Bending and Compressive Properties of Crystallized TCP/PLLA Composites

$\beta$-Tricalcium phosphate ($\beta$-TCP) particles reinforced bioresorbable plastics poly-L-lactide (PLLA) composites were prepared by injection molding. The nominal weight ratio of $\beta$-TCP was selected as 5, 10 and 15%. In order to clarify effects of the PLLA crystallinity on the mechanical properties, the specimens were heat treated isothermally. Results of differential scanning calorimetry indicated that the PLLA crystallinity increased with increasing heat treatment temperature. Bending and compressive tests were conducted on the specimen with different $\beta$-TCP contents and crystallinities. The results show that the bending and compressive moduli increased with increasing $\beta$-TCP contents and crystallinity. On the other hand, bending strength decreased with increasing $\beta$-TCP contents. Maximum bending strength was obtained at the heat treatment of $70^{\circ}C$ for 24 h, whereas compressive 0.2% proof strength increased with increasing heat treatment temperature. This difference is attributed to the difference in the microscopic damages.

This paper deals with the study of four-dimensional automata. Recently, due to the advances in many application areas such as dynamic image processing, computer animation, augmented reality (AR), and so on, it is useful for analyzing computation of four-dimensional information processing (three-dimensional pattern processing with time axis) to explicate the properties of four-dimensional automata. From this point of view, we have investigated many properties of four-dimensional automata and computational complexity. On the other hand, the class of sets accepted by probabilistic machines have been studied extensively. As far as we know, however, there is no results concerned with four-dimensional probabilistic machines. In this paper, we introduce four-dimensional probabilistic finite automata, and investigate some accepting powers of them.

Purpose: Few reports have been made on the therapeutic effects as well as pathological features of an antithrombin preparation in patients diagnosed with septic disseminated intravascular coagulation (DIC) by the diagnostic criteria for acute DIC. Materials and Methods: A total of 88 sepsis patients who had received inpatient hospital care during the period from January 2000 through December 2008 were divided into two groups, an antithrombin group and a non-antithrombin group, to study the outcomes. Furthermore, the relationship between sepsis-related factors and DIC in 44 patients was studied. Results: The antithrombin group contained 34 patients, and the non-antithrombin group contained 54 patients. The outcomes were significantly better in the antithrombin group. The levels of protein C were low in DIC patients. Conclusion: Our results suggest that early administration of antithrombin might improve outcomes of septic DIC patients in the diagnostic criteria for Japanese Association for Acute Medicine acute DIC.

Usefulness of the Endotoxin Activity Assay to Evaluate the Degree of Lung Injury

Purpose: It has been reported that the Pulse Contour Cardiac Output (PiCCO) is very useful mainly in the field of intensive care and treatment to grasp the pathophysiologicalconditions of pulmonary edema because of its capability of obtainingdata such as Pulmonary Vascular Permeability Index (PVPI) and Extra VascularLung Water (EVLW). Furthermore, a high degree of usability of various markers has been reported for better understanding of the pathological conditions in cases with septicemia. Materials and Methods: The correlation between the cardiorespiratory status based upon the PiCCO monitor (EVLW and PVPI) and inflammatorymarkers including C reactive protein, procalcitonin (PC), and EndotoxinActivity Assay (EAA) were evaluated in 11 severe cases that required treatmentwith a respirator in an intensive care unit. Results: The EAA values were significantly higher in patients with abnormal EVLW at 0.46±0.20 compared to the normal EVLW group at 0.21±0.19 (p=0.0064). In a similar fashion, patients with abnormal PVPI values tended to have higher PC levels at 18.9±21.8 comparedto normal PVPI cases at 2.4±2.2 (p=0.0676). On the other hand, PVPI was significantly higher in the abnormal EAA group at 3.55±0.48 in comparison with the normal EAA group at 1.99±0.68 (p=0.0029). The abnormal EAA group tended to have higher PVPI values than the normal EAA group. Conclusion: The EAA is a measurement method designed to estimate the activity of endotoxins in the whole blood. Our results suggest that the EAA value, which had the greatest correlationwith lung disorders diagnosed by the PiCCO monitoring, reflects inflammatoryreactions predominantly in the lungs.
Persistent activation of Nrf2 through p62 in hepatocellular carcinoma cells

<P>Suppression of autophagy is always accompanied by marked accumulation of p62, a selective autophagy substrate. Because p62 interacts with the Nrf2-binding site on Keap1, which is a Cullin 3–based ubiquitin ligase adapter protein, autophagy deficiency causes competitive inhibition of the Nrf2–Keap1 interaction, resulting in stabilization of Nrf2 followed by transcriptional activation of Nrf2 target genes. Herein, we show that liver-specific autophagy-deficient mice harbor adenomas linked to both the formation of p62- and Keap1-positive cellular aggregates and induction of Nrf2 targets. Importantly, similar aggregates were identified in more than 25% of human hepatocellular carcinomas (HCC), and induction of Nrf2 target genes was recognized in most of these tumors. Gene targeting of <I>p62</I> in an HCC cell line markedly abrogates the anchorage-independent growth, whereas forced expression of p62, but not a Keap1 interaction-defective mutant, resulted in recovery of the growth defect. These results indicate the involvement of persistent activation of Nrf2 through the accumulation of p62 in hepatoma development.</P>

Purpose: The International Organization for Standardization-5fluorouracil (FU) 10 trial found that bolus 5-FU and l-leucovorin was not inferior to S-1 in the treatment of gastric cancer (GC). Continuous 5-FU and the rapid injection of 5-FU have different anti-cancer effects. Thus, bolus 5-FU and l-leucovorin treatment might be useful for oral FU-resistant GC. Materials and Methods: We retrospectively analyzed the medical records of all patients with S-1 or capecitabine-resistant, unresectable, or recurrent GC treated with bolus 5-FU and l-leucovorin between January 2010 and December 2015 at Hokkaido University Hospital. The bolus 5-FU and l-leucovorin regimen consisted of intravenous l-leucovorin ($250mg/m^2/2h$) and bolus 5-FU ($600mg/m^2$) administered once weekly followed by a 2-week rest period; each cycle was repeated every 8 weeks. Results: A total of 14 patients were identified. The disease control rate was 35.7%. The median progression-free survival was 1.6 months (95% confidence interval [CI], 1.3~2.0 months), and the median overall survival was 6.3 months (95% CI, 4.7~7.9 months). No patient died from treatment-related causes. The most common severe adverse event associated with bolus 5-FU and l-leucovorin was neutropenia, which occurred in 21.4% of patients. Conclusions: Bolus 5-FU and l-leucovorin treatment might be useful for oral FU-resistant GC. We are planning a multi-center prospective phase II trial to evaluate the efficacy and safety of bolus 5-FU and l-leucovorin treatment for pre-treated unresectable or recurrent GC to confirm the results of this limited, retrospective study.
