We have developed system biology methods to investigate the relationship between disease-associated genes and their network positions. Genes associated with phenotypically similar diseases are more likely connected in the biological network. We applied the network position to predict candidate disease makers and proved that the method is useful to identify the associated genes of major diseases. Furthermore, two diseases displayed high comorbidity when disease-associated genes were tightly connected in the biological network. Our result establishes a direct correlation between network position and disease association, and helps to understand the mechanism of human disease progression.
