        Biological effect of recombinant IL-1β on the expression of antiviral genes in the gill of rainbow trout, Oncorhynchus mykiss

        We have investigated the biological effects of recombinant IL- 1β (rIL-β) on the expression of antiviral genes such as Myxovirus-3 (MX-3) and Interferon regulating factor-Ⅰ (IRF-l), which are related to type Ⅰ interferon. When ten micrograms of rIL-1β were treated, the stimulatory effect we observed on the expression of these antiviral genes. Interestingly, at the early stage of stimulation, these genes were down-regulated and then up-regulated by the results obtained that the expressions of these genes were decreased at day Ⅰ post-injection and gradually increased at day 3 post-injection. Thus, the stimulatory effect of rIL-β on the expression of MX-3 and IRF-3 gene might be an indirect stimulatory effect because significant up-regulation was delayed until day 3 post-injection.

        Development of piezoelectric immunosensor for the rapid detection of marine derived pathogenic bacteria, Vibrio vulnificus

        Biosensors consist of biochemical recognition agents like antibodies immobilized on the surfaces of transducers that change the recognition into a measurable electronic signal. Here we report a piezoelectric immunosensor made to detect Vibrio vulnificus. A 9MHz AT-cut piezoelectric wafer attached with two gold electrodes of 5mm diameter was used as the transducer of the QCM biosensor with a reproducibility of ${\pm}0.1Hz$ in frequency response. We have tried different approaches to immobilize antibody on the sensor chip. Concerning the orientation of antibody for the best antigen binding capacity, the antibody was immobilized by specific binding to protein G or by cross-linking through hydrazine. In addition, protein G was cross-linked on glutaraldehyde activated immine layer (PEI) or EDC/NHS activated sulfide monolayer (MPA). PEI was found to be more effective to immobilize protein G following glutaraldehyde activation than MPA. However, hydrazine chip showed a better capability to immobilize more IgG than protein G chip and a higher sensitivity. The sensor system was able to detect V. vulnificus in dose dependent manner and was able to detect bacterial cells within 5 minutes by monitoring frequency shifts in real time. The detection limit can be improved by preincubation to enrich the bacterial cell number.

        Survey of the heterogeneous gene expression in olive flounder muscle using the luciferase reporter gene system

        The CMV promoter driven luciferase reporter gene coding plasmid (pcDNA-luc) was constructed and used as a model for DNA immunization study. Expression of the recombinant luciferase protein was confirmed in vitro in RTG-2 cell line before using in vivo study in olive flounder. In dose response study, the maximum expression of the luciferase gene was found in the group injected with 10-l5㎍ of plasmid DNA. The kinetic study showed that the luciferase gene expression was reached at the maximum level at one day after injection and slightly decreased after then but significantly high level of expression was sustained until the conducted experiment of 7 days. In the study of tissue distribution of gene expression, it was found that luciferase gene was expressed at the significant level in immune organs such as gill and spleen, located far from the injected site, suggesting the systemic distribution of the intramuscularly injected DNA in olive flounder.

        Proinflammatory Effects of Bacterial Lipopolysaccharide (LPS) in Rainbow Trout (Oncorhynchus mykiss) Macrophage Cells

        Proinflammatory effects of bacterial lipopolysaccharide (LPS) have been assessed by analysing the induction of two inflammatory genes, interleukin-lP (IL-1β ) and cyclooxygenase-2 (COX-2), in rainbow trout (Oncorhynchus mykiss) macrophage cells. Production of a metabolite of arachidonic acid by COX-2, prostaglandin E₂ (PGE₂)), was also analysed in macrophage cells after LPS stimulation. Northern blot analysis revealed that LPS ( 5 ㎍/ mL) significantly upregulated IL-Iβ (54 times) and COX-2 (40.7 times) gene expression in macrophage cells after 4 h stimulation. According to RT-PCR (Reverse Transcription Polymerase Chain Reaction) analysis, IL-1P gene induction in LPS stimulated macrophage cells was started within I h and significantly increased thereafter until 4 h. Meanwhile, COX-2 gene induction by LPS was delayed in comparison with IL-13 gene induction as a faint band was observed after 4 h stimulation in head kidney macrophagc cells. LPS also significantly increased PGE: production in head kidney leucocytes, presumably via activating COX-2 expression that metabolites arachidonic acid to PGE₂. In conclusion, it was demonstrated that LPS could induce two main inflammatory and immune related genes, IL-1β and COX-2, and increase PGE₂ production in trout head kidney macrophage cells, representing a strong inflammatory activity.

