        Transcriptome analysis of gene expression patterns during embryonic development in golden cuttlefish (Sepia esculenta)

        Golden cuttlefish (Sepia esculenta) is an important economic species in China. Because of the rapid decline of its natural resource, researchers are exploring breeding technique for this species. The major obstacle that hinders artificial breeding of S. esculenta is the low larvae survival rate. Mortality is especially high during the mouth-opening stage. Investigating the embryogenesis before the first feed could provide theoretical guidance for reproduction control and breeding of S. esculenta and other Sepia species. In this study, we analyzed the dynamics of the S. esculenta transcriptome along different stages of embryonic development by mRNA-sEq. Our bioinformatics protocol identified 1492 differentially expressed genes (DEGs) across the early developmental stages. Gene ontology enrichment analysis showed that the DEGs were significantly involved in developmental processes and molecular functions, including chitin metabolic process, peptidase activity, catalytic activity, and calcium ion binding. Our results indicated that genes related to cuttlebone development and gene regulation functions were active during the early life phase of S. esculenta. Hierarchical clustering of the DEGs reflected the successiveness of the developmental stages, revealing that gene expression patterns of neighboring stages were similar. The DEG analysis allowed us to identify specific genes and relevant biological pathways to better understand the molecular mechanisms during each developmental stage. This study provides novel insights into the processes underlying the early developmental stages of S. esculenta. The transcriptomic data and identified genes will serve as valuable references for the developmental biology of this species and will help promote its aquaculture research.

        Chimerism Evaluation of ‘Hongrou Huyou’, a Grafted Chimera between Citrus changshan-huyou and Citrus unshiu

        Chimeras occur spontaneously or artificially and are valuable in horticultural crop breeding. A new diploid citrus chimera, named ‘Hongrou Huyou’ (abbreviated HH) (Citrus changshan-huyou + C. unshiu), was found during a bud sports investigation in China. The morphology, flesh carotenoid content, and molecular markers were evaluated in HH and the two grafted donors. The results showed that characteristics derived from L2/L3, such as the fruit size, winged leaf, seed, pollen, and rind aroma, were similar to those of C. changshan-huyou (CH), whereas the juice sac and stomatal characteristics originating from L1 were the same as those of the satsuma mandarin. High-performance liquid chromatography analysis of carotenes from the flesh of HH showed that the content was the same as that of the satsuma mandarin, with β-cryptoxanthin producing the main carotenoid spectrum, whereas lutein and violaxanthin were the main carotenoids in CH. Nuclear simple sequence repeat, chloroplast simple sequence repeat, and mitochondrial simple sequence repeat analyses showed that the leaves, outer pericarp (epidermis and flavedo), segment wall, and juice sac of HH contained the nuclear, chloroplast, and mitochondrial genomes of both donors; however, the albedo of HH only contained the genetic material of CH. Thus, HH is confirmed to be a periclinal chimera that consists of L1 from C. unshiu and L2/L3 from CH.

        Adsorption of Pb(II) from aqueous solution to Ni-doped bamboo charcoal

        Bamboo charcoal (BC) obtained by pyrolysis of Makino bamboo in the absence of oxygen was used as support for the preparation of Ni-doped adsorbent (Ni-BC). The low-cost composite was characterized and used as an adsorbent for Pb(II) removal from water. The results showed that both BET surface area and total pore volume of Ni-BC increased. The adsorption of Pb(II) strongly depended on solution pH,temperature and ionic strength. The adsorption isotherms followed Langmuir isotherm model well, and the maximum adsorption capacities of Pb(II) at 298 K were 25.0 and 142.7 mg/g for BC and Ni-BC,respectively. The adsorption processes were well fitted by pseudo-second-order kinetic model. Thermodynamic parameters showed that the adsorptions of Pb(II) onto both adsorbents were feasible,spontaneous, and exothermic under the studied conditions. The spent Ni-BC could be readily regenerated for reuse.
