항원의 투여방법 및 사육환경 변화에 따른 넙치의 특이항체 반응

본 연구에서는 백신을 넙치 양식 현장에 적용하기 위한 기초연구로서 항원의 투여방법 및 항원 투여 후 사육환경 변화에 따른 넙치의 특이 항체 반응에 대한 영향을 조사하였다. 항원의 투여방법에 따른 항체 반응을 조사하기 위해 넙치에 BSA를 복강 및 근육 주사한 후 주기적으로 혈액을 취해 BSA에 대한 항체가를 측정한 결과, 근육 주사법이 복강 주사법보다 항체형성 초기시점에서 약간 높은 항체가를 나타났으나 그 이후의 항체 반응에는 유의적인 차이가 관찰되지 않아, 백신을 투여하는 방법으로 근육 주사법뿐만 아니라 복강 주사법도 유용하게 사용될 수 있을 것으로 사료되었다. 사육환경 변화에 따른 백신의 영향을 조사하기 위해 BSA를 면역시킨 넙치에 사육수온의 급하강, handling 및 중유에 노출시킨 후 항체가를 측정한 결과, 수온변화 및 handling 실험구가 양성 대조구보다 항체형성 초기시점에서 약간 높은 항체가를 나타났으나, 그 이후의 항체 반응에는 서로간의 유의적인 차이는 관찰되지 않아, 위의 환경변화는 넙치의 특이 항체반응에 영향을 거의 끼치지 않는 것으로 확인되었다. 이상의 결과, 넙치 양식현장에서 백신은 일시적인 수온변화, 중유노출 및 handling에 의한 환경변화에서도 유용하게 사용 될 수 있을 것으로 사료되었다. The specific antibody response of olive flounder, Paralichthys olivaceus to change in rearing-environmental conditions post immunization with antigen (bovine serum albumin, BSA) and different routes of antigen administration were investigated by enzyme-linked immunosorbent assay (ELISA). To test the effect of routes of antigen administration, flounder were injected intraperioneally or intramuscularlly with 1 mg of BSA. In addition, to test the effect of change in environmental condition post immunization, flounder were injected intraperioneally with 1 mg of antigen, and then were exposed to acute thermal change (the water temperature (WT) was decreased from 21℃ to 15℃ within 30 min and maintained at 15℃ for 3 h), handling (fish were caught and subsequently held out of water for 1 min) or heavy oil (76 g/ 200 L for 2 days). Consequently, there was no significant difference between intraperioneal (IP) and intramuscular (IM) injections except at 10 days post-immunization. With these results, it suggests that both IM and IP injections may be used as route of vaccination. Futhermore, no significant difference was observed in the antibody response among the groups exposed to heavy oil, handling, sudden drop of WT and positive control except at 10 days post-immunization. From these results, it was confirmed that specific antibody response was not affected by the above mentioned rearing-environmental conditions, suggesting that vaccination can be employed at changing rearing-environmental conditions.

Farnesylcysteine Methyltransferase Activity and Ras Protein Expression in Human Stomach Tumor Tissue

The processing pathway of G-proteins and Ras family proteins includes the isoprenylation of the Cysteine residue, followed by proteolysis of three terminal residues and α-carboxyl methyl esterification of the cysteine residue. Farnesylcysteine methyltransferase(FCMT) activity is responsible for the methylation reaction which play a role in the membrane attachment of a variety of cellular proteins. Four kinds of Ras protein (c-Ha-ras,c-N-Ras,c-Ki-Ras,pan-Ras) expression were detected in adenocarcinoma of human tissue by immunohistochemical method, and hematoxylin and eosin staining. The level of Ras protein in human stomach tumor tissues was much higher than in normal and peritumoral regions of the same biopsy samples. The FCMT activities of each cellular fractions were high in mitochondrial fraction followed by microsomal fraction, whole homogenate and cytosolic fraction. The inhibitory effect on FCMT activity on stomach tumor tissue was determined after treatment with 0.25㎛ of S-adenosyl-_L-homocysteine. S-adenosyl-_L-homocysteine inhibited FCMT activity from 11.2% to 30.5%. These results suggested that FCMT might be involved in Ras proteins activity.

We report on mycobacteriosis in an imported tropical ornamental fish Macropodus opercularis commonly known as the paradise fish. Mass mortality occurred in paradise fish imported to Korea from Southeast Asia in 2008. The affected fish did not show any outward clinical signs, but enlargement of the spleen, kidneys, and liver was observed on dissection. Histopathological examination revealed numerous granulomas in the spleen, and acid-fast bacilli were observed in the centers of the granulomas. About 65% of spleen DNA samples were PCR positive using mycobacteria-specific primers targeting the 16S rRNA and hsp65 genes. The nucleotide identities of the 16S rRNA and hsp65 genes with those of Mycobacterium marinum were 99.5% and 99.4%, respectively. Although the bacterium was not cultured, the molecular diagnosis and histopathological findings were consistent with mycobacteriosis in paradise fish.