        CSR성과정보에 대한 자본시장의 반응 -KRX SRI 지수 편입․제외 종목을 중심으로-

        본 연구에서는 국내의 대표적인 사회책임투자 지수인 KRX SRI을 대상으로 KRX SRI 편입기업과 제외기업의 주가 및 거래행태 변화를 분석하고, CSR 성과 정보가 투자자들에게 기업이 사회적 책임을 다하고 있다는 신호로 작용하여 투자의사결정에 반영되고 있는지 알아보고자 하였다. 실증분석 결과, 지수설정 및 변경 효과의 사전 반영가능성이 있으며, 지수설정일과 신규편입일 전후로 누적초과수익률이 증가하였으나, 주식거래량은 지수설정 전과 지수변경 후에 음의 초과거래량이 나타났 다. 또한 KRX SRI 지수에 대한 투자자들의 인식이 지수의 최초 설정보다 신규편입에 대한 공시에 더욱 긍정적인 평가를 하고 있으며, 그 효과가 장기적으로 영향을 미친다는 사실을 알 수 있었다. 그러나 KRX SRI 지수편입 제외 기업의 경우에는 지수변경일 전 3일에 일시적으로 상승하였다가 감소하여 지수변경일 이후 균형가격으로 회귀하였으며, 주식거래율의 변화도 없는 것으로 나타났다. 또한 KRX SRI 지수설정 및 편입종목과 제외종목에 대한 누적초과수익률을 상호비교해본 결과, 지수설정 및 편입 공시 시점을 전후로 유의한 양의 누적초과수익률이 나타났으나, 지수제외 시점 전후의 누적초과수익률은 음의 누적초과수익률이 발생하지 않는 자본시장의 비대칭적 반응이 나타났다. 이는 신규편입 후에 투자자의 인지가 증가하고 정보비대칭의 완화에 따른 주가상승이 발생하지만 지수 제외기업의 경우에는 투자 자의 인지가 변화하지 않기 때문에 시장에서 주가가 반응을 보이지 않는 것으로 설명된다. This paper empirically examines market response following additions or deletions of KRX SRI Index revisions in Korea. The index includes companies on the basis of pre-determined social responsibility criteria. Event study methodology, which relies on the notion of market efficiency, is used to examine the relationship between positive and negative announcement and changes in share price or trading volume. Inclusion in and deletion from SRI index is used as a proxy measure for good and poor corporate responsibility and abnormal or unexpected daily returns and trading volume ratios associated with an event are calculated and their significance tested. The results show as followings;First, a trend that positive announcements lead to positive result for inclusion in the index, this means we could cautiously accept the 1st hypothesis. Second, deletion announcements were met with negative changes in share price but were not statistically significant. Thirdly, there was asymmetry between the effects of positive and negative events on share price and trading volume. In the case of Korea, investors perceptions about SRI are not high yet. In addition, the accumulation of statistics for empirical studies is not enough. Therefore, such related data and various methodologies, including the accumulation of data over the follow-up studies should be done continually.

