Quantum chemical analysis of laccase-mediator system on degradation of environmental pollutants.

Many researchers reported biodegradation of environmental pollutants by white-rot fungi. Toward in situ bioremediation, we have investigated biodegradation of environmental pollutants by litter-decomposing fungi. In our results, lignin-degrading enzymes produced from litter-decomposing fungi are thought to participate in degradation of pentachlorophenol (PCP) and 2,4-dichlorophenoxy acetic acid (2,4-DA). Then, we examined the biodegradation of PCP, 2,4-DA and the analogs of 2,4-DA by purified laccase and the role of redox mediator on laccase reaction. Laccase purified from Calvatia craniiformis decomposed phenolic compounds, PCP and 2,4-dichlorophenol (2,4-DP), but not non-phenolic compounds, 2,4-DA and 2,4-dichlorophenoxy ethanol (2,4-DE), even in the presence of redox mediators. To clarify the reaction mechanism between the substrates and redox mediators, quantum chemical analysis was applied using MOPAC 2009 and Gaussian 03. The results of the heat of formation and the perturbation energy showed that even redox mediator radicals could not oxidize the non-phenolic compounds. Previously several reports showed that laccase-redox mediator systems decomposed non-phnolic compounds, but we propose that the system could not react on the chlorinated aromatic compounds based on the result of quantum analysis.
A PARTIAL SKELETON OF TITANOSAURIAN SAUROPOD FROM THE EARLY CRETACEOUS OF TOBA CITY, CENTRAL JAPAN

일본 중앙의 Toba시에 있는 전기 백악기 (Valanginian~Barremian) Matsuo Group에서 용각류 골격 일부가 발굴되었다. 4개의 꼬리 척추뼈, 오른쪽 왼쪽 상박골과 대퇴골이 인지되었으며 왼쪽 요골과 오른쪽 경골, 오른쪽 비골, 그리고 왼쪽 좌골(?)의 동정은 뼈가 불완전하여 잠정적이다. 이 공룡은 다음과 같은 형태적 특징에 기초해 Upchurch et al. (2004)의 분류를 따르면 Titanosauria에 속한 것으로 확인된다. 특징은 (1) 대퇴골의 4th trochanter가 shaft의 안뒤쪽에 위치하며 (2) 상완골과 대퇴골 길이의 비는 대략 0.89 이고 (3) 대퇴골의 상부 좌측 표면이 뚜렷하게 부풀어 있으며 (4) 중간 미추의 신경궁은 추체의 앞쪽에 위치하고 (5) 대퇴골의 단면은 매우 비대칭 (= 2.3)이다. 게다가 미추는 다음과 같은 특징이 있다. (1) 앞쪽 미추의 추체는 amphicoelous이며 (2) 앞쪽 미추체의 길이와 높이의 비는 1.0이 넘고 (3) 앞쪽 미추체의 하부 excavation은 각 양쪽 ventrolateral ridge에 둘러싸여있으며 (4) 중간 미추체의 단면은 위아래로 납작하다. 원시적인 특징 (1)과 (2)와 함께 진화된 특징인 (3)과 (4)의 조합은 Titanosauria 그룹 내에서 독특하며 이 공룡이 새로운 속과 종일 가능성을 암시한다. 일본의 전기 백악기에서 산출된 새로운 기능성이 있는 티타노사우루스류 용각류의 발견은 원시적인 티타노사우루스류가 아시아에 더 다양했으며 이들의 진화가 이전에 생각했던 것보다 더 복잡했다는 것을 지시한다. A partial skeleton of a sauropod dinosaur was excavated from the Lower Cretaceous Matsuo Group (Valanginian to Barremian) in Toba City, central Japan. It preserves four caudal vertebrae, right and left humeri, and right and left femora. Other recovered bones are tentatively identified as left radius, right tibia, right fibula, and left ischium because of their fragmental preservation. This dinosaur is identified as a member of Titanosauria because it represents synapomorphies of the clade: (1) femoral fourth trochanter located on the caudomedial margin of the shaft, (2) approximately 0.89 of the humerus/femur length ratio, (3) presence of prominent "bulge" on the proximal lateral surface of the femur, (4) neural arch of middle caudal vertebra situated on the cranial half of the centrum, and (5) extreme eccentricity of the femur cross section (=2.3). The combination of other characters may suggest that this dinosaur is a new taxon. The discovery of a possibly new titanosaurian dinosaur from the Early Cretaceous of Japan indicates that titanosaurians were more diverse in Asia, and their evolution was more complicated than previously thought.