<P>The dynamic interplay between the intramolecular hydrogen bonding and intramolecular vibrational redistribution is found to be critical in nonadiabatic reaction dynamics. Herein, it has been demonstrated that the molecular planarity, directed by the intramolecular hydrogen bonding, plays an important role in the nonadiabatic passage of the reactive flux at the conical intersection in the photodissociation reactions of 2-fluorothiophenol and 2-chlorothiophenol. As the internal energy increases in the excited state, the intramolecular hydrogen bonding of 2-fluorothiophenol loosens. The floppiness brought into the molecular structure then modifies the dynamic path of the reactive flux, leading to the diminishment of the nonadiabatic transition probability at the conical intersection. On the contrary, for 2-chlorothiophenol having the relatively stronger intramolecular hydrogen bonding, the reactive flux seems to retain the molecular planarity even with the increase of the internal energy as manifested by the constant nonadiabatic transition probability over the wide range of the S<SUB>1</SUB> internal energy. Objective: To quantify the effects of midline-related landmark identification on midline deviation measurements in posteroanterior (PA) cephalograms using a cascaded convolutional neural network (CNN). Methods: A total of 2,903 PA cephalogram images obtained from 9 university hospitals were divided into training, internal validation, and test sets (n = 2,150, 376, and 377). As the gold standard, 2 orthodontic professors marked the bilateral landmarks, including the frontozygomatic suture point and latero-orbitale (LO), and the midline landmarks, including the crista galli, anterior nasal spine (ANS), upper dental midpoint (UDM), lower dental midpoint (LDM), and menton (Me). For the test, Examiner-1 and Examiner-2 (3-year and 1-year orthodontic residents) and the Cascaded-CNN models marked the landmarks. After point-to-point errors of landmark identification, the successful detection rate (SDR) and distance and direction of the midline landmark deviation from the midsagittal line (ANS-mid, UDM-mid, LDM-mid, and Me-mid) were measured, and statistical analysis was performed. Results: The cascaded-CNN algorithm showed a clinically acceptable level of point-to-point error (1.26 mm vs. 1.57 mm in Examiner-1 and 1.75 mm in Examiner-2). The average SDR within the 2 mm range was 83.2%, with high accuracy at the LO (right, 96.9%; left, 97.1%), and UDM (96.9%). The absolute measurement errors were less than 1 mm for ANS-mid, UDM-mid, and LDM-mid compared with the gold standard. Conclusions: The cascaded-CNN model may be considered an effective tool for the auto-identification of midline landmarks and quantification of midline deviation in PA cephalograms of adult patients, regardless of variations in the image acquisition method.

Aims: This study compared the survival benefits between surgical resection and radiofrequency ablation (RFA) in single small hepatocellular carcinoma (HCC). Methods: Patients registered in the database of Korea central cancer registry for HCC between 2008 and 2014 who received surgical resection or RFA for a single HCC of maximum size 3cm were reviewed. The cumulative overall survival (OS) and retreatment-free survival (RFS) were compared according to the type of treatment. Results: A total of 1426 patients were included, 662 (46.4%) in the resection group and 764 (53.6%) in the RFA group. The 5-year OS rates were 91.8% for resection, and 84.9% for RFA, respectively (P<0.001). The 5-year RFS rates were 77.8% for the resection and 70.5% for RFA, respectively (P=0.002). In patients with maximum tumor size <2cm, there was no significant difference in 5-year OS rates between the two groups (91.8% for resection, and 87.5% for RFA, respectively; P=0.094), while resection was superior to RFA in the 5-year RFS rates (81.0% for resection, and 72.3% for RFA, respectively; P=0.017). Resection was superior in both 5-year OS and RFS rates in those with maximum tumor size ≥2cm and <3cm (91.9% for resection, and 80.2% for RFA, respectively; P<0.001; 76.2% for resection, and 67.2% for RFA, respectively; P=0.007). In the entire cohort, type of treatment (HR 1.457, P=0.032), age (HR 1.031, P<0.001), serum albumin (HR 0.466, P<0.001), and serum creatinine level (HR 1.282, P<0.001) were independently associated with survival. Type of treatment (HR 1.544, P<0.001) and maximum size of tumor (HR 1.353, P=0.002) were independent predictors of RFS. Conclusions: In patients with maximum tumor size <2cm, OS is comparable between resection and RFA groups, while RFS is better in resection group. Surgical resection is superior in both OS and RFS in patients with maximum tumor size ≥2cm and <3cm.