        의결권 행사와 기업지배구조가 기업가치에 미치는 영향

        본 연구는 국민연금이 5% 이상 지분을 가지고 있는 기업을 장기간 보유하면서 적극적 의결권을 행사한 기업과 아닌 기업을 기업지배구조수준별로 비교해보고, 적극적 의결권 행사와 국민연금 보유 기간이 기업가치에 미치는 영향을 분석해보고자 하였다. 그 결과, 기업지배구조수준이 높은 집단과 낮은 집단 간에 적극적 의결권 행사를 하는데 있어서 유의한 집단 간 차이가 있는 것으로 나타났다. 또한, 적극적 의결권 행사와 보유 기간 등이 해당 기업의 가치에 미치는 영향을 살펴보고자 다중회귀분석을 실시한 결과, 국민연금이 오랜 기간 보유한 기업이 유의적으로 기업가치가 높은 것으로 나타났으며, 대주주지분율은 높을수록 기업가치가 낮은 것으로 나타났다. 본 연구는 스튜어드쉽 코드 시행이 시작된 현 시점에서 우리나라에서 주주행동주의의 현 상황을 파악해보고, 추후 진행 방향에 대한 시사점을 찾을 수 있는데 그 의의가 있다. This study was aimed at analyzing the effects of the active exercises of voting rights on the corporate value by comparing the companies for which the National Pension has actively exercised its voting rights and those for which it has not, according to the level of corporate governance while having shares of more than 5% for a long time. It was conducted multiple regression analysis to examine the impact of the active exercise of voting rights and the holding period of shares by the National Pension on corporate values. The analysis results showed the significantly differences in both group. Furthermore, it was shown that the period of shareholding by the National Pension had a significant positive impact on the corporate value while the ratio of major shareholder's holding had a significant negative impact on the corporate value. This study is meaningful in that it could determine the current situation of shareholder activism in Korea and suggest implications for future direction at the current commencing stage of implementation of the stewardship code.

        환경우수기업의 경영성과 관련성에 대한 연구

        본 연구는 환경우수기업과 일반기업의 재무비율 및 경영성과를 비교하고, 환경우수기업의 경영성과에 주된 영향을 미치는 재무비율특성을 검토하기 위해 평균차이분석과 다중회귀분석을 실시하였다. 환경우수기업이 부채비율과 활동성비율면에서 일반기업에 비해 더 우수한 것으로 나타났고, 경영성과면에서는 두 기업집단간에 차이가 없는 것으로 나타났다. 재무비율이 기업가치에 미치는 영향을 살펴본 결과, 자산의 규모는 환경우수기업의 경우 토빈큐에만 음(-)의 유의적인 영향을 미쳤으며, 일반기업의 경우는 ROA와 토빈큐에는 음(-)의 영향을 EPS에는 양(+)의 영향을 미치는 것으로 나타났다. 수익성지표인 매출액영업이익률은 두 집단의 기업가치에 모두 유의적인 양(+)의 영향을 미쳤으나, 유동비율의 경우는 환경우수기업의 EPS와 토빈큐에 음(-)의 유의적인 영향을 미쳤다. 본 연구를 계기로 기업의 환경책임이행에 대한 객관적인 측정지표가 개발되고, 이러한 내용들이 투자의사결정에 반영된다면 기업은 기후위기에 더욱 적극적으로 대응하고자 할 것이다. This study conducted mean difference analysis and multiple regression analysis to compare the financial ratio and management performance of environmentally superior companies and general companies, and to examine the financial ratio characteristics that mainly affect the management performance of environmentally superior companies. Excellent environmental enterprises were found to be better than ordinary companies in terms of debt ratio and activity ratio, and there was no difference between the two groups in terms of business performance. Looking at the impact of financial ratios on corporate value, the size of assets had a significant negative impact on Tobin’s Q for excellent environmental entities, and negative impact on ROA and Tobin’s Q for general entities had a positive (+) impact on EPS. The profitability indicator, the operating profit margin of sales, had a significant positive (+) effect on the corporate value of both groups, but in the case of current ratio, it had a negative (-) impact on the EPS and Tobin’s Q of the excellent environmental entities. This study will develop objective metrics for companies' environmental responsibility, and if these are reflected in investment decisions, companies will be more willing to respond more actively to the climate crisis.
