      • Expression and Clinical Significance of Sushi Domain-Containing Protein 3 (SUSD3) and Insulin-like Growth Factor-I Receptor (IGF-IR) in Breast Cancer

        Background: To investigate the expression of insulin-like growth factor-I receptor (IGF-IR) and sushi domain containing protein 3 (SUSD3) in breast cancer tissue, and analyze their relationship with clinical parameters and the correlation between the two proteins. Materials and Methods: The expression of IGF-IR and SUSD3 in 100 cases of breast cancer tissues and adjacent normal breast tissues after surgery was detected by immunohistochemical technique MaxVisionTM, and the relationship with clinical pathological features was further analyzed. Results: The positive rate of IGF-IR protein was 86.0% in breast cancer, higher than 3.0% in adjacent normal breast tissue (P<0.05). The positive expression rate of SUSD3 protein was 78.0% in breast cancer, higher than 2.0% in adjacent normal breast tissue (P<0.05). The expression of IGF-IR and SUSD3 was related to estrogen receptor and pathological types (P<0.05),but not with age, stage, the expression of HER-2 and Ki-67 (P>0. 05). The expression of IGF-IR and SUSD3 in breast cancer tissue was positively related (r=0.553, P<0.01). Conclusions: The expression of IGF-IR and SUSD3 may be correlated to the occurrence and development of breast cancer. The combined detection of IGF-IR, SUSD3 and ER may play an important role in judging prognosis and guiding adjuvant therapy after surgery of breast cancer.

      • Effects of Parafibromin Expression on the Phenotypes and Relevant Mechanisms in the DLD-1 Colon Carcinoma Cell Line

        Background: Parafibromin is a protein encoded by the HRPT2 (hyperparathyroidism 2) oncosuppressor gene and its down-regulated expression is involved in pathogenesis of parathyroid, breast, gastric and colorectal carcinomas. This study aimed to clarify the effects of parafibromin expression on the phenotypes and relevant mechanisms of DLD-1 colon carcinoma cells. Methods: DLD-1 cells transfected with a parafibromin-expressing plasmid were subjected to examination of phenotype, including proliferation, differentiation, apoptosis, migration and invasion. Phenotype-related proteins were measured by Western blot. Parafibromin and ki-67 expression was detected by immunohistochemistry on tissue microarrays. Results: The transfectants showed higher proliferation by CCK-8, better differentiation by electron microscopy and ALP activity and more apoptotic resistance to cisplatin by DNA fragmentation than controls. There was no difference in early apoptosis by annexin V, capase-3 activity, migration and invasion between DLD-1 cells and their transfectants. Ectopic parafibromin expression resulted in down-regulated expression of smad4, MEKK, GRP94, GRP78, $GSK3{\beta}$-ser9, and Caspase-9. However, no difference was detectable in caspase-12 and -8 expression. A positive relationship was noted between parafibromin and ki-67 expression in colorectal carcinoma. Conclusions: Parafibromin overexpression could promote cell proliferation, apoptotic resistance, and differentiation of DLD-1 cells.

        Adsorption behavior and mechanism of multiple Mg atoms on the surface of AlNi compound at Mg alloy/steel interface

        The interface between Mg alloy and the intermetallic compounds (IMCs) is the weak point during the welding of Mg alloy and steel. The models of AlNi surface under different number of absorbed magnesium atoms were established. The absorption energy and electronic properties of the Nth (N represents the number of Mg atoms, which may be 1, 2, 3, and 4 in this paper) Mg atom on the AlNi surface were calculated using first-principles calculations. The results show that the previously absorbed Mg atoms will promote the absorption of the latter Mg atom when the distance between them is equal or less than 2.89 Å. The absorption energy of the Mg atoms on the AlNi surface is related to the electronic exchange between different atoms. The results will provide a new perspective for understanding the atomic scale mechanism of the bonding between the Mg atom and AlNi IMCs.

      • Prognostic Value of CD44 Variant exon 6 Expression in Non-Small Cell Lung Cancer: a Meta-analysis

