        Effect of Light with Different Wavelengths on Nostoc flagelliforme Cells in Liquid Culture

        The effects of lights with different wavelengths on the growth and the yield of extracellular polysaccharides of Nostoc flagelliforme cells were investigated in a liquid cultivation. N. flagelliforme cells were cultured for 16 days in 500 ml conical flasks containing BG11 culture medium under 27 μmol·m-2·s-1 of light intensity and 25oC on a rotary shaker (140 rpm). The chlorophyll a, phycocyanin, allophycocyanin, and phycoerythrin contents in N. flagelliforme cells under the lights of different wavelengths were also measured. It was found that the cell biomass and the yield of polysaccharide changed with different wavelengths of light. The biomass and the yield of extracellular polysaccharides under the red or violet light were higher than those under other light colors. Chlorophyll a, phycocyanin, and allophycocyanin are the main pigments in N. flagelliforme cells. The results showed that N. flagelliforme, like other cyanobacteria, has the ability of adjusting the contents and relative ratio of its pigments with the light quality. As a conclusion, N. flagelliforme cells favor red and violet lights and perform the complementary chromatic adaptation ability to acclimate to the changes of the light quality in the environment.

      • Enantioselective Synthesis of <i>anti</i>–<i>syn</i>-Trihalides and <i>anti</i>–<i>syn</i>–<i>anti</i>-Tetrahalides via Asymmetric β-Elimination

        <P>Structural motifs containing contiguous halide bearing stereocenters are common in natural products as well as bioactive molecules. A few successful examples have been reported in the area of asymmetric vicinal dihalogenation of alkenes for accessing dihalogenated products; in this report, an alternative generation method of contiguous halide-bearing stereocenters alpha,beta,gamma,delta relative to carbonyl group in excellent enantioselectivity is proposed by utilizing a Song's oligoEG catalyst-catalyzed asymmetric beta-elimination. According to this methodology, a wide range of anti-syn-trihalides and anti-syn-anti-tetrahalides with high levels of enantioselectivity were synthesized. The synthetic utility of the contiguous halide-bearing stereocenters was demonstrated by several transformations. The results of high-resolution mass spectrometry indicated that the favorable interaction between catalyst and one of the enantiomers of racemic contiguously multihalogenated ketone contributed to the original enantioselectivity of dehydrohalogenation. A deuterium kinetic isotope effect experiment revealed that this beta-elimination reaction proceeds by the E2 mechanism. This strategy opens a new pathway for the asymmetric synthesis of contiguous halide bearing stereocenters of great complexity.</P>

        A heat shock protein 70 protects the green peach aphid (Myzus persicae) against high-temperature stress

        Heat shock proteins (HSPs) are encoded by Hsp genes and are important in insect tolerance to heat stress. The green peach aphid, Myzus persicae, is an important agricultural pest. The functions of Hsp genes in the thermal tolerance of M. persicae are unknown. This study identified an Hsp70 gene (MpHsp70a) and analyzed its role in protection against high-temperature stress. MpHsp70a encoded a protein consisting of 659 amino acid residues. The protein had three signature motifs of the HSP70 family and was predicted to be localized in the cytoplasm. The highest expression level of MpHsp70a was in adults, and differences in the mRNA levels between apterous and alate adults were not significant. Exposure to high temperatures (30, 35 and 40 ◦ C) for one hour and treatment with 40 ◦ C for different times (0.5, 1 and 2 h) all resulted in a greatly elevated expression level of MpHsp70a, suggesting that the gene is heat-inducible. The transcriptional level of MpHsp70a was suppressed by injection with double-stranded RNA (dsRNA), and knockdown of MpHsp70a significantly increased the suscep tibility of apterous adults to 40 ◦ C. These results indicate that MpHsp70a is required for tolerance to hightemperature stress in M. persicae. Our findings highlight the molecular mechanism underlying Hsp70-mediated thermal adaptation in M. persicae.

      • Laboratory Measurement of Mechanical and Hydraulic Apertures of Smooth Aluminum Joints under Different Confining Stresses

        Fluid flow through rock mass is a critical issue for rock engineering practice and geoscience from shallow to great depth. Among others, the stress-dependent aperture of single fracture is a key for predicting the hydro-mechanical coupling effect of rock mass. To understand the reducing flow rate along a smooth joint with a reduction of aperture, laboratory measurements of mechanical/hydraulic apertures of sawed, polished aluminum samples via YOKO 2 system under a confining stress up to 120 MPa were conducted. The closure of smooth aluminum joints (reduction of mechanical aperture, E) under different normal stress was characterized based on the volume reduction measurements. The hydraulic aperture (e) was determined based on measured flow rate and a cubic law. The hysteresis of the mechanical/hydraulic aperture was observed. However, the permanent aperture closure decreases significantly after the first loading/unloading cycle. The stress dependency of apertures fit the hyperbolic function and semi-logarithmic closure law well. Based on the measurements, we found the ratio of mechanical aperture to hydraulic aperture (E/e) decreased with decreasing hydraulic aperture, which the existing model cannot predict the observed trend. The results of this study provide an important base line for depicting the relation between mechanical aperture and hydraulic aperture, which is widely used to predict the hydraulic aperture from mechanical aperture.