The dynamic process of gas absorption from a CO₂ bubble into a liquid is examined in the presence of satellite bubbles. The bubble under consideration is held stationary, except its jittering, by the liquid flowing downward. The mass transfer rate is determined by monitoring the rate of reduction in the equivalent bubble diameter during the initial absorption process. It is found that the interaction with the satellite bubbles generally hampers the dissolution of the primary bubble. The extent of reduction in the dissolution rate increases with the net contacting time during the interaction. When the secondary bubbles interact with the primary bubble mainly outside of its wake, however, the dissolution tends to be enhanced due to induced turbulence in the surrounding liquid flow. A simple theoretical model is developed to simulate the observed results as well as the basic features prevailing in a recently proposed scheme, called the GLAD system, for shallow injection of CO₂ gas into seawater.

Background: Goshajinkigan (GJG) is used for the treatment of several neurological symptoms. We investigated the efficacy of GJG and mecobalamin (B12) against neurotoxicity associated with docetaxel (DOC) in breast cancer patients. Materials and Methods: Sixty breast cancer patients were treated with DOC. Thirty-three patients (GJG group) received oral administration of 7.5 g/day GJG and 27 patients (B12 group) received oral administration of 1500 ${\mu}g/day$ B12. Neuropathy was evaluated according to DEB-NTC (Neurotoxicity Criteria of Debiopharm), Common Terminology Criteria for Adverse Events (NCI-CTC) ver. 3.0, and a visual analogue scale (VAS). This study employed a randomized open design. Results: The incidence of neuropathy was 39.3% in the GJG group, and 88.9% in the B12 group (p<0.01). In the GJG group, grade 1 DEB-NTC was observed in 2 cases, grade 2 in 5 cases and grade 3 in 5 cases. Grade 1 NCI-CTC was observed in 7 cases, grade 2 in 6 cases, and VAS was $2.7{\pm}2.2$. In the B12 group, grades 1, 2 and 3 DEB-NTC were observed in one case, 12 cases and 12 cases, respectively; and grades 1, 2 and 3 NCI-CTC were observed in 11 cases, 12 cases and one case, and VAS was $4.9{\pm}2.4$. Conclusions: Concomitant administration of GJG is useful in preventing neuropathy in breast cancer patients treated with a DOC regimen.

Background: We compared treatment completion rates and safety of docetaxel and cyclophosphamide sixcycle therapy (TC6) with docetaxel followed by 5FU, epirubicin and cyclophosphamide (T-FEC) therapy in Japanese patients with human epidermal growth factor receptor 2 (HER2)-negative breast cancer. Materials and Methods: We administered TC6 q3w or T-FEC q3w to HER2-negative breast cancer patients. The primary endpoint of this trial was toxicity. As second endpoints, the treatment completion rate and relative dose intensity were evaluated. Results: The TC6 and T-FEC group consisted of 22 and 21 patients, respectively. Concerning hematological toxicity, grade 3 or higher adverse reactions included neutropenia and febrile neutropenia. As non-hematological adverse events, exanthema and peripheral neuropathy were frequently reported in the TC6 group, whereas more patients of the T-FEC group reported nausea and vomiting. In TC6, the treatment completion rate was 86.4% and the relative dose intensity of docetaxel was 93.2%. In T-FEC, the values were 95.2% and 98.9%, respectively. Conclusions: These results suggest that TC6 is tolerable in Japanese, and that this regimen can also be performed in outpatient clinics. However, with the TC6 regimen, the compliance was slightly lower than with the T-FEC regimen, and supportive therapy needs to be managed appropriately.

Background: This study evaluated the chronological changes in physical and cognitive functions in middle-aged and older adults with and without rheumatoid arthritis (RA). Methods: This population-based case-control longitudinal study included individuals aged 40–79 years at baseline who agreed to participate. We identified 42 participants with RA and randomly selected 84 ageand sex-matched controls. Physical function was assessed according to gait speed, grip strength, and skeletal muscle mass. Cognitive function was assessed based on the information, similarities, picture completion, and digit symbol substitution test scores of the Wechsler Adult Intelligence Scale-Revised Short Form. The general linear mixed models comprised the fixed effects of the intercept, case, age, time in years since baseline, and case×time interaction, which were used to examine longitudinal changes in physical and cognitive functions. Results: Regardless of RA status, grip strength decreased and the picture completion score increased in the group aged <65 years, while skeletal muscle mass index and gait speed decreased in the group aged ≥65 years. The interaction of case×follow-up years for grip strength in the group aged ≥65 years was significant (p=0.03). The decline in grip strength in the control group (slope=-0.45) was greater than that in the RA group (slope=-0.19). Conclusion: Chronological changes in physical and cognitive functions were comparable between participants with and without RA; however, the decline in grip strength in the control group was greater among older adults with RA.