        Background: CD44v6 (CD44 variant exon 6) is the chief CD44 variant isoform regulating tumor invasion, progression, and metastasis. The prognostic value of CD44v6 expression in non small cell lung cancer (NSCLC) has been evaluated in many studies, but the results have remained controversial. Thus, we performed a meta-analysis of currently available studies to investigate the prognostic value of CD44v6 expression in NSCLC patients and the relationship between the expression of CD44v6 and clinicopathological features. Materials and Methods: Two independent reviewers searched the relevant literature in Pubmed, Medline and Embase from 1946 to January 2014. Overall survival (OS) and various clinicopathological features were collected from included studies. This meta-analysis was accomplished using STATA 12.0 and Revman 5.2 software. Pooled hazard ratios (HRs) with 95% confidence intervals (95%CIs) were calculated to estimate the effects. Results: A total of 921 NSCLC patients from ten studies met the inclusion criteria. The results showed that CD44v6 high expression was a prognostic factor for poor survival (HR=1.91, 95%CI=1.12-3.26, p<0.05). With respect to clinicopathological features, CD44v6 high expression was related to histopathologic type (squamous cell carcinoma versus adenocarcinoma: OR=2.72, 95%CI=1.38-5.38, p=0.004), and lymph node metastasis (OR=3.02, 95%CI=1.93-4.72, p<0.00001). Conclusions: Our results suggested CD44v6 high expression as a poor prognostic factor for NSCLC, and CD44v6 expression is associated with lymph node metastasis and histopathologic type. Therefore, CD44v6 expression can be used as a novel prognostic marker in NSCLC cases.

      • Wind-induced mechanical energy analyses for a super high-rise and long-span transmission tower-line system

        This study aimed to analyze the wind-induced mechanical energy (WME) of a proposed super high-rise and long-span transmission tower-line system (SHLTTS), which, in 2021, is the tallest tower-line system with the longest span. Anew index – the WME, accounting for the wind-induced vibration behavior of the whole system rather than the local part, was first proposed. The occurrence of the maximum WME for a transmission tower, with or without conductors, under synoptic winds, was analyzed, and the corresponding formulae were derived based on stochastic vibration theory. Some calculation data, such as the drag coefficient, dynamic parameters, windshielding areas, mass, calculation point coordinates, mode shape and influence function, derived from wind tunnel testing on reducedscale models and finite element software were used in calculating the maximum WME of the transmission tower under three cases. Then, the influence of conductors, wind speed, gradient wind height and wind yaw angle on WME components and the energy transfer relationship between substructures (transmission tower and conductor) were analyzed. The study showed that the presence of conductors increases the WME of transmission towers and changes the proportion of the mean component (MC), background component (BC) and resonant component (RC) for WME; The RC of WME is more susceptible to the wind speed change. Affected by the gradient wind height, the WME components decrease. With the RC decreasing the fastest and the MC decreasing the slowest; The WME reaches the its maximum value at the wind yaw angle of 30o . Due to the influence of three factors, namely: the long span of the conductors, the gradient wind height and the complex geometrical profile, it is important that the tower-line coupling effect, the potential for fatigue damage and the most unfavorable wind yaw angle should be given particular attention in the wind-resistant design of SHLTTSs.

        Faecal Microbiota Microsphere Contributed to Relieving Gut Barrier Damage in Colitis

        Gut microbiota play an important role in many diseases. However, few researches evaluated the effect of faecal bacteria transplantation in protecting gut barrier during colitis. In this study, we focused on the effect of faecal microbiota encapsulated alginate microsphere on dextran sulfate sodium (DSS) induced colitis mice. Bacteria are sensitive to gastric juice and digestive enzymes, which is the dilemma of utilizing microbiota. Through taking advantage of alginate, microspheres that could protect its contents were fabricated. In vitro drug release experiment demonstrated their ability in protecting contents in simulated gastric fluid and release contents in simulated intestinal fluid. Thus, the resultant microsphere could be used in faecal microbiota transplantation. In vivo, the transplantation of faecal microbiota from healthy mice showed protection against DSS induced oxidative stress and gut barrier damage. Our findings suggest that the faecal microbiota transplantation could be a potential effective treatment for protecting gut barrier in inflammatory bowel diseases (IBD) colitis with faecal microbiota encapsulated alginate microsphere.