        This paper relates to the principle of PIECI controlled by single chip microcomputer. It is widely used in person's life for its portable, small and lower price. It is the result of computer technics combined with experience in clinic. The structure diagrams and some mathematics modes are given in this paper. Also, the software system function, pseudo difference reconizing and processing technics is dis-cussed in the article.

        A sigma class glutathione S-transferase gene regulated by the CncC pathway is required for phytochemical tolerance in the red flour beetle, Tribolium castaneum

        Insect glutathione S-transferases (GSTs) play a crucial role in the detoxification of exogenous compounds, especially insecticides and plant allelochemicals. A sigma class GST gene, TcGSTS7, mediates the response to eugenol in Tribolium castaneum. However, the mechanism underlying this effect remains largely unknown. In this study, TcGSTS7, which exhibits a structural motif and domain organization characteristic of GSTs, was cloned from the T. castaneum genome. Spatiotemporal expression analysis revealed that TcGSTS7 was most highly expressed at the late larva stage and was mainly expressed in the fat body and epidermis of larvae and adults, suggesting that TcGSTS7 may play a potential role in the protection against toxic xenobiotics in T. castaneum. Furthermore, the expression of TcGSTS7 was significantly induced after exposure to eugenol, while RNA inter ference (RNAi) targeting TcGSTS7 enhanced the sensitivity of the beetle to eugenol, indicating that TcGSTS7 is involved in the tolerance of T. castaneum to this insecticide. Interestingly, the depletion of TcCncC, which encodes a transcription factor of the CncC pathway that has been associated with the regulation of detoxification-related genes in insects, led to a reduction in the TcGSTS7 transcript level following exposure to eugenol, which suggests that TcGSTS7 acts downstream of the CncC pathway. Combined, these results indicated that TcGSTS7 partici pates in the tolerance of T. castaneum to phytochemicals in a CncC pathway-dependent manner. These findings have implications for the development of novel drugs for use in pest control.

        Single-cell RNA sequencing reveals B cell–related molecular biomarkers for Alzheimer’s disease

        In recent years, biomarkers have been integrated into the diagnostic process and have become increasingly indispensable for obtaining knowledge of the neurodegenerative processes in Alzheimer’s disease (AD). Peripheral blood mononuclear cells (PBMCs) in human blood have been reported to participate in a variety of neurodegenerative activities. Here, a single-cell RNA sequencing analysis of PBMCs from 4 AD patients (2 in the early stage, 2 in the late stage) and 2 normal controls was performed to explore the differential cell subpopulations in PBMCs of AD patients. A significant decrease in B cells was detected in the blood of AD patients. Furthermore, we further examined PBMCs from 43 AD patients and 41 normal subjects by fluorescence activated cell sorting (FACS), and combined with correlation analysis, we found that the reduction in B cells was closely correlated with the patients’ Clinical Dementia Rating (CDR) scores. To confirm the role of B cells in AD progression, functional experiments were performed in early-stage AD mice in which fibrous plaques were beginning to appear; the results demonstrated that B cell depletion in the early stage of AD markedly accelerated and aggravated cognitive dysfunction and augmented the Aβ burden in AD mice. Importantly, the experiments revealed 18 genes that were specifically upregulated and 7 genes that were specifically downregulated in B cells as the disease progressed, and several of these genes exhibited close correlation with AD. These findings identified possible B cell-based AD severity, which are anticipated to be conducive to the clinical identification of AD progression.

      • Prognostic Role of Hepatoma-derived Growth Factor in Solid Tumors of Eastern Asia: a Systematic Review and Meta-Analysis

        Hepatoma-derived growth factor (HDGF) is a novel jack-of-all-trades in cancer. Here we quantify the prognostic impact of this biomarker and assess how consistent is its expression in solid tumors. A comprehensive search strategy was used to search relevant literature updated on October 3, 2014 in PubMed, EMBASE and WEB of Science. Correlations between HDGF expression and clinicopathological features or cancer prognosis was analyzed. All pooled HRs or ORs were derived from random-effects models. Twenty-six studies, primarily in Eastern Asia, covering 2,803 patients were included in the analysis, all of them published during the past decade. We found that HDGF overexpression was significantly associated with overall survival (OS) ($HR_{OS}=2.35$, 95%CI=2.04-2.71, p<0.001) and disease free survival (DFS) ($HR_{DFS}=2.25$, 95%CI =1.81-2.79, p<0.001) in solid tumors, especially in non-small cell lung cancer, hepatocellular carcinoma and cholangiocarcinoma (CCA). Moreover, multivariate survival analysis showed that HDGF overexpression was an independent predictor of poor prognosis ($HR_{OS}=2.41$, 95%CI: 2.02-2.81, p<0.001; $HR_{DFS}=2.39$, 95%CI: 1.77-3.24, p<0.001). In addition, HDGF overexpression was significantly associated with tumor category (T3-4 versus T1-2, OR=2.12, 95%CI: 1.17-3.83, p=0.013) and lymph node status (N+ versus N-, OR=2.37, 95%CI: 1.31-4.29, p=0.03) in CCA. This study provides a comprehensive examination of the literature available on the association of HDGF overexpression with OS, DFS and some clinicopathological features in solid tumors. Meta-analysis results provide evidence that HDGF may be a new indicator of poor cancer prognosis. Considering the limitations of the eligible studies, other large-scale prospective trials must be conducted to clarify the prognostic value of HDGF in predicting cancer survival.
