characterized by hepatic and neurologic manifestations due to defect in copper metabolism. The progression of hepatic damage and critical neurologic complication can be prevented by early diagnosis and treatment. Our study investigated the usefulness of clinical manifestations and laboratory findings in Wilson's disease to early diagnosis of wilson's disease. Method : We investigated the 10 patients who are diagnosed as Wilson's disease at Chungnam national university Hospital during the recent 5 years for clinical manifestation, laboratory findings and treatment. Results : 1. Females are 9 patients(90%) among 10 objective patients and the median age of patients is 30.2±14 years. The second and third decade are the more common prevalent and 5 patients(50%) have family history. 2. The clinical manifestations of Wilson's disease at time of diagnosis are hepatic manifestation in 9 patients (90%) except presymptomatic patient and the psycho-neurologic manifestations are appeared in 5 patients(50%). Three patients have the secondary amenorrhea, hemolytic anemia and recurrent spontaneous abortions in each case. 3. The serum ceruloplasmin concentrations were lower than 20mg/dl in 8 patient(80%) including presymptomatic patient and the 24 hours urine copper excretion is above 100μg/24hrs in 7 patients(70%). 4. Seven patients(70%) with symptomatic Wilson's disease have the detectable Kayser-Fleischer ring in slit lamp examinations but one presymptomatic patient has no Kayser-Fleischer ring. 5. The serum level of ceruloplasmin is increased in 3 patients (42.8%) among 7 patients who are had been treated by penicillamine(one patient is not followed up serum ceruloplasmin) and the 24 hours urine copper excretion was decreased in 6 patient(75%) among 8 patients treated by penicillamine. 6. Five patients died with hepatic failure related to uncompensated liver cirrhosis. Conclusion : The slit lamp examination should be recommended in patients with hepatic and neurologic disease of unknown etiology and sibling of the patients wilson's disease. The irrerersible damage of organs would be prevented by aggressive treatment at early state in patients diagnosed as Wilson's disease.
만성 활동성 C형 간염 환자에서 알파 인터페론 2b 투여에 의한 HCV-RNA의 소실

Background/Aims : Interferon-alpha is the most effective agent for the treatment of chronic active hepatitis C(CAH-C). The purpose of this study are to evaluate the efficacy of alpha-interferon therapy for the CAH-C and to clarify the correlations between the biochemical and anti-viral responses. Methods : The presence of serum hepatitis C virus(HCV) with increased serum ALT(Alanine aminotransferase) activities were monitored in 18 patients with CAH-C confirmed by serologic studies and liver biopsy from July 1992 to July 1998. The patients were treated with interferon alpha 2b for 6 months and the other eight patients were selected as the control group. Results : The serum ALT levels became within normal limit in 9 of 10 patients(90%) after interferon treatment (End-of-treatment response;ETR). Sustained responses(SR) were achieved in 7 cases(70%) for follow-up period of 18 to 24 months (SR-12month). Two cases were relapsed responders who showed elevated ALT levels within 1-2 months after cessation of interferon therapy and the other case was a non-responders (NR). In all 7 sustained responders (SR), HCV-RNAs were negative at the end of the treatment. But HCV-RNA reappeared in the serum in the case of the relapsed responders during follow-up period after treatment. While none of 8 patients, control group showed normal ALT levels during 18-24 months follow-up period, and the HCV-RNA states did not change in all the control group. Conclusion : This study suggest that the 3 million units of interferon alpha 2b, T.I.W.(three times a week) for 6-month course is effective in the treatment of CAH-C, especially in younger age and shorter duration of disease. The disappearance of serum HCV-RNA is closely associated with the normalization of serum ALT levles, so that the serial measurement of HCV-RNA may be a reliable marker in the efficacy of interferon treatment and predicting relapse.