        중국 SNS 인플루언서의 인상관리가 유저의 몰입도와 구매의도에 미치는 복합적 관계에 관한 연구

        본 연구는 인터넷 기반의 SNS 플랫폼 환경에서 유명인으로 영향력이 커지고 있는 중국 SNS 인플루언서의 인상관리가 유저의 몰입도와 구매의도에 영향을 미치는지에 연구문제 제기에서 시작되었다. 본 연구의 목적은, 첫째, 중국 1인 미디어 인플루언서, 인상관리, SNS 몰입도, 구매의도에 대한 이론적 이해와 용어 정립을 통해 변수 선정의 이론적 근거를 마련 한다. 둘째, SNS 플랫폼 기반에서 중국 SNS 인플루어서와 유저와의 상호작용에서 인상관리의 영향을 알아보기 위하여 몰입도와 구매의도 변수에 대한 실증연구 기반을 마련한다. 셋째, 중국 SNS 인플루언서의 인상관리가 유저의 몰입도와 구매의도에 미치는 영향 관계를 도출하는 것이다. 따라서 본 논문은 중국 SNS 인플루언서의 인상관리가 유저의 몰입도와 구매의도에 영향을 미치는지 살펴보기 위해 가설을 설정하고 설문 조사를 통해 검증을 시도하였다. 이에 따른 연구 결과 및 내용은 다음과 같다. 첫째, 중국 SNS 인플루언서의 인상관리에서 자기중심적, 타인중심적, 비언어적 인상관리는 유저의 인플루언서에 대한 몰입도에 유의미한 영향을 미치는 것으로 나타났다. 둘째, 인터넷 SNS 플랫폼을 활용하는 유저의 몰입도는 구매의도에 유의미한 영향을 주는 것으로 나타났다. 셋째, 중국 SNS 인플루언서의 인상관리에서 자기중심적, 타인중심적, 비언어적 인상관리는 유저의 구매의도에 유의미한 영향을 미쳤다. 이와 같은 연구 결과를 바탕으로 몰입도와 구매의도 변수의 중요성을 반영한 유저와의 커뮤니케이션 전략을 수립한다면 효과적인 마케팅 방안이 되리라 기대한다. The study is about the effect of impression management of Chinese SNS influencers whose influence is growing as the celebrities in the SNS platform environment based on internet, on the users" flow and purchase intension. The purposes of this study, firstly, is to find the theoretical basis of variable selection through theoretical understanding and terms establishment of Chinese personal media influencers, impression management, SNS flow and purchase intension; Secondly, is to provide empirical research of the variables-flow and purchase intention, in order to inquiry the effect of impression management during the interaction between Chinese SNS influencers and users based on SNS platform; Thirdly, is to deduce the effect relationship of Chinese SNS influencers" impression management on users" flow and purchase intension. The conclusion of this study are as follows: firstly, the self-focused, other-focused and non-verbal impression management of Chinese SNS influencers have a meaningful effect on users‘ flow in influencers; secondly, the flow of Internet SNS platform users has a meaningful influence on purchase intension; thirdly, the self-focused, other-focused and non-verbal impression management of Chinese SNS influencers have a meaningful effect on users" purchase intension. Based on other research results like this, it would be an effective marketing measure if we establish the communication strategy with users which reflects the importance of the variables-flow and purchase intension.

        An Aqueous Extract of Yunnan Baiyao Inhibits the Quorum-Sensing-Related Virulence of Pseudomonas aeruginosa

        Yunnan Baiyao is a famous Chinese medicine that has long been directly applied to wounds to reduce bleeding, pain, and swelling without causing infection. However, little is known about its ability to prevent infection. The present study aimed to assess in vitro the anti-virulence activity of an aqueous extract of Yunnan Baiyao (YBX) using Pseudomonas aeruginosa as a pathogenic model. We found that a sub-MIC (2.5 mg/ml) of YBX can efficiently interfere with the quorum-sensing (QS) signaling circuit. Real-time polymerase chain reaction analysis showed that a sub-MIC of YBX downregulated the transcriptions of lasR, lasI, rhlR, and rhlI,which resulted in global attenuation of QS-regulated virulence activities, such as biofilm formation, and secretion of LasA protease, LasB elastase and pyocyanin. Further, YBX reduced the motility of P. aeruginosa related to QS, and impaired the formation of biofilms. These results suggest that YBX may possess global inhibitory activity against the virulence of P. aeruginosa and that YBX may also exhibit antimicrobial activity in vivo. The present study suggests that Yunnan Baiyao represents a potential source for isolating novel, safe, and efficacious antimicrobial agents.

        Screening and Anti-virulent Study of N-Acyl Homoserine Lactones DNA Aptamers against Pseudomonas aeruginosa Quorum Sensing

        In Pseudomonas aeruginosa, a quorum sensing (QS) system regulates the expression of many virulence factors. N-acyl homoserine lactone (HSL) is the signal molecule of QS system. In order to find a novel HSL binder to interfere with QS signaling and to attenuate P. aeruginosa virulence, an amino lactam surrogate (ALS) of HSL was used as a target to screen HSL aptamers with the technique of systematic evolution of ligands by exponential enrichment (SELEX). Eight HSL aptamers with high affinities for 3O-C12-HSL (20 nM ≤ Kd < 35 nM) or C4-HSL (25 nM < Kd < 50 nM) were finally obtained. In vitro QS-inhibiting study of P. aeruginosa showed that HSL aptamers could inhibit virulence in a dose-dependent manner. ALSap-8 which bound C4-HSL primarily acted on the rhl system and inhibited the secretion of pyocyanin. ALSap-5 which bound 3O-C12-HSL not only showed strong inhibitory activity on biofilm formation as well as secretions of LasA protease and LasB elastase, but also reduced pyocyanin secretion. Since the las system is capable of activating the rhl system mildly, we speculated that ALSap-5 can simultaneously interfere with the las and rhl systems. High-affinity aptamers against HSL in this study are novel QS and virulence-inhibitors, and may have potential as drug candidates for the treatment of P. aeruginosa infection.
